Functional Analysis of ESTs from the 14-year Root of Korean Ginseng

  • To assist genetic study of the root development in Panax ginseng, which is one of the most important medicinal plant, expressed sequence tags (EST) analysis was carried out. We constructed a cDNA library using the 14-year ginseng root. Partial sequences were obtained from 2,975 clone. The ESTs could be clustered into 1,991 (70.2%) non-redundant groups. Similarity search of the non-redundant ESTs against public non-redundant databases of both protein and DNA indicated that 1,553 groups show similarity to genes of blown function. These ESTs clones were divided into sixteen categories depending upon gene function. The most abundant transcripts were ribonuclease 1 (67) and ribonuclease 2 (65). Our extensive EST analysis of genes expressed in 14-year ginseng root not only contributes to the understanding of the dynamics of genome expression patterns in root organ but also adds data to the reperoire of all genomic genes.

  • 현대과학에 의하여 밝혀진 고려인삼의 약리효능을 보면 뇌 기능 개선, 통증감소, 암 예방 및 항암작용, 면역기능 항진, 간 기능 개선, 항당뇨, 혈압 조절기능, 항피로, 항 스트레스, 여성 갱년기장애 개선, 남성 발기부전 개선, 항 산화, 항 노화 등에 관한 효능을 들 수가 있으며, 고려인삼의 효능성분으로서는 ginsenosides(인삼사포닌), 페놀화합물, 폴리아세틸렌화합물, 다당체 특히 산성다당체, 펩타이드 등이 주류를 이루고 있다. 효능성분 중에는 특히 ginsenosides에 관한 연구가 많이 수행되었는 바, 고려인삼이 타국삼에 비하여 다양한 종류의 ginsenosides가 함유되어 있다는 것이 판명되었다.

Physiological Activities of Rheum undulatum and Rheum palmatum Extracts as Affected by Solvents

  • Rheum undulatum and Rheum palmatum have been widely used as food material as well as medicinal ingredients for their therapeutic effects in oriental countries. Many kinds of herbs are being used in the manufacture of functional foods. The objective of this study was to determine polyphenol content, superoxide dismutase (SOD)-like activity, tyrosinase activity, and electron donating ability of R. undulatum and R. palmatum. Total polyphenol content were most effective in 50 and 100% ethanol extracts from R. undulatum and R. palmatum. SOD-like activities of R. undulatum extracts were higher than those of R. palmatum extracts, and water extracts of samples were highest. EDAs of R. undulatum extracts were higher (26.76-44.46%) than those of R. palmatum extracts, while those of both extracts were lower than 1.0 and 0.1% L-ascorbate. And these suggest that the extracts of R. undulatum and R. palmatum can be used as a material in functional food.

Characteristics of Photosynthesis with Growing Stages by different Shading Materials in Panax Ginseng C. A. Meyer (차광재료에 따른 생육시기별 인삼의 광합성작용의 특성)

  • To investigate the influence of shading materials with growing stages in Panax Ginseng C. A. Meyer, the diurnal change of photosynthesis, stomatal conductance, transpiration and its any correlation were measured. The net photosynthetic rate and stomatal conductance of ginseng were higher in the morning than in the broad day. The net photosynthetic rate was increased as the PAR (Photosynthetically Action Radiation) was increased and it was reached the maximum at the $200\;{\mu}mol/m^2/s$ of PAR in overall leaves. Transpiration rate was increased in the afternoon compared to in the morning. The transpiration rate was higher in rain shelter shading plate than in polyethylene net. A linear equation was obtained between net photosynthetic rate and stomatal conductance in the morning. SPAD was higher in rain shelter shading plate than in polyethylene net through all growth stages. It may result from the decrease of growth progress. From investigating photosynthetic characteristics, we concluded that shading plate of rain shelter was more an efficient material to ginseng growth.

