Protective effect of Korean Red Ginseng extract against Helicobacter pylori-induced gastric inflammation in Mongolian gerbils

  • Helicobacter pylori-induced gastric inflammation includes induction of inflammatory mediators interleukin (IL)-8 and inducible nitric oxide synthase (iNOS), which are mediated by oxidant-sensitive transcription factor NF-${\kappa}B$. High levels of lipid peroxide (LPO) and increased activity of myeloperoxidase (MPO), a biomarker of neutrophil infiltration, are observed in H. pylori-infected gastric mucosa. Panax ginseng Meyer, a Korean herb medicine, is widely used in Asian countries for its biological activities including anti-inflammatory efficacy. The present study aims to investigate whether Korean Red Ginseng extract (RGE) inhibits H. pylori-induced gastric inflammation in Mongolian gerbils. One wk after intragastric inoculation with H. pylori, Mongolian gerbils were fed with either the control diet or the diet containing RGE (200 mg RGE/gerbil) for 6 wk. The following were determined in gastric mucosa: the number of viable H. pylori in stomach; MPO activity; LPO level; mRNA and protein levels of keratinocyte chemoattractant factor (KC, a rodent IL-8 homolog), IL-$1{\beta}$, and iNOS; protein level of phospho-$I{\kappa}B{\alpha}$(which reflects the activation of NF-${\kappa}B$); and histology. As a result, RGE suppressed H. pylori-induced mRNA and protein levels of KC, IL-$1{\beta}$, and iNOS in gastric mucosa. RGE also inhibited H. pylori-induced phosphorylation of $I{\kappa}B{\alpha}$ and increases in LPO level and MPO activity of gastric mucosa. RGE did not affect viable H. pylori colonization in the stomach, but improved the histological grade of infiltration of poly-morphonuclear neutrophils, intestinal metaplasia, and hyperplasia. In conclusion, RGE inhibits H. pyloriinduced gastric inflammation by suppressing induction of inflammatory mediators (KC, IL-$1{\beta}$, iNOS), MPO activity, and LPO level in H. pylori-infected gastric mucosa.

The effects of Naetakchunkeum-san on the Allergic Contact Dermatitis induced by DNCB (내탁천금산(內托千金散)이 DNCB로 유발된 알레르기성 접촉피부염에 미치는 영향)

  • Background and Objectives : Allergic Contact Dermatitis is the disease affected by industrialization. The more industrialization advanced, the more materials that could induce the allergic contact dermatitis have been increased. Therefore in oriental medicine, various studies have been performed. The objective of this study is to investigate the effects of Naetakchunkeum-san on the Allergic Contact Dermatitis induced by 2,4-dinitro-chlorobezene(DNCB). Meterial and Methods : Twenty eight mice were divided into four groups ; normal, control, experimental group A and B. Control and experimental group were induced allergic contact dermatitis by DNCB. Experimental group A was orally administered the Naetakchunkeum-san and experimental group B was orally administered the prednisolone. In this study, ear thickness measurement, observation auricle microphotograph, Myeloperoxidase(MPO) activity measurement, Reverse transcription-polymerase chain reaction(RT-PCR) analysis of the mRNA level of $TNF-\alpha$, $IL-1\beta$, $INF-\gamma$ were performed on these four groups. In addition, the effect of Naetakchunkeum-san on cell viability and the effect of Naetakchunkeum-san on the compound 48/80-induced histamine release from HMC and RPMC were measured, Results : 1. In contact hypersensitivity assay, experimental group A and B showed decreased ear thickness compared with control group, 2. In experimental group A, pathological lesion of dermatitis were alleviated. In addition, the numbers of infiltrated cells were reduced, and cleft was not shown compared with control group, In experimental group B, similar results were shown. 3. There was a significant increase in MPO activity in control group compared with normal group, Experimental group A and B significantly inhibited the increase in MPO activity compared with control group. 4, The level of expression of $TNF-\alpha$, $IL-1\beta$, $INF-\gamma$ in experimental group A and B were significantly lower than those in control group. As the internal control, cyclophilin mRNA was also reverse-transcribed and amplified. 5, In MTT assay, there were no statistically significant differences in 100 ${\mu}g/ml$, 200 ${\mu}g/ml$, 500 ${\mu}g/ml$, 1000 ${\mu}g/ml$ Naetakchunkeum-san treated group from 0 ${\mu}g/ml$ Naetakchunkeum-san treated group as determined by the Tukey test. 6. Naetakchunkeum-san dose-dependently inhibited the compound 48/80-induced histamine release from both HMC and RPMC. Conclusions : According to above experiments, Naetakchunkeum-san may be applied to allergic contact dermatitis.

