Effect of Methionine on Heart Lipid Peroxidation in Rat with Alcohol Administration (Ethanol 급여 흰쥐의 심장 지질과산화에 미치는 Methionine의 영향)

  • This study was designed to investigate the effects of methionine(Met) on the activities of heart lipid peroxidation related enzymes in ethanol administrated rats. Male Sprague-Dawley rats were fed on diets containing one of the three levels of Met(0%, 0.3%, 0.9% of kg diet) and ethanol(2.5g/kg of body weight) was administrated as 25v/v% ethanol to ethanol treated groups orally. The rats were sacrificed after 5 and 10 weeks of feeding. Xanthine oxidase(XO) and catalase activities increased with ethanol administration and those activities were higher n Met excessive and deficiency group than those of Met normal group at 5 and 10 weeks dieting. Superoxide dismutase(SOD) activity in heart decreased significantly in Met deficiency and Met excessive group as compared to that of control. Glutathione peroxidase(GSH-Px) activity in heart significantly decreased in Met deficiency group as compared to that of Met excessive and normal group. Glutathione S-transferase(GST) activity of heart tissue significantly increased by ethanol administration. Glutathione(GSH) content in heart decreased with ethanol administration and shwoed no significant differences with Met levels. Ethanol administration increased the content of lipid peroxide(LPO).

The Effects of MET and ICT in Patients with Lumbago by Meridian Muscle Electrography (근에너지기법과 경근간섭파요법이 요통환자의 요방형근 경근전도에 미치는 영향)

  • Objectives The purpose of this study is to compare the effects of MET (muscle energy technique) and ICT (interferential current therapy) on lumbago by meridian muscle electrography (MMEG). We performed MET and ICT on quadratus lumborum muscle. Methods This study was carried out on 30 lumbago patients. 30 patients were randomly divided into MET group and ICT group. MET group take treatment 10 minutes. ICT group take treatment 15 minutes. After performing MET and ICT twice, we compared both in terms of root mean square (RMS), special edge frequency (SEF), VAS and ODI. Results 1. Both MET group and ICT group showed significant improvement in VAS and ODI after treatment. 2. Both MET group and ICT group showed significant improvement in special edge frequency (SEF) of the left side after treatment. But SEF of the right side showed insignificant improvement. 3. Both MET group and ICT group showed insignificant improvement in root mean square (RMS) for both sides. 4. There are no significant differences between both groups. Conclusions According to above results, we found out that performing MET or ICT on quadratus lumborum muscle has effect in terms of VAS and ODI. But, in terms of SEF, we found out different result for both side. And we can't found out effect in terms of RMS.

XRCC3 Thr241Met Gene Polymorphism and Risk of Colorectal Cancer in Kashmir: a Case Control Study

  • XRCC (X-ray cross-complementing group) genes contribute to important DNA repair mechanisms that play roles in the repair of single strand breaks (SSBs) induced by a variety of external and internal factors, including ionizing radiation, alkylating agents and reactive oxygen species. These repair genes have a pivotal role in maintaining genomic stability through different pathways of base excision repair (BER). The aim of this study was to investigate the XRCC3 Thr241Met gene polymorphism in colorectal cancer (CRC) in Kashmir. We investigated the genotype distribution of XRCC3 gene in 120 CRC cases in comparison with 150 healthy subjects and found a significant association between XRCC3 genotypes and CRC ($p{\leq}0.05$). Both heterozygous genotype (Thr/Met) as well as homozygous variant genotype (Met/Met) were moderately associated with elevated risk of CRC [OR=2.53; OR=2.29 respectively]. Also, Thr/Met and Met/Met genotypes demonstrated a significant association with the risk of CRC (p = 0.003). This study displayed a significantly elevated risk for CRC in individuals with XRCC3 Thr/Met and Met/Met Genotype of about 2.5 times that with the Thr/Thr wild genotype.

