치과용 폴리머 기구의 세포독성(MTT) 평가 (Cytotoxicity(MTT) evaluation of dental instruments made of polymers)

  • 최근 치과에서 사용되는 10종의 폴리머 기구에 대한 세포독성을 평가하기 위하여 L-929 세포를 이용한 MTT 시험을 시행하였다. 검체를 4g 당 20mL의 비율로 제조한 37℃의 RPMI 1640 용액에서 24시간 동안 용출한 후 식품의약품안전처 고시 제2020-12호 의료기기 생물학적 안전에 관한 공통기준규격에 따라 검체 용출액과 공시험액, 음성 및 양성대조를 사용하여, 37℃, 5% CO2 Incubator에서 24시간 배양하여 ELISA reader로 판정한 결과, Intraoral camera는 용출물 농도 약 50%에서 약 70% 이상의 세포 생존율을 나타냈나 Plastic impression tray, 3D printing tweezer, Impression disposable syringe, Dental floss holder, Hand implant scaler, Surgical retractor, Oral scanner tip, Dental mirror, Water pick tip은 모두 용출물 농도 100%에서 70% 이상의 세포 생존율로 세포독성을 나타내지 않아 구강점막에 직접 접촉하여 사용이 가능한 기구로 평가되었다.

산자고가 위암세포에 미치는 영향 (Study of Anti-Cancer Effects of Cremastrae Appendiculatae Tuber on Stomach Cancer Cells)

  • In order to investigate anti-cancer effect of Cremastrae Appendiculatae Tuber, this experiment was performed, in vitro. The results are as follows: 1. The MIT assay demonstrated that cell viability was decreased by Cremastrae Appendiculatae Tuber without statistical significance 2. The Apoptosis assay demonstated that apoptosis was induced by Cremastrae Appendiculatae Tuber without statistical significance 3. In quantitative RT-PCR analysis, there were no remarkable changes in expression levels of apoptosis-related genes, Bcl- 2, Bax, P53.

막걸리 분획물의 항산화활성 및 미백효능 (Antioxidant Activity and Whitening Efficacy of Makgeolli Fractions)

  • 본 연구에서는 식품으로 널리 애용되고 있는 막걸리의 항산화효능 및 티로시나아제 활성 억제효과를 연구하여 기능성 화장품 소재로서의 가능성을 평가하고자 하였다. 시료는 시판 막걸리 잔사에 MeOH 3 L를 가한 후 얻어진 현탁액을 농축한 후 EtOAc 3 L을 가하고 분획하여 상층을 최종시료로 사용하였다. MTT assay 평가에서는 $100{\sim}1,000{\mu}g/mL$ 범위의 농도에서 세포독성이 전혀 나타나지 않았고, 항산화활성은 농도 의존적 경향을 보였으며, EC50은 7.008 mg/mL로 높은 활성을 보였다. 또 티로시나아제 저해 활성 결과 추출물의 IC50 값은 49.22 mg/mL로 나타났다. 이상의 결과로 본 시료는 기능성화장품 원료로서의 가능성이 있음을 확인하였다.

대장암 세포주에 대한 만형자(Vitex rotundifolia) 추출물의 항암 효과 (The Anticancer Effect of Extracts from Vitex rotundifolia on Human Colon Carcinoma Cell Lines)

  • 본 연구는 만형자(Vitex rotundifolia)의 세포독성을 확인하기 위한 목적으로 methanol, ethanol, acetone으로 추출한 뒤, 인간 유래의 대장암 세포주인 HT-29에 처리하여 MTT reduction assay를 이용하여 실험을 하였다. 그 결과, 모든 만형자 추출물에서 농도 의존적으로 성장 저해 효과가 나타났으며, 그 중에서도 acetone 추출물을 50 ${\mu}g/ml$ 농도로 처리 시 18%로 가장 높은 항암 효과를 보였다. 또한 각 용매별 추출물을 50 ${\mu}g/ml$를 처리하여 HT-29 세포주의 성장을 관찰한 결과, acetone 추출물은 대조구에 비하여 암세포의 수축 등 형태변화가 두드러지게 나타났으며, Hoechst 33342 staining을 통하여 apoptotic body가 형성된 것을 확인 하였다. 그리고 가장 활성이 좋은 acetone 추출물을 용매 분획하여 얻은 분획물 중, n-hexane 분획물에서 농도 의존적으로 세포독성을 나타냄을 알 수 있었다. 또한 만형자의 n-hexane 분획물은 다른 대장암 세포주 SW620에 대해서도 50 ${\mu}g/ml$ 농도에서 대조군에 비하여 7배 정도의 높은 세포독성을 나타내고 있음을 MTT reduction assay와 LDH release assay를 통하여 확인 할 수 있었다.

