Metabolic Pharmacokinetics in Rats: Differences between Pure Amygdalin and Amygdalin in a Decoction of Peach Seeds

  • The pharmacokinetics of prunasin after oral administration of amygdalin or a decoction of peach seeds was determined and compared in rats. A $C_{18}$ column was used for separation at a column temperature of $25^{\circ}C$. The mobile phase consisted of 20% aqueous acetonitrile, and the flow rate was 0.5 mL/min. After oral administration of a decoction of peach seeds, prunasin was absorbed rapidly, reaching a maximum plasma concentration ($C_{max}$) of 62.1 mg/L within 45 min. After oral administration of amygdalin, the absorption of prunasin was delayed. The $C_{max}$ of prunasin was 42.9 mg/L and was reached at 60 min. Values for the pharmacokinetic parameters of prunasin, including $T_{max}$, $C_{max}$, AUC, $T_{1/2}$, CL/F, and V1/F, were significantly different for the oral administration of amygdalin compared with that of a decoction of peach seeds.

  • A simple and sensitive high-performance liquid chromatographic method for quantitation of hydrochlorothiazide in human plasma was developed and bioavailability parameters of hydrochlorothiazide were assessed in Korean healthy male volunteers. Caffeine was used as an internal standard. Hydrochlorothiazide and internal standard in plasma sample were extracted using tert-butylmethylether (TBME). A centrifuged upper layer was then evaporated and reconstituted with mobile phase of acetonitrile-25 mM phosphate buffer (20/80, pH 2.5). The reconstituted samples were injected into a Luna C18 column $(250{\times}4.6\;mm,\;5{\mu}m)$ at a flow-rate of 1.0 ml/min. The wavelength was set at 230 nm and no endogenous substances were found to interfere, A linear relationship for hydrochlorothiazide was found in the range of $10{\sim}300\;ng/ml$. The lower limit of quantitation (LLOQ) was 10 ng/ml with acceptable precision and accuracy. Assayed in plasma, the intra- and inter-day validation for all coefficients of variation (R.S.D.%) were found less than 15%. Main pharmacokinetic parameters of 50mg of hydrochlorothiazide were revealed as follows: $AUC_t\;1761{\pm}509.0\;ng{\cdot} hr\;ml,\;C_{max}\;296.5{\pm}95.5\;ng/ml,\;T_{max}\;1.94{\pm}0.85hr,\;K_{el}\;0.12{\pm}0.04\;hr^{-1}\;and T_{12}\;6.81{\pm}2.92\;hr.\;C_{max}\;and\;T_{max}$ were in accordance with the values $(270{\sim}350\;ng/ml\;and\;1.9{\sim}2.7\;hr)$ of Caucasian.

  • 3GPP evolved packet system (EPS) is an all-IP based system that supports various access networks such LTE, HSPA/HSPA+, and non-3GPP networks. Recently, the support of IP flows with packet level QoS profiles was added to the requirements of the EPS. This paper proposes an adaptive modulation and coding (AMC) scheme that supports the QoS of such IP flows in the 3G LTE access network of the EPS. Defining the retransmission as a critical factor for QoS, the proposed scheme applies different maximum packet error probability $P_{max}$ to each packet when selecting the AMC transmission mode. In determining $P_{max}$, the QoS constraints and NACK-to-ACK error as well as channel condition are considered, balancing two objectives: the satisfaction of the QoS and the maximization of spectral efficiency. The simulation results show that it is able to reduce both delay violation and status report by 10%, while improving the throughput 10% in comparison with an existing scheme.

Effects of Verapamil in Cardioplegic Perfusates on the Ischemic Myocardium in Isolated Rat Heart (흰쥐의 적출된 심장에서 Verapamil이 허혈성 심근에 미치는 효과)

