Adsorptive Stripping Voltammetry of Ge(IV)-Mercaptoacetic Acid Complex (Ge(Ⅳ)-Mercaptoacetic Acid 착물에 의한 흡착벗김 전압-전류법)

  • The adsorptive stripping voltammetric determination method of trace germanium (IV) using mercaptoacetic acid as a ligand was studied. Optimal conditions were found to be 0.25 M NaCl solution (pH 6.0) containing mercaptoacetic acid concentration of $5.0{\times}10^{-6}M$. The peak potential appeared at - 1.402 V vs. Ag/AgCl. Effects of sodium chloride concentration, mercaptoacetic acid concentration, and accumulation time for the complex of Ge(IV)-Mercaptoacetic acid on the peak current were studied. Amberlite IRC-718 chelating resin was applied to the separation of Ge(IV) from other metal ions.

Nutritional value of calcium acetate obtained from sea-mussel shell (진주담치 껍질을 이용한 아세트산 칼슘의 제조와 영양학적 가치)

  • The present studies were undertaken to prepare calcium acetate from sea-mussel shell and then, to investigate the calcium absorption ratio for calcium acetate by using young albino rate male. Purities such as chloride, nitrate, nitrate, sulfate, phosphate and heavy metal passed to test as reagent grade and calcium acetate assay was 99.0%. No significant differences in the body weight gain between calcium acetate group, calcium gluconate group and calcium carbonate group were not recognized. Diet consumptions of calcium acetate group was almost similar with calcium gluconate group and calcium carbonate group. It was found that absorption rate of calcium acetate was $57.68{\pm}0.83%,\;58.08{\pm}0.94%$ and was $2.0{\sim}3.0%$ high than calcium gluconate and calcium carbonate group.

  • Cadmium is considered one of the most toxic, non biodegradable heavy metal for the human and animals. The purpose of the present study was to investigate the changes in biochemical parameters of blood and accumulation of cadmium in various tissue caused by various levels of dietary cadmium chloride ($CdCl_2$) in broiler chicks. $CdCl_2$ was administered through drinking water to broiler chicks. In spectral analysis, $CdCl_2$ treatment caused a significant increase in Glutamate pyruvate transaminase (GPT), creatinine and uric acid levels in all treated groups. Intriguingly, the GPT, creatinine, and uric acid levels were significantly higher at 75 mg/kg as compared to the groups treated with high doses (100, 125 and 150 mg/kg) of $CdCl_2$. Atomic Absorption Spectrophotometer (AAS) was used for the determination of Cd accumulation in kidney, liver and Breast muscles. AAS analysis revealed that Cd accumulation is increased in breast muscles as compared to liver and kidney at higher doses of Cd than 75 mg/kg.

  • THF-based crown-4 of 16-membered rings having tetrahydrofuran unit was synthesized by an acid-catalyzed condensation of furan and acetone followed by hydrogenation in an effort to obtain highly elective ionophores for lithium ions. The new ionophore was compared with previously reported ionophores under similar measurement conditions with the same plasticizer, tris(2-ethylhexyl) phosphate in poly(vinyl chloride)(PVC) membrane electrodes. Separate solution method was used to determine relative selectivity coefficients for the electrode. The selectivity coefficients($K_{LiM}^{POT}$) of lithium over ammonium, alkali and alkaline earth metal ions go from about $2.4{\times}10^{-1}$ to $2.3{\times}10^{-4}$ to working range and pH dependence have also been studied.

Enhancement of Calcium-Binding Quality of Proglycinin Peptides by Chemical Phosphorylation

  • Glycinin, one of the predominant storage proteins in soybeans, was examined as to whether it could be used as a calcium-binding mediator after chemical phosphorylation and enzymatic hydrolysis. Glycinin is composed of six subunits. One of the proglycinin subunits $(A_{la}B_{lb})$ was overexpressed in E. coli to obtain nonphosphorylated proteins with homogeneity. To investigate the enhanced calcium-binding properties of the phosphopeptides, the proglycinin was purified, phosphorylated, and hydrolyzed with trypsin. The proglycinin expressed in E. coli was purified by ammonium sulfate precipitation, ion-exchange chromatography, and cryoprecipitation. Chemical phosphorylation by sodium trimetaphosphate was performed to obtain phosphorylated proglycinin. After the phosphorylation, one-dimensional isoelectric focusing gel electroanalysis confirmed the phosphorylation of the proglycinin. The phosphorylated peptides were then hydrolyzed with trypsin, followed by a binding reaction with calcium chloride. The calcium-bound phosphopeptides were finally separated using immobilized metal $(Ca^{2+})$ chromatography. Consequently, a limited tryptic hydrolysate of the isolated phosphopeptides exhibited an enhanced calcium-binding ability, suggesting the potential of glycinin phosphopeptides as a calcium-binding mediator with greater availability.

Synthesis and kinetic of ultrafine titanium carbide particles by Mg-thermal reduction of liquid metal chlorides (마그네슘의 금속염 환원에 의한 초미립 탄화티탄 분말 합성거동)

  • Ultrafine titanium carbide particles were synthesized by the reaction of liquid-magnesium and vaporized TiCl$_{4}$+C$_{x}$Cl$_{4}$(x = 1 and 2) solution. Fine titanium carbide particles with about 50 nm were successfully produced by combining Ti and C atoms released by chloride reduction of magnesium, and vacuum was then used to remove the residual phases of MgCl$_{2}$ and excess Mg. Small amounts of impurities such as O, Fe, Mg and Cl were detected in the product, but such problem can be solved by more precise process control. The lattice parameter of the product was 0.43267 nm, near the standard value. With respect to the reaction kinetics, the activation energy for the reactions of TiCl$_{4}$+C$_{2}$Cl$_{4}$and Mg was found to 69 kJ/mole, which was about half value against the use of TiCl$_{4}$+CCl$_{4}$, and such higher reactivity of the former contributed to increase the stoichiometry until the level of TiC$_{0.96}$ and decrease the free carbon content below 0.3 wt.%.