  • Background : Neutrophils are considered to play critical roles in the development of acute respiratory distress syndrome. Histamine, which is distributed abundantly in lung tissue, increases the rolling of neutrophills via increase of P-selectin expression on the surface of endothelial cells and is known to have some interrelationships with IL-1, IL-8 and TNF-$\alpha$. We studied to investigate the effect of the histamine on the acute lung injury of the rats induced by intratracheal insufflation of TNF-$\alpha$ which has less potency to cause lung injury compared to IL-1 in rats. Methods : We intratracheally instilled saline or TNF(R&D, 500ng), IL-1(R&D, 50ng)or histamine of varius dose(1.1, 11 and $55\;{\mu}g/kg$) with and without TNF separately in Sprague-Dawley rats weighing 270-370 grams. We also intratracheally treated IL-1(50ng) along with histamine($55\;{\mu}g/kg$). In cases, there were synergistic effects induced by histamine on the parameters of TNF-induced acute lung injury, antihistamines(Sigma, mepyramine as a $H_1$ receptor blockade and ranitidine as a $H_2$ receptor blockade, 10 mg/kg in each)were co-administered intravenously to the rats treated TNF along with histamine($1.1\;{\mu}g/kg$) intratraeheally. Then after 5 h we measured lung lavage neutrophil numbers, lavage cytokine-induced neutrophil chemoattractants(CINC), lung myeloperoxidase activity(MPO) and lung leak. We also intratracheally insufflated TNF with/without histamine($11\;{\mu}g/kg$), then after 24 h measured lung leak in rats. Statistical analyses were done by Kruskal-Wallis nonparametric ANOVA test with Dunn's multiple comparison test or by Mann-Whitney U test. Results : We found that rats given TNF, histamine alone(11 and $55\;{\mu}g/kg$), and TNF with histamine(l.1, 11, and $55\;{\mu}g/kg$) intratracheally had increased (p<0.05) lung MPO activity compared with saline-treated control rats. TNF with histamine $11\;{\mu}g/kg$ had increased MPO activity (P=0.0251) compared with TNF-treated rats. TNF and TNF with histamine(1.1, 11, and $55\;{\mu}g/kg$) intratracheally had all increased (p<0.05) lung leak, lavage neutophil numbers and lavage CINC activities compared with saline. TNF with histamine $1.1\;{\mu}g/kg$ had increased (p=0.0367) lavage neutrophil numbers compared with TNF treated rats. But there were no additive effect of histamine with TNF compared with TNF alone in acute lung leak on 5 h and 24 h in rats. Treatment of rats with the $H_1$ and $H_2$ antagonists resulted in inhibitions of lavage neutrophil accumulations and lavage CINC activity elevations elicited by co-treated histamine in TNF-induced acute lung injury intratracheally in rats. We also found that rats given IL-1 along with histamine intratracheally did not have increase in lung leak compared with IL-1 treated rats. Conclusion : Histamine administered intratracheally did not have synergistic effects on TNF-induced acute lung leak inspite of additive effects on increase in MPO activity and lavage neutrophil numbers in rats. These observations suggest that instilling histamine intratracheally would not play synergistic roles in neutrophil-mediated acute lung injury in rats.

Effect of Pioglitazone on Excitotoxic Neuronal Damage in the Mouse Hippocampus

  • Pioglitazone (PGZ), a synthetic peroxisome proliferator-activated receptor ${\gamma}$ agonist, is known to regulate inflammatory process and to have neuroprotective effects against neurological disorders. In the present study, we examined the effects of 30 mg/kg PGZ on excitotoxic neuronal damage and glial activation in the mouse hippocampus following intracerebroventricular injection of kainic acid (KA). PGZ treatment significantly reduced seizure-like behavior. PGZ had the neuroprotective effect against KA-induced neuronal damage and attenuated the activations of astrocytes and microglia in the hippocampal CA3 region. In addition, MPO and $NF{\kappa}B$ immunoreactivities in the glial cells were also decreased in the PGZ-treated group. These results indicate that PGZ had anticonvulsant and neuroprotective effects against KA-induced excitotocix injury, and that neuroprotective effect of PGZ might be due to the attenuation of KA-induced activation in astrocytes and microglia as well as KA-induced increases in MPO and $NF{\kappa}B$.

The Effect of Tokoro Rhizoma Pharmacopuncture at $KI_{10}$ in Lipopolysaccharide Induced Acute Nephritis in Rats (음곡에 시술한 비해약침이 Lipopolysaccharide로 유도된 흰쥐의 신장염에 미치는 영향)

  • Objectives : This study was designed to evaluate the effects of Tokoro Rhizoma pharmat $KI_{10}$ in nephritis induced by lipopolysaccharide(LPS) in rat. Methods : Rats were divided into 4 groups and 3 groups(LPS, saline, Tokoro Rhizoma pharmacopuncture(TR-P) group) were injected LPS to induce nephritis. TR-P group was treated with TR at $KI_{10}$ three times for a week, saline group with normal saline. To evaluate the effects of TR at $KI_{10}$ on nephritis in rats, white blood cell(WBC), neutrophil in blood, creatinine, cytokine-induced neutrophil chemoattractant-1(CINC-1) in serum and urinary volume, creatinine in urine, renal myeloperoxidase(MPO) were measured and renal tissue was analyzed. Results : TR-P group significantly reduced WBC, neutrophil in blood, creatinine, CINC-1 in serum, creatinine in urine and renal MPO than LPS group. TR-P group increased urinary volume but, not significant. Conclusion : TR at $KI_{10}$ has a therapeutic effect on nephritis in LPS stimulated rat. Therefore, it is suggested that TR at $KI_{10}$ may be an useful therapeutics for nephritis in clinical field after further researches.