五子 추출물이 Leydig 세포 내 testosterone 합성에 미치는 영향 (Effects of Extracts from Oja on Testosterone Synthesis in Leydig Cells)

  • Traditionally, 5 kinds of fruits with "ja(子)" in their name, including Rubus coreanus, Schisandra chinensis, Lycinum chineuse, Torilidis fructus, and Cuscuta seed, collectively called Oja(五子), are long known to enhance stamina. In the present study, we replaced tosaja with gyeolmyeongja(Cassiae semen ) and examined the effects of extracts from these fruits on andropause. This study investigated the antioxidant effect and testosterone synthesis of Oja water extract on Leydig TM3 cells. To investigate whether hydrogen peroxide induces oxidative stress in Leydig cells, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay, nitric oxide assay, and testosterone assay were performed on mouse Leydig TM3 cells. The results were obtained as follows: Leydig TM3 cells viability was assessed by a modified MTT assay, and the protection effect of Oja water extract against hydrogen peroxide-induced cell oxidative stress were examined by mitochondrial activity. Oja water extract could efficiently protect cytotoxicity induced by H2O2. Oja water extract promoted testosterone synthesis. These results suggest that Oja water extract has protective roles and promotes steroidogenesis in Leydig cells through its anti-oxidant action.

복분자 딸기(Rubus coreanum) 에탄올 및 열수추출물의 항돌연변이 활성과 암세포 성장 억제 효과 (Antimutagenic and Cytotoxic Activity of Ethanol and Water Extracts from Rubus coreanum)

  • The antimutagenic and cytotoxic activities of ethanol and water extracts from Rubus coreanum were investigated in this study. Their antimutagenic activities were measured by the Ames test and their cytotoxic activities were evaluated by the growth inhibition of cancer cells via the 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide (MTT) assay and the sulforhodamine B (SRB) assay. In the results, the inhibition rates of the ethanol and water extracts toward mutagenicity induced by 4-NQO were 95.0% and 93.6% at 5 mg/plate, respectively, while their inhibition rates against mutagenicity induced by sodium azide were 27.2% and 40.8%, respectively. According to MTT assay, the cytotoxicity values of the ethanol extract against Hep3B and HeLa cells were 67.2% and 68.5%, respectively, and the values for the water extract were 65.8% and 66.4%, respectively. In the SRB assay, the ethanol and water extracts inhibited over 60% of cancer cell growth. In conclusion, both the ethanol and water extracts of Rubus coreanum offer potentially good antimutagenic and anticancer effects.

Anticarcinogenic and Antigenotoxic Effects of Bacillus polyfermenticus

  • The morbidity and mortality of colon cancer are increasing, because of the westernization of food habit. Probiotics such as lactic acid bacteria (LAB) have been known to play an important role in retarding colon carcinogenesis by possibly influencing metabolic, immunologic, and protective functions in the colon. In this study, we evaluated the effect of B. polyfermenticus SCD on N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) induced DNA damage in CHO-K, cells and human lymphocytes, and on proliferation of human colon cancer cell. Using the Comet assay to detect DNA damage, we found that B. polyfermenticus SCD protected cells from the DNA damage induced by MNNG in $CHO-K_1$ cells and in human lymphocytes. B. polyfermenticus SCD was also found to inhibit the growth of colon cancer cells in a dose-dependent manner, detected by the MTT assay. These results indicate that B. polyfermenticus SCD has the potential to inhibit not only DNA damage induced by a carcinogen, but also the proliferation of colon cancer cells.