  • Using isolated rat heart preparations, we observed the protective effe ts of verapamil cardioplegia on ischemic myocardial injury. Isolated rat hearts were subjected to global ischemia at $25^{\circ}C$ Twenty four isolated Sprague Dawley rat hearts underwent 30 minutes of the retrograde nonworking perfusion with Krebs-Henseleit buffer solution followed by $25^{\circ}C$ cardioplegic solution (St. Thomas'Hospital Cardioplegic Solution) for 60 minutes. Before ischemic arrest, rat hearts were treated with cold cardioplegic solution in control group (n=12) and cold cardioplegic solution with verapamil (1 mg/L) in experimental group (n=12). After 60 minutes of ischemia, hemodynamic and biochemical parameters such as heart rate, left ventricular pressure (LVP), + dp/dt max, coronary flow and creatine phosphokinase (CPK) were measured before giving cardioplegia and 30 minutes after reperfusion. Verapamil group exhibited greater recovery of heart rate, LVP, +dpldt max, coronary flow and CPK than control group (p < 0.05).

Maximum Capacity-based Minimum Cut Algorithm (최대 수용량-기반 최소절단 알고리즘)

  • The minimum cut problem is to minimize c(S,T), that is, to determine source S and sink T such that the capacity of the S-T cut is minimal. The flow-based algorithm is mostly used to find the bottleneck arcs by calculating flow network, and does not presents the minimum cut. This paper suggests an algorithm that simply includes the maximum capacity vertex to adjacent set S or T and finds the minimum cut without obtaining flow network in advance. On applying the suggested algorithm to 13 limited graphs, it can be finds the minimum cut value $_{\min}c$(S, T) with simply and correctly.

Turbulent Wake Flow around Tubes in Single Row Tube Banks (일렬관군에서의 난류 후류특성에 관한 연구)

  • An experimental study is conducted to investigate the turbulent wake flow around tubes in vertical single row tube banks. All measurements are performed at Reynolds number(Re$_{max}$) 4.2*10$_{3}$ - 2.5*10$_{4}$ with varying tube spacings from the wide pitch ratio(H/D=3.07) to the very narrow one(H/D=1.23). Flow patterns are visualized using the smoke-wire method. Mean static pressures, velocity components, and various statistical quantities of turbulence are obtained by the computer on-line technique. In the case of wide tube spacings, the near wakes of tube show similar trends to those of a single tube, and their flow indicats an anisotropic turbulence. However, as the pitch ratio decreases, wide and narrow wakes appear alternately behind adjacent tubes due to the deflected flow. Also, in the case of H/D .leq. 1.54, Karman vortex is not formed at the side of relatively wide wake.e.

  • Fischer-Tropsch synthesis reaction converts syngas (mixture of CO and H2) to valuable hydrocarbon products. Simulation of low temperature Fischer -Tropsch Synthesis reaction and heat transfer at intensified process condition using catalyst filled single and multichannel microchannel reactor is considered. Single channel model simulation indicated potential for process intensification (higher GHSV of $30000hr^{-1}$ in presence of theoretical Cobalt based super-active catalyst) while still achieving CO conversion greater than ~65% and $C_{5+}$ selectivity greater than ~74%. Conjugate heat transfer simulation with multichannel reactor block models considering three different combinations of reactor configuration and coolant type predicted ${\Delta}T_{max}$ equal to 23 K for cross-flow configuration with wall boiling coolant, 15 K for co-current flow configuration with subcooled coolant, and 13 K for co-current flow configuration with wall boiling coolant. In the range of temperature maintained (498 - 521 K), chain growth probability calculated is desirable for low-temperature Fisher-Tropsch Synthesis.

  • The roots of teeth exposed by gingival recession, may be successfully covered by various type of gingival grafting procedures. Vascularization of the recipient site is an essential determinant of the grafts' survival during the first healing stages. It has been suggested that a procedure by which they stimulate the periosteum presurgically will induce the proliferation of neo-endothelium in the site to be operated. The purpose of this study is to evaluate the variations in the gingival blood flow during 4weeks after periosteal stimulation in patient scheduled to receive gingival grafts and to compare variations in the gingival blood flow between smoker and non-smoker. Laser Doppler Flowmetry(floLAB(R), Moor Instruments Ltd, England ; wave length = 780mm. Max. power =l.6mW) was used to measure the gingival blood flow. 112 sites of 68 male patients (32 smokers and 36 non-smoker), aged between 23 and 48 years (smoker : 24-44 years. mean=32.6, non-smoker : 23-48 years, mean=28.5) were monitored for the blood flow. Gingival blood flow measured at before periosteal stimulation, 1-, 2-, 3-, and 4-weeks after periosteal stimulation from 10 a.m. to 2 p.m. The difference of blood flow in each measuring time, each measuring site and between smokers and non-smokers were statistically analyzed by MANOVA. The results were as follows : (1) Blood flow stayed increased for 2 weeks, and then, it was a tendency to decrease(p<0.05). (2) There was no statistically significant difference of blood flow change between smokers and non-smokers. (3)The blood flow at middle site had lower than mesial and distal site during the measuring periods(p<0.05). The present study suggested that blood flow change following periosteal stimulation was significant difference, thus periosteal stimulation before gingival graft might induce favorable results in gingival recession patient.

