A NOTE ON ENDOMORPHISMS OF LOCAL COHOMOLOGY MODULES

  • Let I denote an ideal of a Noetherian local ring (R, m). Let M denote a finitely generated R-module. We study the endomorphism ring of the local cohomology module $H^c_I(M)$, c = grade(I, M). In particular there is a natural homomorphism $$Hom_{\hat{R}^I}({\hat{M}}^I,\;{\hat{M}}^I){\rightarrow}Hom_R(H^c_I(M),\;H^c_I(M))$$, $where{\hat{\cdot}}^I$ denotes the I-adic completion functor. We provide sufficient conditions such that it becomes an isomorphism. Moreover, we study a homomorphism of two such endomorphism rings of local cohomology modules for two ideals $J{\subset}I$ with the property grade(I, M) = grade(J, M). Our results extends constructions known in the case of M = R (see e.g. [8], [17], [18]).

  • An apparatus for continuous measurements of sorption isotherm of dried food was manufactured to shorten the time required for equilibration. The apparatus was so designed that the temperature, air velocity and relative humidity in the experimental chamber could be controlled. The use of dynamic stream of conditioned air with a velocity of 0.2m/sec, instead of static atmosphere, allowed a faster equilibration of dried filefish muscle at $25^{\circ}C$. The mean time necessary for the equilibration of dried filefish muscle at the water activity of a given state to a higher water activity was about 45 hours. The monolayer moisture content of dried filefish muscle calculated from BET-equation was 0.092 kg water /kg dry matter at $25^{\circ}C$.

Heteroexpression and Functional Characterization of Glucose 6-Phosphate Dehydrogenase from Industrial Aspergillus oryzae

  • The engineered Aspergillus oryzae has a high NADPH demand for xylose utilization and overproduction of target metabolites. Glucose-6-phosphate dehydrogenase (G6PDH, E.C. 1.1.1.49) is one of two key enzymes in the oxidative part of the pentose phosphate pathway, and is also the main enzyme involved in NADPH regeneration. The open reading frame and cDNA of the putative A. oryzae G6PDH (AoG6PDH) were obtained, followed by heterogeneous expression in Escherichia coli and purification as a his6-tagged protein. The purified protein was characterized to be in possession of G6PDH activity with a molecular mass of 118.0 kDa. The enzyme displayed maximal activity at pH 7.5 and the optimal temperature was $50^{\circ}C$. This enzyme also had a half-life of 33.3 min at $40^{\circ}C$. Kinetics assay showed that AoG6PDH was strictly dependent on $NADP^+$ ($K_m=6.3{\mu}M$, $k_{cat}=1000.0s^{-1}$, $k_{cat}/K_m=158.7s^{-1}{\cdot}{\mu}M^{-1}$) as cofactor. The $K_m$ and $k_{cat}/K_m$ values of glucose-6-phosphate were $109.7s^{-1}{\cdot}{\mu}M^{-1}$ and $9.1s^{-1}{\cdot}{\mu}M^{-1}$ respectively. Initial velocity and product inhibition analyses indicated the catalytic reaction followed a two-substrate, steady-state, ordered BiBi mechanism, where $NADP^+$ was the first substrate bound to the enzyme and NADPH was the second product released from the catalytic complex. The established kinetic model could be applied in further regulation of the pentose phosphate pathway and NADPH regeneration of A. oryzae to improve its xylose utilization and yields of valued metabolites.

Purification and Characterization of Myrosinase in Dolsan Leaf Mustard(Brassica juncea) and Changes in Myrosinase Activity during Fermentation of Leaf Mustard Kimchi (돌산갓의 Myrosinase 분리 정제 및 갓김치 숙성 중 Myrosinase 활성도의 변화)

  • Myrosinase in leaf mustard was purified and characterized to furnish a grounding information for utilizing the pungent taste and the potential antimicrobial capability of Dolsan leaf mustard to enhance the taste and storage life of kimchi. When myrosinase was purified from leaf mustard through a series of DEAE Sephadex, chromatofocusing and Con A Sepharose column chromatography, specific activity of the enzyme increased 7107-fold compared with that of crude enzyme preparation, and 18.8% yield was obtained. The purified myrosinase showed the optimum pH of 5.9, isoelectric point of 4.6, molecular weight of 129 kD, Km of 0.206 mM, and Vmax of $2.039\;{\mu}M{\cdot}min^{-1}{\cdot}mg\;protein^{-1}$, respectively. The optimum concentration of L-ascorbic for the maximum activity of the enzyme was 0.6 mM, and the enzyme activity decreased at a higher concentration of L-ascorbic acid than 0.6 mM, showing almost no enzyme activity at a L-ascorbic acid concentration of higher than 2.0 mM. Myrosinase activity in leaf mustard kimchi immediately after the kimchi was formulated was shown to be about 70 nmol/min/mg protein which decreased rapidly after 3 days of storage at $20^{\circ}C$, showing that less than half and almost none of the enzyme activity was retained in 4 and 10 days of storage, respectively.

