Surface Display of Organophosphorus Hydrolase on E. coli Using N-Terminal Domain of Ice Nucleation Protein InaV

  • Recombinant Escherichia coli displaying organophosphorus hydrolase (OPH) was used to overcome the diffusion barrier limitation of organophosphorus pesticides. A new anchor system derived from the N-terminal domain of ice-nucleation protein from Pseudomonas syringae InaV (InaV-N) was used to display OPH onto the surface. The designed sequence was cloned in the vector pET-28a(+) and then was expressed in E. coli. Tracing of the expression location of the recombinant protein using SDS-PAGE showed the presentation of OPH by InaV-N on the outer membrane, and the ability of recombinant E. coli to utilize diazinon as the sole source of energy, without growth inhibition, indicated its significant activity. The location of OPH was detected by comparing the activity of the outer membrane fraction with the inner membrane and cytoplasm fractions. Studies revealed that recombinant E. coli can degrade 50% of 2 mM chlorpyrifos in 2 min. It can be concluded that InaV-N can be used efficiently to display foreign functional protein, and these results highlight the high potential of an engineered bacterium to be used in bioremediation of pesticide-contaminated sources in the environment.

Effect of PCB on the Oocyte Maturation and Proges- terone Production of Frog, Rana dybowskii in Vitro

  • In order to know the effects of PCB(Arochlor 1248) on the oocyte maturation and proges-terone(P$_4$) production by FPH(Frog pituitary homogenate: 0.01 p.e/$m\ell$) of frog in vitro, the oocytes were cultured for 20 hours in presence of the PCB at various concentrations and exam-ined their maturation(germinal vesicle breakdown: GVBD) rates and P$_4$ levels secreted by the oocyte in the culture medium. The results show that PCB concentration of 10 ppb suppressed the maturation of the oocytes and secretion of P$_4$. To examine the reversibility of the inhibitory effects, the oocytes were exposed to the PCB only for 3 hours, and then transferred to plain medium and cultured further for 17 hours. The oocytes were recovered from the toxic effect of the PCB when they were exposed to 2.5 ppb, but not to 5 ppb of the PCB. These results indi-cate that PCB suppress the maturation of oocytes and secretion of p, at low concentration, sug-gesting that the frog oocyte culture system can be used as a useful tool to evaluate the toxicity of the pollutants in the environment.

Development of a Measuring Instrument for the Coefficient of Luminous Intensity of Retro-Reflection of Miniature Telecentric Cameras (소형 텔레센트릭 카메라 광학계의 재귀반사 측정 방법 연구 및 측정 장치 개발)

  • In this paper, the optical properties of a miniature telecentric camera are studied analytically and experimentally. By means of optical properties, the luminous intensity of retro-reflection of a miniature telecentric optical system is investigated. First, from a simulation the theoretical value of the coefficient of luminous intensity of retro-reflection of a miniature telecentric system is estimated. Second, a miniature telecentric camera device is designed, and the value of the coefficient of luminous intensity of retro-reflection of the actual device is measured through experiment. From simulation and experiment, we found that both the estimated and observed values of the coefficient of luminous intensity of retro-reflection range from 0.002 to $0.03m^2/sr$ (P384200CPH: $0.0042-0.018m^2/sr$, P285200CPH: $0.0045-0.0297m^2/sr$, P321450S: $0.0021-0.00963m^2/sr$) at 1.4 to 4.9 arcminute. The results prove the significance of the mechanisms used to design the miniature telecentric camera, and the experimental method. Accordingly, with these significant results, this study contributes to the development of theory and practice in optical engineering science.

  • Terra MODIS is one of the few space-borne sensors currently capable of acquiring radiometric data over the range of view angles. Institute of Industrial Science, University of Tokyo, has been receiving Terra MODIS data at Tokyo since May 2001 and Asian Institute of Technology at Bangkok since May 2001. They can cover whole East Asia and is expected to monitor environmental changes regularly such as deforestation, forest fires, floods and typhoon. Over eight hundred scenes have been archived in the storage system and they occupy 2 TB of disk space so far. In this study, MODIS data processing system on WWW is developed including following functions: spectral subset (250m, 500m, 1000m channels), radiometric correction to radiance, spatial subset of geocoded data as a rectangular area with latitude-longitude grid system in HDF format, generation of a quick look file in JPEG format. Users will be notified just after all the process have finished via e-mail. Using this system enables us to process MODIS data on WWW with a few input parameters and download the processed data by FTP access. An easy to use interface is expected to promote the use of MODIS data. This system is available via the Internet on the following URL from September 1 2002, "http : //webmodis.iis.u-tokyo.ac.jp/".