The Wine Yeast Strain-Dependent Expression of Genes Implicated in Sulfide Production in Response to Nitrogen Availability

  • Sulfur metabolism in S. cerevisiae is well established, but the mechanisms underlying the formation of sulfide remain obscure. Here, we investigated by real-time RT-PCR the dependence of expression levels of MET3, MET5/ECM17, MET10, MET16, and MET17 along with SSU1 on nitrogen availability in two wine yeast strains that produce divergent sulfide profiles. MET3 was the most highly expressed of the genes studied in strain PYCC4072, and SSU1 in strain UCD522. The strains behaved differently according to the sampling times, with UCD522 and PYCC4072 showing the highest expression levels at 120 h and 72 h, respectively. In the presence of 267 mg assimilable N/l, the genes were more highly expressed in strain UCD522 than in PYCC4072. MET5/ECM17 and MET17 were only weakly expressed in both strains under any condition tested. MET10 and SSU1 in both strains, but MET16 only in PYCC4072, were consistently upregulated when sulfide production was inhibited. This study illustrates that strain genotype could be important in determining enzyme activities and therefore the rate of sulfide liberation. This linkage, for some yeast strains, of sulfide production to expression levels of genes associated with sulfate assimilation and sulfur amino acid biosynthesis could be relevant for defining new strategies for the genetic improvement of wine yeasts.

Function of Microbial Electrochemical Technology in Anaerobic Digestion using Sewage Sludge (하수슬러지를 이용한 혐기성소화조에서 미생물 전기화학기술의 역할)

  • Microbial electrochemical technology (MET) has recently been studied to improve the efficiency of a traditional anaerobic digestion (AD). The purpose of this study was to investigate the impact of MET in the system when MET was combined with traditional AD (i.e., AD-MET). Electrodes used in the MET were Cu coated graphite electrodes. They were supplied with a voltage of 0.3 V. AD started to generate methane in 80 days. But AD-MET started to generate methane from the initial operation after the system started. It was observed that AD-MET reached steady state faster and produced higher methane yield than AD. During the steady state, the average daily methane productions in AD and AD-MET were 2.3L/d and 4.9L/d, respectively. Methane yields were 0.07-CH4/g‧CODre in AD and 0.25L-CH4/g‧CODre in AD-MET. In AD-MET, the production rates of total volatile fatty acids (TVFAs) and soluble chemical oxygen demand (SCOD) were 0.12 mg TVFAs/mg VS‧d and 0.35 mg SCOD/mg VS‧d, respectively. They were significantly (p < 0.05) higher than those in AD. However, the concentrations of residual TVFAs in both systems were not significantly (p > 0.05) different from each other, confirming that methane conversion in AD-MET was greater than that in AD.

  • S-Adenosyl-L-methionine synthetase (ATP: methionine S-Adenosyltransferase, EC 2.5.1.6; AdoMet synthetase) catalyzes the biosynthesis of S-Adenosyl-L-methionine(AdoMet) from methionine in the presence of ATP. To elucidate the role of transmethylation reaction in the pancreatic tissues, we examined AdoMet synthetase and isozyme activities, and AdoMet contents in the various tissues. The activities of AdoMet synthetase marked the highest in the kidney, and the lowest in the testis among the various tissues of rat. Considerable amounts of AdoMet synthetase activities were detected in the pancreatic tissues of various animals except for those of frog. The level of ${\alpha}$ and ${\gamma}$ isozyme activities were present in the pancreatic tissues of various animals, while ${\beta}$ isozyme activities were detected as trace. AdoMet synthetase activities of rat brain, liver, testis were decreased with growth. In the rat pancreatic tissues, AdoMet synthetase activities were increased during 16 days after birth and then decreased between 16 and 47 days of age. Levels of AdoMet contents of rat brain and testis were decreased with growth. However, AdoMet contents of rat pancreas were decreased until 26 days of age, and then increased thereafter. AdoMet synthetase isozyme patterns did not vary with growth in the pancreas and testis. But, in the liver, ${\beta}$ form is strikingly increased with growth.

  • Solution conformation of cyclo-($Gln^1-Trp^2-Phe^3-{\beta}Ala^4-Leu^5-Met^6$), new NK-2 antagonist in dimethyl sulfoxide solution, has been determined by the use of two-dimensional nuclear magnetic resonance spectroscopy combined with simulated annealing calculations. The peptide exhibited converged structures with the atomic root-mean-square difference for the backbone atoms ($N,\;C^{\alpha},\;C'$) of all residues being 0.02${\AA}$ in the 25 annealed structures. The analysis of the structures indicated that the cyclic peptide has three intramolecular hydrogen bonds between $Met^6NH$ and ${\beta}Ala^4CO$, ${\beta}Ala^4NH$ and $Met^6CO$, $Phe^3NH$ and $Met^6CO$, and contain a type-I ${\beta}$-turn with Gln and Trp and ${\gamma}$-turn with Leu. The addition of an extra methylene group to Gly, i.e. P-Ala residue, may relax some unfavorable restraints in the cyclic backbone structure, hence enabling an additional hydrogen bond, which results in stabilizing one conformation.

