Bone Mineral Density and Breast Cancer Risk Factors among Premenopausal and Postmenopausal Women - A Systematic Review

  • Background: Bone mineral density (BMD) is a lifetime marker of estrogen in a woman's body and has been associated with increased breast cancer risk. Nonetheless the actual association is still debatable. Furthermore, estrogen is very crucial in maintaining human bone density and gradually decreases over age. A systematic search was conducted to assess any association of BMD with breast cancer risk factors among premenopausal and postmenopausal women. Materials and Methods: Review identification was performed through databases searching on MEDLINE, CINAHL and SCOPUS and 19 qualified studies were elected. The keywords used were "bone mineral density", "breast cancer", and "breast density". Results: A total of 19 articles showed variation with the majority of the studies focused on postmenopausal and a few focused on premenopausal women. Overall there was no concensus on effects. Conclusions: An enormous effort is being undertaken by researchers to prove that BMD might be one of the significant risk factors for breast cancer.

  • 선택적 표식인자가 전위인자일때, 표식인자의 선택 순서에 따라 동시형질도가 달라진다. 본 연구에서는 이 현상을 설명하기 위하여 아래와 같은 수학적 접근을 시도하였다. 먼저, transducing particle 들의 형성과정을 다섯종류로 유별하였다. 그런다음, 하나의 표지가 고정될 가능성을 의미하는 확률함수들을 확률밀도 개념에 근거하여 수학적으로 정식화였다. 그리고 실제 값들과 유용한 가정들을 이식들에 결합하여 그로부터 얻어진 이론적인 빈도를 실측 값들과 비교하였다. 이려한 분석에 의하여 그 빈도의 상이성이 일반 재조합에 필요한 상용성을 전위인자가 제공하지 봇하고, 그 주변에 재조합 빈도를 거리에 의존하는 형태로 감소시키기 때문이라는 결론을 얻었다.

Feasibility of Serum Pentosidine Level as a Potential Risk Factor for Osteoporotic Vertebral Compression Fracture

  • Study Design: Feasibility study. Purpose: To evaluate the feasibility of using serum pentosidine level as a potential marker for osteoporotic vertebral compression fracture (OVCF). Overview of Literature: A review of previous studies suggests a negative correlation between serum pentosidine concentration and bone strength. However, it is unclear whether serum pentosidine level might be a potential marker of OVCF in Koreans. Methods: Forty patients who underwent bone mineral density examination were included in this study, and their serum pentosidine levels were prospectively analyzed. Serum pentosidine level was evaluated using enzyme-linked immunosorbent assay. Among all the patients, 11 with OVCF were assigned to the vertebral fracture group and 29 who did not have vertebral fracture were included in the non-fracture group. In addition, we used the Fracture Risk Assessment (FRAX) tool Korean version for assessing the 10-year probability of fracture. Results: There was a statistically significant difference in the mean serum pentosidine level (p=0.04) of the vertebral fracture group (110.8 ng/mL) and the non-fracture group (64.3 ng/mL). Logistic regression analyses showed that serum pentosidine was significantly associated with OVCF. The vertebral fracture group had significantly higher 10-year probability of major osteoporotic fracture as per FRAX than the non-fracture group. There was a positive correlation between pentosidine level and FRAX results (r=0.35, p=0.02). Conclusions: These results suggest that increased serum pentosidine level could be a potential marker for OVCF.

Effects of Colostrum Basic Protein from Colostrum Whey Protein: Increases in Osteoblast Proliferation and Bone Metabolism

  • Colostrum basic proteins (CBP) (MW 1$\sim$30 kDa) were isolated from bovine colostrum using a series of ultrafiltration processes and their effects on osteoblast cell proliferation and bone metabolism were investigated in cell line and animal models. Treatments with CBP (1, 10, 100 $\mu$g/mL) dose-dependently increased cell proliferation of osteoblastic MC3T3 cells. Alkaline phosphatase activity, a marker of osteoblastic phenotype, in the cells was also increased after treatments with CBP in a dose-dependent manner. Significant increases in bone density were observed in femur of ovariectomized rats which were fed a diet with 1% and 10% CBP, compared to rats fed a normal diet. These results suggest that CBP may increase bone mass and density and be useful for the prevention of bone-related diseases.

  • A Digital Wave Tank simulation technique, based on a finite-difference method and a modified marker-and-cell (MAC) algorithm, is applied in order to investigate the characteristics of nonlinear Tsunami propagations and their interactions with a 2D sloping beach, Ohkushiri Island, and to predict maximum wove run-up around the island. The Navier-Stokes (NS) and continuity equation are governed in the computational domain, and the boundary values are updated at each time step, by a finite-difference time-marching scheme in the frame of the rectangular coordinate system. The fully nonlinear, kinematic, free-surface condition is satisfied by the modified marker-density function technique. The near shore Tsunami is assumed to be a solitary wave, and is generated from the numerical wave-maker in the developed Digital Wave Tank. The simulation results are compared with the experiments and other numerical methods, based on the shallow-water wave theory.

