Whitening effect of extracts from Matricaria chamomilla L. with B16F10 melanoma cells (B16F10 melanoma cell을 이용한 캐모마일(Matricaria chamomilla L.) 추출물의 미백 효과)

  • Matricaria chamomilla L. has been used as a bath agent in Europe because of its sterilization effect on the skin. Flowers contain terpenes, flavonoids are effective in relieving inflammation. Matricaria chamomilla L. has been reported to have various drug efficacies such as sedation, anti-diabetic effect and anti-arthritic effect, but there is little research on the scientific efficacy of whitening effect. The purpose of this study was to examine the whitening effect of Matricaria chamomilla L. extract and to investigate the mechanism of inhibition of melanogenesis. The extracts were used to determine tyrosinase inhibitory activity. The tyrosinase inhibitory activity of extracts was ineffective for water extract but in 60% ethanol extract was shown in a concentration-dependent manner. B16F10 melanoma cell was measured using a powder obtained by lyophilization of 60% ethanol extract. The toxicity was observed at a concentration of $75{\mu}g/mL$. And concentration range was selected to be at most $50{\mu}g/mL$. The effect of tyrosinase, MITF, TRP-1 and TRP-2 on the expression of melanin protein was investigated in melanoma cells of B16F10 melanoma cells. As a result, it was confirmed that as the concentration of the extract increased, the melanogenesis level decreased and the protein expression level also decreased in a concentration dependent manner. Therefore, it was concluded that Matricaria chamomilla L. extract inhibited melanogenesis in cells. Based on the above results, it is expected that it will be used as a useful basic data for industrialization of whitening functional food of Matricaria chamomilla L.

In Silico Analysis of Potential Antidiabetic Phytochemicals from Matricaria chamomilla L. against PTP1B and Aldose Reductase for Type 2 Diabetes Mellitus and its Complications

  • Type 2 diabetes mellitus (T2DM) and its complications are important noncommunicable diseases with high mortality rates. Protein tyrosine phosphatase 1B (PTP1B) and aldose reductase inhibitors are recently approached and advanced for T2DM and its complications therapy. Matricaria chamomilla L. is acknowledged as a worldwide medicinal herb that has many beneficial health effects as well as antidiabetic effects. Our research was designed to determine the most potential antidiabetic phytochemicals from M. chamomilla employing in silico study. 142 phytochemicals were obtained from the databases. The first screening employed iGEMdock and Swiss ADME, involving 93 phytochemicals. Finally, 30 best phytochemicals were docked. Molecular docking and visualization analysis were performed using Avogadro, AutoDock 4.2., and Biovia Discovery Studio 2016. Molecular docking results demonstrate that ligand-protein interaction's binding affinities were -5.16 to -7.54 kcal/mol and -5.30 to -12.10 kcal/mol for PTP1B and aldose reductase protein targets respectively. In silico results demonstrate that M. chamomilla has potential antidiabetic phytochemical compounds for T2DM and its complications. We recommended anthecotulide, quercetin, chlorogenic acid, luteolin, and catechin as antidiabetic agents due to their binding affinities against both PTP1B and aldose reductase protein. Those phytochemicals' significant efficacy and potential as antidiabetic must be investigated in further advanced research.

  • Chlorsulfuron (chemical name : 2-Chloro-N-(4-methoxy-6-methyl-1,3,5-triazin-2-yl)-aminocarbonyl-benzenesulfonamide) is a herbicidally active ingredient which shows effect against susceptible weeds already at such low rates like 5-20g active ingredient per hectare. In the here reported trials metabolism in several sensitive cultivated plants and weeds have been analysed using ^{14}C-labelled active ingredient. The uptake of chlorsulfuron by leaves or the root system is good in all plants species, and translocation takes place either symplasmatically or apoplasmatically. Metabolism takes place in all investigated plant species by development of hydrophile suhstances in roots and shoots. Decomposition of chlorsulfuron in roots and shoots of tolerant species (Triticum aestivum and Hordeum vulgare) to polare substances takes place quantitatively faster and quicker than in susceptible species (Beta vulgaris and Matricaria chamomilla).