  • Objectives : To evaluate the clinical utility of MET in elector spinae muscle on low back pain patients. Methods : We compared electrical activity between a before MET and a after MET in elector spinae muscle on low back pain patients in same group(n=30) in dynamic flexion-reextension state during five seconds. We analyzed amplitudes and areas of electrical activity and asymmetry index(AI). Results : 1. After MET in elector spinae muscle on low back pain patients were significantly decreased in Visual Analogue Scale(VAS) than before MET in elector spinae muscle on low back pain patients(p<0.05). 2. After MET in elector spinae muscle on low back pain patients were lower electrical activity than before MET in elector spinae muscle on low back pain patients but it is not a pointless observation(p<0.05). 3. AI of the after MET in elector spinae muscle on low back pain patients significantly decreased compared with before MET in elector spinae muscle on low back pain patients(p<0.05). Conclusions : According to above results, there is clinical effect MET on low back pain patients.

Discovery of an Indirubin Derivative as a Novel c-Met Kinase Inhibitor with In Vitro Anti-Tumor Effects

  • The c-Met protein is a receptor tyrosine kinase involved in cell growth, proliferation, survival, and angiogenesis of several human tumors. Overexpression of c-Met has been found in gastric cancers and correlated with a poor prognosis. Indirubin is the active component of Danggui Longhui Wan, which is a traditional Chinese antileukemic recipe. In the present study, we tested the anti-cancer effects of an indirubin derivative, LDD-1937, on human gastric cancer cells SNU-638. When we performed the in vitro kinase assay against the c-Met activity, LDD-1937 inhibited the activity of c-Met. This result was confirmed by immunoblot and immunofluorescence of phosphorylated c-Met. Immunoblot analysis showed that LDD-1937 decreased the expression of the Erk1/2, STAT3, STAT5, and Akt, downstream proteins of c-Met. In addition, LDD-1937 reduced the cell viability and suppressed colony formation and migration of SNU-638 cells. Furthermore, LDD-1937 induced $G_2/M$ phase arrest in the SNU-638 cells by decreasing the expression levels of cyclin B1 and CDC2. Cleaved-PARP, an apoptosis-related protein, was up-regulated in cells treated with LDD-1937. Overall, this study suggests that LDD-1937 may be a novel small-molecule with therapeutic potential for selectively inhibiting c-Met and c-Met downstream pathways in human gastric cancers overexpressing c-Met.

Picropodophyllotoxin Inhibits Cell Growth and Induces Apoptosis in Gefitinib-Resistant Non-Small Lung Cancer Cells by Dual-Targeting EGFR and MET

  • Patients with non-small-cell lung cancer (NSCLC) with epidermal growth factor receptor (EGFR) amplification or sensitive mutations initially respond to the tyrosine kinase inhibitor gefitinib, however, the treatment becomes less effective over time by resistance mechanism including mesenchymal-epithelial transition (MET) overexpression. A therapeutic strategy targeting MET and EGFR may be a means to overcoming resistance to gefitinib. In the present study, we found that picropodophyllotoxin (PPT), derived from the roots of Podophyllum hexandrum, inhibited both EGFR and MET in NSCLC cells. The antitumor efficacy of PPT in gefitinib-resistant NSCLC cells (HCC827GR), was confirmed by suppression of cell proliferation and anchorage-independent colony growth. In the targeting of EGFR and MET, PPT bound with EGFR and MET, ex vivo, and blocked both kinases activity. The binding sites between PPT and EGFR or MET in the computational docking model were predicted at Gly772/Met769 and Arg1086/Tyr1230 of each ATP-binding pocket, respectively. PPT treatment of HCC827GR cells increased the number of annexin V-positive and subG1 cells. PPT also caused G2/M cell-cycle arrest together with related protein regulation. The inhibition of EGFR and MET by PPT treatment led to decreases in the phosphorylation of the downstream-proteins, AKT and ERK. In addition, PPT induced reactive oxygen species (ROS) production and GRP78, CHOP, DR5, and DR4 expression, mitochondrial dysfunction, and regulated involving signal-proteins. Taken together, PPT alleviated gefitinib-resistant NSCLC cell growth and induced apoptosis by reducing EGFR and MET activity. Therefore, our results suggest that PPT can be a promising therapeutic agent for gefitinib-resistant NSCLC.