Evaluation of Anti-inflammatory activity of Fissistigma poilanei and Rubus laxus Focke

  • The aim of the study was to determine the anti-inflammatory activities of the plants with origin of China. The Fissistigma poilanei, which is a species of plant in the family Annonaceae and Rubus laxus Focke, which is a species of plant in the family Rosaceae from China were tested for anti-inflammatory activities. Samples were prepared using 95% ethanol using nitric oxide (NO) assay for assessing the anti-inflammatory activity. NO assay experiment showed that extracts of the Fissistigma poilanei give 28.3% increases in anti-inflammatory activity and extracts of Rubus laxus Focke give 57.1% decreases in anti-inflammatory activity, compared to control. To determine the cell toxicity, MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay was used. MTT assay experiment showed that Fissistigma poilanei and Rubus laxus Focke might have less toxicity of 60.5% and 62.3%, respectively, compared to control. Taken together, these experiments showed that Fissistigma poilanei extracts might have significantly higher anti-inflammatory activities and relatively lower toxicity, compared to control. This implies that this study might give a tool to develop a new potential anti-inflammatory therapeutic candidate.

Studies on Anti-inflammatory Activities of Extracts from Manglietia insignis and Tirpitzia sinensis

Blume, which is a species of plant in the family Magnoliaceae and Tirpitzia sinensis (Hemsl.) Hallier f., which is a species of plant in the family Linaceae were tested for antioxidant activities. Samples were prepared using 95% ethanol using Nitric Oxide (NO) assay for assessing the anti-inflammatory activity. NO assay experiment showed that extracts of the Manglietia insignis (Wall.) Blume and Tirpitzia sinensis (Hemsl.) Hallier f. might have the 36.2% more anti-inflammatory activity and 59.5% more anti-inflammatory activity, respectively, compared to control. To determine the cell toxicity, MTT (3-(4,5-dimethylthiazol-2-yl) -2,5-diphenyltetrazolium bromide) assay was used. MTT assay experiment showed that the Manglietia insignis (Wall.) Blume and Tirpitzia sinensis (Hemsl.) Hallier f. might have the 31.0% less toxicity and 8.52% more toxicity, respectively, compared control. Taken together, these experiments showed that Manglietia insignis (Wall.) Blume extracts might have significantly higher anti-inflammatory activities and relatively lower toxicity, compared to control.

Cytotoxicity of Nigella Sativa Seed Oil and Extract Against Human Lung Cancer Cell Line

  • Nigella sativa (N sativa), commonly known as black seed, has been used in traditional medicine to treat many diseases. The antioxidant, anti-inflammatory, and antibacterial activities of N sativa extracts are well known. Therefore, the present study was designed to investigate the anticancer activity of seed extract (NSE) and seed oil (NSO) of N sativa against a human lung cancer cell line. Cells were exposed to 0.01 to 1 mg/ml of NSE and NSO for 24 h, then percent cell viability was assessed by 3-(4, 5-dimethylthiazol-2yl)-2, 5-biphenyl tetrazolium bromide (MTT) and neutral red uptake (NRU) assays, and cellular morphology by phase contrast inverted microscopy. The results showed NSE and NSO significantly reduce the cell viability and alter the cellular morphology of A-549 cells in a concentration dependent manner. The percent cell viability was recorded as 75%, 50%, and 26% at 0.25, 0.5, and 1 mg/ml of NSE by MTT assay and 73%, 48%, and 23% at 0.25, 0.5, and 1 mg/ml of NSE by NRU assay. Exposure to NSO concentrations of 0.1 mg/ml and above for 24 h was also found to be cytotoxic. The decrease in cell viability at 0.1, 0.25, 0.5, and 1 mg/ml of NSO was recorded to be 89%, 52%, 41%, and 13% by MTT assay and 85%, 52%, 38%, and 11% by NRU assay, respectively. A-549 cells exposed to 0.25, 0.5 and 1 mg/ml of NSE and NSO lost their typical morphology and appeared smaller in size. The data revealed that the treatment of seed extract (NSE) and seed oil (NSO) of Nigella sativa significantly reduce viability of human lung cancer cells.
