The Study of Sagunja-tang, Ijin-tang, Yukgunja-tang on the Change of Cerebral Hemodynamics in Rats (사군자탕, 이진탕, 육군자탕이 뇌혈류역학변동에 미치는 실험적 연구)

  • This experimental study was designed to investigate the effects of Sagunja-tang(SGJT), Ijin-tang(IJT), Yukgunja-tang(YGJT) on the change of cerebral hemodynamics [regional cerebral blood f1ow(rCBF), mean arterial blood pressure(MABP), and pial arterial diameter (PAD)] in normal rats, and further to determine the mechanism of action of YGJT. And, this Study was designed to investigate whether YGJT inhibit lactate dehydrogenase(LDH) activity in neuronal cells. The results were as follows ; 1. SGJT significantly increased rCBF but MABP was not changed comparing with normal MABP(l00 %). This results were suggested that SGJT significantly increased rCBF by dilating PAD. 2. IJT significantly decreased rCBF in a dose-dependent, but significantly increased MABP in a dose-dependent. This results were suggested that IJT significantly decreased rCBF by contracting PAD. 3. YGJT significantly increased rCBF and PAD in a dose-dependent, and YGJT increased MABP compared with normal MABP(100 %). This results were suggested that YGJT significantly increased rCBF by dilating PAD. 4. The YGJT-induced increase in rCBF was significantly accelerated by pretreatment with indomethacin (IDN, 1 mg/kg, i.p.), an inhibitor of cyclooxygenase but was significantly inhibited by methylene blue (MTB, 10 ㎍/㎏ i.p.), an inhibitor of guanylate cyclase. 5. The YGJT-induced increase in PAD and MABP were accelerated by pretreatment with IDN but was significantly inhibited by MTB. This results suggested that the mechanism of YGJT is mediated by guanylate cyclase. 6. YGJT inhibited significantly LDH activity in neuronal cells. This results were suggested that YGJT prevented the neuronal death. I thought that YGJT should have improvement of cerebral hemodynamics and inhibitive effect on the brain damage.

Effect of Cheonmabanhwa-Tang on the Changes in Cerebral Hemodynamics in Cerebral Ischemia of Rats (천마반하탕이 뇌허혈로 인한 뇌혈류역학적 변동에 미치는 영향)

  • Cheonmabanhwa- Tang (CBT) has been used in the Oriental Medicine for many centuries as a therapeutic agent for dizziness due to Poong-Dam, We reported that CBT had effects on the cerebral hemodynamics [regional cerebral blood flow (rCBF), pial arterial diameter (PAD), and mean arterial blood pressure (MABP)] in normal and cerebral ischemic rats. Therefor we designed to determine the mechanism of action of CBT. The changes of rCBF and MABP were determinated by laser-Doppler flowmetry(LDF), and the change of PAD was determinated by video-microscopy. The results were as follows: The CBT-induced increase in rCBF was significnatly inhibited by pretreatment with indomethacin (IDN, 1 ㎎/㎏, i.p.), an inhibitor of cyclooxygenase, or methylene blue (MTB, 10 ㎍/㎏, i.p.), an inhibitor of guanylate cyclase. The CBT-induced increase in PAD was also significantly inhibited by pretreatment with IDN or MTB. The CBT-induced increase in MABP was also significantly inhibited by pretreatment with IDN or MTB. In conclusion, it is suggested that CBT causes a diverse effect on cerebral hemodynamics via mediation of cyclooxygenase and guanylate cyclase.

A Comparative Study of action Mechanism on the Cerebral Hemodynamics by Cheonghunhwadam-tang and Cheonghunhwadam-tang adding Gastrodiae Rhizoma in Rats (청훈화담탕 및 청훈화담탕가천마에 의한 뇌혈류역학의 작용기전에 대한 비교연구)

  • Cheonghunhwadam-tang(CHT) have been used in oriental medicine for many centuries as a therapeutic agent of vertigo by wind, fire and phlegm. CHTGR was CHT adding Gastrodae Rhizoma. The effects of CHTGR on the regional cerebral blood flow(rCBF) and mean arterial blood pressure(MABP) is not known. A comparative Study of action-mechanism of CHT and CHTGR on the cerebral hemodynamics is not known too. Therefore, purpose of this Study was to investigate effects of CHT and CHTGR on the rCBF and MABP, compare action-mechanism of CHT and CHTGR on the rCBF and MABP. The changes of rCBF and BP was determinated by Laser-Doppler Flowmetry(LDF). The results were as follows ; CHT extract was increased rCBF in a dose-dependent, but was not changed MABP compared with CHT non-treated group. CHTGR extract was decreased rCBF and MABP compared with CHTGR non-treated group in a dose-dependent. Action of CHT is not related with adrenergic β-receptor, cyclooxygenase and guanylate cyclase, but action of CHTGR is related with guanylate cyclase.

