Study on Geochemical Behavior of Heavy Metals by Indigenous Bacteria in Contaminated Soil and Sediment (국내 일부 오염 토양 및 퇴적물 내 토착 미생물에 의한 중금속의 지구화학적 거동 연구)

  • Microbial control of the geochemical behavior of heavy metals (Cd, Cu, Pb, and Zn) and As in contaminated subsurface soil and sediment was investigated through activation of indigenous bacteria with lactate under anaerobic condition for 25 days. The results indicated that dissolved Cd, Pb and Zn were microbially removed from solutions, which was likely due to the formation of metal sulfides after reduction of sulfate by indigenous sulfate-reducing bacteria. Soils from the Dukeum mine containing a large amount of sulfate resulted in complete removal of dissolved As after 25 days by microbial activities, while there were gradual increases in dissolved As concentration in soils from the Hwabuk mine and sediments from the Dongducheon industrial area which showed low $SO_4{^2-}$ concentrations. Addition of appropriate carbon sources and sulfate to contaminated geological media may lead to activation of indigenous bacteria and thus in situ stabilization of the heavy metals; however, potential of As release into solution after the amendment should be preferentially investigated.

Impact of Application Rates of Pre-planting Liming Fertilizers on Changes in Soil Chemical Properties and Growth of 'Melody Yellow' Pansy in Plug Production (팬지 'Melody Yellow'의 플러그 육묘시 석회질 비료의 시비수준이 토양화학성 변화와 식물생장에 미치는 영향)

  • Objective of this research was to investigate the influence of liming fertilizers on changes in soil chemical properties and growth of pansy 'Melody Yellow' in plug production. To achieve this, dolomite (DO) with 0, 1.0, 3.5, 8.0 or $13.0g{\cdot}L^{-1}$ and calcium carbonate (CC) with 0, 2.0, 2.5, 3.0, 3.5, or $4.0g{\cdot}L^{-1}$ in application rate were incorporated into peatmoss + vermiculite (1:1, v/v) during the formulation. The treatments of $3.5g{\cdot}L^{-1}$ of DO and 2.5 or $3.0g{\cdot}L^{-1}$ of CC had acceptable range of soil solution pH such as 5.6-6.2. Faster rising of pH was observed in root media containing CC rather than those of DO, indicating higher solubility of CC. The soil Ca concentrations in all treatments of CC were 2 times as high as those of DO. The treatments of 3.5 or $8.0g{\cdot}L^{-1}$ of DO had the highest soil Mg concentrations, but all treatments of CC had lower soil Mg concentrations than control indicating that additional applications of Mg fertilizers are required. The elevated application rate of DO or CC resulted in the increase of fresh and dry weight; however, plant heights were not influenced by application of liming fertilizers. The results of tissue analysis showed that application of DO or CC influenced the Ca and Mg contents, but did not influence the contents of other nutrients such as N, P, Fe, Mn, Zn and Cu.

Influence of Interferon-${\tau}$ on the Production of Prostaglandins, Cyclooxygenase-2 Expression In Vitro and Release of Progesterone in Bovine Endometrial Cells

  • The purpose of the present study was to investigate the effect of IFN-${\tau}$ on prostaglandin synthesis, cyclooxygenase-2 (COX-2) gene expression in vitro and concentration of progesterone (P4) in endometrial cells. Epithelial and stromal cells cultured in vitro were isolated from bovine endometrium and stimulated with increasing doses of IFN-${\tau}$ (0, 0.02, 0.2 and 2 ug/ml). Human chorionic gonadotropin (hCG, 1.5 IU/ml) was used as a positive control. Prostaglandin $E_2$ and $F_{2{\alpha}}$ levels in the culture media were analyzed by enzyme immunoassays and total RNA was extracted from the cells for RT-PCR. P4 concentrations of blood samples were assayed by chemiluminescent immuno assays system. In epithelial cells, COX-2 gene expression was increased in the presence of IFN-${\tau}$ (p<0.05), but it was not significantly different in all groups of stromal cells except for 2 ug/ml IFN-${\tau}$ group (p<0.05). Although IFN-${\tau}$ did not affect $PGE_2$ and $PGF_{2{\alpha}}$ production in epithelial cells, it decreased $PGE_2$ and $PGF_{2{\alpha}}$ production significantly in stromal cells (p<0.05). In vivo experiment, blood concentration of P4 was significantly increased after addition of IFN-${\tau}$ (1 ug/ml). The results indicate that PG production was mediated by COX-2 expression in stromal cells but it was not affected in epithelial cells and this suggest that treatment of IFN-${\tau}$ could improve the implantation environment of uterine by maintenance of high P4 concentration.

