Metabolic Changes on Occipital Cortex during Visual Stimulation with Functional MR Imaging and H MR Spectroscopy (기능적 자기공명영상법과 양성자 가지공명분광법을 이용한 시각자극에 의한 후두염 피질의 대사물질 변화)

  • Purpose : The purpose of this study was aimed to evaluate the BOLD(blood oxygen level dependent) contrast fMRI(functional MR imaging) in the occipital lobe and to compare with the metabolic changes based on H MRS (MR spectroscopy) and MRSI (MR spectroscopic imaging) before and after visual stimulation Materials and Methods : Healthy human volunteers (eight males and two females with 24-30 year age) participated in this study. All of the BOLD fMRI were acquired on a 1.5T MR with EPI during supervised visual stimulation in the occipital lobe. The red flicker with 8Hz was used for visual stimulation. After imaging acquisition, the MR images were transferred into unix workstation and processed with acquired from the same location based on the activation map. MRSI (magnetic resonance spectroscopic imaging) was also acquired to analyze the lactate changes before and after stimulation. Results : The activation maps were successfully produced by BOLD effect due to visual stimulation. NAA (N-acetyle aspartate)/Cr (creatine) ratio varied only from $1.79{\pm}0.28{\;}to{\;}1.88{\pm}0.20$ in activation area before and after stimulation. However, the signal intensity of lactate was elevated $9.48{\pm}4.38$ times higher than before activation. Lactate metabolite images were consistent with the activation maps. Conclusion : The BOLD contrast fMRI is enough sensitive to detect the activated area in human brain during the visual stimulation. Lactate metabolite map presents the evidence of lactate elevation on the same area of activation.

A Stochastic User Equilibrium Transit Assignment Algorithm for Multiple User Classes (다계층을 고려한 대중교통 확률적사용자균형 알고리즘 개발)

  • The object of this study is a development of a stochastic user equilibrium transit assignment algorithm for multiple user classes considering stochastic characteristics and heterogeneous attributes of passengers. The existing transit assignment algorithms have limits to attain realistic results because they assume a characteristic of passengers to be equal. Although one group with transit information and the other group without it have different trip patterns, the past studies could not explain the differences. For overcoming the problems, we use following methods. First, we apply a stochastic transit assignment model to obtain the difference of the perceived travel cost between passengers and apply a multiple user class assignment model to obtain the heterogeneous qualify of groups to get realistic results. Second, we assume that person trips have influence on the travel cost function in the development of model. Third, we use a C-logit model for solving IIA(independence of irrelevant alternatives) problems. According to repetition assigned trips and equivalent path cost have difference by each group and each path. The result comes close to stochastic user equilibrium and converging speed is very fast. The algorithm of this study is expected to make good use of evaluation tools in the transit policies by applying heterogeneous attributes and OD data.

Isolation and Characterization of Exopolysaccharide Producing Lactic Acid Bacteria from Korean Soy Sauce and Soybean Paste (전통 장류로부터 Exopolysaccharide 생성 유산균의 분리 및 특성)

  • Three slime-forming lactic acid bacteria were isolated from traditional Korean fermented soy sauce and soybean paste and shown to produce exopolysaccharides (EPS) in sucrose media. By isolating the strains, examining their morphological characteristics and determining their 16S rDNA sequences, N58-5 and K6-7 were identified as Leuconostoc mesenteroides and N45- 10 as Leuconostoc citreum. The acid and bile tolerances of these three strains were investigated. Amongst the three lactic acid bacteria, Leuc. citreum N45-10 exhibited the highest viability ($10^5-10^6$ CFU/ml) in 0.05 M sodium phosphate buffer (pH 0.3) for 2 h, in artificial gastric juice for 2 h and in 0.3%, 0.5% oxgall for 24h. Leuc. mesenteroides K6-7, N58-5 and Leuc. citreum N45- 10 were grown in sucrose liquid medium and 8.16 g/L, 3.65 g/L, 16.17 g/L of EPS was collected, respectively. The hydrolyzed EPS was analyzed by HPLC in order to determine the sugar composition of EPS. Leuc. mesenteroides K6-7 and N58-5 showed two peaks indicating glucose and fructose, thus they were determined to be hetero-type polysaccharides. Leuc. citreum N45-10 showed only the glucose polymer, indicating it to be a homo-type polysaccharide. In addition, all three lactic acid bacterial hemolysis did not demonstrate a clear zone in blood agar in the area surrounding a lactic acid bacteria colony.