  • A digital wave tank (DWT) simulation technique has been developed by authors to investigate the interactions of fully nonlinear waves with 3D marine structures. A finite-difference/volume method and a modified marker-and-cell (MAC) algorithm have been used, which are based on the Navier-Stokes (NS) and continuity equations. The fully nonlinear kinematic free-surface condition is implemented by the marker-density function (MDF) technique or the Level-Set (LS) technique developed for one or two fluid layers. In this paper, some applications for various engineering problems with free-surface are introduced and discussed. It includes numerical simulation of marine environments by simulation equipments, fully nonlinear wave motions around offshore structures, nonlinear ship waves, ship motions in waves and marine flow simulation with free-surface. From the presented simulations, it seems that the developed DWT simulation technique can handle various engineering problems with free-surface and reliably predict hydrodynamic features due to the fully-nonlinear wave motions interacting with such marine structures.

Localization of Quantitative Trait Loci for Bone Mineral Density on Chromosome 13 in the Mongolian Population

  • Although the genetic basis for bone mineral density (BMD) has been studied by many groups so far, genes responsible for this complex trait has not been completely revealed. In order to localize quantitative trait loci (QTLs) for BMD variation in Asian population, the study was designed using a group of Mongolian population, a genetically closed population with a homogeneous lifestyle. BMD was measured at the left and right wrists and ankles using DEXA in 1,082 participants from 142 families. Genotyping of 13 polymorphic microsatellite markers on chromosome 13 (average spacing 8-9 cM) and two-point and multipoint linkage analysis were performed. In two-point linkage analysis, we identified two markers, D13S175 (6.03 cM) and D13S265 (68.73 cM) that had LOD scores greater than 1 for left ankle (LOD=2.09, LOD=1.49, respectively). We also found a marker D13S175 (6.03 cM) with a high LOD for left wrist (LOD=1.49) and the markers D13S265 (68.73 cM) and D13S217 (17.21 cM) for the right wrist (LOD= 1.82, LOD= 1.62, respectively). Among these significant marker regions, only two regions at 17 cM (13p11) and 65 cM (13q21) for the right wrist overlapped with major QTLs reported in following multipoint linkage analysis (LOD= 1.7549, LOD=1.4462, respectively). This study provides the possible evidence of the presence of QTLs affecting right wrist BMD in Mongolian populations on 13p11 and 13q21. Modest evidence was also found for genes affecting left ankle and left wrist BMD on 13p13.

Effects of Single Nucleotide Polymorphism Marker Density on Haplotype Block Partition

  • Many researchers have found that one of the most important characteristics of the structure of linkage disequilibrium is that the human genome can be divided into non-overlapping block partitions in which only a small number of haplotypes are observed. The location and distribution of haplotype blocks can be seen as a population property influenced by population genetic events such as selection, mutation, recombination and population structure. In this study, we investigate the effects of the density of markers relative to the full set of all polymorphisms in the region on the results of haplotype partitioning for five popular haplotype block partition methods: three methods in Haploview (confidence interval, four gamete test, and solid spine), MIG++ implemented in PLINK 1.9 and S-MIG++. We used several experimental datasets obtained by sampling subsets of single nucleotide polymorphism (SNP) markers of chromosome 22 region in the 1000 Genomes Project data and also the HapMap phase 3 data to compare the results of haplotype block partitions by five methods. With decreasing sampling ratio down to 20% of the original SNP markers, the total number of haplotype blocks decreases and the length of haplotype blocks increases for all algorithms. When we examined the marker-independence of the haplotype block locations constructed from the datasets of different density, the results using below 50% of the entire SNP markers were very different from the results using the entire SNP markers. We conclude that the haplotype block construction results should be used and interpreted carefully depending on the selection of markers and the purpose of the study.

Analysis of the oxidized low density lipoprotein receptor 1 gene as a potential marker for carcass quality traits in Qinchuan cattle

  • Objective: The oxidized low density lipoprotein receptor 1 (OLR1) gene plays an important role in the degradation of oxidized low-density lipoprotein and adipocyte proliferation in mammals. For this reason, we aimed at investigating the association of OLR1 gene polymorphisms with carcass quality traits in Chinese Qinchuan cattle. Methods: The single nucleotide polymorphism (SNP) was identified in the 3' untranslated region of bovine OLR1 gene by DNA sequencing. In addition, the haplotype frequency and linkage disequilibrium estimates of three SNPs were evaluated in 520 individuals. Results: Results indicated that the studied three SNPs were within the range of moderate genetic diversity (0.25< polymorphism information content<0.5). Haplotype analysis of three SNPs showed that ten different haplotypes were identified, but only five haplotypes were listed as those with a frequency of <0.05 were excluded. The Hap3 ($-G_1T_2C_3-$) had the highest haplotype frequency (42.10%). Linkage disequilibrium analysis showed that the three SNPs had a low linkage ($r^2<0.001$). The T10588C and C10647T were significantly associated with backfat thickness and intramuscular fat content in Qinchuan cattle. Conclusion: Based on our results, we believe that the OLR1 gene could be a strong candidate gene for influencing carcass quality traits in Qinchuan cattle.
