일본목련의 분산 및 식물군집 특성에 관한 연구 -한국유네스코평화센터 주변을 대상으로- (The Dispersal and Plant Community Characteristics of Magnolia obovata - Focused on Case of Korea UNESCO Peace Center area in Gyeonggi-do -)

  • 본 연구는 초본에 한하여 진행되고 있는 귀화식물 연구 영역을 확대하여 목본 귀화식물인 일본목련을 대상으로 산림군집, 분산범위, 산포방법 등을 조사하였다. 대상지는 경기도 이천시 호법면 매곡리 산 27번지 일대 한국유네스코평화센터 주변 산림지역이다. 일본목련의 분산특성 조사결과 조경 식재된 일본목련이 모수를 중심으로 산림내부까지 확산되었으며, 거리별 분산은 모수로부터 $20{\sim}100m$ 에서 가장 많은 개체수가 관찰되었고 최대 420m 까지 분포하였다. 이러한 일본목련의 분산은 중력산포에 의한 경향이 두드러진 것으로 보이며, 그 밖에 어치, 까마귀, 까치, 청설모, 다람쥐 등의 야생동물에 의한 동물산포가 이루어진 것으로 보인다. 산림군집조사 결과 리기다소나무-일본목련-신갈나무 군집에서는 일본목련이 리기다소나무를 피압 가능한 상태에 있었으며, 신갈나무-일본목련 군집은 중 하층에서 신갈나무와 경쟁 중인 것으로 보인다. 생장량 및 울폐도 분석결과 일본목련은 신갈나무와 졸참나무보다 생장이 빠르며, 내음성 수종으로서 신갈나무, 졸참나무와의 경쟁에서도 뒤지지 않는 것으로 분석되었다. 결과적으로 일본목련은 출현한 군집내 층간우점도, 내음성, 속성수 등의 특징으로 인하여 차후에 산림생태계의 교란을 야기할 것으로 보인다. 특히 본 대상지 산림 내부에서 관찰된 수고 10m, 흉고직경 10cm 이상인 일본목련이 교목층에 분포하고 있는 지역은 일본목련 성목을 대상으로 벌채 등을 통한 관리가 필요한 것으로 판단된다.

A Short Review on the Chemistry, Pharmacological Properties and Patents of Obovatol and Obovatal (Neolignans) from Magnolia obovata

  • This short review on the chemistry, pharmacological properties and patents of obovatol and obovatal from Magnolia obovata is the first publication. Pharmacological properties are focused on anti-cancer, anti-inflammatory, anti-platelet and neuroprotective activities. Obovatol and obovatal were first isolated from the leaves of M. obovata. Also reported in the bark and fruits of M. obovata, obovatol and obovatal are neolignans i.e., biphenolic compounds bearing a C-O coupling. Other classes of compounds isolated and identified from M. obovata include sesquiterpene-neolignans, dineolignans, trineolignan, lignans, dilignans, phenylpropanoids, phenylethanoid glycosides, flavonoids, phenolic acids, alkaloids, sesquiterpenes, ketone and sterols. The anti-cancer properties of obovatol and obovatal involve apoptosis, inhibition of the growth, migration and invasion of cancer cell lines. However, obovatol displays cytotoxicity against cancer cells but not obovatal. Similarly, anti-inflammatory, anti-platelet, neuroprotective, anxiolytic and other pharmacological activities were only observed in obovatol. The disparity in pharmacological properties of obovatol and obovatal may be attributed to the -CHO group present in obovatal but absent in obovatol. From 2007 to 2013, eight patents were published on obovatol with one mentioning obovatal. They were all published at the U.S. Patent and Trademark Office by scientists of the Korea Research Institute of Bioscience and Biotechnology (KRIBB) as inventors and assignee, respectively. Some future research and prospects are suggested.

화장품 소재로서 후박 에틸아세테이트 분획물의 미백활성에 관한 효과 (A Study of the Whitening Activities of Magnolia obovata Bark Ethyl Acetate Fractions as Cosmetic Ingredient)

