Studies on the Safety Assessment of Red-ginseng Radix Extract Solution for Herb-acupuncture (약침용 홍삼추출액의 급성독성시험에 관한 연구)

  • The components of Red-ginseng radix extract solution for herb- acupuncture were analyzed by HPLC. According to the Medical Product Safety Administration Guidelines for safety assessment, mice and rats were used for acute toxicity test. The results were summarized as follows; 1. In the Saponin contents(%) of Red-ginseng radix extract, Ginsenoside $Rb_1$ Saponin was 0.27% in raw material, 1.67% in extract powder and Ginsenoside Rc Saponin was 0.16% in raw material, 1.12% in extract powder and Ginsenoside Rd Saponin was 0.08% in raw material, 0.54% in extract powder. 2. There were no abnormal findings in acute toxicity test treated with Red-ginseng radix extract solution for herb-acupuncture and $LD_{50}$ could not be measured.

Determination of Buprenorphine in Raw Material and Pharmaceutical Products Using Ion-pair Formation

  • A simple and sensitive extractive spectrophotometric method has been described for the determination of buprenorphine either in raw material or in pharmaceutical formulations. The developed method is based on the formation of a colored ion-pair complex (1 : 1 drug/dye) of buprenorphine and bromocresol green (BCG) in buffer pH 3 and extracting in chloroform. The extracted complex shows absorbance maxima at 415 nm. Beer's law is obeyed in the concentration range of 1.32-100.81 μ g mL-1. The proposed method has been applied successfully for the determination of drug in commercial sublingual tablets and injectable dosage form. No significant interference was observed from the excipients commonly used as pharmaceutical aids with the assay procedure.

Hibiscus hamabo Exerts Anti-inflammatory Effects in Lipopolysaccharide-induced RAW 264.7 Cells

  • Hibiscus hamabo is a deciduous shrub that grows around salt marshes and is considered a semi-mangrove plant found in Asia. There are no studies on the biological activity of H. hamabo except for studies on the anthocyanin content. We investigated the anti-inflammatory effects of H. hamabo extract (HHE) on lipopolysaccharide (LPS)-induced RAW 264.7 cells. As nuclear factor-kappa B (NF-kB) induced by LPS moves into the nucleus, inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), and inflammatory cytokines are promoted and the inflammatory reaction begins. The nitric oxide (NO) production decreased by the treatment of HHE. Moreover, mRNA levels of inflammation-related cytokines, such as tumor necrosis factor-α (TNF-α), interleukin (IL)-6, and IL-1β, were significantly suppressed by HHE. Similarly, the expressions of iNOS and COX-2 were also decreased. The phosphorylation of p65, a subunit of NF-κB, was suppressed by HHE. As a result, HHE can be used as an effective natural material for the anti-inflammatory agent.

Inhibitory Effects of Soyeum Pharmacopuncture (SPP) on LPS-induced Inflammation Related Cytokine Expressions of RAW 264.7 cells (소염약침의 LPS로 유도된 RAW 264.7 세포의 염증성 사이토카인 발현에 미치는 영향)

  • Aim This study was done to investigate whether SPP has inhibitory effects on the activation of RAW 264.7 cells. Method In tumor necrosis factor-a (TNF-a)/ interleukin-1b (IL-1b) and IL-6, the mRNA expression of molecular indicators related to inflammatory changes of the Reumatoid Arthritis (RA) were examined using quantitative real-time PCR. Results The treatment of SPP significantly suppressed the expression of proinflammatory cytokines and chemokines such as TNF-a, IL-1b, IL-6 compared with the control. The expression of NOS-II was considerably reduced, which was accompanied by a reduction in the production of nitric oxide (NO). It also reduced the expression of $TNF-{\alpha}$ in serum of Balb/c mice compared with control group. Conclusion SPP is an effective herbal material for suppressing the inflammation related cytokines of RAW 264.7 cells.