Cheonan Patrol Combat Corvette Sinking and Media Control: A Production Study on Investigative Programs (천안함 침몰 사건과 미디어 통제: 탐사보도 프로그램 생산자 연구)

  • This study examines why follow-up investigative programs about the sinking of the Cheonan Patrol Combat Corvette(ROKS Cheonan) have not been produced, despite much speculation surrounding the cause of the sinking since November 2010, when KBS investigative program, In-Depth 60 Minutes which had covered the incident was aired. We have uncovered four reasons through a series of in-depth interviews with producers and reporters, as follows. First, Korean military authorities monopolized relevant information, while the producers could not prove or confirm the validity of the findings of the JIG(Joint Civilian-Military Investigation Group)'s report because the facts had been revealed partially and selectively by the authorities and the press' scientific investigation were rejected by the authorities. Second, the crew of In-Depth 60 Minutes was subjected to severe disciplinary action by the Korea Communications Commission. This caused a chilling effect, - it broke the producers' resolve to further explore the reason behind the sinking. Furthermore, the screening of the investigative documentary, Project Cheonan Ship was cancelled without prior notice, presumably by the power of the State and markets. Third, the reorganization and shake-up of personnel by broadcasting stations' presidents appointed by the power of the State crushed the spirit of PD Journalism. Finally, the "red purge" factions stigmatized by the political or military, or the mainstream press, caused the producers or broadcasters to engage in self-censorship. The idea has been used in Korea as a smoke screen to deflect public attention. Nevertheless, the producers hope to shed light on the Cheonan sinking incident and to prove reasonable doubts by pursuing follow-up investigations.

Assessment for Effect of Water Environment by Addition of Improvement Agents on Sediments (저질 개선제의 주입에 의한 수 환경에 미치는 영향 평가)

  • Control if Sediment is very important in prawn farm due to the eruption of toxic materials such as unionized $H_{2}S,\;NH_{3}\;and\;NO_3$. In this study, column test was conducted with filter media such as activated carbon, zeolite, oyster shell and iron chloride to evaluate the reduction of toxicity from sediment. ammonia-N($NH_3$) was effectively removed by Zeolite and oyster shell. It was indicated that ammonium ion($NH_4^+$) was removed by ion exchange of zeolite. And the ammonia in the column of oyster shell was existed as the form of $NH_4^+$, which is not toxic for prawn because oyster shell was stably kept at $8{\sim}9g$ of pH. Therefore, some of ammonia($NH_4^+$) was removed by oyster shell. Hydrogen sulfide and COD were effectively removed by adsorption of activated carbon and a partial removal of hydrogen sulfide was accomplished by Oyster shell. Phosphorous was removed by activated carbon, oyster shell and iron chloride. In prawn farm, the concentration of ammonia was increased with increase of pH by algae photosynthesis in the column of activated carbon, zeolite and iron chloride, but it was revealed that pH was stably kept in the column of oyster shell.

Propagation of Cutting Method of a Rare Endemic Juniperus chinensis var. sargentii Henry in Korea (희귀수종 눈향나무(Juniperus chinensis var. sargentii Henry)의 삽목증식)

  • Juniperus chinensis var. sargentii Henry is a short and creeping evergreen shrub which reaches about 60 cm in height and only occurs in the northeast Asia and in the top of high mountains over the Korea. The Korea Forest Service protects it strictly by law since J. chinensis var. sargentii is an eccentric plant and possibly may be exterminated soon in Korea. This study was carried out to develop the propagation technique by cutting for conservation of genetic resources of J. chinensis var. sargentii. The rooting responses of branch cuttings, obtained from hard(May) and semi-hard wood shoots (August) to four growth regulators, namely, IAA, IBA, NAA and Rooton(exceptionally powder method) applied at various concentrations(0, 100, 200, 500, 1000 and 2000 $mgL^{-1}$) were examined in mixed soil media. Rooting rate showed significant difference between cutting times, among kinds and among concentrations of growth regulators. The optimum cutting time was April to May in hardwood cutting. The application of IBA 1000 $mgL^{-1}$(rooting rate : 36.4%) was most effective in callus formation and rooting of cutting. Relatively, rooting of cutting of the control taken in May was 30.4%. Root characteristics such as number, length and diameter of root were not significantly affected by kinds and concentrations of growth regulators in hardwood cutting.

Bacterial Contamination of Dental Unit Water Systems in a Student Clinical Simulation Laboratory of College of Dentistry (치과대학 임상 시뮬레이션 실습실 치과용 유니트 수계의 세균 오염도 조사)

  • The water supplied from dental unit water systems (DUWS) in dentistry may be heavily contaminated with bacteria and thus may be a potential source of infection for both practice staff and patients. The aim of this study was to evaluate the level of heterotrophic bacteria and to confirm the presence of opportunistic pathogens from DUWS in student clinical simulation laboratory of college of dentistry. Water samples were collected from 36 ultrasonic scalers in student clinical simulation laboratory. The levels of heterotrophic bacteria in water samples were quantified by counting colony forming units (CFUs) on R2A agar media. In addition, opportunistic pathogens were detected by using the polymerase chain reaction (PCR) method. The mean CFUs were 16,095 CFU/ml for water samples and all of water samples exceeded current American Dental Association recommendations of 200 CFU/ml. Pseudomonas species and non-tuberculous Mycobacterium species were detected in the one sample and two samples, respectively, among the 36 water samples by the PCR with specific primers for these bacteria. Our study indicated that DUWS in student clinical simulation laboratory can cause potential infection in students and participants. This study suggested the dental unit water line management and wearing personal protective equipment in student clinical simulation laboratory will be needed to reduce bacterial contamination.