Optimization and Flavor Quality of Enzymatic Hydrolysate from Dark Muscle of Skipjack

  • Enzymatic hydrolysis of dark muscle of skipjack was optimized by using response surface methodology. Three factors of independent values were pH (4.2 to 9.8), time (0.6 to 3.4 hrs) and temperature (34℃ to 76℃), and independent values were optical density and brix. The optimum conditions for enzymatic hydrolysis were pH 7.0 to 8.0, 55℃ and 3 hrs. The headspace volatile compounds of reaction flavors using the enzymatic hydrolysate, cysteine and xylose were identified by using the combination of a canister system, gas chromatography and mass selective detector. Among 67 compounds, we identified 8 sulfur-containing compounds and 7 furans which were thought to be highly related to meat-like flavors.

Evaluation of circulating PD-1 and PD-L1 as diagnostic biomarkers in dogs with tumors

  • Background: Programmed cell death protein-1 (PD-1) and programmed cell death ligand-1 (PD-L1) have important roles in tumor evasion of the immune system. Objectives: This study aimed to assess the diagnostic utility of circulating PD-1 and PD-L1 levels in healthy dogs and dogs with tumors. Methods: Circulating PD-1 and PD-L1 levels in the serum of 71 dogs with tumors were compared with those of 52 healthy dogs by performing enzyme-linked immunosorbent assay (ELISA). Results: The ELISA results revealed higher circulating PD-1 and PD-L1 levels in dogs with tumors (2.9 [2.2-3.7] ng/mL; median [IQR] and 2.4 [1.4-4.4] ng/mL, respectively) than in healthy dogs (2.4 [1.9-3.0] ng/mL; p = 0.012 and 1.4 [0.9-2.1] ng/mL; p < 0.001, respectively). Especially, there was a significant difference in circulating PD-1 levels between healthy dogs and dogs with malignant epithelial tumors (2.4 [1.9-3.0] ng/mL and 3.1 [2.6-4.4] ng/mL, respectively; p < 0.01). In addition, there was a significant difference in circulating PD-L1 levels between healthy dogs and dogs with lymphomas (1.4 [0.9-2.1] ng/mL and 2.7 [1.6-5.8] ng/mL, respectively; p < 0.001). Conclusion: This study indicates that circulating PD-1 and PD-L1 have potential as tumor diagnostic biomarkers in dogs with tumors.

The Effect of NaOH treatment-Citric acid neutralization of Korean Standard Operating Procedure (SOP) on Physicochemical Compositions of Domestic Swine Excrement for the Connection to Thermophilic Aerobic Oxidation (TAO) (구제역 긴급행동지침(SOP)상의 화학처리방법으로 처리된 양돈분뇨의 고온호기산화공정 적용 연구)

  • We planned to study how the chemical disposal designed by SOP can affect on physicochemical compositions of the livestock excrement. According to Livestock Manure Management Scheme, we experimented in two steps; the first step, NaOH treatment-Citric acid neutralization, and then the second procedure, Thermophilic Aerobic Oxidation (TAO) system. Physicochemical compositions of the 3-days-old samples after NaOH treatment were pH 10.31, EC 24.54 mS/cm, SCOD 3,022 mg/L, T-N 4,315 mg/L, $NH{_4}^+-N$ 1,960 mg/L, and not detected E. coli.. And those of one-day-old samples after citric acid neutralization were pH 7.36, EC 32.89 mS/cm, $SCOD_{Mn}$ 12,733 mg/L, T-N 4,787 mg/L, $NH{_4}^+-N$ 2,450 mg/L, and E. coli. not detected. In contrast, the physicochemical compositions of the treatment plots after the second treatment with TAO system (72hr) were pH 9.42 EC 24.21 mS/cm, $SCOD_{Mn}$ 3,660 mg/L, T-N 3,616 mg/L, $NH{_4}^+-N$ 1,190 mg/L, and no detection of E. coli.

