Biofilm Formation, Antimicrobial Peptide Resistance, and Hydrogen Peroxide Resistance in Livestock-Associated Staphylococcus aureus Isolates

  • Human infections with livestock-associated methicillin-resistant/-susceptible Staphylococcus aureus (LA-MRSA/LA-MSSA) have recently been increasing significantly. These LA-MRSA and LA-MSSA strains can be transmitted to individuals who have frequent contact with livestock animals and foods of animal origin. In this study, major virulence potentials of S. aureus such as biofilm formation, antimicrobial peptide resistance, and in vitro hydrogen peroxide (H2O2) resistance were assessed using 20 MRSA and MSSA strains isolated from raw milk, beef cattle, and workers in the livestock industry. Static biofilm formation assays revealed that there is no difference in levels of biofilm production between MRSA versus MSSA or bovine- versus human-associated strains. In vitro BMAP (bovine myeloid antimicrobial peptide)-28 susceptibility assays also revealed no difference in the resistance to the antimicrobial peptide between MRSA versus MSSA or bovine- versus human-associated S. aureus strains. However, LA-MRSA strains displayed increased resistance to H2O2, which may play an important role in survival and dissemination of the pathogen in livestock. These results provide an important basis for understanding pathogenic potentials of LA-MRSA and LA-MSSA strains in human and animal hosts.

Identification of Fuzzy Inference Systems Using a Multi-objective Space Search Algorithm and Information Granulation

  • We propose a multi-objective space search algorithm (MSSA) and introduce the identification of fuzzy inference systems based on the MSSA and information granulation (IG). The MSSA is a multi-objective optimization algorithm whose search method is associated with the analysis of the solution space. The multi-objective mechanism of MSSA is realized using a non-dominated sorting-based multi-objective strategy. In the identification of the fuzzy inference system, the MSSA is exploited to carry out parametric optimization of the fuzzy model and to achieve its structural optimization. The granulation of information is attained using the C-Means clustering algorithm. The overall optimization of fuzzy inference systems comes in the form of two identification mechanisms: structure identification (such as the number of input variables to be used, a specific subset of input variables, the number of membership functions, and the polynomial type) and parameter identification (viz. the apexes of membership function). The structure identification is developed by the MSSA and C-Means, whereas the parameter identification is realized via the MSSA and least squares method. The evaluation of the performance of the proposed model was conducted using three representative numerical examples such as gas furnace, NOx emission process data, and Mackey-Glass time series. The proposed model was also compared with the quality of some "conventional" fuzzy models encountered in the literature.

Occurrence and Characteristics of Methicillin-Resistant and -Susceptible Staphylococcus aureus Isolated from the Beef Production Chain in Korea

  • The emergence and persistence of methicillin-susceptible Staphylococcus aureus (MSSA) and methicillin-resistant S. aureus (MRSA) in livestock animals have been reported as a potential risk factor for transmission to humans. In this study, we investigated the nationwide prevalence and characteristics of MRSA and MSSA in the Korean beef production system, including retail markets, slaughterhouses, and cattle farms. From a total of 1,285 samples, only 5 MRSA strains were isolated: from a farmer (1 ST72 MRSA), a carcass sample from a slaughterhouse (1 ST72 MRSA), and beef cattle (3 ST5 MRSA). In addition, 11 MSSA strains were isolated from beef cattle (n=3), humans (1 farmer, 1 slaughterhouse worker, and 4 retail market workers), and carcass samples (n=1) and slaughterhouse environment (n=1). Although the prevalence of MRSA and MSSA in beef cattle was much lower than that reported in pigs, 5/5 MRSA and 2/11 MSSA strains displayed multiple drug resistance (MDR) phenotypes. Unlike the swine-associated MRSA, no correlation was found between tetracycline/zinc resistance and MDR phenotype. However, MRSA strains had an identical set of staphylococcal enterotoxins and exhibited enhanced levels of resistance to antimicrobial peptides (PMAP-36 and LL-37) compared to the MSSA strains. In conclusion, continued and systemic surveillance of livestock, meat products, and humans in close contact with livestock/meat products is necessary to prevent the transmission of MRSA and MSSA to humans.

Photodynamic Therapy for Methicillin-resistant Staphylococcus aureus Strain using Photogem and 630 nm LED

  • Methicillin-resistant Staphylococcus aureus (MRSA) is one of the most common nosocomial pathogens. It is associated with hospitals is now being isolated in the community. The aim of this study was to evaluate the antibacterial effect of photodynamic therapy using Photogem and 630 nm LED on MRSA and methicillin-sensitive Staphylococcus aureus (MSSA). The broth cultured MRSA and MSSA incubated with various concentrations of Photogem (500,50,5 and $0.5{\mu}g/mL$) for 4 h. Then 630 nm LED was given at $9\;J/cm^2$, $20{\mu}l$ of the exposed bacteria solution was inoculated onto agar plate. Plates were incubated for 24 hand colonies were counted. The PDT group was effective in killing MRSA and MSSA at the Photogem dose of $50{\mu}g/mL$. But MSSA is more sensitive than MRSA in photodynamic effect. Other groups (light only, sensitizer only, or no treatment) observed no bacterial cell killing. These results raise the possibility of using PDT with or without antimicrobial drugs to eradicate MRSA and MSSA. In order to confirm this result, we need to further study bacterial death mechanism and in vivo study.