Identification of electron beam-resistant bacteria in the microbial reduction of dried laver (Porphyra tenera) subjected to electron beam treatment (전자선 처리에 따른 마른 김(Porphyra tenera)의 미생물 저감화 효과와 저항성 세균의 동정)

  • This study investigated the effect of electron beam (EB) treatment on the microbial reduction of dried laver (Porphyra tenera) and identified EB-resistant bacteria from the treated dried laver. After EB treatments of 4 kGy and 7 kGy, the numbers of total bacteria and EB-resistant bacteria were measured using tryptic soy agar and mannitol salt agar, respectively. The morphological and biochemical characteristics of each isolated EB-resistant bacteria were investigated and these bacteria were identified. Compared to the control ($1.5{\pm}0.2){\times}10^6CFU/g$, the total bacterial number was significantly decreased to ($5.4{\pm}0.5){\times}10^4CFU/g$ and ($1.1{\pm}0.6){\times}10^4CFU/g$ after EB treatments of 4 kGy and 7 kGy, respectively. With a higher EB dosage, the number of red colonies was almost same, whereas the number of yellow colonies was significantly decreased to ($3.3{\pm}1.2){\times}10^3CFU/g$ and 0 CFU/g for 4 kGy and 7 kGy, respectively. All red and yellow colonies were gram-positive cocci, catalase-positive, and resistant to 3% and 5% NaCl media. From the 16S rDNA sequence analysis, yellow and red colonies were identified as either Micrococcus flavus or M. luteus, with 99% similarity for the yellow colonies, and Deinococcus proteolyticus and D. piscis, with 99% and 97% similarity for the red colonies, respectively.

Physiological Characteristics of Actinomycetes Isolated from Turfgrass Rhizosphere (잔디 근권에서 분리된 Actinomycetes균주의 생리학적 특성)

  • Total 443 isolates of actinomycetes were isolated from turfgrass rhizosphere as potential biological control agents. The two isolates (S11 and S4) showed highest cellulase activity with compared to the other isolates that exhibited a clear zone of 1.2 mm around the colony on cellulose agar medium. S12 strain appeared the most active chitin degrading, which exhibited a 1.2 mm of clear zone. The highest proteolytic activity on skim milk agar was which exhibited a 7.5 mm of clear zone by S2 strain. S1 strain from the soli showed siderophore production ability, which exhibited a 0.6 mm of large clear zone on chrome azurol S agar. The antifungal activity of the volatile compound producing by 4 selected actinomycetes was investigated that inhibition rate against Rhizoctonia solani AG2-2 and Sclerotinia homoeocarpa. Growth inhibition effect of S8 isolate against S. homoeocarpa was appeared to 94.8%, S2 to 76.9%, S5 to 46.1% and S12 to 43.5%. The significant inhibition effects on mycelial growth of S. homoeocarpa were shown on media with four strains. The inhibition effect was the highest with S8 strain treatment at 94.8%.

The Inhibitory Effects of Intestine-oriented Lactobacillus sp. KP-3 on the Accumulation of Heavy Metals in Sprague Dawley rats (Sprague Dawley 쥐에서 장내 유래 Lactobacillus sp. KP-3의 중금속 축적 저해 효과)

  • To investigate the effect of lactic acid bacteria on the heavy metal adsorption from internal organs and blood, lactic acid bacteria were isolated from human feces. Some strains resistant to heavy metals were selected by incubation in agar media containing each of chrome and cadmium salts. Among them, a strain named KP-3 was ultimately chosen due to its higher growth rate in selective broth medium containing the heavy metals at the concentration of 0.01%. The strain was identified as Lactobacillus sp. based on its morphological, cultural and physiological characteristics. For evaluating the ability to prevent accumulation of heavy metals by selected Lactobacillus sp. strain in vivo, Sprague Dawley rats were fed with heavy metal salts (cadmium, chrome and lead) with or without cultured whole cells for 7 days. The amounts of heavy metals accumulated in liver, kidney and blood were analyzed. As a result, chrome was accumulated to kidney mostly, and lead was frequently found in liver and kidney. Experimental group (rats fed with lactic acid bacteria) showed less accumulation of heavy metal than control group (rats fed with saline solution). The inhibition rates of heavy metal accumulation were calculated to 41.8% (Cd), 33.4% (Cr) and 44.2% (Pb). Especially, feeding lactic acid bacteria strongly reduced accumulation of cadmium in blood. The results showed that feeding Lactobacillus sp. KP-3 could prevent the bioaccumulation of heavy metals in the living body.