Inhibitiory Activity of Lactic Acid Bacteria against Hazardous Microbes

  • One hundred of lactic cultures were evaluated for their ability to inhibit hazardous microbes, such as Salmonella enteritidis, Salmonella typhimurium, Escherichia coli, Listeria monocytogenes, and Bacillus cereus by agar well diffusion method. None of them showed inhibitory halo against S. enteritidis, while 27 strains showed inhibitory activity against S. typhimurium, 6 against E. coli, 9 against ampicillin resistant E. coli, 31 against L. monocytogens, 10 against B. cereus. pH of the culture does not explain for the inhibitory activity except against B. cereus. A neutralized culture from corn silage showed highest inhibitory activity against S. typhimurium, and the size of inhibitory halo was same as 10 ug/mL of ampicillin. The culture was identified to be Lactobacillus buchneri on the basis of biochemical characteristics and utilization of substrates. Using the culture as probiotics could be expected to reduce antibiotics for animal feeding.

The Introduction of Polychaetes Hydroides elegans (Haswell), Polydora limicola Annenkova, and Pseudopotamilla occelata Moore to the Northwestern Part of the East Sea

  • The polychaeta fauna of the benthos and fouling of the northwestern part of the East Sea was studied during the period of 1971-1998. Three introduced species of polychaetes: Hydorides elegans (Haswell), Polydora limicola Annenkova, and Pseudopotamilla occelata Moore were found. H. elegans was discovered only on the artificial surfaces in Golden Horn Inlet (port Vladivostok), where this species may occur because of hermal pollution due to the discharge of warm waters of the water cooling system of Thermal-Electric Power Station-2 (TEPS-2) in Vladivostok which has been in function since 1971. The abundant population of H. elegans exists in the bay throughout the year and is capable of reproduction. The biomass of H. elegans may reach several $kg/m^2$ in August-September. P. limicola was found at the same time in the fouling of hydrotechnical structures of Vladivostok, Nakhodka, Holmsk and Uglegorsk ports with a biomass of $1-3kg/m^2$. Slow introduction of P. limicola occurs by coastal sail ships at present. The invasion of P. occelata into Peter the Great Bay may be an example of introduction and subsequent naturalization, which produced considerable changes in the structure of benthic communities. The three species of polychaetous sessile organisms and their invasion occurred by ocean and coasters sea-going ships (unintentional transport vectors). H. elegans and P. occelata were most probably transported to the northwestern part of the East Sea from Japan, and P.

Technical Improvement Using a Three-Dimensional Video System for Laparoscopic Partial Nephrectomy

  • Background: Laparoscopic partial nephrectomy is one of the major surgical techniques for small renal masses. However, it is difficult to manage cutting and suturing procedures within acceptable time periods. To overcome this difficulty, we applied a three-dimensional (3D) video system with laparoscopic partial nephrectomy, and evaluated its utility. Materials and Methods: We retrospectively enrolled 31 patients who underwent laparoscopic partial nephrectomy between November 2009 and June 2014. A conventional two-dimensional (2D) video system was used in 20 patients, and a 3D video system in 11. Patient characteristics and video system type (2D or 3D) were recorded, and correlations with perioperative outcomes were analyzed. Results: Mean age of the patients was $55.8{\pm}12.4$, mean body mass index was $25.7{\pm}3.9kg/m^2$, mean tumor size was $2.0{\pm}0.8cm$, mean R.E.N.A.L nephrometry score was $6.9{\pm}1.9$, and clinical stage was T1a in all patients. There were no significant differences in operative time (p=0.348), pneumoperitoneum time (p=0.322), cutting time (p=0.493), estimated blood loss (p=0.335), and Clavien grade of >II complication rate (p=0.719) between the two groups. However, warm ischemic time was significantly shorter in the 3D group than the 2D group (16.1 min vs. 21.2min, p=0.021), which resulted from short suturing time (9.1 min vs. 15.2 min, p=0.008). No open conversion occurred in either group. Conclusions: A 3D video system allows the shortening of warm ischemic time in laparoscopic partial nephrectomy and thus may be useful in improving the procedure.