Sensitivity Analysis of the SWMM Model Parameters Based on Design Rainfall Condition (설계강우조건에 따른 SWMM모형 매개변수의 민감도 분석)

  • This study is a sensitivity analysis of the parameters which affect the simulation results under various design rainfall conditions, using the SWMM model, for three selected basins in urban areas. The sensitivity of the peak flow rate is defined by $S_Q$ (=1.0 - (min. ratio of peak flow rate/max. ratio of peak flow rate)), and the rainfall conditions are classified in terms of design rainfall frequency, duration, and distribution. The simulation results show that in most conditions the parameters - the impermeable area ratio, the sewer slope, and the initial infiltration capacity - have more significant effects on the results than other parameters. As the design rainfall frequency increases, the sensitivity of the sewer slope and sewer roughness increases, while the parameters related with the surface runoff decrease. When the rainfall duration increases, the sensitivities of most parameters of surface runoff and sewer flow decrease. Also, at the 1st quarterly Huff rainfall distribution condition, the impermeable area ratio has high sensitivity, but at the 4th quarterly condition the parameters related with sewer flow show higher sensitivities. These tendencies can be explained by considering the procedure for computing the effective rainfall and kinematic wave on the surface and sewer flow.

Peak Expiratory Flow(PEF) Measured by Peak Flow Meter and Correlation Between PEF and Other Ventilatory Parameters in Healthy Children (정상 소아에서 최고호기유량계(peak flow meter)로 측정한 최고호기유량(PEF)와 기타 환기기능검사와의 상관관계)

  • Background : In diagnosis or monitor of the airway obstruction in bronchial asthma, the measurement of $FEV_1$ in the standard method because of its reproducibility and accuracy. But the measurement of peak expiratory flow(PEF) by peak flow meter is much simpler and easier than that of $FEV_1$ especially in children. Yet there have been still no data of the predicted normal values of PEF measured by peak flow meter in Korean children. This study was conducted to provide equations to predict the normal value of PEF and correlation between PEF and $FEV_1$ in healthy children. Method : PEF was measured by MiniWright peak flow meter, and the forced expiratory volume and the maximum expiratory flow volume curves were measured by Microspiro HI 501(Chest Co.) in 346 healthy children(age:5-16 years, 194 boys and 152 girls) without any respiratory symptoms during 2 weeks before the study. The regression equations for various ventilatory parameters according to age and/or height, and the regression equations of $FEV_1$ by PEF were derived. Results : 1. The regression equation for PEF(L/min) was: $12.6{\times}$age(year)+$3.4{\times}$height(cm)-263($R^2=0.85$) in boys, and $6{\times}$age(year)+$3.9{\times}$height(cm)-293($R^2=0.82$) in girls. 2. The value of FEFmax(L/sec) derived from the maximum expiratory flow volume curves was multiplied by 60 to compare with PEF(L/min), and PEF was faster by 125 L/min in boys and 118 L/min in girls, respectively. 3. The regression equation for $FEV_1$(ml) by PEF(L/min) was:$7{\times}$PEF-550($R^2=0.82$) in boys, and $5.8{\times}$PEF-146 ($R^2=0.81$) in girls, respectively. Conclusion : This study provides regression equations predicting the normal values of PEF by age and/or height in children. And the equations for $FEV_1$, a gold standard of ventilatory function, was predicted by PEF. So, in taking care of children with airway obstruction, PEF measured by the peak flow meter can provide useful information.
