This study was conducted to investigate the hematological changes of the dogs when administrated excessive amount of the Artemisia capillaris extract for a short period. In this experiment, clinically healthy dogs(n = 20, average weight = 7.68 kg) were divided into 2 groups : Artemisia capillaris rude juice extract group(CR) and concentrated extract group(CE). They were inserted stomach tube and administrated the extracts (5 ml/kg) for 8 days. The followings are the results of this experiment.: 1). The number of red blood cell(RBC) was significantly (p<0.05) decreased in CE group on day 4, 5, 6, whereas CJE group showed significantly (p<0.05) difference on day 5. 2). The packed cell volume(PCV) and hemoglobin concentration were significantly (p<0.05) decreased in both groups during experimental periods. 3). The met hemoglobin was significantly (p < 0.05) increased from 12 hrs after administration to day 8 in CJE group wheres, it was significantly (p<0.05) increased on day 6,7 in CE group.
Another application of the new quantitative analytical method of prussic acid by "Microdiffusion analysis" for the determination of prussic acid in human blood was studied. The blood containing potassium cyanide was dropped in outer room of unit, and then N-sulfuric acid was added. The liberated HCN gas was absorbed into nickel sulfate solution of inner room, afterward, absorbed prussic acid was determined with EDTA by residual titration. The result was coincided with the result of Liebig Denigs' method at ordinary temperature.
A new exposure system was developed to generate p-dinitrobenzene (p-DNB) containing atmosphere. A glass column was filled with small glass beads coated with the chemical. The p-DNB containing medium was heated to a temperature beyond the boiling point of p-DNB. A stream of air flow was forced to pass through the column and let it mixed with fresh air before introducing into an inhalation chamber. The concentration of p-DNB in the chamber air was measured by direct assaying the air directly and by sampling the chemical using a microfilter installed in the chamber.
Effects of ambient nitrite, NO$_2$$\^$-/, at 1, 3, 10 and 30 mg/1, on the changes of plasma nitrite/nitrate and on hepatic drug - metabolizing enzyme activity were examined in the juvenile Israeli carp, Cyprinus carpio. When the fish were exposed to 1 and 3 mg/1 NO$_2$$\^$-/, there was an exposure duration-dependent increase in plasma NO$_2$$\^$-/ over the 96-hr period reaching 6∼7 fold excess the ambient concentration. In the fish exposed to 10 mg/1, a plateau concentration of less than 2-fold of the environment was attained in 12 hr. With 30 mg/1, however, the maximal plasma NO$_2$$\^$-/ was 41.25 mg/1 at 12 hr followed by a gradual decline. There was a concentration-dependent increase in methemoglobin (metHb) level in all NO$_2$$\^$-/ -exposed groups and a significant decrease in hematocrit value in 30 mg/l group after 96-hr exposure. Apart from the blunted increase in plasma NO$_2$$\^$-/ with higher NO$_2$$\^$-/ (10 and 30 mg/1) exposure, the ratio of plasma NO$_3$$\^$-/ to NO$_2$$\^$-/ was signifirantly higher in these groups compared to 1 and 3 mg/1. The imbalance in the plasma NO$_3$$\^$-//NO$_2$$\^$-/ at higher NO$_2$$\^$-/ exposure suggests a possible accelerated conversion of NO$_2$$\^$-/ to NO$_3$$\^$-/. Nitrite exposure did not affect the hepatic drug-metabolic activities in juvenile Israeli carp. All these data indicate that disposition of NO$_2$- differ depending upon exposed concentration and that metHb production may not be the exclusive toxic mechanism in carp.
$\alpha $-Arabinofuranosidase was produced by E. coli HB101 haboring the recombinant plasmid pKMG11 which contained the arfI gene of Bacillus stearothermophilus. The maximum production of the enzyme was observed when E. coli HB101 cells were grown at 37$\circ$C for 20 hours in the medium containing 0.5% arabinose, 1.0% tryptone, 0.5% yeast extract, and 1% NaCl. The $\ALPHA $-arabinofuranosidase produced was purified to homogeneity using a combination of 20-50% ammonium sulfate precipitation, DEAE-Sepharose CL-6B ion exchange column chromatography and Sepharose 6B-100 gel filtration. The purified enzyme was most active at 55$\circ$C and pH 6.5. The K$_{m}$ and V$_{max}$ values of the enzyme on $\rho $-nitrophenyl-$\alpha $-arabinofuranoside was determined to be 2.99 mM and 0.43 $\mu $mole/min (319.74 $\mu $mole/min/mg), respectively. The pI value was 4.5. The molecular weight of the native protein was estimated to be 289 kDa. The SDS-polyacrylamide gel clectrophoresis analysis suggested that the functional protein was a trimer of the 108 kDa identical subunits. The N-terminal amino acid sequence of the a-arabinofuranosidase was identified as X-Ser-Thr-Ala-Pro-Arg( \ulcorner )-Ala-Thr-Met-Val-Ile-Asp-X-Ala-Phe.
Background: The aim of this study was to compare the effects of different cupping therapy methods combined with Korean medicine treatments for peripheral facial paralysis. Methods: 105 patients treated for peripheral facial paralysis with cupping therapy and other Korean medicine treatment at Gil Korean medicine hospital, Gachon University between May 19, 2014 and June 30, 2018 were selected and their medical charts retrospectively analyzed. 48 patients who met the inclusion criteria were divided into 2 groups: wet cupping (WC) therapy, or dry cupping (DC) therapy combined with Korean medicine treatment. The duration of treatment ranged from 2 weeks for inpatient treatment to 2 months for outpatient treatment. Both WC and DC therapy were performed on TE13, 3 times per week during the treatment period. The effect of cupping therapy was evaluated by using the Gross Grading System of the House-Brackmann (HB score) and the Yanagihara's Unweighted Grading System (Y score). Results: For both WC and DC treatment of symptoms related to peripheral facial paralysis, HB scores showed a significant decrease and Y scores showed a significant increase from baseline to end of treatment, indicating a beneficial improvement in patient symptoms for both WC and DC. Conclusion: In this study, both DC and WC treatment had significant improvements on peripheral facial paralysis symptoms, with WC having significantly greater beneficial effects than DC.