Oral Exposure to Mercury Alters T Lymphocyte Phenotypes and Augments LPS-induced Cytokine Expressions in Spleen and Thymus (비장과 흉선의 림프세포와 LPS에 의해 유도된 사이토카인의 발현에 대한 수은의 영향)

  • Mercury is a widespread metal and consequently there are large populations that currently exposed to low levels of mercury. Endotoxin is a component of the gram-negative bacteria and promotes inflammatory responses. The present study was designed to determine the impact of mercury on lymphocytes phenotype populations and endotoxin-induced inflammatory cytokine expressions in immune organ, spleen and thymus. Male BALB/c mice were exposed continuously to 0, 0.3, 1.5, 7.5, or 37.5 ppm of mercuric chloride in drinking water for 14 days and at the end of the treatment period, lipopolysaccharide (LPS, 0.5 mg/kg) was injected intraperitoneally 2 h prior to euthanasia. The dose-range of mercury used did not cause hepatotoxicity. Mercury at 7.5 and 37.5 ppm dose-dependently decreased CD3$^{+}$ T lymphocytes in spleen; both CD4$^{+}$ and CD8$^{+}$ single positive lymphocyte populations were decreased. Exposure to 7.5 and 37.5 ppm of mercury decreased the CD8$^{+}$ T lymphocyte population in the thymus, whereas double positive CD4$^{+}$ / CD8$^{+}$ and CD4$^{+}$ thymocytes were not altered. Mercury altered LPS-induced inflammatory cytokine gene expressions such as, tumor necrosis factor $\alpha$, interferon ${\gamma}$, and interleukin-12 in spleen and thymus. Results indicated that decreases in T lymphocyte populations in immune organs and altered cytokine gene expression may contribute to the immune-modulative effects of inorganic mercury.ganic mercury.

Fibrinolytic Enzyme Production by Bacillus subtilis KH-4 Isolated from Deonjang

  • A strong fibrin-specific fibrinolytic enzyme was produced from Bacillus subtilis KH-4 isolated from Deonjang, a Korean fermented soybean paste similar to Japanese miso. The addition of glucose as a carbon source resulted in the highest levels of caseinolytic and fibrinolytic activities. Likewise, the addition of yeast extract as the nitrogen source resulted in the highest caseinolytic and fibrinolytic activities (3473.2 unit and 47.4 munit, respectively), It was observed that out of all metal ion sources only calcium (chloride) enhanced caseinolytic and fibrinolytic activities, with increases of 4949.3 unit and 58.2 unit/mg, respectively. The optimal temperature for the production of the enzyme was found to be 4$0^{\circ}C$ in the optimal medium (glucose 20 g, yeast extract 5 g, CaCl$_2$l g, and NaCl 2 g). The maximum fibrinolytic activity was observed at the late stationary phase. B. subtilis KH-4 produced a fibrinolytic enzyme at 4$0^{\circ}C$, after 30 h growth, which increased up to 54 h and then remained constant. These results suggest that Deonjang has potential as a source of physiologically active anti-thromotic enzymes.

Effect of Welding Processes on Corrosion Resistance of UNS S31803 Duplex Stainless Steel

  • An attractive combination of corrosion resistance and mechanical properties in the temperature range -50 to $250^{\circ}C$ is offered by duplex stainless steel. However, undesirable secondary precipitation phase such as $\sigma$, $\gamma_2$ and $Cr_2N$ may taken place at the cooling stage from the welding processes. Therefore, this paper describes the influence of different welding procedures such as manual metal arc welding (MMA), tungsten inert gas welding (TIG) and vacuum brazing on corrosion resistance of the welded joint for UNS S31803 duplex stainless steel. Microstructure and chemical compositions of the welded joint were examined. The weight loss of specimens immersed in 6% $FeCl_3$ solution at $47.5^{\circ}C$ for 24-hours was determined and used to evaluate the pitting resistance of duplex stainless steel and their welds. The region of heat-affected zone of specimen obtained by the MMA is much wider than that resulted from TIG, therefore, the weight loss of welds by MMA was larger than that of weld by TIG. The weight loss of brazed specimens cooled from slow cooling rate was larger than those of specimens cooled from high cooling rate, because the precipitation of $\sigma$ phase. Beside that, the weight loss of brazed specimen is greater than those of the welded specimens. The galvanic corrosion was observed in brazed duplex stainless steel joints in the chloride solution.

Properties and Photocatalytic Activity of Pitch-binded ACF/TiO2Composites

  • Pitch-binded activated carbon fiber(ACF)/$TiO_2$ composite photocatalysts were prepared by Carbon Tetra Chloride (CTC) solvent mixing method with different mixing ratios of anatase to ACF. The result of the textural surface properties demonstrated that there is a slight increase in the Brunauer, Emmett and Teller (BET) surface area of composites with an increase of the amount of ACF. The surfaces structure morphologies of the composites were observed using an Scanning Electron Microscope (SEM). In the XRD patterns for all ACF/$TiO_2$ composites, the diffraction peaks showed the formation of anatase crystallites. The EDX spectra showed the presence of C, O and Si with strong Ti peaks. Most of these samples were richer in carbon and major Ti metal than any other elements. From the photo-decomposition results, the excellent activity of the ACF/$TiO_2$ composites between c/$c_0$ for methylene blue and UV irradiation time could be attributed to both the effects of the photocatalysis of the supported $TiO_2$ and adsorptivity of activated carbon fiber and another carbon derived from pitch.
