Anaerobic co-digestion of swine manure and food waste for biogas production was performed in serum bottles at various volatile solids(VS) contents and mixing ratios of two substrates(swine manure:food waste=$100:0{\sim}0:100$). Through kinetic mode of surface methodology, the methane production was fitted to a Gompertz equation. The ultimate methane production potential of swine manure alone was lower than that of food waste regardless of VS contents. However, it was appeared that maximum methane production potentials in 80 : 20 of the mixing rate at VS 3% was enhanced at 144.7%, compared to its only swine manure. The potential increased up to 815.71 ml/g VS fed as VS concentration and food composition increased up to 3.0% and 20%, respectively. The ultimate amount of methane produced had significantly a positive relationship with that of methane yield rate. Overall, it would be strongly recommended that feeding stocks use 20% of mixing ratio of food waste based on VS 3% contents when operating the anaerobic reactor on site at $35^{\circ}C$ if not have treatment of its anaerobic waste water.
1. Objectives This study was aimed to evaluate marker substances in Gamijowi-tang by high performance liquid chromatography(HPLC) 2. Methods The seperation was performed an AquasilC18(4.6X250mm)column by gradient elution with 0.1N H3PO4 in H2O - acetonitrile(0min 100:10,20min60:40, 40min 80:20, 60min 100:0) as the mobile phase at a flow-rate of 1.0 ml/min with detection at 190-800nm 3. Results Ephedrine and schizandrin were established as marker substances in Gamijowi-tang and its qualitative analysis was carried out. Analyzed was similarity of marker substances between in standard article and in Gamijowi-tang. The result of the analysis was that ephedrine and schizadrin showed 0.9999 of similarity, which meant they were really ephedrine and schizadrin. In addition, ephedrin content was 12.16mg, pseudoephedrine conrenr was 5.09mg, and schizandrin content was 26.6mg in 76g of Gamijowi-tang. 4. Conclusions Qualitative and quantitative analyses of ephedrine and schizandrin in Gamijowi-tang were conducted, and it was identified thar they could be used as marker substances in standardizing quality of Gamijowi-rang.
We investigated the optimal conditions for keratinase production by feather-degrading Pseudomonas geniculata H10 using one variable at a time (OVT) method. The optimal medium composition and cultural condition for keratinase production were determined to be glucose 0.15% (w/v), beef extract 0.08% (w/v), $KH_2PO_4$ 0.12% (w/v), $K_2HPO_4$ 0.02% (w/v), NaCl 0.07% (w/v), $MgSO_4{\cdot}7H_2O$ 0.03%, $MgCl_2{\cdot}6H_2O$ 0.04% along with initial pH 10 at 200 rpm and $25^{\circ}C$, respectively. The production yield of keratinase was 31.6 U/ml in an optimal condition, showing 4.6-fold higher than that in basal medium. The strain H10 also showed plant growth promoting activities. This strain had ammonification activity and produced indoleacetic acid (IAA), siderophore and a variety of hydrolytic enzymes such as protease, lipase and chitinase. Therefore, this study showed that P. geniculata H10 could be not only used to upgrade the nutritional value of feather wastes but also useful in situ biodegradation of feather wastes. Moreover, it is also a potential candidate for the development of biofertilizing agent applicable to crop plant soil.

Porous Co-P catalysts electroplated on Cu in chloride based solution with an addition of $NaH_2PO_2$ and glycine were developed for hydrogen generation from alkaline $NaBH_4$ solution. The microstructures of the Co-P catalysts and their hydrogen generation properties were analyzed as a function of cathodic current density and plating time during the electrodeposition. Amorphous Co-P electrodeposits with porous structure was formed on Cu at cathodic current density of $0.05\;A/cm^2$, and showed very high hydrogen generation rate in alkaline $NaBH_4$ solution due to an increase in the surface area of the catalyst as well as the catalytic activity. The Co-P catalyst, which was obtained at cathodic current density of $0.05\;A/cm^2$ for 5 min, exhibited the best hydrogen generation rate of 2290 ml/min.g-catalyst in 1 wt. % NaOH+10 wt. % $NaBH_4$ solution at $30^{\circ}C$.