  • 천연물인 후박(Magnolia obovata Bark, M. obovata Bark)을 에탄올 추출 및 농축하여 에틸아세테이트 층으로 분획한 분획물이 피부에 안전하고 효과적인 미백 활성을 확인함으로써 미백 화장품 원료 소재로서 가능성을 확인하였다. 후박의 에탄올 농축물을 에틸아세테이트 분획하여 유효물질 honokiol을 HPLC로 정량하였다. 후박 에틸아세테이트 분획물에 대한 in vitro 미백활성 결과, 농도 의존적으로 세포 외 멜라닌 분비를 감소시켜 $IC_{50}=11.05{\mu}g/mL$임을 확인하였다. 또한 세포에 독성을 가지지 않는 최대농도인 $12.5{\mu}g/mL$ 처리 시 최대 약 60%의 멜라닌 분비 억제로 세포 외로 분비되는 멜라닌의 양을 효과적으로 억제함을 확인할 수 있었다. 또한 ${\alpha}-MSH$ (50 nM) 처리한 그룹과 비교하여 $IC_{50}=10.85{\mu}g/mL$로 우수한 세포 내 멜라닌 생성억제 효과를 보였으며 세포에 자극이 되지 않는 최대농도인 $12.5{\mu}g/mL$ 처리 시 최대 약 59%의 멜라닌 생성 억제를 확인하여 세포 외 멜라닌 분비 뿐만 아니라 세포 내에 존재하는 멜라닌의 양 또한 효과적으로 억제함을 확인할 수 있었다. 양성대조군으로 사용된 ${\alpha}-arbutin$의 경우, $IC_{50}=59.99{\mu}g/mL$로 후박 에틸아세테이트 분획물이 양성대조군 ${\alpha}-arbutin$과 비교하여 우수한 세포 내 멜라닌 생성 억제 효과를 가짐을 확인하였다. 임상연구의 경우, 후박 에틸아세테이트 분획물을 적용한 화장품 크림은 (주)대한피부과학연구소(KDRI) 윤리위원회의 IRB 승인(KDRI-IRB-1536) 후 반복 첩포를 통한 인체 누적첩포시험을 진행하여 무감작 물질로 확인하였다. 또한 후박 에틸아세테이트 분획물을 적용한 화장품 크림은 대조군 대비 국소적 미백효과와 신뢰성 있는 피부 안전성을 보여주었다. 최종적으로 후박 에틸아세테이트 분획물이 안전하고 효과적인 미백효과를 가지는 화장품 소재로 충분한 이용 가능성이 있음을 확인하였다.

Synergistic Antimicrobial Effect of Lonicera japonica and Magnolia obovata Extracts and Potential as a Plant-Derived Natural Preservative

  • Most people use cosmetics to protect their skin. Preservatives are often used to prevent their contamination upon use. There has been a great demand for natural preservatives due to recent reports on the side effects of parabens. Therefore, we evaluated the antimicrobial activities of Lonicera japonica and Magnolia obovata extracts and determined their potential as natural preservatives. We found that the 50% ethanol extract from L. japonica had antibacterial activity only against S. aureus and P. aeruginosa, while the ethyl acetate fraction showed antimicrobial activity against all six microbial strains tested. On the other hand, the 70% ethanol extract and the ethyl acetate fraction from M. obovata showed antimicrobial activity against all six strains. A synergistic effect against S. aureus, B. subtilis, and C. albicans was confirmed when two ethyl acetate fractions having antimicrobial activity against all six strains were used in combination. Synergistic activity against B. subtilis was also confirmed through kill-time analysis. High-performance liquid chromatography was performed to identify the components of each extract. Based on the minimum inhibitory concentration and the results of a disc diffusion assay, we confirmed that caffeic acid and luteolin influenced the antimicrobial activity of L. japonica and that the antimicrobial activity of M. obovata was influenced by the interaction of magnolol and honokiol with other components. Therefore, this study suggests that the combination of L. japonica and M. obovata extracts may be used as a plant-derived natural preservative.

후박(厚朴)의 외.내부형태 및 이화학패턴연구 (A Study of Morphology and Pattern Analysis in Magnoliae Cortex)

  • Objectives: To determine the standards for discrimination of Magnoliae Cortex, the experiment of specific external-internal characters and the physicochemical pattern analysis were performed. Methods: External characteristics was observed using a stereoscope. Paraffin-mediated sectioned materials were stained by Ju's method. Physicochemical patterns of materials were analyzed using HPLC. Results: 1. Botanical characteristics: Magnolia officinalis had one seed and a white flower, while M. obovata had two seeds and a white flower. Machilus thunbergii had berry and spherical fruits and yellowish green panicles. 2. External characteristics: M. officinalis and M obovata were dark and thick. M. officinalis was gray brown and greasy while M. obovata was light-gray, less oily and smoothly sectioned. Machilus thunbergii was thin and relatively light or yellow-brown, coarsely sectioned and faintly specific scents. 3. Internal characteristics: The bast parts of M. officinalis and M. obovata were commonly wider than Machilus thunbergii The cork cortex of M. officinalis was $10{\sim}mg/L$ cell layers with many oil cells, while that of M. obovata was $4{\sim}7$ cell layers with less oil cells. Machilus thunbergii's xylem which consisted of ring-shaped cambium at 1st and 2nd part was occupied in large portion. 4. Physicochemical pattern: Both M. officinalis and M. obovata involved honokiol and magnolol. All kinds of M. officinalis involved Magnatriol B but one kind of M. ovobata and all of Machilus thunbergii didn't. Machilus thunbergii showed different pattern of chromatogram from that of 2 species above. Conclusions: These results could be used as standards for discrimination of Magnoliae Cortex and as the method of objectification in medicinal herbs giving the basic resource for bioactivity research.

Antimicrobial Activities of Hydroxybiphenyl Derivatives (I)

  • It was revealed that magnolol and honokiol isolated from the stem bark of Magnolia obovata, had potent antibacterial activity against Bacillus anthracis. A quantitative analytical method of magnolol and honokiol by HPLC has been established, and the amounts of the two components in the dried stem bark of M. obovata were 1, 94% and 0.44%, respectively.

