꾸지뽕나무 열매에서 추출한 4'-O-methylalpinumisoflavone의 항산화 및 미백 효과 (Anti-oxidative and Whitening Efects of 4'-O-methylalpinumisoflavone Isolated from Fruit of Maclura Tricuspidata Carrière)

  • The anti-inflammatory effects of 4'-O-methylalpinumisoflavone (OMAI) has been reported in recent years. To develop effective and safe skin whitening agents, we investigated the anti-oxidative and melanogenic effects of OMAI isolated from fruit of Maclura tricuspidata Carrière (Cudrania tricuspidata) in macrophage and melanoma cell lines. In our results, OMAI showed effective superoxide scavenging activity and suppressed production of lipopolysaccharide (LPS)-induced intracellular reactive oxygen species (ROS) in RAW264.7 cells. In addition, α-melanocyte stimulation hormone (MSH)-induced production of melanin was also reduced by OMAI in B16F10 cells. Finally, OMAI significantly inhibited tyrosinase activity in B16F10 cells. These results suggest that OMAI suppressed melanin production via scavenging reactive oxygen species and inhibition of tyrosinase activity.

Xanthone and Flavonoid Derivatives from the Leaves of Maclura tricuspidata with Antioxidant and Anti-tyrosinase Activity

  • Masclura tricuspidata, also called as Cudrania tricuspidata, is one of the most common Moraceae family plants in East Asia. Its trivial name follows mulberry due to the similar morphology. Investigation of the bioactive constituents of M. tricuspidata leaves yielded a new xanthone derivative along with twenty known compounds through various chromatographic techniques. A new compound was defined as mascluraxanthone (3), a prenylated xanthone glucoside on the basis of 1D and 2D NMR and MS data. Twenty known compounds were identified as four xanthone derivatives (1-2 and 4-5), two flavans (6-7), six flavanol derivatives (8-13), a flavonone (14) and seven flavonol derivatives (15-21). Among the isolated compounds, flavanol and flavonoid derivatives with 3',4'-OH groups showed antioxidant and anti-tyrosinase activities. Conclusively, the leaves of M. tricuspidata are rich in aromatic compounds including xanthones and flavonoids. In addition, these constituents showed antioxidant and anti-tyrosinase potentials, which might be useful for oxidative stress related diseases.

영지버섯 균사체 발효 꾸지뽕 잔가지 추출물의 3T3-L1 지방전구세포 분화 억제 및 항염증 효과 (Effect of extract from Maclura tricuspidata twig fermented with Ganoderma lucidum mycelium on adipocyte differentiation and inflammation in 3T3-L1 cells)

  • 본 연구에서는 영지버섯 균사체 발효 꾸지뽕 잔가지 추출물의 지방세포 분화 억제 및 염증 억제 효과를 확인하고자 하였다. 세포독성을 확인한 결과, 최대 1,000 ㎍/mL 농도까지 세포에 처리하여도 세포 생존율이 감소하지 않음을 확인되었다. 분화과정 동안 발효 꾸지뽕 잔가지 추출물을 처리하였을 때, 세포 내 지방 축적이 감소되었다. 분화과정에 관여하는 대표적인 adipokine인 leptin과 adiponectin의 생성량을 확인한 결과, 발효 꾸지뽕나무 잔가지 추출물에 의해 adiponectin의 생성 증가와 leptin의 생성 감소를 확인하였다. 지방세포 분화 관련 유전자의 발현은 분화 유도인자만 처리한 군에 비해 발효 꾸지뽕나무 잔가지 추출물이 함께 처리된 군에서 추출물의 농도가 높아질수록 지방분화 전사인자인 PPARγ와 C/EBPα의 발현량이 감소하는 것을 확인하였다. 분화된 3T3-L1 세포에서 LPS에 의해 분비되는 염증성 사이토카인(TNF-α, IL-6, MCP-1)의 생성량 변화를 측정한 결과, LPS 처리로 증가된 염증성 사이토카인의 생성을 억제하였으며 염증 관련 유전자인 iNOS, COX-2의 발현도 감소되었다. 또한, 발효 꾸지뽕나무 잔가지 추출물과 발효되지 않은 꾸지뽕나무 잔가지 추출물을 비교하였을 때, 발효 꾸지뽕나무 잔가지 추출물이 지방세포 분화 및 염증 억제 효과에서 더 높은 활성을 보였으며, 이들 결과를 종합적으로 볼 때, 영지버섯 균사체를 이용한 발효가 꾸지뽕나무 잔가지의 활성 증가에 기여하는 것으로 보인다. 따라서 발효 꾸지뽕나무 잔가지 추출물은 항비만, 항염증 소재로서 비만뿐만 아니라 대사성 질환 개선을 위한 기능성 소재로서 활용이 가능할 것으로 판단되며, 향후 발효 꾸지뽕나무 추출물의 명확한 항비만, 항염증 효능을 확인하기 위해서는 유용 물질 분리 및 작용기전 등 추가적인 연구가 필요할 것으로 판단된다.

