The Study on the Foot Type of Female Farmers (여성 농업인 발 유형에 관한 연구)

  • This study was performed to offer the basic data for the design of farm shoes. 265 Korean female farmers aging between the 40s to the 80s volunteered for this study and we measured 40 items on each foot with the 3D foot scanner. First, the differences between farmers' feet and non-farmers' feet were analyzed. Farmers' feet were thicker in the instep areas, but had lower arch height than non-farmers' feet. In addition, farmer's feet were tilted to the inside. Next, eight factors were extracted among the 40 measuring items, and the classification criteria of the foot shape was analyzed. The important factors were: size of foot length and volume of ankle, malleolus height and size, volume of the front part of ankle, medial & lateral ball width, and vertical size of foot. Third, three clusters according to the foot shapes were categorized by cluster analysis of eight factor scores. Foot type 1 was medium in foot length, big in thickness, large in lateral ball width, small in toe 1 angle, and tilted to the inside. Foot type 2 was long and slim, and big in toe 5 angle. Foot type 3 was short in foot length, medium in volume of the front part of ankle, large in medial ball width, and big in toe 1 angle. Despite its shortness, foot type 3 was thick and showed severe deformation in toe 1. Lastly, the frequency distributions of the foot types in each age group were analyzed. Female farmers of the forties showed high frequency in type 1 and other age groups showed high frequency in type 2. The older female farmers showed higher frequency of type 3.

High Frequency Somatic Embryogenesis and Plant Regeneration from Cultured Immature Seeds of Ostericum koreanum Kitagawa and Angelica purpuraefolia Chung (강활(Ostericum koreanum)과 지리강활(Angelica purpuraefolia)의 미숙종자로부터 고빈도의 체세포배 발생과 식물체 재분화)

  • The objective of this study is to establish an efficient celll culture system for somatic embryogenesis in Ostericum koreanum and Angelica purpuraefolia. The highest frequency of embryogenic callus on immature seeds of O. konanum and A, purpuraefolia. was obtained when seeds were cultured on MS medium containing 0.1 mg/L 2,4-D and 0.1 mg/L BA. However somatic embryos were formed directly from the edge of cotyledon and hypocotyl of plant which regenerated on medium supplemented with 0.1-3.0 mg/L NAA. Immature seed explane cultured at 25$\pm$2$^{\circ}C$ after 10 days treatment at 5$^{\circ}C$ produced embryogenic callus and somatic embryos, and these differentiated into whole plane. Addition of glutamine and coconut milk to media did not enhance the frequency of somatic embryogenesis in immature seed cultures of A. purpuraefolia. However in immature seed culture of O. koreanum, the frequency of somatic embryogenesis were increased on media supplemented with glutamine and 10% coconut milk. Especially addition of glutamine to the medium substituted effect of NH$_4$N0$_3$ in constast to coconut milk. The highest frequency of conversion somatic embryos into plantlet was 89.1% on MS basal medium Embryogenic calli were grown vigorously when maintained on medium with 0.01 mg/L 2,4-D and 0.01 mg/L BA.

Efficient Callus Culture and Plant Regeneration from Mature Seed of Tall Fescue (Festuca arundinacea Schreb.) (톨 페스큐의 성숙종자로부터 효율적인 캘러스 배양 및 식물체 재분화)

  • In an effort to optimize tissue culture conditions for genetic transformation of tall fescue (Festuca arundinacea Schreb.), an efficient plant regeneration system from seed-derived calli was established. MS medium containing 6 mg/L 2,4-dichlorophenoxy acetic acid (2,4-D) and 0.1 mg/L benzyladenine (BA) were optimal for embryogenic callus formation from mature seed and had a strong effect on successive plant regeneration. The plant regeneration frequency above 50% was observed when embryogenic calli induced in this medium were transferred to N6 medium supplemented with 1 mg/L 2,4-D and 3 mg/L BA. Among several basic media, MS and N6 medium were optimal for callus induction and plant regeneration, respectively. 'Kentucky-31' showed to have high frequencies of embryogenic callus induction and plant regeneration up to 58.3 and 50%, respectively. Addition of sucrose to the regeneration medium as a carbon source increased regeneration frequency up to 55%. A short tissue culture period and high-frequency regeneration system established in this study will be useful for molecular breeding of tall fescue through genetic transformation.