Purification and Enzymatic Characteristics of the Bacillus pasteurii Urease Expressed in Escherichia coli (Escherichia coli에서 발현된 Recombinant Bacillus pasteurii Urease의 정제 및 효소학적 특성)

  • The gene coding for urease of alkalophilic Bacillus pasteurii had been cloned in Escherichia coli previously. The urease protein was purified 63.1-fold by TEAE-cellulose, DEAE-Sephadex A-50, Sephadex G-150 and Sephadex G-200 chromatographies with a 7.3% yield from the sonicated fluid of the E. coli HB1Ol(pBUll) encoding B. pasteurii urease gene. The ureases of E. coli (pBUll) and B. pasteurii possessed as a $K_m$ for urea, 42.1 mM and 40.4 mM, respectively. They hydrolyzed urea with $V_{max}$ of 86.9$\mu$mol/min and 160$\mu$mol/min, respectively. Both ureases were composed with four subunits (Mrs 67,000) and a subunit (Mr 20,000). The molecular weight of both native enzymes was Mr 280,OOO$pm$10,000 determined by gel filtration chromatography and Coomassie blue staining of the subunits. The optimal reaction pH of both ureases were pH 7.5. The ureases were stabled in pH 5.5-10.5. The optimal reaction temperature of both ureases were $60^{\circ}C$, and the ureases were stable for an hour at $50^{\circ}C$, 40min at $60^{\circ}C$ and 10 min at $70^{\circ}C$ The activity of both enzymes were inhibited completely by $Ag^{2+}$, $Hg^{2+}$, $Zn^{2+}$, $Cu^{2+}$, and were inhibited 60% by CoH, 30% by $Fe^{2+}$ and 10% by $Pb^{2+}$. However it was increased by the addition of $Sn^{2+}$, $Mn^{2+}$, $Mg^{2+}$ at concentration of $1{\times}10^{-3}$M. Both ureases were inhibited completely by p-CMB and acetohydroxamic acid. The urease expressed in E. coli (pBU11) was inhibited 70% by SDS. The urease of B. pasteurii was inhibited 40% by hydroxyurea, whereas the recombinant urease of E. coli strain was inhibited 17%. Both enzymes were not inhibited by cyclohexanediaminetetraacetic acid (CDTA) and ethylendiaminetetraacetic acid (EDTA).

Edible Culture Media from Cereals and Soybeans for Pre-cultivation of Lactic Acid Bacteria (곡류 및 두류를 이용한 젖산균 전배양용 식용 배지의 제조)

  • This study was conducted to develop an edible culture media with various types of cereals and soybeans for the pre-cultivation of lactic acid bacteria (LAB). To manufacture the edible culture media, LAB enrichment media were prepared using cereals such as brown rice (including germinated brown rice, glutinous brown rice, and germinated glutinous brown rice), yellow soybeans (including yellow soybeans, hulled yellow soybeans, germinated yellow soybeans, hulled and germinated yellow soybeans), and black soybeans (black soybeans, hulled black soybeans, germinated black soybeans, hulled and germinated black soybeans). Seven species of LAB were used in the experiment: Lactobacillus (Lb.) farciminis, Lb. homohiochii, Lb. pentosus, Lb. plantarum, Leuconostoc (Leu.) paramesenteroides, Leu. citreum, and Leu. lactis. For edible culture media from cereals, the average viable cell count of the seven starter cultures was 7.6~8.0 log CFU/mL, while that of the MRS culture medium, a synthetic medium, was 9.2 log CFU/mL; thus proliferation was lower by about 1~2 log CFU/mL in starter cultures from cereals compared to the synthetic medium. In the case of the edible culture media from soybeans, most bacteria showed higher proliferation in the hulled and germinated soybean media. In particular, Lb. plantarum showed the highest cell count at 10.08 log CFU/mL. In the case of edible culture media from black soybeans, the proliferation rate was higher in the hulled and germinated black soybean medium. Lb. homohiochii showed the highest proliferation in the hulled and germinated black soybean medium at 9.90 log CFU/mL. All results show that edible culture media using cereals and soybeans are generally good for LAB. Especially, hulled and germinated black soybeans are optimal for the pre-cultivation of LAB medium.