  • 본 연구에서는 여러 기상지수들과 우리나라 기온, 강수량에 대해서 Multi-Channel Singular Spectrum Analysis(MSSA)를 실시함으로써 상호영향에 따른 주성분을 분석하였다. Window length가 150일 때 SOI 등의 기상지수와 기온, 강수량의 MSSA를 실시하였으며 이 때 각각의 eigenvalue는 전체 공분산에 대한 각 요소의 비율을 설명한다. Window length는 Vautard 등(1992)이 제시한 $N/5{\sim}N/3$의 값을 사용하였다. 기상요소들과 기온, 강수량의 MSSA를 이용한 기후변화에 따른 국내 수문변수의 변화 상관분석은 기온과 각 기상요소들과의 분석결과에 비해 강수와 각 기상요소들의 분석결과가 기상요소들에 대한 주기패턴을 잘 따르지 못하고 약한 진폭을 나타내며 특히 SOI와 Rainfall의 경우 첫 번째 주성분에서의 상관분석결과 3개월 지체 시 상관계수 0.8410의 상관성이 높은 장주기 변화 쌍을 가짐에도 불구하고 자료의 변화도에 대하여 각 요소가 설명하는 비중이 매우 낮았다.

Changes in Coagulase Serotype of Staphylococcus aureus Isolates in Busan, 1994-2005 (부산지역에서 분리된 Staphylococcus aureus의 Coagulase 혈청형 변화)

  • We analyzed the phenotypic changes in coagulase serotype of S. aureus isolated from clinical sources and nasal cavities of healthy persons, $1994{\sim}2005$. A total of 715 isolates, 408 methicillin resistant S. aureus (MRSA) from clinical sources and 307 methicillin-susceptible S. aureus (MSSA), were classified into eight coagulase sero-types, I to VIII. The most prevalent serotype in MRSA was type II (54.3%, 222/408) and followed IV (24.7%), III (10.9%), and V (5.2%), whereas the majorities in MSSA were type VII (30.9%, 95/307), IV (22.2%), V (22.2%) and II (7.1%). Among the isolates collected periods, significant changes of coagulase serotypes in both strains were observed. In MRSA strains, the serotype V was not detected until 1997, but rapidly increased to 18.5% (20/108) in 2005, and the serotypes III decreased from 27% (31/115) in 1994 to 0.9% (1/108) in 2005. A similar trend in MSSA strains was observed but serotype II strain was not detected in 2005. The antigenic shift and changes in the coagulase of S. aureus were confirmed.

Staphylococcal Scalded Skin Syndrome in Children : Comparison of the Clinical Features of That Isolated Methicillin-resistant and Methicillin-sensitive Staphylococcus aureus (포도상 구균성 열상 피부 증후군의 임상적 고찰 : Methicillin 내성과 감수성 Staphylococcus aureus 예의 비교)

  • Purpose : The purpose of this study was to compare the clinical features of staphylococcal scalded skin syndrome(SSSS) isolated methicillin-resistant Staphylococcal aureus(MRSA) with those of SSSS isolated methicillin-sensitive Staphylococcal aureus(MSSA) in children. Methods : We retrospectively reviewed the medical records and microbiological results of 36 cases of SSSS who were admitted to the Pediatric Department of Ulsan Dong-Kang General Hospital from Jan. 1999 to Dec. 2003. Results : Among 36 cases, there were 7 generalized types, 1 abortive type in MRSA group and 5 generalized types, 6 abortive types in MSSA group. The peak incidence of age was between 2 and 5 years in both groups. SSSS were diagnosed with increasing frequency. Most of MRSA group were diagnosed in 2003(6/8). The peak seasonal incidence was fall and winter in both groups. Facial lesion was observed in all cases followed by flexural lesion in both groups. Staphylococci were isolated from the skin and throat most frequently in both groups. Upper respiratory illness and conjunctivitis were most common preceding infections in both groups. Among 8 cases of MRSA group, 4 cases were treated completely with ampicillin-sulbactam. There was no significant difference of mean duration of admission between MRSA group and MSSA group. Conclusion : There was no significant difference of clinical features between MRSA group and MSSA group although generalized types of SSSS were more common in MRSA group than in MSSA group.

