Proinflammatory Effects of High Mobility Group B1 (HMGB1) Versus LPS and the Mechanism of IL-8 Promoter Stimulation by HMGB1 (High mobility group B1(HMGB1)과 LPS의 염증유발효과 차이의 비교 및 HMGB1에 의한 IL-8 promoter 자극 기전의 규명)

  • Background: High mobility group box 1 (HMGB1) is a novel, late mediator of inflammation. This study compared the pro-inflammatory effects of LPS and HMGB1. The transcriptional factors that play an important role in mediating the HMGB1-induced stimulation of IL-8 were also evaluated. Methods: RAW264.7 cells were stimulated with either LPS (100 ng/ml) or HMGB1 (500 ng/ml). The $TNF-{\alpha}$, MIP-2 and $IL-1{\beta}$ levels in the supernatant were evaluated by ELISA at 0, 2, 4, 8, 12 and 24h after stimulation. An acute lung injury was induced by an injection of LPS (5 mg/kg) or HMGB1 (2.5 mg/kg) into the peritoneum of the Balb/c mice. The lung cytokines and MPO activity were measured at 4h (for LPS) or 24h (for HMGB1) after the injection. The transcriptional factor binding sites for NF-IL6, $NF-{\kappa}B$ and AP-1 in the IL-8 promoter region were artificially mutated. Each mutant was ligated with pIL-6luc and transfected into the RAW264.7 cells. One hour after stimulation with HMGB1 (500 ng/ml), the cell lysate was analyzed for the luciferase activity. Results: The expression of MIP-2, which peaked at 8h with LPS stimulation, increased sequentially until 24h after HMGB1 stimulation. An intraperitoneal injection of HMGB1, which induced a minimal increased in $IL-1{\beta}$ expression, provoked the accumulation of neutrophils the lung. A mutation of AP-1 as well as $NF-{\kappa}B$ in the IL-8 promoter region resulted in a lower luciferase activity after HMGB1 stimulation. Conclusion: The proinflammatory effects of HMGB1, particularly on IL-8, are mediated by both $NF-{\kappa}B$ and AP-1.

  • As UNFCC(United Nations Framework Convention on Climate Change) is enhanced, Korea will perform a CO2 reduction duty. The CO2 reduction duty will effect Korea power system because coal and oil thermal generations emit large CO2 form about 46% of total CO2 emission. Moreover various alternatives should be designed to comply with CO2 reduction duty. In this paper, we analysis resource planning considering CO2 emission constraints and emission trading. And we analysis resource planning under using LP(Linear Programing) and MIP(Mixed Integer Programing).

  • Recently, there are many efforts to support seamless mobility in 802.11 WLANs using IP Layer mobility protocols. The IP layer mobility protocols are the most efficient mechanism to guarantee the service session continuity when IP subnet is changed during handover. Even if the IP layer mobility protocols are quite efficient, the feature of the protocols that had been designed to consider only L3 layer makes it difficult to improve the performance of hand over more and more. Nowadays, to overcome this limitation of IP mobility protocols, many researchers have worked on the mobility protocols integration of different layers (e.g., L2 layer). In this paper, we propose the enhanced Proxy MIPv4 to minimize the latency of handover using MIH protocol in 802.11 WLANs. The proposed mechanism minimizes the latency of authentication by exchanging security keys between Access Routers during handover. Moreover, it also minimizes packet losses by Inter-AP Tunneling and data forwarding.

  • CCFH(Candidatecasting Fast Handoff) algorithm excels the performance of existing fast handoff schemes using multicasting in terms of handoff latency and B/W efficiency. The scheme uses L2 information(BSSID) at Wireless LAN and starts multicasting before L2 handoff. So it can reduce L3 handoff latency. At this article, we would show that Candidatecasting Fast Handoff Scheme is clearly new scheme using L2 information and has the good performance.