중국 및 일본산 후박 잎의 형태 및 성분 비교 (The Comparison of Morphological and Constituents of the Leaves of Magnolia officinalis, M. biloba and M. obovata)

  • In this study, we analyzed and quantified the contents of magnolol (1), honokiol (2), and obovatol (3) in the leaves of Magnolia species by HPLC. HPLC methods were developed and validated by ICH guide lines comparing the linearity, intraday precision, inter-day precision. The optimal conditions were as follows; wavelength: 288 nm, column: Zorbax Eclipse XDBC18 $(2.1\times150\;mm,\;5{\mu}m)$, mobile phase: Acetonitril - 0.1 % $H_3PO_4$ (45:55, v/v) buffer, flow rate: 0.5 ml/min. The contents of magnolol, honokiol and obovatol were 1.08%, 2.37%, and 0.57% in the leaves of M. officinalis, 0.50%, 0.03%, and 1.54% in the leaves of M. biloba, 0.37%, 0.03%, and 0.76% in the leaves of M. obovata. The contents of effective ingredients in the leaves of M. officinalis and M biloba were higher than that of M. obovata. Especially, the content of obovatol in the leaves of M. biloba was remarkably high. This result showed that the shapes of leaves have a certain relationship with the content of obovatol.

형태특징 및 RAPD분석에 의한 후박(厚朴) 약재의 진위감별 (Discrimination of Genuines and Misuses of Hu-Bak medicines by Morphological Characters and Random Amplified Polymorphic DNA Analysis)

  • For identification of genuine and misuse samples of Hu-Bak medicines, an morphological characters, and RAPD (random amplified polymorphic DNA) were performed. In this study, three samples were collected, of which two were genuine and one was misuse sample. Genuine samples of 'Hu-Bak' were Magnolia obovata Thunberg and Magnolia officinalis Rehder et Wilson. Misuse samples was Machilus thunbergii S. et Z. In the morphological characters, M. obovata Thunberg and M. officinalis Rehder et Wilson were similar, but M. thunbergii S. et Z. was different with cortex, cambium, and fibrous layer. The result of RAPD analysis, seven primers can distinguish between genuines and counterfeit samples of Hu-Bak medicines.

승마, 황련, 황백, 후박 추출 혼합물의 항노화 및 멜라닌 생성 저해효과 (Anti-aging and Anti-melanogenesis Effect of Cimicifuge dahurica, Coptis chinensis, Phellodendri amurense and Magnol obovata Extracts Mixture)

  • 한의학에서 승마(Cimicifuge dahurica, C. dahurica), 황련(Coptis chinensis, C. chinensis), 황백(Phellodendri amurense, P. amurense) 추출물은 해독작용을 하며 후박(Magnol obovata, M. obovata) 추출물은 항균 효과가 있다고 알려져 있다. 본 연구에서는 승마, 황련, 황백, 후박 추출물을 혼합하여 기능성 화장품 소재로써의 이용 가능성을 검토하였다. MTT assay를 이용한 세포 독성평가와 DPPH와 ABTS를 이용한 항산화 효과를 측정한 결과, 단일 추출물보다 추출혼합물이 더 안전함을 확인 하였으며 농도 의존적으로 항산화 활성이 증가됨을 확인하였다. 뿐만 아니라 NO 생성을 억제함으로써 우수한 항염증 효과가 있을 것으로 예상되며, 섬유아세포에서 UVB로 유도된 세포 손상을 보호하면서 type 1 pro-collagen 합성량이 증가되는 것을 관찰하였다. 또한 추출 혼합물은 melanin 합성과 tyrosinase 활성 및 melanin 합성 관련 인자 및 효소들(MITF, tyrosinase, TRP-1, TRP-2)의 발현을 효과적으로 억제하였다. 이러한 연구결과로 보아 승마, 황련, 황백, 후박 추출 혼합물은 항노화 및 미백 등 기능성 화장품 소재로써 개발 가능성이 높을 것으로 사료된다.

Anti-complement Activity of Phenolic Compounds from the Stem Bark of Magnolia obovata

  • Five neolignans (1 - 4, 8), two sesquiterpene-lignans (5 - 6), and two phenylpropanoids (7, 9) were isolated from the stem bark of Magnolia obovata Thunberg (Magnoliaceae) by repeated column chromatography. The structures of isolated compounds were identified as 4-methoxyhonokiol (1), obovatol (2), magnolol (3), honokiol (4), eudeshonokiol B (5), eudesobovatol B (6), coumaric acid (7), magnaldehyde B (8), and ${\rho}-coumaric$ acid (9) on the basis of spectroscopic analysis including 2D-NMR and MS data. Compounds 1 - 9 were evaluated for their anti-complement activities against the classical pathway of the complement system. Of them, compound 8 showed significant anti-complement activity on the classical pathway with $IC_{50}$ value of 102.7 ${\mu}M$, whereas compounds 1 - 7 and 9 were inactive. This result indicated that an aldehyde group in the neolignan is important for the anti-complement activity against the classical pathway.