건강식품 소비자의 한약 및 천연물 온라인 구입 검색 동향 분석 및 고찰 (Analysis of health food consumers' online purchase search trend of herbal medicines and natural products)

  • Objectives : The purpose of this study was to confirm the consumption trends of Korean medicine for health food consumption of consumers by using the Naver DataLab Shopping Insight service. Methods : In this study, the search data for the category of Korean herbal ingredients in the health food field of Naver Datalab shopping insight site was collected and sorted in order of frequency from August 1st, 2017 to June 22nd, 2022. The frequently searched keywords were organized based on the inclusion of Korean Pharmacopoeia (KP), Korean Herbal Pharmacopoeia (KHP), and Food Code. Results : 67,804 keywords were collected, and the most frequent keywords appearing for more than 200 days among the top 500 were 827 (1.184%). Among the frequent keywords, there were 149 keywords related to traditional medicine names included in the KP and KHP, and five prescriptions were included. 60 keywords were not included in the KP and KHP, and the keyword with the highest search frequency was "kujibbongnamu" (Maclura tricuspidata). Conclusions : The findings of this study provide information on the consumer's interest in traditional korean medicine (TKM) and natural products (NP), and can be used as a basis for understanding the demand for TKM and NP in the online shopping market.

종 분포 모형을 활용한 제주고사리삼의 복원 및 보전 대상지 선정과 관리방안 (Selection and Management Strategies for Restoration and Conservation Target Sites of Mankyua chejuense using Species Distribution Models)

  • As the destruction of habitats due to recent development continues, there is also increasing interest in endangered species. Mankyua chejuense is a vulnerable species that is sensitive to changes in population and habitat, and it has recently been upgraded from Endangered Species II to Endangered Species I, requiring significant management efforts. So in this study, we analyzed the potential habitats of Mankyua chejuense using MaxEnt(Maximum Entropy) modeling. We developed three models: one that considered only environmental characteristics, one that considered artificial factors, and one that reflected the habitat of dominant tree species in the overstory. Based on previous studies, we incorporated environmental and human influence factors for the habitats of Mankyua chejuense into spatial information, and we also used the habitat distribution models of dominant tree species, including Ulmus parvifolia, Maclura tricuspidata, and Ligustrum obtusifolium, that have been previously identified as major overstory species of Mankyua chejuense. Our analysis revealed that rock exposure, elevation, slope, forest type, building density, and soil type were the main factors determining the potential habitat of Mankyua chejuense. Differences among the three models were observed in the edges of the habitats due to human influence factors, and results varied depending on the similarity of the habitats of Mankyua chejuense and the dominant tree species in the overstory. The potential habitats of Mankyua chejuense presented in this study include areas where the species could potentially inhabit in addition to existing habitats. Therefore, these results can be used for the conservation and management planning of Mankyua chejuense.
