Designing Materials for Hard Tissue Replacement

  • In last two decades, an impressive progress has been recorded in terms of developing new materials or refining existing material composition/microstructure in order to obtain better performance in biomedical applications. The success of such efforts clearly demands better understanding of various concepts, e.g. biocompatibility, host response, cell-biomaterial interaction. In this article, we review the fundamental understanding that is required with respect to biomaterials development, as well as various materials and their properties, which are relevant in applications, such as hard tissue replacement. A major emphasize has been placed to present various design aspects, in terms of materials processing, of ceramics and polymer based biocomposites, Among the bioceramic composites, the research results obtained with Hydroxyapatite (HAp)-based biomaterials with metallic (Ti) or ceramic (Mullite) reinforcements as well as $SiO_2-MgO-Al_2O_3-K_2O-B_2O_3-F$ glass ceramics and stabilized $ZrO_2$ based bioinert ceramics are summarized. The physical as well as tribological properties of Polyethylene (PE) based hybrid biocomposites are discussed to illustrate the concept on how can the physical/wear properties be enhanced along with biocompatibility due to combined addition of bioinert and bioactive ceramic to a bioinert polymeric matrix. The tribological and corrosion properties of some important orthopedic metallic alloys based on Ti or Co-Cr-Mo are also illustrated. At the close, the future perspective on orthopedic biomaterials development and some unresolved issues are presented.

MECHANICAL PROPERTIES AND CORROSION BEHAVIORS OF AGED Ti-4Mo-4Cr-X (X = Sn, V, Zr) ALLOYS FOR METALLIC BIOMATERIALS

  • The purpose of this study was to investigate the mechanical properties of beta type aged Ti-4Mo-4Cr-X (X = V, Sn, Zr) quaternary alloy for use as a cardiovascular stent. Titanium (Ti) alloys were fabricated using a vacuum arc remelting furnace process. To homogenize the specimens of each composition and remove the micro segregation, all cast specimens were subjected to homogenization at 850℃ for 4 h, which was 100℃ higher than the β-transus temperature of 750℃. The tensile strength and elongation of the aged Ti-4Mo-4Cr-X (X = V, Sn, Zr) alloys were increased as compared to the homogenized alloys. In addition, many α/β interface boundaries formed after aging treatment at 450℃, which acted as inhibitors of strain and caused an increase in tensile strength. The elongation of Ti-4Mo-4Cr-X alloys consisting of α + β phases after aging treatment was improved by greater than 30%. Results of a potentiodynamic polarization test showed that the lowest current density of Ti-4Mo-4Cr-4Sn with 1.05 × 10-8 A/cm2 was obtained. The present Ti-4Mo-4Cr-X alloys showed better corrosion characteristics as compared to the 316L stainless steel and L605 (Co-Cr alloy) cardiovascular stent alloys.

Hydroxyapatite-Based Biomaterials for Hard Tissue Applications

  • Over the past few decades, much effort has been made to improve the mechanical and biological performance of HA, in order to extend its range of applications. As a major inorganic component of human hard tissues, hydroxyapatite bioceramic is regarded as being one of the most biocompatible materials. Numerous in vitro and in vivo studies have confirmed its excellent bioactivity, osteoconductivity and bone forming ability. However, because of its poor mechanical properties, its use in hard tissue applications has been restricted to those areas in which it can be used in the form of small sized powders/granules or in the non-load bearing sites. A number of researchers have focused on improving the mechanical and biological performance of HA, as well as on the formulation of hybrid and composite systems in order to extend its range of applications. In this article, we reviewed our recent works on HA-based biomaterials; i) the strengthening of HA with ceramic oxides, ii) HA-based bioactive coatings on metallic implants, iii) HA-based porous scaffolds and iv) HA-polymer hybrids/composites.

  • Zn/HA coating on the Ti-xNb alloys after nanotube formation for dental lmplant was researched using various experimental methods. Due to g ood biocompatibility and osteoconductivity, hydroxyapatite (HA) coating s on metallic biomedical implants were widely employed in orthopedic and dental applications. To improve biocompatibilities, Zinc (Zn) plays very important roles in the bone formation and immune reg ulations. The nanotube formed Zn-HA films were characterized with X-ray diffraction (XRD), field emission scanning electron microscopy (FE-SEM), energy dispersive X-ray spectroscopy (EDS).

Physico-mechanical properties and prosthodontic applications of Co-Cr dental alloys: a review of the literature

  • Cobalt-Chromium (Co-Cr) alloys are classified as predominantly base-metal alloys and are widely known for their biomedical applications in the orthopedic and dental fields. In dentistry, Co-Cr alloys are commonly used for the fabrication of metallic frameworks of removable partial dentures and recently have been used as metallic substructures for the fabrication of porcelain-fused-to-metal restorations and implant frameworks. The increased worldwide interest in utilizing Co-Cr alloys for dental applications is related to their low cost and adequate physico-mechanical properties. Additionally, among base-metal alloys, Co-Cr alloys are used more frequently in many countries to replace Nickel-Chromium (Ni-Cr) alloys. This is mainly due to the increased concern regarding the toxic effects of Ni on the human body when alloys containing Ni are exposed to the oral cavity. This review article describes dental applications, metallurgical characterization, and physico-mechanical properties of Co-Cr alloys and also addresses their clinical and laboratory behavior in relation to those properties.