Effects of Geopungjeseub-tang(Gufengchushi-tang) on the Changes of Cerebral Blood Flow in Rats (거풍제습탕이 뇌허혈이 유발된 백서의 뇌혈류 변화에 미치는 영향)

  • Objectives : Geopungjeseub-tang(Gufengchushi-tang) has been used in oriental medicine for many centuries as a therapeutic agent for hemiplegia caused by deficiency of qi(氣虛) and damp phlegm(濕痰). This study was performed to evaluate effects of Geopungjeseub-tang extract(GJT) on hemodynamics[regional cerebral blood flow(rCBF), pial arterial diameter(PAD), mean arterial blood pressure(MABP), heart rate(HR)] in normal rats and in rats with cerebral ischemia by middle cerebral artery(MCA) occlusion. Also, effects of adrenergic ${\beta}-receptor$, cyclooxygenase on response to GJT were evaluated. Methods : Laser-doppler flowmetry(LDF) measured changes of rCBF, MABP and HR. Video microscope and width analyzer measured changes in PAD. Results : rCBF and PAD increased after treatment with GJT(10mg/kg, i.v.) during the period of cerebral reperfusion, and pretreatment with indomethacin raised rCBF and PAD increased after treatment with GJT during the same period as above. Pretreatment with propranolol decreased rCBF, but increased after GJT treatment, but raised PAD increased after GJT treatment during this period of reperfusion. Conclusion : CR caused diverse responses were observed in rCBF and PAD after treatment with GJT. ACF action is mediated by adrenergic ${\beta}-receptor$ and cyclooxygenase. Result suggest that GJT has an anti-ischemic effect through the improvement of cerebral hemodynamics and has theraputic potential for cerebral apoplexy.

Effects of Gleditsiae Spina(GS) Extract in Cerebral Blood Flow in Rats (조각자(皂角刺) 추출물(抽出物)이 정상 흰쥐의 뇌혈류(腦血流) 변화(變化)에 미치는 영향(影響))

  • Objectives : This study was designed to investigate the effects of Gleditsiae spina(GS) on changes in cerebral blood flow in rats. Methods : The present study was investigated regional cerebral blood flow(rCBF) and mean arterial blood pressure(MABP) in rats. In addtion, the present study also investigated action mechanisms of GS on changes in rCBF and MABP by Pre-treatment with indomethacin(IDM) and methylene blue(MTB), an inhibitor of a vasodepressor material. Results : Treatment with GS elevated rCBF in dose-dependent manner, but MABP levels were not affected by treatment with GS. Pre-treatment with indomethacin(IDM), an inhibitor of cyclooxygenase, inhibited increase of rCBF effectively. And pre-treatment with methylene blue(MTB), an inhibitor of guanylate cyclase, inhibited increase of rCBF induced by GS too. In addition, Pre-treatment with MTB inhibited increase of MABP too. But, pre-treatment with IDM did not affect MABP levels. Conclusions : These results suggest that GS is effective to treat patient with disease related to cerebral ischemia, because GS can increase rCBF. In addition, the mechanisms are thought to be related to both of cyclooxygenase and guanylate cyclase pathways.

The Effect of Jaeumgeonbitang adding Evodiae Fructus Extract on the Cerebral Hemodynamics in Rats (자음건비탕가오수유(滋陰健脾湯加吳茱萸) 추출물이 흰쥐의 뇌혈류역학에 미치는 영향)