Adventitious Shoots Regeneration from Seed Explants of Xanthoceras sorbifolium

  • Xanthoceras sorbifolium Bunge (yellowhorn) is a woody tree in the soapberry family, Sapindaceae, native to northern China. This species has been identified as a major woody bioenergy plant for bio-diesel production because of high oil content in seed. But the flowers do not bear fruit well while the many flowers blooming. This study was performed to regenerate in vitro plantlet using adventitious shoot formation. To establish the protocol of plant regeneration, adventitious shoots formation rate in the culture of cotyledon of immature zygotic embryos was 68.6% in 1/2 MS medium with 0.5 mg l-1 BA and 3% sucrose (w/v). In the culture of cotyledons of mature zygotic embryos, induction of adventitious shoots was needed to contain high sucrose in pre-culture medium and the frequency of shoot induction was 64.4%. Multiple shoots were induced in 0.5 mg l-1 TDZ, and rooting of shoot was induced 4.0 mg l-1 IBA. Flow cytometry analysis revealed that all the regenerated plantlets were diploid.

  • To establish high frequency shoot formation from two cultivars (Cheongyangjaerae and Myungan) of boxthorn (Lycium chinense Mill.), hypocotyl segments with cotyledons from seedlings were used as explants. High frequency adventitious shoot formation (more than 80%) were obtained from hypocotyl segments with cotyledon on MS medium supplemented with 1.0 mg/L zeatin, when precultured for 3 weeks under dark conditions followed by transfer to light conditions. But there was no shoot induction in the explants cultured without preculture under dark conditions. Roots were induced from the shoots when transferred to rooting medium supplemented with 1.0 mg/L IAA for 4 weeks. Regenerated plantlets were grown to normal mature plants in soil.

The embryogenic competency and morphological changes during somatic embryogenesis in Iris pseudacorus

  • Embryogenic callus was obtained from bulb segments of Iris pseudacorus on Murashige and Skoog (MS) medium with 2,4-dichlorophenoxyacetic acid (2,4-D) alone or in combination with kinetin. When early globular somatic embryos were subcultured onto MS medium with $4.52{\mu}M$ 2,4-D, high frequency of somatic embryogenesis was obtained. Deprivation of 2,4-D was required for maturation. Mature somatic embryos had an elongated scutellum with a notch on the base of scutellum. Separation of embryos from embryo clusters was necessary to enhance the frequency of germination. Germination was stimulated by separation of embryos from embryo clusters and transfer onto fresh half-strength MS medium with 3% sucrose. After acclimation in artificial soil in greenhouse for 2 months, 96.4% of plantlets survived.

  • Multiple attribute for decision making including user preference will increase the complexity of route selection process. Various approaches have been proposed to solve the optimal route selection problem. In this paper, multi attribute decision making (MADM) algorithms such as Simple Additive Weighting (SAW), Weighted Product Method (WPM), Analytic Hierarchy Process (AHP) method and Total Order Preference by Similarity to the Ideal Solution (TOPSIS) methods have been proposed for acoustic signature based optimal route selection to facilitate user with better quality of service. The traffic density state conditions (very low, low, below medium, medium, above medium, high and very high) on the road segment is the occurrence and mixture weightings of traffic noise signals (Tyre, Engine, Air Turbulence, Exhaust, and Honks etc) is considered as one of the attribute in decision making process. The short-term spectral envelope features of the cumulative acoustic signals are extracted using Mel-Frequency Cepstral Coefficients (MFCC) and Adaptive Neuro-Fuzzy Classifier (ANFC) is used to model seven traffic density states. Simple point method and AHP has been used for calculation of weights of decision parameters. Numerical results show that WPM, AHP and TOPSIS provide similar performance.