  • The purpose of this study is to develope prototype EPID system and improve image quality of radiation therapy field imaging system using CCD camera. For this research we used Linac(Clinac 4/100), Copper metal plate, $Gd_2O_2S_2$ phosphor and CCD camera(Photronic). In this study we find best thickness of buil-up metal plate and acquired projection image of humanoid head phantom. Also we enhanced raw image data using superposition and histogram stretching method. Through the thickness optimized of metal plate and image processing, we confirmed of an improved image quality of an EPID system using CCD camera. As result, highest quality image was acquired at 1mm thickness of Copper metal plate and improved image quality by image processing methods.

  • In this study, effective cosmetic raw material weighing and bulk manufacturing management system were constructed by using bar code or quick response code (QR code) and database. Raw material labels and weighing labels for bulk manufacturing were published in web environment using the information entered in the database using ScriptX, a print component of Medi&Co. By checking the weighing and manufacturing process by using scanner, tablet and PC, it was possible to remarkably improve the product error caused by erroneous amount or misapplication which is the most cause of error in the production of cosmetic bulk. In conclusion, applying a database that utilizes bar code and QR code to cosmetics manufacturing can reduce the various problems in the process, thereby improving quality control and productivity of cosmetics.

  • We have in vestigated on physico-chemical and biological test of plastic transfusion set which was manufactured from the three companies after $Co^{60}\;{\gamma}-radiation$. The results obtained were as follows ; 1) In physico-chemical test generally showed a tendency to decrease of pH and tensile strength, and increase of oxidizable matter, phosphoric acid-phosphosphate and heavy metals in its eluted solution by irradiation of 2,5 Mrad. 2) In biological test including toxicity and safety test were passed but indicating haemolysis range the haemolysis test were failed to pass from 30% to 50%. 3) We have found that the raw materials of plastic transfusion set according to manufactures are all different and especially the most radioresistant raw material wss found in the product which was manufactured from C company.

Antiaging Effects of Musa sapientum L. (Banana) Leaf Extract (바나나 잎 추출물의 주름개선 효과)

  • To examine the possibility of using Musa sapientum L. (Banana) leaf extract as a cosmetic raw material, banana leaves grown in Jeju Island were extracted with 70% ethanol. Polysaccharides present in banana leaf extract were discarded by precipitation with cold ethanol. Polysaccharide-discarded banana leaf extract promoted procollagen and COL1A1 gene expression, but inhibited matrix metalloproteinase (MMP)-1 and MMP-2 gene expression in human skin fibroblasts when examined by real-time reverse-transcription polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA). The active compound in banana leaves was identified by fractionation with various solvents. The chloroform fraction showed the highest anti-wrinkle efficacy and the active compound of chloroform fraction was identified as corosolic acid by NMR, FT-IR, EA, and HPLC-MS. In addition, banana leaf extract showed anti-oxidative efficacy with an IC50 value of 67.91 ppm, as determined by DPPH free radical scavenging assay. Finally, the anti-wrinkle efficacy of banana leaf extract-containing cream was confirmed by clinical tests. Based on these results, banana leaves could have an application as a cosmetic raw material with anti-wrinkle efficacy.

  • In order to examine anti-inflammatory effect of Cinnamomi Ramulus (CR) alcohol extract on rheumatoid arthritis, the present study investigated the viability and TNF-${\alpha}$ production in Raw264.7 cells treated with CR and collagen induced arthritis in DBA/1J mice which were orally administered with CR prior to immunization. The results are as follows: CR extract at $20{\mu}g/ml$ inhibited the viability of Raw264.7 by 50%. CR at 15 and $20{\mu}g/ml$ showed a significant decrease in TNF-${\alpha}$ levels from Raw264.7 cells treated with LPS. CR administration decreased arthritic index in DBA/1J mice immunized with bovine collagen type II but it did not reach statistical significance. CR administration significantly decreased spleen weights obtained from mice in 6 weeks after immunization. CR administration significantly decreased serum anti-type II collagen antibody levels compared with control group. CR administration decreased serum IL-6 levels compared with control group but it did not reach statistical significance.

  • From the root of Angelica gigas Nakai, decursinol angelate was isolated, which is structural isomer of decursin. Raw material contained average 2.44% of decursinol angelate and 3.30% of decursin by the GC analysis.
