High Resolution Ocean Color Products Estimation in Fjord of Svalbard, Arctic Sea using Landsat-8 OLI (Landsat-8 OLI를 이용한 북극해 스발바드 피요르드의 고해상도 Ocean Color Product 산출)

  • Ocean Color products have been used to understand marine ecosystem. In high latitude region, ice melting optically influences the ocean color products. In this study, we assessed optical properties in fjord around Svalbard Arctic sea, and estimated distribution of chlorophyll-a and suspended sediment by using high resolution satellite data, Landsat-8 Operational Land Imager (OLI). To estimate chlorophyll-a and suspended sediment concentrations, various regression models were tested with different band ratio. The regression models were not shown high correlation because of temporal difference between satellite data and in-situ data. However, model-derived distribution of ocean color products from OLI showed a possibility that fjord and coastal areas around Arctic Sea can be monitored with high resolution satellite data. To understand climate change pattern around Arctic Sea, we need to understand ice meting influences on marine ecosystem change. Results of this study will be used to high resolution monitoring of ice melting and its influences on the marine ecosystem change at high latitude. KOPRI (Korea Polar Research Institute) has been operated the Dasan station on Svalbard since 2002, and study was conducted using Arctic station.

A comparison of the hydrolase activities of excretory-secretory products and somatic extracts from fish parasitic nematodes, Anisakis simplex sensu stricto and Anisakis pegreffii larvae (어류 기생성 선충 Anisakis simplex sensu stricto와 Anisakis pegreffii 유충의 excretory-secretory products 및 somatic extracts의 가수분해효소 활성 비교)

  • Hydrolase activities of excretory-secretory products (ESP) and somatic extracts (SE) from Anisakis simplex sensu stricto (s.s.) and Anisakis pegreffii larvae were investigated by using API ZYM kit. In esterase group, acid phosphatase showed high activity from both of A. simplex (s.s.) and A. pegreffii. Esterase (C4) showed activity only from SE and A. simplex (s.s.) showed higher activity than A. pegreffii. Alkaline phosphatase, acid phosphatase and naphthol-AS-BI-phosphohydrolase showed higher activity in 3rd stage larvae than in 4th stage larvae of both species. In aminopeptidase group, only leucine arylamidase showed remarkable activity in SE of both anisakid species, and A. simplex (s.s.) SE showed higher activity than A. pegreffii SE. In glycosidase group, N-acetyl-${\beta}$-glucosaminidase, ${\alpha}$-mannosidase, ${\alpha}$-fucosidase showed higher activity in A. simplex (s.s.) than A. pegreffii, and 4th larvae showed higher activity than 3rd larvae. These differences in hydrolase activity of anisakid nematodes larvae are thought to be due to different metabolism such as growth, moulting, digestion and feeding.

Differences by RAPD-PCR Analysis within and between Rockfish (Sebastes schlegeli) Populations from the Yellow Sea and the Southern Sea in Korea (황해 및 남해산 조피볼락 (Sebastes schlegeli) 개체군 사이의 RAPD-PCR 분석에 의한 차이)

  • Polymerase chain reaction (PCR) amplification of DNA as 30 different arbitrary primers and random amplified polymorphic DNAs (RAPD) analysis were performed on genomic DNA extracted from the blood of the marine rockfish (Sebastes schlegeli) from the Yellow Sea and the Southern Sea. The unique properties of the genomic DNA were used to investigate the features of the population dynamics and origins of the species. Out of 30 primers, seven generated 207 highly reproducible RAPD polymorphic products, producing approximately 2.7 polymorphic bands per primer. About 67.4% of total amplified products (307) were either polymorphic (207) to rockfish. The degree of similarity varied from 0.22 to 0.63 as calculated by bandsharing analysis. Also, the average level of bandsharing was 0.39$\pm$0.02 within the rockfish strains. The electrophoretic analysis of RAPD-PCR products showed the relatively high levels if variation between different individuals in rockfish from the Yellow Sea. However, the RAPD outlines obtained with DNA of different rockfish strains from the Yellow Sea and the Southern Sea in Korea were very similar. Also, a small number of polymorphic bands were identified. Even if further analyses or more rockfish populations are required, this result implies RAPD analysis reflects genetic differences between the geographical strains of the rockfish.