Studies on Symptom, Pathology, Composition and Clinical Applications of Woman-related Disease Prescriptions in Bang Yahk Hap Peun (방약합편 중 부인과에 관련된 방제의 활용에 대한 고찰)

  • In this study, symptoms, pathologies, compositions and clinical applications of 52 prescriptions for woman-related disease treatment descripted in Bang Yahk Hap Peun were investigated. The results were same as following; Classification of prescriptions for woman-related disease treatments was 23 prescriptions in high-chepter, prescriptions in 16 medium-chepter, and prescriptions in 13 low-chepter prescriptions and the frequency of clinical applications for high-chepter, medium-chepter and low-chepter prescriptions was 44%, 31% and 25%, respectively. Clinical applications of woman-related disease prescriptions were identified aocording to pre-pregnancy and post-birth, uterus, breast disease, before pregnancy. Infertile, emmeniopathy, uterus Blooding made practical application to high frequency in the treatments. Oriental Medical Pathology applied with prescriptions were Biheo(脾虛), Ganbinoyool(肝脾怒鬱), Infirmity(虛弱), 熱入血室, Jungpung(中風), Gihyulguheo(氣血俱虛), hyulheo(血虛), Aeohyul(瘀血) etc. Prescriptions applied with the most frequency in woman-related disease were Samuoltang(四物湯), Sagunjatang(四君子湯), Gungkuitang(芎歸湯), Boanbaekchuolsan(保安白朮散), Dangguibohyeltang(當歸補血湯), Sanhyointang(酸棗仁湯), Dohongsamuoltang(桃紅四物湯), Jipeisan(芷貝散), Silsosan(失笑散).

Efficient Callus Induction and Plant Regeneration from Immature and Mature Embryo Culture of Korean Wheat Genotypes

  • Immature and mature embryos of 18 Korean wheat genotypes were cultured in vitro to develop an efficient method of callus formation and plant regeneration, and to compare the responses of both embryo cultures. Immature and mature embryos were placed on a solid agar medium containing the MS salts and vitamins, 30g/l maltose, 2 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D), and amino acids. The developed calli were maintained on regeneration medium containing MS salts and B5 vitamins, 20 g/l sucrose, and the combination of two plant growth regulators, 6-benzylaminopurine (BAP) and indole-3-acetic acid (IAA). Immature embryos in most genotypes showed high efficiency of callus induction except three genotypes; Eunpamil, Chunggemil, and Namhaemil, and significant differences among the genotypes. Plant regeneration of calli induced from immature embryos showed high efficiency in Geurumil (56.5%), Tapdongmil (50.5%), Gobunmil (45.5%), and Urimil(42.2%). The analysis of variance showed significant differences for regeneration frequency among the genotypes. Mature embryos showed low callus induction frequency compared with that in immature embryos, and significant differences among the genotypes. Plant regeneration of calli induced from mature embryos showed high efficiency in Keumkangmil (33.33%), Tapdongmil(28.13%), and Geurumil (27.78%). The analysis of variance showed significant differences for plant regeneration frequency among the genotypes.

Somatic Embryogenesis and Plant Regeneration in Mature Seed Cultures and Seed-Derived Embryogenic Suspension Cultures of Yuzu (유자의 성숙종자 배양 및 종자유래 배발생 현탁배양으로부터 체세포배발생을 통한 유자의 식물체 재생)

  • Off-white, friable embryogenic calluses were formed on the internal integument of mature seeds of yuzu (Citrus junos) cultured on Murashige and Skoog's basal medium at a frequency of 1.2%. Embryogenic calluses were proliferated when cultured on medium with 1 mg/L 2,4-D. Upon transfer to medium with 0.1 mg/L kinetin, embryogenic calluses produced numerous somatic embryos. Embryogenic suspension cultures were established by placing embryogenic calluses into liquid medium with 1 mg/L 2,4-D. When plated onto medium with 0.5 mg/L ABA, embryogenic cells developed into somatic embryos at a high frequency, and then regenerated into plantlets. Plantlets were successfully transplanted to potting soil and grown in a greenhouse.