  • Chamomile (Matricaria chamomilla), a member of the Asteraceae family, is a well-known for medicinal plant and can be found in India and Europe. Chamomile is an effective sedative and various medical effects. But, the effects of acne treatment by chamomile were not investigated. Therefore, we assessed the anti-oxidant effects, anti-microbial activity and anti-inflammatory effects by chamomile extracts in HaCaT keratinocyte cells. Anti-oxidant effects of chamomile extracts were investigated by DPPH assay. Also, results of MTT assay was demonstrated that chamomile extracts did not have a cytotoxic effect in HaCaT cells. To assess the antimicrobial activity, we determined formation of inhibition zone of Propionibacterium acnes by extracts from chamomile. Tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) induces production of inflammatory cytokines such as interleukin-$1{\beta}$ (IL-$1{\beta}$), IL-6 and IL-8 and expression of COX-2. Chamomile extracts could inhibit TNF-${\alpha}$-induced mRNA expression levels of IL-$1{\beta}$, IL-6, IL-8 and COX-2 gene. These results demonstrated the possibility of chamomile for prevention and treatment of skin inflammatory diseases such as acne.

  • The essential oil fraction of Chrysanthemum sibiricum Turcz. (Compositae) was analysed by means of TLC and GLC. Utilizing silicagel column chromatography, a blue-color compound was isolated, and purified by preparative TLC. The obtain compound had the same Rf-value on TLC and exactly same UV-visible and IR spectra with that of chamazulene isolated from Matricaria chamomilla L. The composition of the essential oil was also compared with those from flowers of Chrysanthemum indicum L. and Chrysanthemum morifolim Ramatuelia.

Volatile components of chamomile(Matricaria Chamomilla L.) cultivated in Korea (한국산 카모밀의 향기 성분)

  • The essential oils from whole plant, flower, seed, stem and leaves of domestic chamomile were extracted by simultaneous distillation-extraction and analyzed by GC/ MSD/IRD and retention index matching. The experimental results revealed the presence of over 31 volatile components. Major components were chamazulene, bisabolol, bisabolol oxide A, B. The contents of these major components which possess the pharmacological effects were found to be flower(75.1%), seed(76.6%), stem and leaves(10.09%), whole plant (48.9%), respectively, in domestic chamoile oil, whereas found to be flower(49.2%) in foreign chamomile oil. These results suggest that the usefulness of domestic chamomile is promising because of high contents of these four major components which posses pharmacological effects.

  • 꽃차(Flower tea)는 최근 다양한 제품이 개발되고 소비가 확대되는 등 그 산업적 가치가 증대하고 있다. 꽃차의 향기 특성은 우렸을 때 나타나는 색깔 및 인체에서의 생리활성과 더불어 주요한 꽃차 품질결정 요소이다. 본 연구는 꽃차로의 이용이 활발한 국화과 식물 중 구절초(Dendranthema zawadskii var. latilobum), 국화(Chrysanthemum morifolium), 노랑색 및 주황색 마리골드(Tagetes erecta 'Yellow' and 'Orange'), 그리고 캐모마일(Matricaria chamomilla)의 향기 성분특성을 구명하기 위하여 제조된 꽃차를 headspace-solidphase microextraction/gas chromatography-mass spectrometry (HS-SPME/GC-MS)를 이용하여 분리, 동정하였다. 국화과 꽃차로부터 총 117종의 휘발성 성분이 확인되었는데, 각 꽃차 종류별 동정된 휘발성 성분의 개수, 주요 3개 휘발성 성분과 전체 향기성분 peak중 이들이 차지하는 구성비율(%)은 다음과 같았다: 구절초 64종, camphor (31%), α-pinene(14%), camphene(14%); 국화 60종, camphor(15%), chrysantheny acetate(13%), eucalyptol (11%); 마리골드 '옐로우' 53종, 2,4-heptadienal(26%), trans-isocarveol(21%), cis-β-Copaene(18%); 마리골드 '오렌지' 61종, β-caryophyllene(16%), β-ocimene epoxide(12%), β-ocimene(12%); 캐모마일 50종, β-farnesene(63%), nonane(9%), spathulenol(5%). 국화과 꽃차 5종 모두에서 공통적으로 검출된 성분은 β-caryophyllene, α-pinene, β-farnesene 등 10종이었으며 마리골리 '옐로'는 '오렌지'와 주요 향기성분의 조성에서 뚜렷한 차이를 나타내었다. 비록 그 함량은 낮았으나 구절초, 국화, 마리골드 '오렌지', 그리고 캐모마일은 각각 10종, 12종, 3종 및 13종이었다. 마리골드 '엘로'의 경우 검출된 모든 향기성분은 마리골드 '오렌지'나 다른 국화과 식물의 꽃차에서도 검출된 바, 향기 성분이 다양성이 다소 낮게 나타났다.