High cell density cultivation of microalga Dunaliella bardawil using nitrogen fed-batch cultures was studied in batch flask. Optimum environmental conditions include concentrated nutrients except NaCl and carbon sources, carbon sources, pH, light, agitation, nitrate and phosphate ions. Cell growth, consumption rates of nitrate and phosphate ions were monitored. Optimal conditions for higher cell density were found to be(in the range tested): 5 times concentrated media(1 times-10 times concentrated media) pH 8.0 (7.0-9.0) white light(blue and red light) 15mM of nitrate (0.94-15mM) 250mM $NaHCO_3$ and $CO_2$ gas. However, the addition of phosphate ions did not enhance the algal maximum cell density and specific growth rate. Nitrate was found to be effective for the cell growth. The maximum cell density of fed-batch culture using nitrate ions in $8.955{\times}106$cells/ml after 189hr incubation.
Daily variation of phytoplankton community and environmental parameters were investigated at the lower Nakdong River (Mulgum) from January 2002 to December 2003 to investigate the dynamics of a phytoplankton community in detail. The daily results of water quality in this investigation showed pH (8.1 $\pm$ 0.7), DO (10.3 $\pm$ 2.7 $mg{\cdot}l^{-1}$), water temp. (18.8 $\pm$ 7.4°C), BOD (2.4 $\pm$ 1.0 $mg{\cdot}l^{-1}$), COD (5.3 $\pm$ 1.2 $mg{\cdot}l^{-1}$) and chl. a (43.5 $\pm$ 35.1 mg ${\cdot}m^{-3}$). The results of nutrient factors were the following: TN (3.1 $\pm$ 0.8 $mg{\cdot}l^{-1}$), NO3-N (2.5 $\pm$ 0.5 $mg{\cdot}l^{-1}$), TP (90 $\pm$ 48 ${\mu}g\;{\cdot}\;l^{-1}$), PO4-P (43 $\pm$ 30 ${\mu}g\;{\cdot}\;l^{-1}$). Dominant phytoplankton species during the study period were diatom (Stephanodiscus hantzschii, Aulacoseira granulata var. angustissima and A. italica) and cyanobacteria (Microcystis aeruginosa, Aphanizomenon flos-aquae). The small centric diatom, Stephanodiscus hantzschii, was repeatedly dominant from late fall to the following spring (mean and maximum cell density, 2.3 × 103 $\pm$ 3.8 × 103, 4.5 × 105 cells $ml^{-1}$, respectively). Pinnate diatom, Aulacoseira granulata var. angustissima and A. italica, were frequently observed all season except January to March. Cyanobacteria, Microcystis aeruginosa and Aphanizomenon flos-aquae, proliferated in summer of 2002 except in 2003 due to heavy precipitation. The dominant zooplankton species (March-early May) was rotifer (Brachionus, Keratella, Polyarthra) and cladocerans (Diaphanosoma). The daily observed dynamics of the phytoplankton community in the lower Nakdong River in this study may play an important role in increasing the detailed resolution of limnological information and serving as ecological data for future studies.

In this study, we analyzed and quantified the contents of magnolol (1), honokiol (2), and obovatol (3) in the leaves of Magnolia species by HPLC. HPLC methods were developed and validated by ICH guide lines comparing the linearity, intraday precision, inter-day precision. The optimal conditions were as follows; wavelength: 288 nm, column: Zorbax Eclipse XDBC18 $(2.1\times150\;mm,\;5{\mu}m)$, mobile phase: Acetonitril - 0.1 % $H_3PO_4$ (45:55, v/v) buffer, flow rate: 0.5 ml/min. The contents of magnolol, honokiol and obovatol were 1.08%, 2.37%, and 0.57% in the leaves of M. officinalis, 0.50%, 0.03%, and 1.54% in the leaves of M. biloba, 0.37%, 0.03%, and 0.76% in the leaves of M. obovata. The contents of effective ingredients in the leaves of M. officinalis and M biloba were higher than that of M. obovata. Especially, the content of obovatol in the leaves of M. biloba was remarkably high. This result showed that the shapes of leaves have a certain relationship with the content of obovatol.