Effect of Water Extract of Omija(Schizandra chinensis) on Growth of Yoghurt Starter (오미자(Schizandra Chinensis) 물 추출액이 Yoghurt Starter의 증식에 미치는 영향)

  • The effect of water extract of Schizandra chinensis on the growth of yoghurt starter was investigated in order to manufacture the drink type yoghurt added with water extract of Schizandra chinensis. It was the most desirable extraction conditions for Schizandra chinensis to soak in 50 times of water for 15 hours at 20$^{\circ}C$. The water extract of Schizandra chinensis showed pH 3.07, 2.39% acidity, 1.10% total sugars, and 0.15 optical density for color. The water extract of Schizandra chinensis was added to MRS broth medium from 0.1% to 1.0% and the medium was fermented by 4 types of lactic acid bacteria such as Lactobacillus acidophilus, Lactobacillus casei, Lactobacillus bulgaricus, and Streptococcus thermophilus. The addition of water extract of Schizandra chinensis inhibited the growth of the lactic acid bacteria. The maximum addition amounts of water extract of Schizandra chinensis was 0.9% for Lac. acidophilus, 0.8% for Lac. casei, 0.2% for Lac. bulgaricus and 0.1% for Str. thermophilus in order to maintain the propagation of the lactic acid bacteria. When the drink type yoghurts added with 0.4%, 0.6%, 0.8% and 1.0% water extract of Schizandra chinensis were kept at 4$^{\circ}C$ for 15 days, it was showed that the number of lactic acid bacteria was not significantly changed during the storage. The viable cell counts of the drink type yoghurts by addition of 0.4∼l.0% of water extract of Schizandra chinensis were 1.13${\times}$10$\^$9/∼2.29${\times}$10$\^$9/ CFU/mL, and these bacterial counts were still more than the legal standard(1.0${\times}$10$\^$8/ CFU/mL) even at 15 days of storage.

Isolation and Characterization of Lactobacillus Strains Isolated from Korean Feces (한국인에서 분리된 Lactobacillus 속 젖산균의 동정 및 생리적 특성)

  • Lactobacilli have been generally recognized as an important lactic acid bacteria present in the normal human intestinal flora. Fifty two Lactobacillus isolates were recovered from the feces of healthy Koreans whose age ranged from thirteen days after birth to 37 years. Among the isolates above, 17 isolates were tentatively identified as strains of Lactobacillus acidophilus and 3 isolates as strains of Lactobacillus casei. For their characterization, these twenty isolates were subjected to the experiments for the resistance to the artificial gastric juice, pH2.5 and bile salt. Interestingly, 3 strains survived pH 2.5 after 3 hour incubation in the artificial gastric juice with more than 75% of survival rate. L. acidophilus a-4 had the highest survival rate of 100%. Four strains including L. acidophilus a-3 grew gradually in MRS broth in the presence of the artificial bile salt. Cholesterol assimilation was also tested for the 20 isolates. The result showed that cholesterol concentration of the medium was reduced by 10 Lactobacillus isolates with more than 60%, as compared to the control. L. acidophilus a-2 had the highest reduction rate of 77%. Judging from these data obtained in vitro, some isolates ware likely to reach the lower small intestine after consumption without a significant loss of viability, suggesting that they had the potential to be developed as a probiotic culture which might lower the cholesterol level in human.

Assessment of Applicability of a Calcium Carbonate-Alginate Beads as Neutralizer for the High Cell Density Cultivation of Isolated Sourdough Lactic Acid Bacteria (Sourdough에서 분리된 유산균의 고농도 배양을 위한 중화제로서 Calcium Carbonate-Alginate Bead의 이용가능성 평가)

  • Lab scale experiments were conducted in order to assess the applicability of $CaCO_{3}$-alginate beads as neutralizer for the high cell density cultivation and prepare the direct vat inoculation cultures of isolated sourdough lactic acid bacteria. With increasing the amount of bead and decreasing the diameter of bead in acidic solution, the neutralizing effect of $CaCO_{3}$-alginate bead became higher. In batch process with $CaCO_{3}$-alginate beads, Lactobacillus amylovorus DU-21 isolated from sourdough showed the highest viable cell counts and optical density in MRS broth. The values of viable cell counts and optical density were 9.996 log CFU/mL and 3.97, respectively. Experiments on the conditions which increase viability during lyophilization were carried out and the following results were obtained; 15% glycerol revealed the high cryoprotective effect on the concentrated cultures during lyophilization among the two cryoprotective agents. Consequently, $CaCO_{3}$-alginate beads and 15% glycerol were found to be useful not only to cultivate Lactobacillus amylovorus DU-21 but also to preserve strain.