Anti-Oxidative and Anti-Inflammatory Effects of Green Tea Mixture and Dietary Fiber on Liver of High Fat Diet-Induced Obese Rats (고지방 식이로 유도된 비만 흰쥐의 간에서 식이섬유와 녹차혼합물의 항산화 및 항염증 효과)

  • This study was conducted to investigate anti-oxidative and inflammatory inhibition effects of green tea and dietary fiber mixture on liver of high fat diet-induced obese rats. 21 male rats were divided into 3 dietary groups and control group (A), high fat diet-induced group (B), and high fat (HF) diet-induced + EQ diet-$S^{(R)}$ diet group (C). Immunoblotting and RT-PCR analysis revealed protein expression, and anti-oxidant analysis revealed MDA (malondialdehyde), GSH (glutathione), and free DPPH radical. As a results, Body weight and food consumption were not significantly different between groups. The levels of MDA and GSH were lower in HF + $EQS^{(R)}$ group than in HF group. Also, the $EQS^{(R)}$ demonstrated to be more effective than HF group for a DPPH radicals scavenging activities. In addition, protein and mRNA level of TNF-$\alpha$ in HF + $EQS^{(R)}$ group showed relatively more potent pro-inflammatory activity inhibition compared to HF group. These results suggest that green tea mixture (EQ diet-$S^{(R)}$) provide positive effects on anti-oxidative and inflammatory inhibition effects on obese animal model or obesity related diseases.

Enhancement of Anticancer Activities from Lithospermum erythrorhizon Extracts by Ultra High Pressure Process (초고압 가공 공정을 통한 지치 추출물의 항암 활성 증진)

  • This study was performed to enhance anticancer activities of Lithospermum erythrorhizon by eluting high amount of shikonin through ultra high pressure process. Extraction yield was increased up to 5~10% by ultra high pressure process, compare to the normal extraction processes such as water solvent extraction, 70% ethyl alcohol solvent extraction. The cytotoxicity of the extracts ($1.0{\mu}g/m{\ell}$) from ultra high pressure process was showed the lowest cytotoxicity 13.4% for human lung cell (HEL299). The anticancer activities showed 80~85% by adding $1.0{\mu}g/m{\ell}$ of the extracts from ultra high pressure process in several cancer cell lines such as AGS, Hep3B, MCF-7 and HeLa cells. Among them, MCF-7 cell of the endocrine system was highest inhibited than other cells. The anticancer activities of the extracts from ultra high pressure extraction process showed 10~15%, which was higher than the extracts from normal extraction processes. From HPLC analysis of the extracts, the contents of shikonin in the extracts from ultra high pressure process was 11.42% (w/w), which was 20% higher than others. This results indicate that ultra high pressure process could increase the extraction yield of shikonin and other contents, which resulted in higher anticancer activities.

Immunoregulatory Effects of Water Extracts of Inonotus obliquus in Carbon Tetrachloride-Induced Liver Damage Animal Model (사염화탄소로 유도된 간 손상 동물모델에서 차가버섯 열수 추출물이 면역 조절 기능에 미치는 영향)

  • Inonotus obliquus is one of the immune-regulatory substances and is recognized to play the role in the metabolic process of inflammation, allergy and immuntiy. The purpose of this study was to evaluate the effects of water extracts of Inonotus obliquus (IOW) on the liver lymphocyte immune function in the Sprague-Dawley male rats treated with carbon tetrachloride ($CCl_4$) to induce liver damage. Rats were fed with each experimental diet and water for 4 weeks. We found that effects of IOW on interferon-gamma (IFN-$\gamma$), signal transducer and activator of transcription 1 (STAT1), phospho-signal transducer and activator of transcription 1 (pSTAT1) and GATA-binding protein 3 (GATA-3) were decrease in vivo. Interleukin-4 (IL-4), STAT6, pSTAT6 and T-box expressed in T-cells (T-bet) decreased significantly lower in $CCl_4$+IOW group than the $CCl_4$ group. Our data indicated that cytokine protein production were increased in $CCl_4$ group and $CCl_4$+IOW group. As a result of this study, we assume that IOW fed could regulate the immuno-modulating functions through regulate the cytokine production capacity activated by liver damage.