This experiment was carried out to study formation of fruitbody with Cordyceps scarabaeicola (EFCC C-252) isolate. This isolate was the one of best fruitbody formation on brown rice 60 g plus 30 g silkworm pupa media among EFCC C-251, EFCC C-252, EFCC C-1092 from EFCC (Entomopathogenic Fungal Culture Collection) of Kangwon National University. Fruiting body was formed only isolate EFCC C-252 among tested isolates on the medium of brown rice (60 g) and silkworm pupae (30 g). The optimal temperature and light for fruiting body formation were $25^{\circ}C$ and fluorescent light (300 lux). The maximal fruiting body formation was observed at 70 g of brown rice and 80 g of silkworm pupa medium which was treated separately. Fruiting body was formed maximally by 2 days interval of irrigation.

관상동맥은 전기적으로 자극하여도 활동전위가 쉽게 유발되지 않는 조직인데, 이는 세포내의 $Ca^{2+}$에 의해서 활성화되는 $K^{+}$통로때문이라고 여겨지고 있다. 저자들은 단일 이온통로 수준에서 이러한 관상동맥 $K^{+}$ 통로의 성질을 조사하고, 아울러 $K^{+}$ 통로 개방제를 포함한 몇가지 약물이 $K^{+}$ 통로에 미치는 영향을 알아보고자 본 연구를 수행하였다. 막소포는 돼지의 심장에서 관상동맥을 분리하여 균질화 및 원심분리하여 준비하였으며, sucrose 20/30%에 있는 층을 취하여 분주한 후 -7$0^{\circ}C$로 보관하여 사용하였다. 평지방막은 1-palmitoyl, 2-oleoyl-phosphatidyl ethanolamine과 1-palmitoyl, 2-oleoyl phosphatidylcholine (4:1)을 n-decane에 녹인지방액(25mg/m1)을 200 $\mu\textrm{m}$의 구멍에 칠하여 만들었다. 단일 $K^{+}$ 통로는 막소포 (1-5$\mu\textrm{g}$/ml)를 평지방막의한쪽에 추가하고 자석으로 저어 융합이 되도록 하였으며, 통로의 활성은 막의 양쪽에 150 mM KCI, 10 mM HEPES-KOH, 0.5 mM EGTA, 0.6 mM $CaCl_2$ (pH 7.2)로 된용액이 존재하는 상태에서 막전압을 다르게 하며 기록하였다. 단일 이온 통로 전류는 amplifier를 통하여 VCR tape에 녹화하여 후에 분석에 이용하였으며, 동시에 종이기록지에 기록하였다. 단일이온통로 활성의 분석은 pClamp (Ver5.5)를 이용하여 수행하였다. 관상동맥에서 가장 흔히 기록되는 이온통로는 막전압과 $Ca^{2+}$의 농도에 의하여 변하는 $K^{+}$ 통로이었는데 pCa 4.0과 -10 mV에서 초기 활성도(Po)는 0.1에서 0.8까지 다양하였다. 전류와 전압과의 관계는 -60 - +60mV의 전압범위에서 직선형이었다.

Objectives : Polygoni Multiflori Ramulus has been widely used as a traditional medicinal herb for the treatment of insomnia, limb pain and itch. The extract of Polygoni Multiflori Ramulus (PMRE) is known to have a modulatory effect of many inflammatory responses. This study was performed to investigate the hepatoprotective effect of PMRE against arachidonic acid (AA) + iron-induced oxidative stress on HepG2 cell and carbon tetrachloride ($CCl_4$)-induced liver injury on mice. Methods : The effects of PMRE on cell viability was assessed by MTT assay. And flow cytometric analysis was performed to estimate the effects on mitochondrial function. To investigate its underlying mechanism, apoptosis-related proteins were analysed by using immunoblot analysis. In addition, ICR mouse were administrated (po) with the PMRE (30, 100 mg/kg) for 3 days and then, injected (ip) with $CCl_4$ (0.5 ml/kg body weight) to induce acute liver damage. The level of pro-caspase-3 was measured. Results : Treatment of PMRE increased relative cell viability, prevented a cleavage of poly (ADP ribose) polymerase and pro-caspase-3, and also reduced mitochondrial membrane permeability against AA + iron-induced oxidative stress. In addition, PMRE treatment decreased liver injury induced by $CCl_4$, as evidenced by increases in pro-caspase-3 level. Conclusions : These results demonstrate that PMRE has an ability to anti-oxidant and hepatoprotective effect against AA + iron-induced oxidative stress and $CCl_4$-induced liver injury.