Hepatoprotective and Anti-diabetic Effects of Pelvetia siliquosa, a Marine Algae, in Rats

  • The effects of various fractions from the whole plant of Pelvetia siliquosa Tseng et Chang (Fucaceae) on the $CCl_4$-induced hepatotoxicity as well as on streptozotocin (STZ)-induced diabetes in rats were investigated. The ether fraction exhibited a potent rat lens aldose reductase (RLAR) inhibition in vitro and showed a significant inhibition of not only serum glucose concentrations but also sorbitol accumulations in the lens, red blood cells and sciatic nerves in the STZ-induced diabetic rats. When administered orally in Sprague-Dawley rats, $H_{2}O$ fraction was found to cause a significant inhibition of the rise in the serum transaminase activities in $CCl_4$-intoxicated rats. These results suggested that this plant might possess constituents with hepatoprotective, anti-diabetic effects and those effects on diabetic complications.

Sanitary Quality Characterization of Commercial Salted Semi-dried Brown Croaker (시판 반염건 민어의 위생학적 품질 특성)

  • Salted semi-dried brown croaker, Miichthys miiuy, has been proposed as a palatable seafood in Korea due to its specific texture, taste, and nutritional content. This study was conducted to characterize the sanitary quality of commercial salted semi-dried brown croaker for the development of high quality salted semi-dried products. The chemical properties ranged from 64.2 to 77.1% (mean $74.7{\pm}3.5%$) for moisture content, from 2.1 to 9.5% for salinity, from 14.1 to 58.1 mg/100 g for volatile basic nitrogen, and from 19.1 to 107.2 meq/kg for peroxide values. The viable cell counts and Escherichia coli were 4.2~8.3 log(CFU/g) [mean: 6.2 log(CFU/g)] and 18>~4.6 log(MPN/100 g), respectively. The chemical and microbial results of commercial salted semi-dried brown croaker suggest that provisions should be established for development of high quality salted semi-dried products.

Component characteristics of fish bone as a food source (식품소재로서 어류뼈의 성분 특성)

  • As a part of basic investigation for utilizing by-products derived from seafood processing as a food source, a chemical characteristics of fish bones (cod bone, Alaska pollack bone, yellowfin sole bone, hoki bone, conger eel bone and mackerel bone) were investigated. The crude protein (40.7% on the dry basis) and collagen contents (5.86%, on the dry basis), imino acid composition (189 residues/1,000 residues) of hoki bone were higher than those of the other fish bones, but were lower than those of the animal bone. The crude lipid contents and EPA and DHA compositions of yellowfin sole, conger eel and mackerel bones were $22.8{\sim}43.9%$ on the dry basis and $15.6{\sim}23.8%$, respectively and were lower than those of squid viscera. The major ash components of the fish bones were found to be calcium and phosphorus and the contents in 100 g crude ash were $37.1{\sim}38.6%$ and $18.0{\sim}18.5%$ respectively. The calcium and phosphorus contents in 100 g crude ash of cod and Alaska pollack bones were more than those of the animal bones, as well as the others. It may be concluded, front these results, hoki bone can be effectively utilized as a processing materal of collagen or gelatin and cod and Alaska pollack bones as a calcium source.

Processing of Enzymatic Hydrolysates from Conger eel Scrap (붕장어 가공잔사를 이용한 효소분해소재의 가공)

  • In order to develope nutritional and flavoring intermediate products, the optimal processing conditions for two stage enzyme hydrolysate (TSEH) from low-utilized conger eel scrap such as head and intestine were investigated. The optimal processing conditions for TSEH were revealed in temperature at $55^{\circ}C$ 3$\~$4 hours digestion with alcalase at the 1st stage, and 4 hours at $45{\~}50^{\circ}C$ digestion with neutrase at the 2nd stage. Among water extract, steam extract and enzyme hydrolysates of conger eel scrap, the present TSEH was superior to other extracts in terms of yield ana organoleptic taste such as harmonic umami and inhibition of fishy and greasy taste formation. From the results of chemical experiments and sensory evaluation, we may conclude that TSEH of conger eel scrap could be utilized as the flavoring intermediate materials for the fisheries products such as flavoring sauces, drinkable beverage and instant food materials.