Zizyphus jujube-based Edible Film Development by the Depolymerization Processes (고분자 분쇄 공정을 이용한 대추 소재 가식성 필름 개발)

  • Edible films were developed from jujube (Zizyphus jujube Miller) using depolymerization processes of ultrasound and high-pressure homogenization. A 4.6% (w/w) jujube hydrocolloid was treated by ultrasound (600W, 20 min) or homogenized at high pressure (172 MPa, 6 s) and mixed with whey protein isolate, glycerol, xanthan, and sucrose esters of fatty acids to form film-forming solutions from which films were formed by drying. The film prepared by highpressure homogenization (HPH film) produced more homogeneous films without particles than those prepared without depolymerization or with the ultrasound treatment. HPH films possessed the highest tensile strength (4.7MPa), the lowest water vapor permeability ($2.9g{\cdot}mm/kPa{\cdot}h{\cdot}m^2$), and the most uniform and dense microstructures among the films. Flavor profiles of jujube powder and the films were distinguishable. Heat seal strength and oxygen permeability of the HPH films were 44.4 N/m and $0.025mL{\cdot}{\mu}m/m^2$/day/Pa, respectively. Antioxidant activities of jujube power and HPH films were not significantly different.

Thermal Characteristics of Pellets made of Agricultural and Forest by-products (농림부산물을 이용한 펠릿의 열적 특성)

  • Biomass is considered to be a major potential fuel and renewable resource for the future. In fact, there is high potential to produce the large amount of energy from biomass around the world. In this study, to obtain basic data for practical application of agricultural and forest by-products as fuel of heating system in agriculture, agricultural and forest biomass resources were surveyed, the pelletizer with capacity of $50\;kg{\cdot}h^{-1}$ was designed and manufactured and pellets were made by the pelletizer. High heating value, ash content, etc. of pellets made of agricultural and forest by-products were estimated. Straw of rice was the largest agricultural biomass in 2009 and the total amount of rice straw converted into energy of $299{\times}10^3$ TOE. And in 2009, amount of forest by-product converted into energy of $9,579{\times}10^3$ TOE. High heating values of pellets made of stem and seed of rape, stem of oat, rice straw and rice husk were 16,034, 16,026, 16,089, 15,650, $15,044\;kJ{\cdot}kg^{-1}$ respectively. High heating values of pellets made of agricultural by-products were average 83.6% compared to that of wood pellet. Average bulk density of pellets made of stem and seed of rape, stem of oat, rice straw and rice husk was $1,400\;kg{\cdot}m^{-3}$ ($1.4\;g{\cdot}cm^{-3}$). Ash contents of the pellets were 6.6, 7, 6.2, 5.5, 33% respectively. Rice husk pellet produced the largest ash content compared to other kinds of pellets.

Oxidation Reaction of Hydrazobenzene by Activated Catalysts of Oxygen Adducted Tetradentate Schiff Base Cobalt(Ⅱ) Complexes in Methanol Solvent. (Ⅱ) (메탄올 용매에서 산소 첨가된 네자리 Schiff Base Cobalt(II) 착물들의 활성촉매에 의한 Hydrazobenzene의 산화반응 (제 2 보))

  • Dioxygen binding and homogeneous catalytic oxidation of hydrazobenzene were investigated by employing tetradentate Schiff base Cobalt(II) complexes such as Co(II)(SED)$(Py)_2$, Co(II)(SOPD)$(Py)_2$ and Co(II)(SND)$(Py)_2$ in saturated oxygen methanol solvent. The major product of hydrazobenzene ($H_2$AB) oxidation by catalysts of superoxo type [Co(III)(SED)(Py)$O_2$] and [Co(III)(SOPD)(Py)$O_2$] complexes are trans-azobenzene (t-AB) and rate constants k for oxidation reaction was 7.692 ${\times}$ $10^{-2}$ M/sec for [Co(III)(SED)(Py)$O_2$] and 5.076 ${\times}$ $10^{-2}$ M/sec for [Co(III)(SOPD)(Py)$O_2$]. But cis-azobenzene (c-AB) are obtained as a major product with ${\mu}$-peroxo type [Co(III)(SED)(Py)]$_2O_2$ catalyst, and rate constant k is 1.266 ${\times}$ $10^{-2}$ M/sec. The rate constants of oxidation reaction has been studied spectrophotometrically and the rate law established. A mechanism involving a intermediate activated complexes of catalyst, hydrazobenzene and oxygen has been proposed. $H_2$AB + Co(II)(Schiff base)$(Py)_2$ + $O_2$ ${\rightleftharpoons}_{MeOH}^K$ Co(III)(Schiff base)(Py)$O_2$${\cdot}$$H_2$AB + Py $\longrightarrow^k$ Co(II)(Schiff base)$(Py)_2$ + t-AB + $H_2O_2$(Scchiff base : SED and SOPD). $H_2$AB + 2Co(II)(SND)$(Py)_2$ + $O_2$ ${\rightleftharpoons}_{MeOH}^K$ [Co(III)(SND)(Py)]$_2O_2$${\cdot}$H_2$AB + 2Py ${\longrightarrow}^k$ (Co(II)(SND)$(Py)_2$ + c-AB + $H_2O_2$.