  • Various kinds of water soluble N,N,N-trimethylchitosan ammonium chloride(TMC) with different molecular weights were synthesized to examine the antimicrobial activity against Methicillin Sensitive Staphylococcus aureus(MSSA) and Methicillin Resistant Staphylococcus aureus(MRSA), which causes serious hospital infection, and to apply them to antimicrobial finishing agents for textiles. Chitosan samples were highly deacetylated with sodium hydroxide solution and degraded with hydrogen peroxide to control the molecular weight. TMC has the antimicrobial activities against MRSA and MSSA. TMC showed an excellent antimicrobial activity below the molecular weight of 70,000, especially at 40,000. The minimum inhibitory concentration (MIC) of TMC with optimum molecular weight against MRSA and MSSA was 250ppm. Because MRSA did not resist TMC in the subculture test of bouillon medium, it was expected that the successive use of TMC against MRSA was possible.

  • 최근 이상기후와 같은 기후변화로 인한 기온, 강수 등의 변화는 안정적인 수자원 확보에 큰 영향을 미칠 것으로 판단되고 수자원을 필요로 하는 사회 모든 분야에 있어 큰 영향을 끼친다. 특히 농업, 공업, 도시의 용수 공급에 있어 변화는 더욱 심해질 것으로 판단되며 기후변화로 인한 기온, 강수 등의 변화의 정확한 분석이 필요로 한다. 따라서 본 연구에서는 동아시아 해수면 온도와 우리나라 강수량에 대한 MSSA (Multi-channel Singular Spectrum Analysis)를 실시함으로 두 시계열 사이에 공통적으로 나타나는 변화, 즉 특정 상관 주기 변동을 분석함으로 두 변수 사이에 변화 상관 분석을 실시하였다. 우리나라 강수량 자료로는 현재 기상청에서 운영 중인 지상 기상관측소 76개소 중 가용관측소 61개소 자료에 대하여 1973년 1월부터 2008년 12월까지의 자료를 수집하여 월 평균값을 사용하였고 동아시아 해수면 온도 자료로는 한반도 근해 해수면 온도 변화, 남중국해 해수면 온도 변화, 인도양 해수면 온도 변화, 적도 해수면 온도 변화 등을 선택하여 관측시점부터 2008년 12월까지 자료를 수집하여 사용하였다. 분석 자료에 대해 선형 회귀분석을 통한 선형추세 제거와 정규화한 자료를 사용하여 각각의 지수에 대해 MSSA 분석을 실시하였다. 이때 window length는 Vautard 등(1992)이 제시한 N/5$^{\sim}$N/3의 값인 108의 값을 사용하였고 이때 각각의 고유치는 전체 공분산에 대한 각 요소의 비율을 설명한다. 상관분석 결과는 각 지수와 강수자료 사이에 높은 상관성을 가지는 장단주기 변화가 존재함을 보여주었다. 그럼에도 불구하고 우리나라 월강수자료의 전체 변화는 계절변화를 제외하고도 장단 주기를 가지는 시간변화가 자료 전체 변화의 절반에 해당하며 장주기 변화가 나타내는 부분이 미미하다. 이는 계절 주기를 제외한 자료들 사이의 상관변화가 설명할 수 있는 부분이 미미 하며 여러 기상지수들과 국내 강수량사이의 MSSA 분석을 통하여 제시 할 수 있는 변화의 정량적 정도가 매우 제한됨을 보여준다. 그럼에도 불구하고 이러한 접근을 통하여 강수 변화의 불확실성을 줄여나가는 노력이 필요하다고 하겠다.

Antibiofilm Activity of a Curcuma zedoaria Rosc Rhizome Extract against Methicillin-Resistant and Susceptible Staphylococcus aureus

  • Methicillin-resistant Staphylococcus aureus (MRSA) and methicillin-susceptible S. aureus (MSSA) are major causes of hospital- and community-acquired infections. The treatment of biofilm-related infections caused by these bacteria is a global healthcare challenge. Therefore, the development of alternative therapeutics is required. An essential oil extracted from Curcuma zedoaria (CZ) Rosc, also known as white turmeric, has been reported to possess various antimicrobial activities. In the present study, we evaluated the antibiofilm activities of an ethanolic extract of the CZ rhizome against MRSA and MSSA. The results showed that the CZ extract with the highest sub-minimum inhibitory concentration (sub-MIC), 1/2 MIC (0.312 mg/ml), significantly inhibited biofilm production by up to 80-90% in both tested strains. Subsequently, we evaluated the ability of the CZ extract to prevent cell-surface attachment to a 96-well plate and extracellular DNA (eDNA) release from the biofilm. The CZ extract demonstrated an inhibitory effect on bacterial attachment and eDNA release from the biofilm biomass. The CZ extract may inhibit biofilm formation by preventing eDNA release and cell-surface attachment. Therefore, this CZ extract is a potential candidate for the development of alternative treatments for biofilm-associated MRSA and MSSA infections.