The Newly Identified Goryeo Memorial Inscriptions (새롭게 확인된 고려(高麗) 묘지명(墓誌銘) : 「김용식(金龍軾) 묘지명」·「상당현군(上黨縣君) 곽씨(郭氏) 묘지명」·「민수(閔脩) 묘지명」)

  • In commemoration of the 1,100th anniversary of the foundation of the Goryeo Dynasty, the National Museum of Korea and the National Institute of Korean History are working together on a project to reveal memorial inscriptions(墓誌銘) in the Goryeo Dynasty. So far, It founded that four newly identified memorial inscriptions of Goryeo Dynasty. Among them, I would like to introduce two memorial inscriptions in overseas and a memory inscription that was first identified as being housed in the National Museum of Korea. Kim Yong Sik's memorial inscription is currently housed at the Smithsonian Museum's FreerSackler Gallery in the U.S.A.. Kim Yong Sik(金龍軾, 1129~1197) is a bureaucrat in the middle time of Goryeo Dynasty. He came from a family of influential people in the Andong province. In Goryeo times, provincial figures have been able to make inroads into central politics through the bureaucratic select examination(科擧). Kim's family came from the capital of Goryeo in that way. However, Kim did not rise very high. This inscription is meaningful in that it shows this ordinary middle class's life to study Goryeo history further. Sangdanghyeongoon(上黨縣君) Gwak Ssi's memorial inscription is currently housed at the Kyoto university museum in the Japan. Sangdanghyeongoon Gwak Ssi(郭氏(Mrs. Gwak), ?~1149?) is a bureaucrat class woman in the middle time of Goryeo Dynasty. There is not much information about her. But the method of marking the location of the her tomb is unique. Her tomb is located at the northern foot of the temple, Baekhaksa(白鶴寺, White Crane's temple). That marking method is sometimes confirmed in Goryeo period's historical text. This inscription is significant in that it shows practical example of that methods. Min Su's memorial inscription is missing after Japanese occupation time, but confirmed that currently housed in the National Museum of Korea. Min Su(閔脩, 1067~1122) is a bureaucrat in the middle time of Goryeo Dynasty. Although his track record is partly recorded in Goryeosa(高麗史, Historia of Goryeo Dynasty), the discovery of this inscription has made new research possible. I hope that more and more memory inscription of Goryeo will emerge from somewhere and contribute greatly to the study of Goryeo history.

Comparison of Bioconversion Ability and Biological Activities of Single and Multi-Strain Probiotics for an Active Molecule in Roasted Tartary Buckwheat (단일 및 복합 프로바이오틱스 균주에 의한 쓴메밀 내 Rutin의 Quercetin으로의 생물전환 및 이의 생리활성 비교)

  • In this study, we aimed to evaluate the bioconversion ability of single (Lactiplantibacillus plantarum CBT LP3, Lactococcus lactis subsp. lactis CBT SL6, Streptococcus thermophilus CBT ST3) and multi-strain probiotics to convert rutin to quercetin in roasted tartary buckwheat, and to assess their biological activities. To evaluate the bioconversion efficiency, each strain was cultured for 24 h in MRS media with 5% roasted tartary buckwheat 'Hwangguem-Miso' powder. After then, rutin and quercetin contents were determined by HPLC. Additionally, the biological activities were compared before and after bioconversion of an ingredient. Anti-oxidant effects were measured by DPPH and ABTS assays. Anti-inflammatory effects were determined by measuring NO production, and levels of iNOS, TNF-α, IL-6 and IL-4 using an LPS-induced Raw 264.7 cell model. The bioconversion rate under the combination of three species of probiotics significantly increased more than single species. Antioxidant efficacy results showed the highest activity when the combination of three species of probiotics cultured. The pro-inflammatory factors such as nitric oxide, iNOS, TNF-a, and IL-6 were significantly decreased when the three types of probiotics were combined than single strain was cultured. In addition, level in the anti-inflammatory factor IL-4 was increased. The multi-strain probiotics showed increased bioconversion efficiency, effects of anti-oxidant and anti-inflammatory compared to the single strain. These findings suggest that the fermentation of tartary buckwheat by probiotics may be a valuable candidate for developing functional foods with anti-oxidation and anti-inflammation.