Anti-Inflammatory Effect of Polygoni Avicularis Herba Herbal-Acupuncture at KI10 on LPS-Induced Nephritis in Rats (음곡(KI10) 편축 약침의 LPS 유도 급성신장염 억제 효과)

  • Objectives : This study aimed to evaluate the effects of Polygoni avicularis Herba Herbal-acupuncture( PaH-HA) at KI10(Umgok) on nephritis induced by lipopolysaccharide(LPS) in rats. Methods : Rats were allocated into normal, control, and 3 experimental groups. The rats in the control group were intra-peritoneally injected with LPS for nephritis induction. The rats in the groups of experiment 1, experiment 2, and experiment 3 were treated with single needle prick, saline injection, and PaH-HA, respectively at KI10 three times for a week and then injected with LPS. To evaluate the effects of PaH-HA at HI10, WBC count in blood, serum CINC-1, renal TNF-${\alpha}$ and renal MPO were measured. Results : Needle prick at KI10 suppressed the increase of WBC in blood and CINC-1 in serum of LPS-stimulated rats. Saline injection at KI10 suppressed the increase of WBC in blood. PaH-HA at KI10 suppressed the increase of WBC in blood, CINC-1 in serum, and MPO in kidney of LPS-stimulated rats. Conclusions : PaH-HA at KI10 has an anti-inflammatory effect on LPS-induced nephritis in rats and there may be a synergism between KI10(Umgok) stimulation and PaH-HA Solution.

The Effect of Junci Medulla Herbal-acupuncture at $KI_{10}$ on LPS induced Nephritis in Rats (음곡에 시술한 등심초약침이 LPS로 유도된 흰쥐의 신장염에 미치는 영향)

  • Objectives : This study aimed to evaluate the effects of Junci Medulla Herbal-acupuncture(JM-HA) at $KI_{10}$(Umgok) on nephritis induced by lipopolysaccharide(LPS) in rats. Methods : Rats with nephritis induced by LPS, were treated with JM-HA at $KI_{10}$ 3 times a week. The rats in the NP group and the saline group were treated with a needle prick and a saline injection respectively. To evaluate the effects of JM-HA at $KI_{10}$ on nephritis in rats, WBC, neutrophils in blood, BUN, creatinine, TNF-${\alpha}$ in serum, creatinine, total protein in urine and renal MPO were measured. Results : JM-HA at $KI_{10}$ significantly inhibited the increase of WBC and neutrophils in blood, BUN, creatinine, TNF-${\alpha}$ in serum, and MPO in kidney of LPS-stimulated rats. Conclusion : JM-HA at $KI_{10}$ has therapeutic effects on nephritis in LPS-stimulated rats. Therefore, it is suggested that JM-HA at $KI_{10}$ may be a useful therapy in clinical field after further researches.

Results : WBC in blood, CINC-1 in serum, renal MPO significantly reduced in DH-HA group. Conclusions : DH-HA at $KI_{10}$ is effective for nephritis in LPS-induced rats. Therefore, DH-HA at $KI_{10}$ may be useful for nephritis in clinical field.

Anti-Inflammatory Effects of Glycyrrhiza glabra Linne Extract in a Dextran Sulfate Sodium-Induced Colitis Mouse Model (감초 추출물의 Dextran Sulfate Sodium 유도 마우스 궤양성 대장염 억제 효과)

  • The aim of this study was to evaluate the anti-inlfammatory effects of Glycyrrhiza glabra Linne extract on ulcerative colitis induced by 3% dextran sulfate sodium in mice. The experimental animals were divided into six groups: control(normal), DSS-induced colitis(control), 1 mg/kg, 10 mg/kg, and 100 mg/kg of Glycyrrhiza glabra Linne extract, and 150 mg/kg 5-aminosalicylic acid(5-ASA)(positive control). We evaluated the pathological disease activity index(DAI), change in weight, colon mucosa damage and myeloperoxidase(MPO) in colon mucosa. Treatment with 10 mg/kg and 100 mg/kg of Glycyrrhiza glabra Linne extract led to significant loss of body weight, the decrease of MPO activity and clinical symptoms such as DAI and histological change. In particular, 100 mg/kg Glycyrrhiza glabra Linne extract led to markedly greater improvement than 150 mg/kg 5-ASA treatment. These results suggest that Glycyrrhiza glabra mediated anti-inflammatory action on colorectal sites may be a useful therapeutic approach to ulcerative colitis.