DV-Xα 분자궤도법으로 설계한 생체용 Ti-Ag-Zr 합금 특성 평가 (A Study on the Properties of Design for the Biomaterial Ti-Ag-Zr Alloys Using DV-Xα Molecular Orbital Method)

  • Ti and Ti alloys have been extensively used in the medical and dental fields because of their good corrosion resistance, high strength to density ratio and especially, their low elastic modulus compared to other metallic materials. Recent trends in biomaterials research have focused on development of metallic alloys with elastic modulus similar to natural bone, however, many candidate materials also contain toxic elements that would be biologically harmful. In this study, new Ti based alloys which do not contain the toxic metallic components were developed using a theoretical method (DV-$X{\alpha}$). In addition, alloys were developed with improved mechanical properties and corrosion resistance. Ternary Ti-Ag-Zr alloys consisting of biocompatible alloying elements were produced to investigate the alloying effect on microstructure, corrosion resistance, mechanical properties and biocompatibility. The effects of various contents of Zr on the mechanical properties and biocompatibility were compared. The alloys exhibited higher strength and corrosion resistance than pure Ti, had antibacterial properties, and were not observed to be cytotoxic. Of the designed alloys' mechanical properties and biocompatibility, the Ti-3Ag-0.5Zr alloy had the best results.

금속 생체재료를 위한 Sn 함량에 따른 Zr-7Cu 합금설계 (Zr-7Cu Alloy Design According to Sn Content for Bio-Metallic Materials)

  • The purpose of this study is to develop a zirconium-based alloy with low modulus and magnetic susceptibility to prevent the stress-shielding effect and the generation of artifacts. Zr-7Cu-xSn (x = 1, 5, 10, 15 mass%) alloys are prepared by an arc melting process. Microstructure characterization is performed by microscopy and X-ray diffraction. Mechanical properties are evaluated using micro Vickers hardness and compression test. The magnetic susceptibility is evaluated using a SQUID-VSM. The average magnetic susceptibility value of the Zr-7Cu-xSn alloy is 1.176 × 10-8 cm3g-1. Corrosion tests of zirconium-based alloys are conducted through polarization test. The average Icorr value of the Zr-7Cu-xSn alloy is 0.1912 ㎂/cm2. The elastic modulus value of 14 ~ 18 GPa of the zirconium-based alloy is very similar to the elastic modulus value of 15 ~ 30 GPa of the human bone. Consequently, the Sn added zirconium alloy, Zr-7Cu-xSn, is very interesting and attractive as a biomaterial that reduces the stress-shielding effect caused by differences of elastic modulus between human bone and metallic implants. In addition, this material has the potential to be used in metallic dental implants to effectively eliminate artifacts in MRI images due to low magnetic susceptibility.

생체의료용 임플란트 소재를 위한 Zr-7Si-xSn 합금설계 (Design of Zr-7Si-xSn Alloys for Biomedical Implant Materials)

  • The metallic implant materials are widely used in biomedical industries due to their specific mechanical strenth, corrosion registance, and superior biocompatability. These metallic materials, however, suffer from the stress-shielding effect and the generation of artifacts in the magnetic resonance imaging exam. In the present study, we develope a Zr-based alloys for the biomedical implant materials with low elastic modulus and low magnetic susceptibility. The Zr-7Si-xSn alloys were fabricated by an arc melting process. The elastic modulus was 24~31 GPa of the zirconium-based alloy. The average magnetic susceptibility value of the Zr-7Si-xSn alloy was 1.25 × 10-8cm3g-1. The average Icorr value of the Zr-7Si-xSn alloy was 0.2 ㎂/cm2. The Sn added zirconium alloy, Zr-7Si-xSn, is very interested and attractive as a biomaterial that reduces the stress-shielding effect caused by the difference of elastic modulus between human bone and metallic implant.

바이오 코팅 재료의 습윤 안정성에 대한 카테콜 작용기의 역할 (Role of Catechol in the Stability of Biocoating Materials in Wet Environment)

  • 생체친화성 다당류 코팅은 금속류의 생체재료 자체 염증 반응으로 의한 합병증(예: 스텐트 재협착 등) 문제를 해결하는 데 활용될 수 있지만, 자체적인 친수성으로 인해 체내의 습윤 환경에서 코팅층이 오랫동안 유지되기 어렵다. 본 논문에서는 히알루론산 상에 접착성 카테콜 작용기를 도입하여 습윤 환경에서도 강한 부착력을 유지하는 다당류 기반의 바이오코팅 소재를 합성하고, 폴리젖산으로 구성된 스텐트 상에서의 수중 안정성 테스트를 통해 그 기능성을 확인하고자 하였다. 연구 결과, 1.26%의 낮은 카테콜 작용기 도입만으로도 수중 안정성이 크게 증대되었음을 모세관 실험 및 분광학적 분석 방법을 통해 확인할 수 있었다. 본 연구 결과는 바이오 코팅 재료의 생채 내 안정성 증대를 위해 카테콜 작용기 도입이 유용할 수 있다는 점을 시사한다.