Biological Activity of Chamomile (Matricaria chamomilla L.) Extracts (캐모마일(Martricaria chamomilla L.)의 생리활성)

  • The biological activity of water and ethanol extracts from Chamomaile for functional food source were examined. Total phenol contents in the $60\%$ ethanol extracts $(24.98\pm0.20\;mg/g)$ from Chamomaile leaf was higher than those of water extracts $(23.64\pm0.35\;mg/g)$ The major phenolic compound by HPLC were rosemarinic acid and Quercetin. $60\%$ ethanol extracts had higher content of these phenolics than water extacts. Electron donating ability showed $91.05\%$ in the water extracts and $95.49\%$ in the $60\%$ ethanol extracts. Antioxidant protection factor (PF) showed $0.71\pm0.02 $ in the water extracts and $1.48\pm0.03 $ in the $60\%$ ethanol extracts. The water extracts of Chamomaile leaves did not have antimicrobial activity against H. pylori, but the $60\%$ ethanol extracts revealed the slight antimicrobial activity as 9.42 mm of clear zone. Angiotensin converting enzyme inhibition was $57.98\%$ in water extracts and $91.36\%$ in $60\%$ ethanol extracts. Xanthine oxidase activity was $73.48\%$ in water extracts and $81.96\%$ in $60\%$ ethanol extracts. The results suggest that Chamomailes extract may be useful as potential source as antioxidant, angiotensin converting enzyme and xanthine oxidase inhibitors.

Effect of Ginger and Chamomile on Nausea and Vomiting Caused by Chemotherapy in Iranian Women with Breast Cancer

  • Background: Chemotherapy-induced nausea and vomiting (CINV) places a significant burden on the patient. Herbal agents are the most commonly complementary therapies used among the public. This study was done to determine the effect of ginger and chamomile capsules on nausea and vomiting in cases undergoing chemotherapy for breast cancer (BC). Materials and Methods: In a randomized, double-blind and clinical trial study, 65 women with BC undergoing chemotherapy were referred to Breast Cancer Research Center, Tehran, Iran, between May 2013 to June 2014. Regimen for ginger group for 5 days before and 5 days after chemotherapy was: 2 times a day and 500 mg capsules of powdered ginger root in addition to a routine antiemetic regimen consisting of dexamethasone, metoclopramide and aprepitant (DMA) capsules. Chamomile group similarly was: 2 times a day and 500 mg capsules of Matricaria chamomilla extract in addition to a routine antiemetic regimen consisting of DMA capsules. Control group, routine antiemetic regimen consisting of DMA capsules. Results: There were no significant differences between the ginger, chamomile and control groups regarding age. Drugs used for chemotherapy were identical and duration of disease was also matched (1-4 months). Ginger and chamomile were both significantly effective for reducing the frequency of vomiting, there being no significant difference between the ginger and chamomile groups. Moreover, unlike the chamomile, ginger significantly influenced the frequency of nausea. Conclusions: According to the findings of this study, it should be declared that taking ginger capsules (1 g/day) might relieve CINV safely. Nurses dealing directly with cancer patients should be responsible for providing educational programs for patients and their families about how to deal with their drug regimens and associated side effects.