Preparation of graphene oxide incorporated polyamide thin-film composite membranes for PPCPs removal

  • Incorporating nano-materials in thin-film composite (TFC) membranes has been considered to be an approach to achieve higher membrane performance in various water treatment processes. This study investigated the rejection efficiency of three target compounds, i.e., reserpine, norfloxacin and tetracycline hydrochloride, by TFC membranes with different graphene oxide proportions. Graphene oxide (GO) was incorporated into the polyamide active layer of a TFC membrane via an interfacial polymerization (IP) reaction. The TFC membranes were characterized with FTIR, FE-SEM, AFM; in addition, the water contact angle measurements as well as the permeation and separation performance were evaluated. The prepared GO-TFC membranes exhibited a much higher flux ($3.11{\pm}0.04L/m2{\cdot}h{\cdot}bar$) than the pristine TFC membranes ($2.12{\pm}0.05L/m2{\cdot}h{\cdot}bar$) without sacrificing their foulant rejection abilities. At the same time, the GO-modified membrane appeared to be less sensitive to pH changes than the pure TFC membrane. A significant improvement in the anti-fouling property of the membrane was observed, which was ascribed to the favorable change in the membrane's hydrophilicity, surface morphology and surface charge through the addition of an appropriate amount of GO. This study predominantly improved the understanding of the different PA/GO membranes and outlined improved industrial applications of such membranes in the future.

Pre-Monsoon Dynamics of Zooplankton Community in the Yongwol West River Tributary (갈수기 영월 한반도지형 습지 주변 유수역의 동물플랑크톤 군집 동태)

  • Pre-monsoon dynamics of zooplankton community were investigated in Yongwol West River tributary; wetland shaped Korean Peninsula. Zooplankters were collected at three sites (stn 1: Yongwol West River, stn 2: Pyungchang River, stn 3: Jucheon River) monthly during the period between March and July 2014 except April. Totally 58 taxa of zooplankton occurred, consisting of 27 species of rotifer, 18 species of cladoceran, eight species of copepod, four kinds of aquatic insect larvae and one nematod. Lotic cladocerans of Alona spp. belonging to Chydoridae occurred frequently while typical lentic ones of Bosmina spp. and Daphnia spp. were not distributed, then sampling sites may be located in lotic waters. Zooplankton abundance recorded as $4,451-8,011indiv{\cdot}m^{-3}$ in Jucheon River but never exceeded $500indiv{\cdot}m^{-3}$ in other rivers. In Yongwol West River, dominant taxa were aquatic insect larvae from March through June and succeeded to cladocerans and copepods in July. Similar succession was observed in Jucheon River where aquatic insect larvae dominated in March and changed to cladocerans and copepods from May with high abundance. No such succession of zooplankton community was detected in Pyunchang River. Species diversity indices were the highest in Yongwol West River except in March. High values of dissolved oxygen ($9.0-11.0 mg{\cdot}liter^{-1}$) may show the characteristics of mountain streams. Water temperature increased gradually from $10.1^{\circ}C$ to $27.9^{\circ}C$ and pH varied within generally high range between 7.9-8.9, respectively. High value of pH seems to be derived from lime stone mines and cement factories around the studied area.

Synthesis of zinc oxide nanoparticles via aqueous solution routes (수용액 합성법에 의한 ZnO 나노분말의 합성)

  • ZnO nanoparticles were synthesized by aqueous preparation routes of a precipitation and a hydrothermal process. In the processes, the powders were formed by mixing aqueous solutions of Zn-nitrate hexahydrate ($Zn(NO_3)_2{\cdot}6H_2O$) with NaOH aqueous solution under controlled reaction conditions such as Zn precursor concentration, reaction pH and temperature. Single ZnO phase has been obtained under low Zn precursor concentration, high reaction pH and high temperature. The synthesized particles exhibited flakes (plates), multipods or rods morphologies and the crystallite sizes and shapes would be efficiently controllable by changing the processing parameters. The hydrothermal method showed advantageous features over the precipitation process, allowing the precipitates of single ZnO phase with higher crystallinity at relatively low temperatures below $100^{\circ}C$ under a wider pH range for the Zn precursor concentration of 0.1~1 M.