Preparation of Calcium-based Powder from Fish Bone and its Characteristics (어류뼈를 이용한 칼슘제의 제조 및 특성)

  • As a part of investigation for utilizing seafood processing by-products as a functional food source, we investigated the processing condition (extraction methods and autoclaving time) of a functional calcium-based powder from fish bones and its characteristics. The calcium-based powder from Alaska pollack bone by autoclaving in water showed lower in the calcium content and higher in the yields and the soluble calcium ratio than those from fish bone by the other calcium extraction methods. As the autoclaving time elapsed up to 40 min, the content and soluble ratio of calcium increased, and the yield decreased. On the other hand, there were no difference found in those among calcium-based powders autoclaved for 40 min and over. Judging from the results, the calcium-based powder from Alaska pollack autoclaved in water for 40 min (CAP) was superior to those from fish bone by the other processing condition. The CAP was mostly solublized by shaking $(37^{\circ}C)$ for 1 hr at the acidic condition (pH 2.0). The soluble calcium ratio of acid-solublized CAP was about 59% by shaking $(37^{\circ}C)$ for 3 hr and over at the neutral condition (pH 7.0).

Development and Sensory Characteristics of Seasoned Broughton's Ribbed Ark Scapharca broughtonii Soy Sauce with Added Mustard Leaf Brassica juncea (갓(Brassica juncea)을 첨가한 간장 피조개(Scapharca broughtonii)장의 개발 및 관능특성)

  • The home meal replacement (HMR) market has attracted significant attention due to COVID-19 pandemic and products that utilize the combination of different processed ingredients are now being developed. In this study, Broughton's ribbed ark Scapharca broughtonii was soaked in seasoned soy sauce with the incorporation of mustard leaf Brassica juncea (M-BRA), which is known to have a unique texture as well as excellent functional properties such as antioxidation, to develop a regional specialty product. The optimal conditions for manufacturing M-BRA from the seasoned soy sauce (the sauce to be added [X1] and the soaking time [X2]), were examined using response surface methodology (RSM) to analyze the significance of the salinity (Y1), amino-N content (Y2), and overall acceptance (Y3). The coefficient of determination (R2) between X1-X2 and Y1-Y3 were close to 1, thereby confirming the suitability of the RSM model. The optimal conditions were seasoned soy sauce addition of 140% and soaking time of 156 min. The M-BRA manufactured under these conditions exhibited superior overall acceptance compared to seasoned commercial soy sauce, which was used as a control. We expect that the market for M-BRA manufactured by combining marine and agricultural materials will expand owing to superior overall acceptance compared with commercial products.

Development of Marine Toxicity Standard Method for Marine Luminescent Bacteria: Introduction of N-Tox test (해양성 발광박테리아를 이용한 해양환경 독성평가 시험법 개발: N-Tox test)

  • Luminescent bacterial toxicity test was first introduced in the early 1980s, registered as international standard method in 1998 and now widely used as a common toxicity test method. This toxicity test uses luminescent bacterium, Vibrio fischeri, originated from marine environment as a test organism. The degree of toxicity can be evaluated from the comparison of luminescent emission intensity between control and treatment groups to toxicants and materials from various environmental matrix for 30 min. This test can be carried out by using commercial products and its results are sensitive and precise. This research is on the feasibility of adopting luminescent bacterial test as a domestic standard test protocol. Using commercial products, a series of experiments were conducted to identify the precision and accuracy of injection volume and light emission, and to evaluate concentration-response relationship between chemical concentrations and light emissions. Also, the feasibility of the application to environmental media and quality assurance/quality control were checked. The results of serial toxicity tests revealed that the preliminary luminescent bacterial toxicity test was robust and suitable as a standard method.