This experiment was carried out in order to elucidate the effect of NaCl(0.85%) acclimation on congestion symptoms in central venous sinuses(CVS) of gill filaments and on the alteration of blood constituents in cultured eels for 1, 2, 3, 4, 5, 6 and 7days after handling stress. After giving a handling stress on eels, the frequency of gill congestion was about 90%. It gradually decreased to 40% on the 3rd day, then increased to 60% on the 7th day in NaCl(0.85%) acclimation. The values of haematocrit(Ht), methemoglobin(Met-Hb), glucose (GLU), calcium($Ca^{++}$), lactic dehydrogenase(LDH), glutamic pyruvic transaminase (GPT) and glutamic oxaloacetic transaminase(GOT) were lower than those of the control, whereas magnesium($Mg^{++}$) and haemoglobin(Hb) were higher than the control. However total protein(Tp) and albumin (ALB) levels similar to the control. When the fish after giving a handling stress were cultured in NaCl-free water as a control, the frequency of gill congestion and the value of blood components showed a similar pattern to that obtained from the NaCl acclimated experiments. However, in the early period(1-3day) after handling stress, NaCl acclimating slightly helped the natural tendancy of recovery. These results suggest that the frequency of gill congestion is not always reduced over the whole period of NaCl acclimation. However, the acclimation to NaCl solution may be a somewhat useful therapy for the gill congestion in stressed eels.
The Allium tuberosum is a perennial herb. From the ancient times, it has been used for food or medical purpose. Allium tuberosum is widely distributed in Korea, China, Japan and other countries. As herbal medicine, the Allium tuberosum has medical effects on lumbago, sthenia, diuresis, nocturia, hematemesis, anemia, haemorrhoid, hiccups, diarrhea, eczema, sunstroke, pollution and so on. Recently, the Allium tuberosum has been studied on its components, anti-microorganism action, anti-fungal action, anti-arteriosclerosis action, anti-cancer action. However, there has not been any study on hematological effect of allium tuberosum extract on the blood in large dogs whereas the effects of excess garlic and onion on the blood of cattle, horses, dogs, cats and sheep has been reported by many researchers. Thus, this study was performed to observe the hematological effect of excessive Allium tuberosum extract administrated in large dogs. 1. The Red Blood Cell(R3C) count was significantly decreased(p<0.05), Packed Cell Volume (PCV) count were significantly decreased(P<0.05), and Reticulocyte count was significantly increased(p<0.05) 2. The White Blood Cell(WBC) count was significantly increased(p<0.05).3. The Hemoglobin concentration was significantly decreased(p<0.05) and Mean Corpuscular Hemoglobin Concentration (MCHC) was increased. and Mean Corpuscular Volume(MCV) was significantly increased.(p<0.05). 4. The Reduced glutathione(GSH) and Met-Hemoglobin were significantly increased (p<0.05).
The present study was designed to investigate the anti-diabetic effect and mechanism of Korean red ginseng in C57BL/KsJ db/db mice. The db/db mice were divided into three groups: diabetic control group (DC), Korean red ginseng group (KRG, 100 mg/kg) and metformin group (MET, 300 mg/kg), and treated with drugs once per day for 10 weeks. Compared to the DC group, fasting blood glucose levels were decreased by 19.8% in KRG-, 67.7% in MET-treated group. With decreased plasma glucose and insulin levels, the insulin resistance index of the KRG-treated group was reduced by 27.6% compared to the DC group. The HbA1c levels in KRG and MET-treated groups were also decreased by 11.0% and 18.9% compared to that of DC group, respectively. Plasma triglyceride and non-esterified fatty acid levels were decreased by 18.8% and 16.8%, respectively, and plasma adiponectin and leptin levels were increased by 20.6% and 12.1%, respectively, in the KRG-treated group compared to those in DC group. Histological analyses of the liver and fat tissue of mice treated with KRG revealed significantly decreased number of lipid droplets and decreased size of adipocytes compared to the DC group. From the pancreatic islet double-immunofluorescence staining, we observed KRG has increased insulin contents, but decreased glucagon production. To elucidate action mechanism of KRG, effects on AMP-activated protein kinase (AMPK) and its downstream target proteins responsible for fatty acid oxidation and gluconeogenesis were explored in the liver. KRG activated AMPK and acetyl-coA carboxylase (ACC) phosphorylations, resulting in stimulation of fatty acid oxidation. KRG also caused to down regulation of SREBP1a and its target gene expressions such as FAS, SCD1 and GPAT. In summary, our results suggest that KRG exerted the anti-diabetic effect through AMPK activation in the liver of db/db mice.

In this experiment, clinically healthy dogs(n = 10, average weight = 4.17 kg) were used. They were inserted stomach tube and administrated the extracts(5ml/kg) for 7 days. The followings are the result of this experiment. The red blood cell counts, hemoglobin concentration and mean corpuscular hemoglobin concentration were significantly decreased. (P<0.05) The packed cell volume was slightly decreased. The mean corpuscular volume and methemoglobin concentration were significantly increased.(P<0.05) The white blood cell counts and reduced glutathione were slightly increased. Consequently, We determined that administrated excessive amount of the Korean native chinese chive extracts induced hemolytic anemia.