Comparative Study of Toxic Effects of Anatase and Rutile Type Nanosized Titanium Dioxide Particles in vivo and in vitro

  • Two types of nanosized titanium dioxide, anatase ($anTiO_2$) and rutile ($rnTiO_2$), are widely used in industry, commercial products and biosystems. $TiO_2$ has been evaluated as a Group 2B carcinogen. Previous reports indicated that $anTiO_2$ is less toxic than $rnTiO_2$, however, under ultraviolet irradiation $anTiO_2$ is more toxic than $rnTiO_2$ in vitro because of differences in their crystal structures. In the present study, we compared the in vivo and in vitro toxic effects induced by $anTiO_2$ and $rnTiO_2$. Female SD rats were treated with $500{\mu}g/ml$ of $anTiO_2$ or $rnTiO_2$ suspensions by intra-pulmonary spraying 8 times over a two week period. In the lung, treatment with $anTiO_2$ or $rnTiO_2$ increased alveolar macrophage numbers and levels of 8-hydroxydeoxyguanosine (8-OHdG); these increases tended to be lower in the $anTiO_2$ treated group compared to the $rnTiO_2$ treated group. Expression of $MIP1{\alpha}$ mRNA and protein in lung tissues treated with $anTiO_2$ and $rnTiO_2$ was also significantly up-regulated, with $MIP1{\alpha}$ mRNA and protein expression significantly lower in the $anTiO_2$ group than in the $rnTiO_2$ group. In cell culture of primary alveolar macrophages (PAM) treated with $anTiO_2$ and $rnTiO_2$, expression of $MIP1{\alpha}$ mRNA in the PAM and protein in the culture media was significantly higher than in control cultures. Similarly to the in vivo results, $MIP1{\alpha}$ mRNA and protein expression was significantly lower in the $anTiO_2$ treated cultures compared to the $rnTiO_2$ treated cultures. Furthermore, conditioned cell culture media from PAM cultures treated with $anTiO_2$ had less effect on A549 cell proliferation compared to conditioned media from cultures treated with $rnTiO_2$. However, no significant difference was found in the toxicological effects on cell viability of ultra violet irradiated $anTiO_2$ and $rnTiO_2$. In conclusion, our results indicate that $anTiO_2$ is less potent in induction of alveolar macrophage infiltration, 8-OHdG and $MIP1{\alpha}$ expression in the lung, and growth stimulation of A549 cells in vitro than $rnTiO_2$.

  • This research focused on large variation of the result of Ca(OH)$_2$ quantity by TG/DTA test or micro-pore size by MIP test. Assume that sample postion has influenced on experimental result, therefore, relationship between sampling position and the experimental result was studied As a result of the research, it is obvious that samples from inside(P-3) shows more Ca(OH)$_2$ and less porosity than samples from outside(P-1).

Inter-Domain Mobility Management Based on the Proxy Mobile IP in Mobile Networks

  • System Architecture Evolution (SAE) with Long Term Evolution (LTE) has been used as the key technology for the next generation mobile networks. To support mobility in the LTE/SAE-based mobile networks, the Proxy Mobile IPv6 (PMIP), in which the Mobile Access Gateway (MAG) of the PMIP is deployed at the Serving Gateway (S-GW) of LTE/SAE and the Local Mobility Anchor (LMA) of PMIP is employed at the PDN Gateway (P-GW) of LTE/SAE, is being considered. In the meantime, the Host Identity Protocol (HIP) and the Locator Identifier Separation Protocol (LISP) have recently been proposed with the identifier-locator separation principle, and they can be used for mobility management over the global-scale networks. In this paper, we discuss how to provide the inter-domain mobility management over PMIP-based LTE/SAE networks by investigating three possible scenarios: mobile IP with PMIP (denoted by MIP-PMIP-LTE/SAE), HIP with PMIP (denoted by HIP-PMIP-LTE/SAE), and LISP with PMIP (denoted by LISP-PMIP-LTE/SAE). For performance analysis of the candidate inter-domain mobility management schemes, we analyzed the traffic overhead at a central agent and the total transmission delay required for control and data packet delivery. From the numerical results, we can see that HIP-PMIP-LTE/SAE and LISP-PMIP-LTE/SAE are preferred to MIP-PMIP-LTE/SAE in terms of traffic overhead; whereas, LISP-PMIP-LTE/SAE is preferred to HIP-PMIP-LTE/SAE and MIP-PMIP-LTE/SAE in the viewpoint of total transmission delay.

