The effect of L-carnitine in the expression of matrix metalloproteinases by human dermal fibroblasts

  • L-camitine ($\beta$ -hydroxy-${\gamma}$ -trimethyl-ammoniumbutyric acid) is a small water-soluble molecule important in mammalian fat metabolism. It is essential for the normal oxidation of fatty acids by the mitochondria, and is involved in the trans-esterification and excretion of acyl-CoA esters. In this paper, to investigate the relationship between aging and L-camitine, we investigated the effects of in vitro MMP inhibition and activity and expression of UVA-induced MMP 1 in human skin fibroblasts. Fluorometric assays of the proteolytic activities of MMP-l were performed using fluorescent collagen substrates. ELISA (enzyme linked immuno sorbent assay), gelatin-substrate zymography, and RT-PCR ELISA techniques were used for the effects of L-camitine on MMP expression and activity, MMP mRNA expression in UVA irradiated fibroblast. L-camitine inhibited the activities of MMP-l in a dose-dependent manner and the $IC_{50}$/ values calculated from semi-log plots were 2.45mM, and L-carnitine showed strong inhibition on MMP-2 (gelatinase) activity in UVA irradiated fibroblast by zymography. Also, UVA induced MMP expression was reduced 40% by treated with L-carnitine, and MMP-l mRNA expression was reduced dose-dependent manner. Therefore L-carnitine was able to significantly inhibition the MMP activity, regulation of MMP expression in protein and mRNA level. All these results suggest that L-carnitine may be useful as new anti-aging cofactor for protection against UVA induced MMP expression and activity.

Also, we studied to develop as anti-aging cosmetics with L-carnitine. Fluorometric assays of the proteolytic activities of MMP-1 (collagenase) were performed using fluorescent collagen substrates. ELISA (enzyme linked immune sorbent assay), gelatin-substrate zymography, RT-PCR ELISA techniques were used for the effects of L-carnitine on MMP expression, activity, and MMP mRNA expression in UVA irradiated fibroblast $(5\;J/cm^2)$, respectively. In addition, we performed clinical study with L-carnitine cream. L-carnitine inhibited the activities of MMP-1 in a dose-dependent manner and the $IC_{50}$ values calculated from semi-log plots were 2.45 mM, and L-carnitine showed strong inhibition on MMP-2 (gelatinase) activity in UVA irradiated fibroblast by zymography. Also, UVA induced MMP-1, 2 expression was reduced $43\%,\;53\%$ by treated with L-carnitine at 1.25 mM, and MMP-1 mRNA expression was reduced dose-dependent manner. Therefore L-carnitine was able to significantly inhibit the MMP activity, and regulate MMP expression in protein and mRNA level. The results of clinical study showed that $1.0\%$ L-carnitine treated group reduced wrinkle significantly compared with placebo treated group (P<0.05). All these results suggest that L-carnitine may be useful as new anti-aging cosmetics for protection against UVA induced Mm expression and activity.

The Effects of Marigold(Tagetes L.) Extract and Calendula(Calendula officinalis L.) Extract on Collagen Growth and MMP-1 Expression in Human Dermal Fibroblasts (메리골드(Tagetes L.)와 카렌듈라(Calendula officinalis L.) 추출물이 인간 섬유아세포에서 콜라겐 생성 및 MMP-1 발현에 미치는 영향)

  • To research the effects of marigold extract, which is used mixed with calendula extract, on collagen growth and MMP-1 expression in human fibroblast, we measured cytotoxicity, collagen growth and MMP-1 expression by using HDF cells. The result of measurement showed over 80% cell survival rate in $5{\sim}100{\mu}g/mL$ concentration of marigold extract and calendula extract for HDF cells, which indicates there is no cytotoxicity. The result of measuring collagen synthetic abilities showed both types of extract had collagen synthetic ability increase dose dependently, by 25% in $100{\mu}g/mL$ concentration of marigold extract, and by 7% in $100{\mu}g/mL$ concentration of calendula extract. The result of experimenting the effect on MMP-1 expression showed that both types of extract suppress MMP-1 expression. The result of observing phosphorylation of p-JNK and p-ERK, which are known to be involved with MMP-1 expression, revealed that marigold extract effectively suppresses MMP-1 expression through signaling pathway of p-JNK and p-ERK. The above results confirm the wrinkle improvement effect of marigold extract, and furthermore, it can be used as a cosmetic ingredient for anti-aging.

