Isolation of a Gibberellin-producing fungus (Penicillium sp. MH7) and Growth Promotion of Crown Daisy (Chrysanthemum coronarium)

  • Plant growth promoting fungi (PGPF) are well known for the production of useful secondary metabolites. However, limited information is available on the gibberellin (GA) production capacity of PGPF of endophytic origin. In the current study, 15 fungal endophytes were isolated from the roots of Crown daisy, and then screened on Waito-c rice, in order to identify plant growth promoting fungi. The fungal isolate MH7 significantly increased the shoot length (12.1 cm) of Waito-c in comparison with control treatment (7.9 cm). In a separate experiment, the culture filtrate (CF) of MH7 significantly promoted the growth attributes of Crown daisy. The MH7 CF was analyzed for gibberellins and it contained all physiologically active gibberellins ($GA_1$, 1.37 ng/ml; $GA_3$, 5.88 ng/ml; $GA_4$, 8.62 ng/ml; and $GA_7$, 2.05 ng/ml) in conjunction with physiologically inactive $GA_9$ (0.83 ng/ml), $GA_{12}$ (0.44 ng/ml), $GA_{15}$ (0.74 ng/ml), $GA_{19}$ (1.16 ng/ml), and $GA_{20}$ (0.98 ng/ml). The CF of MH7 produced higher amounts of $GA_3$, $GA_4$, $GA_7$, $GA_9$, and $GA_{12}$ than wild-type Fusarium fujikuroi, which was used as a control for GA production. The fungal isolate MH7 was later identified as a new strain of Penicillium on the basis of its morphological characteristics and phylogenetic analysis of the 188 rDNA sequence.

Growth and changes in the biochemical composition of Isochrysis galbana under different light-emitting diode conditions

  • The marine microalgae Isochrysis galbana was cultured under various light-emitting diode (LED) light conditions with different wavelengths to examine changes in growth and in amino acid and fatty acid profiles. The culture conditions for the microalgae were Conway medium, salinity of 33 psu, temperature of $24^{\circ}C$, and a 16/8 h light/dark photoperiod. Six light sources, including 5 units of 180W LED lamps (peak wavelength: blue [LB] 470 nm; green [LG] 525 nm; yellow [LY] 595 nm; red [LR] 636 nm; white [LW] 442 nm) and 1 unit of a 175W metal halide (MH) lamp, were used for the experiment. The dry cell weights ($gL^{-1}$) of I. galbana under different light conditions were in the order of LW>LB${\geq}$MH>LR>LG>LY. Levels of essential amino acids were revealed to be significantly higher under LW, LG, and MH than under the other wavelengths (P<0.05). The fatty acid, unsaturated fatty acid, and DHA contents of I. galbana were higher under MH, LW, and LG. In addition, the carotenoid content was higher under MH, LW, and LG than under the other wavelengths (P<0.05). The fucoxanthin content was highest under MH (0.28%) and lowest under LY (0.2%), and it was 0.26% under LW and LG. The results indicate that the combined use of LW and LG is effective when using LED lamps for I. galbana cultivation.

  • Positive ion mass spectra of some corticosteroids were obtained by using liquid chromatography-mass spectrometry(LC-MS). The base peak of each compound showed the protonated molecular ion [$MH^+$], ammonium adduct ion [${MNH_4}^+$] or [$MH^+-60$] ion according to its chemical structure and other characteristic mass ions were [$MH^+-18$], [${MNH_4}^+-18$] and so on. Several rat urinary metabolites of methylprednisolone acetate after the oral administration were detected by the thermospray LC-MS. The identified major metabolites were 20-hydroxymethylprednisolone(20-HMP), methylprednisolone(MP) and methylprednisone(11-KMP), which were supposed to be formed by deacetylation at the position of C-21, reduction at C-20, oxidation at C-11, or due to the bond cleavage between C-17 and C-20.