  • Objective : Jaeumgeonbitang have been used in Korean medicine for many centuries as a therapuetic agent of vertigo. JAE was extract of Jaeumgeonbitang adding Evodiae Fructus. The effects of JAE on the cerebral blood flow and blood pressure is not known. This study was designed to investigate the effects of JAE on the ischemic crebral injuries. Method : We performed to investigate effects of JAE on the changes of regional cerebral blood flow(rCBF) and mean arterial blood pressure (MABP) in normal and ischemic rats, and further to determine the mechanism and cytokines production ($IL-1{\beta}$, $TNF-{\alpha}$, IL-10, $TGF-{\beta}$) of JAE. Results : In normal rats, JAE significantly increased rCBF and significantly decreased MABP in a dose-dependent manner. This result suggested that JAE significantly increased rCBF by dilating pial arterial diameter. Increase of JAE-induced rCBF was significantly inhibited by the pretreatment with indomethacin (1 mg/kg, i.p.), an inhibitor of cyclooxygenase, and was significantly inhibited by methylene blue ($10{\mu}g/kg$, i.p.), an inhibitor of guanylate cyclase. Decrease of JAE-induced MABP was significantly increased by the pretreatment with indomethacin (1 mg/kg, i.p.), an inhibitor of cyclooxygenase. So, these results suggested that the mechanism of JAE was mediated by cyclooxygenase. In ischemic rat, the rCBF was significantly and stably increased by JAE (10 mg/kg, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in Control group. In cytokine production of serum by drawing from femoral arterial blood at 1 hr after reperfusion, Sample group (JAE 10 mg/kg treated group) was significantly decreased $IL-1{\beta}$ and $TNF-{\alpha}$ production compared with Control group. In cytokine production of serum by drawing from femoral arterial blood at 1 hr after reperfusion, Sample group was significantly increased IL-10 production compared with Control group. Conclusion : These results suggested that JAE was significantly and stably increased regional cerebral blood flow by inhibited $IL-1{\beta}$ and $TNF-{\alpha}$ production, and increased IL-10 production.

Effects of Folium Perillae on Cerebral Ischemia in Rats (소엽(蘇葉) 추출물이 백서(白鼠)의 뇌허혈에 미치는 효과)

  • Objectives : Folium perillae (FP) can relieve superficial pathogenic factors to dissipate cold and promote the circulation of qi and regulate the function of the stomach and is often used for interior qi-stagnation. We hypothesized that FP could rescue cerebral ischemia in rats. Methods : The present study was carried out to investigate the effects of FP on cerebral ischemia in terms of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in rats. Finally, lactate dehydrogenase (LDH) release was investigated, too. Results : In this study, treatment with FP elevated rCBF and MABP levels in dose-dependent manner. Pre-treatment with indomethacin, an inhibitor of cyclooxygenase, inhibited rCBF increase induced by FP effectively. However, FP did not affect stability during cerebral reperfusion. Finally, FP significantly inhibited LD H activity in vitro Conclusions : These results suggest that FP is useful to treat patient with diseases related to cerebral ischemia, because FP can increase rCBF and MABP.

Both rCBF and PAD were significantly and stably increased by CBT(10 mg/kg, i.p.) during the period of cerebral reperfusion. which contrasted with the findings of rapid and marked increase in control group. In conclusion, it is suggested that CBT causes a diverse effect on cerebral hemodynamics thereby has an anti-ischemic action.

Effects of Onion Vinegar on the Cerebral Blood Flow and the Safety Examination (양파식초가 뇌혈류 및 안전성에 미치는 영향)

  • The aim of this study is to investigate the effects of onion vinegar on the cerebral blood flow by measuring the changes of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) and by observing the recovery of focal ischemic brain injury in rats. Rats are divided into thee groups depending on the medication; control group (no medication), 8.8-OV group (vinegar using 8.8 brix onion medication), 14.6-OV group (vinegar using 14.6 brix onion medication). The medication of onion vinegar significantly increased rCBF but decreased MABP. This result suggests that onion vinegar significantly increased rCBF by dilating arterial diameter. In addition, focal ischemic brain injury is induced in rats by middle cerebral arterial occlusion. The recovery from focal ischemic brain injury is more significantly improved in the groups using onion vinegar compared to the control group. The amount of recovery is measured by the GAP-43 and the medication of onion vinegar significantly increased GAP-43. This result suggests that onion vinegar is effective on the nerve regeneration. After the medication, the change of body weight, outcomes of renal and liver function test, and outcomes of CBC are analysed for safety examination. There are no statistical differences among control group and all experimental groups in the body weight, renal and liver function test, and CBC. In conclusion, these results suggest that onion vinegar can increase rCBF in normal state, and improve the stability of rCBF in ischemic state.