Anti-Oxidative and Inhibitory Effect of Saussurea involucrata on MMP-1 in UVA-irradiated Human Dermal Fibroblast (설련의 항산화능와 사람 섬유아세포에서 UVA에 의한 MMP-1발현 저해효과)

  • In order to investigate the effects of Saussurea involucrata on (relationship between) aging (and Saussurea involucrata), we examined the activities of antioxidation, in vitro MMP inhibition and UVA-induced MMP-1 expression in human dermal fibroblasts. S. involucrata showed scavenging activities radicals and reactive oxygen species (ROS) with the $IC_{50}$ values of $3.89{\mu}g/mL$ against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and $67.29{\mu}g/mL$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. At the concentration of $1000{\mu}g/mL$, S. involucrata showed 93.27% inhibition on lipid peroxidation of linoleic acid. S. involucrata inhibited the activities of MMP-1 in a does-dependent manner and the $IC_{50}$ value calculated from semi-log plots was $97.18{\mu}g/mL$. Also, UVA induced MMP expression in human dermal fibroblasts was reduced 42.86% by treatment with S. involucrata, and MMP-1 mRNA expression was reduced in a dose-dependent manner. Therefore S. involucrata was able to significantly inhibit MMP expression in protein and mRNA level. All these results suggested that S. involucrata might act as an anti-aging agent by antioxidation and reducing UVA-induced MMP-1 production.

Effect of Antioxidation and Inhibition of Matrix Metalloproteinase-1 from Ligularia fischeri (곰취의 항산화와 UVA에 의한 MMP-1 발현 저해효과)

  • In this study, we examined the effects of antioxidant, lipid peroxidation inhibition and suppression of UVA-induced MMP-1 expression in human dermal fibroblasts. Ligularia fischeri extract showed free radical scavenging effect by 82.3% at 5 mg/mL and superoxide radical scavenging effect by 79.3% at 5 mg/mL in the kanthine/xanthine oxidase system, respectively. At the concentration of $500{\mu}g/mL$, L. fischeri extract showed 97% inhibition on lipid peroxidation of linoleic acid. UVA induced MMP expression in human dermal fibroblasts was reduced 35% by treatment with $100{\mu}g/mL$ of L. fischeri extract. These results suggest that L. fischeri extract can be used for an anti-aging agent by antioxidation, lipid peroxidation inhibition and reducing UVA-induced MMP-1 production.

A Study on the Anti-wrinkle Activities of Sesamum indicum L. Ethanol Extracts on CCD-986sk (CCD-986sk 세포 내 참깨 에탄올 추출물의 항주름 활성 연구)

  • In order to investigate the possibility of Sesamum indicum L. (S. indicum) extract as an active ingredient for wrinkle-care cosmetics, we prepared 70% ethanolic extract of S. indicum and measured its elastase inhibitory activity and collagenase inhibitory activity. We also evaluated the effect of S. indicum extract on protein and mRNA expression of MMPs in fibroblast cell (CCD-986sk). For anti-wrinkle effects, elastase inhibition activities and collagenase inhibition activities were 37.8% and 45% at a dose of $1,000{\mu}g/mL$ of S. indicum 70% ethanol extract. For a cell viability test, measured on fibroblast cell by ethanol extract of S. indicum, results showed 96% with cell viability at $100{\mu}g/mL$ concentration. According to the results of western blot of ethanol extract from S. indicum the expression of the matrix metalloproteinase-1 (MMP-1), matrix metalloproteinase-2 (MMP-2), matrix metalloproteinase-3 (MMP-3) protein was decreased by 63%, 43%, 49% at $100{\mu}g/mL$ concentration. Reverse transcription-polymerase chain reaction (PCR) of ethanol extract from S. indicum showed that the expression of MMP-1, MMP-2, MMP-3 mRNA was decreased by 82%, 79%, 82% at $100{\mu}g/mL$ concentration. The findings suggest that 70% ethanol extract from S. indicum has potential as a cosmeceutical ingredient with anti-wrinkle effects.

Excellent Anti-aging Effects of Ursolic acid and Oleanolic acid Present in Ligustrum lucidum (광나무의 주성분, Ursolic acid와 Oleanolic acid의 항노화 효능)

  • Ligustrum lucidum (L. lucidum) contains large quantities of ursolic acid and oleanolic acid. In this study, we evaluated anti-wrinkle effects of three parts of L. lucidum extracts. We found that L. lucidum extracts were not only innocuous to human skin fibroblasts but also significantly decreased the expression of both MMP-1 and MMP-2 and increased the expression of COL1A1. Among the three parts of L. lucidum extracts (i.e., fruit, cane, and leaf extracts), the fruit extract was found to contain the greatest amounts of ursolic acid and oleanolic acid. These three parts of L. lucidum extracts increased the expression of COL1A1 and decreased the expression of MMP-1 and MMP-2 in a dose-dependent manner. Especially, the fruit extract of L. lucidum had the greatest upregulating effect on COL1A1 and the greatest downregulating effect on MMP-1 and MMP-2 in a non-toxic and dose-dependent manner. These results suggest that L. lucidum, especially the fruit, could be used as active ingredients for functional cosmetics.