Inoculation with Bacillus licheniformis MH48 Promotes Nutrient Uptake in Seedlings of the Ornamental Plant Camellia japonica grown in Korean Reclaimed Coastal Lands

  • The objective of this study was to determine whether inoculation with Bacillus licheniformis MH48 as a plant growth-promoting rhizobacterium (PGPR) could promote nutrient uptake of seedlings of the ornamental plant Camellia japonica in the Saemangeum reclaimed coastal land in Korea. B. licheniformis MH48 inoculation increased total nitrogen and phosphorus content in soils by 2.2 and 20.0 fold, respectively, compared to those without bacterial inoculation. In addition, B. licheniformis MH48 produced auxin, which promoted the formation of lateral roots and root hairs, decreased production of growth-inhibiting ethylene, and alleviated salt stress. Total nitrogen and phosphorus uptake of seedlings subjected to bacterial inoculation was 2.3 and 3.6 fold higher, respectively, than the control. However, B. licheniformis MH48 inoculation had no significant effect on the growth of seedlings. Our results suggest that inoculation with B. licheniformis MH48 can be used as a PGPR bio - enhancer to stimulate fine root development, promote nutrient uptake and alleviate salt stress in ornamental plant seedlings grown in the high-salinity conditions of reclaimed coastal land.

Effects of C-MH on Yield and Pure Seed Ratio in Peanut(Arachis hypogaea L.) (개화유제제 C-MH 처리가 땅콩수량과 정입율에 미치는 영향)

  • C-MH(Choline salt of maleic hydrazide, 39%) has been applied on peanut(Arachis hypogaea L.) plant in 1994∼1995 on three different times of 24 days after flowering (DAF), 28 DAF and 31 DAF and on three different levels of 0.2, 0.1. 0.067%, respectively. Main stem length was noticeably retarded by 38∼44% compared to that of non-treatment. Branch numbers were not changed by C-MH treatments. Ratio of flowering inhibition was 38% in 0.2% concentration and 22% in 0.067% at 28 days after flowering treatment compared to that of control. Ratio of pure seed was slightly enhanced by 3% in 28 DAF treatments. Yield productivity was increased by 7% in 0.1%, at 28 DAF treatment compared to that of non-treatment (3.2MT /ha).

  • This study was performed to evaluate the effects of combined treatment of clove bud essential oil (CBEO) and mild heat (MH) on inactivation of Escherichia coli O157:H7 inoculated onto red oak leaves. Combined treatment of 0.2% CBEO with MH at $50^{\circ}C$ exhibited the highest inhibitory effect against E. coli O157:H7 among treatments, resulting in 1.45 log reduction compared with water washing treatment. In addition, inhibitory effect of the combined treatment was maintained during storage of red oak leaves at $4^{\circ}C$ for 9 days, showing 1.67~2.25 log reductions compared with non-treated samples. Thus, these results indicate that combined treatment with CBEO/MH can be used to ensure the microbiological safety of fresh leaf vegetables such as red oak leaves during storage.

A report of six unrecorded radiation-resistant bacterial species isolated from soil in Korea in 2018

  • Six bacterial strains 18JY42-3, 18SH, 18JY76-11, 17J11-11, 18JY14-14, and 18JY15-11 assigned to the phylum Proteobacteria, Firmicutes, and Actinobacteria were isolated from soil samples in Korea. The Cohnella species, strain 18JY42-3 was Gram-stain-positive, short rod-shaped and beige-colored. The Methylobacterium species, strains 18SH and 18JY76-11 were Gram-stain-negative, short rod-shaped and pink-colored. The Microterricola species, strain 17J11-11 was Gram-stain-positive, short rod-shaped and yellow-colored. The Paenarthrobacter species, strains 18JY14-14 and 18JY15-11 were Gram-stain-positive, short rod-shaped and white-colored. Phylogenetic analysis based on 16S rRNA gene sequence showed that strains 18JY42-3, 18SH, 18JY76-11, 17J11-11, 18JY14-14, and 18JY15-11 were most closely related Cohnella rhizosphaerae (MH497628; 98.8%), Methylobacterium goesingense (MH497632; 99.1%), Methylobacterium populi (MH497635; 99.9%), Microterricolagilva (MH504108; 98.4%), Paenarthrobacter nicotinovorans (MH497641; 100%), and Paenarthrobacter nitroguajacolicus (MH497646; 99.2%), respectively. All the six unrecorded strains showed resistance to UV radiation. This is the first report of these six species in Korea.