Effect of Leptin on the Expression of Chemokine Genes in THP-1 Cells (THP-1 세포주에서 Leptin에 의한 케모카인 유전자 발현)

  • Background: Leptin is a 16-KDa non-glycosylated peptide hormone synthesized almost exclusively by adipocytes. The well-known function of leptin is regulation of food intake and energy expenditure. Leptin also plays a regulatory role in immune and inflammatory process including cytokine production. The purpose of this study was to investigate the effect of leptin on the expression of several chemokine genes(RANTES, IL-8, MCP-1, IP-10, Mig, MIP-$1{\alpha}$, MIP-$1{\beta}$, and GRO-${\alpha}$) in THP-1 cells. Materials and Methods: Total RNA of THP-1 cells were prepared by Trizol method, and then stimulated with the leptin(250 ng/$m{\ell}$) or LPS(100 ng/$m{\ell}$). We examined the expression patterns of various chemokine mRNAs in THP-1 cell lines by RT-PCR and Northern blot. Results: Leptin did not induce the expression of chemokine mRNAs in THP-1 cells. The expression patterns of RANTES, IL-8, MCP-1, IP-10, and Mig mRNAs in THP-1 cells stimulated with leptin and LPS simultaneously was almost same to the patterns of LPS alone-induced chemokine mRNAs. RANTES mRNA expression was independent on the concentrations of leptin. Although leptin did not have strong effect on the expression of RANTES, IL-8, MCP-1, IP-10, Mig, MIP-$1{\alpha}$, MIP-$1{\beta}$, and GRO-${\alpha}$ mRNAs in THP-1 cells, leptin could induce the expression of long isoform of leptin receptor(OB-RL) mRNA, and its expression was elevated in simultaneous stimulation of leptin and LPS. Conclusion: These data suggest that leptin is able to induce OB-RL in THP-1 cells, however, leptin has little effect on the expression of pro-inflammatory chemokine genes.

Influence of the lung mechanical ventilation with injurious parameters on 7-ketocholesterol synthesis in Sus Scrofa

  • The aim of work was to investigate changes of 7-ketocholesterol synthesis in alveolar macrophages in the dynamic of lung mechanical ventilation with injurious parameters. The goal of in vitro part of work was to observe influence of 7-ketocholesterol on iNOS and MIP1 $\beta$ production in bronchoalveolar lavage fluid (BALF) cells. We used 17 healthy domestic pigs randomly assigned into two treatment groups: group I with mechanical ventilation with physiological parameters; group II underwent injurious ventilation with high volume tidal (VT) and low positive end expiratory pressure (PEEP). Cells were analyzed for CYP27A1 protein and gene expression levels, 7-ketocholesterol production. In alveolar macrophages of group II, we obtained increase of production of CYP27A1 protein and 7-ketocholesterol, as well as the expression of the CYP27A1 gene at the 2nd hour of ventilation. In the in vitro experiments we show dose-dependent increase of MIP1 $\beta$ and decrease of CYP27A1, iNOS protein production after 7-ketocholesterol treatment.

Effects of Five-month Training of Playing Harmonica on Pulmonary Function in Patients With Neuromuscular Disease: A Pilot Study

  • Background: Progressive muscle weakness is aggravated not only in the skeletal muscles but also in the respiratory muscles in many patients with neuromuscular diseases (NMD). Inspiratory muscle training (IMT) has been reported as therapy for pulmonary rehabilitation to improve respiratory strength, endurance, exercise capacity, and quality of life, and to reduce dyspnea. Objects: The purpose of this study was to determine the effect of playing harmonica for 5 months on pulmonary function by assessing the force vital capacity (FVC), peak cough flow (PCF), maximal inspiratory pressure (MIP), maximal expiratory pressure (MEP), and maximal voluntary ventilation (MVV) in patients with NMD. Methods: Six subjects with NMD participated in this study. The subjects played harmonica once a week for 2 hours at a harmonica academy and twice a week for 1 hour at home. Thus, training was performed thrice a week for 23 weeks. The examiner assessed pulmonary function by measuring FVC in the sitting and supine positions and PCF, MIP, MEP, and MVV in the sitting position at the beginning of training and once a month for 5 months. Results: Both sitting and supine FVC significantly increased after playing harmonica (p=.042), as did MIP (p=.043) and MEP (p=.042). Conclusion: Playing harmonica can be used as an effective method to improve pulmonary function in patients with NMD.