Effect of Phellinus Extracts on Sprouting in Porcine Pulmonary Artery Endothelial Cells (혈관내피세포의 발아에 미치는 상황버섯 추출물의 효과)

  • One of the steps in angiogenesis is the degradation of the underlying basement membrane via proteases. Endothelial cells release proteinases to degrade the extracellular matrix for their sprouting in vivo. In this study, we examined the effect of water extracts of Phellinus linteusis(Phellinus extracts) and combination of Phellinus extracts and fibroblast growth factor(FGF-2) on cultured porcine pulmonary artery endothelial cells(PPAECs). Phellinus extracts induced sprouting of PPAECs, which was inhibited by MMPs and plasmin inhibitors, and induced the secretion of matrix metalloproteinase-3(MMP-3) and plasmin. At high concentration of Phellinus extracts($200{\sim}400{\mu}g/mL$), the active MMP-2 secretion was induced. It is therefore, suggested that Phellinus extracts induces the sprouting of cultured endothelial cells by means of increased active MMP-2 and plasmin secretion. Also, combination with Phellinus extracts and FGF-2 produced an enhanced effect on sprouting and secretion of active MMP-2, and MMP-3 and plasmin from PPAECs.

Regulatory mechanism of Angelica Gigas extract powder on matrix metalloproteinases in vitro and in vivo model (참당귀 추출분말이 in vitro and in vivo model에서 MMPs 조절 기전)

  • The precise mechanism underlying the therapeutic efficacy of an extraction powder of Angelica gigas (AGE) for the treatment of degenerative osteoarthritis was investigated in primary cultured rabbit chondrocytes and in a monosodium-iodoacetate (MIA)-induced osteoarthritis rat model. The treatment with AGE (50 μg/mL) effectively inhibited NF-B activation. The anti-inflammatory mechanism was clarified by gelatin zymography and western blotting measurements of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) activities. The AGE (50 μg/mL) treatment significantly reduced MMP-9 activity. The constituents of AGE— decursinol, decursin, and decursinol angelate—were determined by LC-MS/MS after a 24 hr treatment of rabbit chondrocytes. The contents of the major products, decursin and decursinol angelate, were 3.62±0.47 and 2.14 ±0.36 μg/mg protein, respectively in AGE-treated (50 μg/mL) rabbit chondrocytes. An in vivo animal study on rats fed a diet containing 25, 50, and 100 mg/kg AGE for 3 weeks revealed a significant inhibition of the MMPs in the MIA-induced rat articular cartilage. The genetic expression of arthritic factors in the articular cartilage was examined by RT-PCR of collagen Type I, collagen Type II, aggrecan, and MMP (MMP3, MMP-9, MMP13). Specifically, AGE up-regulated the expression of collagen Type I, collagen Type II, and aggrecan and inhibited MMP levels at all tested concentrations. Collectively, AGE showed a strong specific site of action on MMP regulation and protected against the degeneration of articular cartilage via cellular regulation of MMP expression both in vitro and in vivo.

Inhibitory Efficacy of Smilax china L. on Pro-collagen Type-1 Activity and MMP-1 Gene Expression in Fibroblasts (CCD-986sk) (섬유아세포(CCD-986sk)에서 청미래덩굴의 pro-collagen type-1 활성 및 MMP-1의 유전자 발현 저해능)

  • The purpose of this study was to research the biological activity of ethanol extract from Smilax china L. which is a vine shrub belonging to the lily family. For antiwrinkle effects, elastase inhibition effect of ethanol and water extracts from S. china L. showed 41.1% and 16.3% at $1,000{\mu}g/ml$ concentration. The collagenase inhibition effect of ethanol and water extracts from S. china L. showed more than 96.6% and 60.0% at $1,000{\mu}g/ml$ concentration. As a result of having fibroblast measured cell viability on fibroblast cell of ethanol extract from S. china L., it showed 71.7% with cell viability at $100{\mu}g/ml$ concentration. At $50{\mu}g/ml$ concentration, the procollagen biosynthesis effect of ethanol extract from S. china L. was 139.86%. At the same concentration, the matrix metalloprotease (MMP)-1 inhibition effect of the ethanol extract was 74.9%. According to the results of Western blot of ethanol extract from S. china L., the expression of the MMP-1 protein was decreased by 35% at $50{\mu}g/ml$ concentration. Reverse transcription-polymerase chain reaction (PCR) of ethanol extract from S. china L. showed that the expression of MMP-1 mRNA was decreased by 45% at $50{\mu}g/ml$ concentration. The findings suggest that 70% ethanol extract from S. china L. (SC) has great potential as a cosmeceutical ingredient with antiwrinkle effects.