Dronedarone Attenuates Ang II-Induced Myocardial Hypertrophy Through Regulating SIRT1/FOXO3/PKIA Axis

  • Background and Objectives: Long-term pathological myocardial hypertrophy (MH) seriously affects the normal function of the heart. Dronedarone was reported to attenuate left ventricular hypertrophy of mice. However, the molecular regulatory mechanism of dronedarone in MH is unclear. Methods: Angiotensin II (Ang II) was used to induce cell hypertrophy of H9C2 cells. Transverse aortic constriction (TAC) surgery was performed to establish a rat model of MH. Cell size was evaluated using crystal violet staining and rhodamine phalloidin staining. Reverse transcription quantitative polymerase chain reaction and western blot were performed to detect the mRNA and protein expressions of genes. JASPAR and luciferase activity were conducted to predict and validate interaction between forkhead box O3 (FOXO3) and protein kinase inhibitor alpha (PKIA) promoter. Results: Ang II treatment induced cell hypertrophy and inhibited sirtuin 1 (SIRT1) expression, which were reversed by dronedarone. SIRT1 overexpression or PKIA overexpression enhanced dronedarone-mediated suppression of cell hypertrophy in Ang II-induced H9C2 cells. Mechanistically, SIRT1 elevated FOXO3 expression through SIRT1- mediated deacetylation of FOXO3 and FOXO3 upregulated PKIA expression through interacting with PKIA promoter. Moreover, SIRT1 silencing compromised dronedarone-mediated suppression of cell hypertrophy, while PKIA upregulation abolished the influences of SIRT1 silencing. More importantly, dronedarone improved TAC surgery-induced MH and impairment of cardiac function of rats via affecting SIRT1/FOXO3/PKIA axis. Conclusions: Dronedarone alleviated MH through mediating SIRT1/FOXO3/PKIA axis, which provide more evidences for dronedarone against MH.

Mental Health of Medical Students After Combating the COVID-19 Epidemic: A Cross-sectional Study in Vietnam

  • Objectives: This study was conducted to investigate the prevalence of mental health (MH) symptoms and associated factors among medical students who were engaged in combating the coronavirus disease 2019 (COVID-19) epidemic in 4 provinces/cities of Vietnam. Methods: A cross-sectional study with 580 participants was conducted at a medical university in Northern Vietnam. MH was assessed using the 21-item Depression, Anxiety, and Stress Scale, which was previously standardized in Vietnam. Data were collected through a structured self-administered questionnaire. Multivariate logistic regression was employed to examine the association between MH symptoms and relevant factors. Results: Out of a total of 2703 medical students, 21.5% responded to the questionnaire. Among the 580 respondents, the prevalence rates of depression, anxiety, and stress were 43.3%, 44.0%, and 24.7%, respectively. Factors significantly associated with self-reported depression included being female and having a COVID-19 infection. Similarly, being female and having a COVID-19 infection were significantly associated with self-reported anxiety. Factors associated with self-reported stress included being female, having a personal or family history of MH symptoms, working more than 8 hr/day, and having a COVID-19 infection. Conclusions: COVID-19 has adversely impacted the MH of medical students. Our findings are valuable in their potential to motivate universities, MH professionals, and authorities to offer mental healthcare services to this group. Furthermore, there is a pressing need for training courses designed to equip future healthcare workers with the skills to manage crises effectively.

Development of Battery Management System for Electric Vehicle Applications of Ni/MH Battery

  • Electric vehicle performance is very dependent on traction batteries. For developing the electric vehicles with high performance and good reliability, the traction batteries have to be managed to get maximum performance under various operating conditions. The enhancement of the battery performance can be accomplished by implementing battery management system (BMS) that plays important roles of optimizing the control mechanism of charge and discharge of the batteries as well as monitoring battery status. In this study the battery management system has been developed for maximizing the use of Ni/MH batteries in electric vehicle. This system provides several tasks: the control of charging and discharging, overcharge and over-discharge protection, the calculation and display of state of charge, safety and thermal management. The BMS was installed in and tested using the DEV5-5 electric vehicle developed by Daewoo Motor Co. and Institute for Advanced Engineering in Korea. The 18 modules of Panasonic Ni/MH battery, 12 V-95 Ah, were used in the DEV5-5. The high accuracy within the range of $3\%$ and the good reliability were shown in the test results. The BMS can also improve the performance and cycle life of Ni/MH battery pack as well as the reliability and safety of the electric vehicles (EV).
