Study on Relaxing Effect of Oxybutynin on the Contractile Response of Arterial Smooth Muscle (동맥근 수축에 대학 Oxybutynin의 이완효과에 관한 연구)

  • Pharmacological actions of an antispasmodic agent, oxybutynin were investigated in the isolated procine coronary arteries. The coronary rings were contracted by acetylcholine (ACh) and KCl in a dose-dependent fashion. The ACh-induced contractions were signifcantly potentiated by removal of endothelium and $EC_{50}=0.52\;{\mu}M$ of intact endothelial rings was about 2 times greater than $EC_{50}=0.28\;{\mu}M$ of rings without the endothelium. These results suggest that the endothelium plays an inhibitory role in ACh-induced contraction. Oxybutynin and atropine inhibited dose-dependently $1.0\;{\mu}M$ ACh-induced contraction and atropine inhibited dose-dependently $1.0\;{\mu}M$ ACh-induced contraction and the $IC_{50s}$ were 11.0 nM and 0.47 nM, respectively. Atropine did not affect 35 mM KCl-induced contraction but oxybutynin inhibited the contraction to the basal tension in a dose-dependent manner. The $IC_{50}$ of oxybutynin on the KCl-induced contraction was $49.7\;{\mu}M$. The dose-response curve to ACh was parallelly shifted to the right by pretreating coronary rings with $IC_{50}$ of atropine (0.47 nM) or oxybutynin (11.0 nM) but the curve to KC1 was rightward shifted in a noncompetitive manner under pretreatment with $IC_{50}$ of oxybutynin $(49.7\;{\mu}M$). Oxybutynin inhibited $0.1\;{\mu}M$ Bay K 8644-induced contraction to the basal tension in a dose dependent manner, but $35\;{\mu}M$ histamine-induced contraction was inhibited to only 50e/e of the original level even in maximal concentration $(5{\times}10^{-4}M)$ of oxybutynin. These results suggest that oxybutynin causes antispasmodic action through sensitive blocking action on muscarinic receptors and inhibitory action on calcium influx in the procine coronary artery.

An Analysis of the Impacts of FDI Types on CO2 Emissions - Focus on Scale Effects and Technique Effects - (FDI 유형별 CO2 배출량에 미치는 영향 분석 - 규모효과와 기술효과를 중심으로 -)

  • The purpose of this study is to determine whether there is a significant difference in impacts decomposed into scale effects and technique effects on $CO_2$ emissions between Greenfield FDI and M&A FDI flows into Korean manufacturing sectors, ultimately leading to clarify the relationship between FDI and environmental pollution. To this end, the research constructed a simultaneous model to analyze coincidental relationship of influence and interactions between each variable. Archival data, spanning the 15 years period from 1995 to 2009, is industry-level panel data on 13 Korean manufacturing sectors, and it is empirically analyzed with three-stage least squares (3SLS) method. Key findings can be summarized into two parts. First of all, Greenfield FDI has a greater impact on increasing industrial gross output, resulting in more $CO_2$ emissions than M&A FDI through scale effects. Secondly, technique effects of FDI have a bigger impact on $CO_2$ emissions than scale effects, implying that this inflow of FDI into Korea contributes positively to the reduction of $CO_2$ emissions. These findings are expected to play a meaningful role in establishing FDI policies with consideration of the environment by giving the implication that different incentives for each FDI type should be considered to maximize the effect of environmental protection.

Estimation of Appropriate Infiltration Rate and the Effects of the Flowerbed Type Infiltration System (화단형 침투시설의 단위설계침투량 산정 및 효과분석)

  • This study developed a flowerbed type infiltration system that could control the amount of runoff discharge under a certain level estimated its proper design infiltration rate, and analyzed the effects of its implementation. Analyzing the performance of infiltration system is the one of the essential processes that should be under review to predict its effects after implementation when a rainwater infiltration system is included in a district-based plan. To estimate the unit design infiltration rate of this system, the runoff decrease effect was analyzed by varying the unit infiltration rate of the system applied to the parking lot adjacent to the Korea Institute of Construction Technology laboratory building by using a water balance analysis program. After varying the unit design infiltration to $0.1{\sim}3m^3/m^2.day$ to analyze the variation in the rate of runoff, 80% of the runoff was infiltrated at $1.0m^3/m^2.day$, and the unit infiltration design rate at the time was 0.0416(m3/m2.hr). It was also found that the unit design infiltration rate obtained from a field infiltration test of the developed system was about $0.045m^3/hr$. Based on this study, it was possible that infiltration rate is estimated to consider the economic scale and environmental effect. It is significant to apply the spatial plan of rainwater infiltration system as green infrastructure.

Antiproliferative and Antioxidative Activities of Methanol Extracts of Echinacea angustifolia (Echinacea angustifolia 메탄올 추출물의 암세포 증식억제 및 항산화 효과)

  • Echinacea, also blown as the purple coneflower, is a herbal medicine that has been used for centuries, customarily as a treatment for the common cold, coughs, bronchitis, upper respiratory infections, and some inflammatory conditions. We investigated the effects of methanol extracts of Echinacea angustifolia on the cytotoxicity against cancer cells $(HepG_2,\;3LL,\;HL60,\;L1210)$ and antioxidative activity. From the test results, each part of Echinaceashowed a cytotoxic effect against the cancer cell lines, and this cytotoxic effect increased with increasing sample concentration. At 1.0 mg/mL concentration the relative cytotoxic activities of the flower bud, leaf, stern and root parts were $90.5\%,\;52.7\%,\;37.1\%\;and\;19.2\%$, respectively, in $HepG_2$ cells, and $75.5\%,\;93.3\%,\;81.2\%,\;and\;75.1\%$ respectively, in HL60 cells, as evaluated by MTT assay. $IC_{50}(50\%\;inhibitory\;concentration)$ of the methanol extracts of the Echinacea flower bud was 0.214 mg/mL on /$HepG_2$ cells, and that of the Echinacea leaf and root was 0.166 mg/mL and 0.210 mg/mL, respectively, on HL60 cells. After /$HepG_2$ cells were incubated for 6 days at $37^{\circ}C$ with various concentrations of each part, the cell number increased while the inhibition rate on the /$HepG_2$ cell growth decreased. The antioxidative activities of the flower bud, leaf, stem and root parts were $59.0\%$ (0.75 mg/mL), $80.76\%$ (0.5 mg/mL), $95.5\%$ (0.25mg/mL) and $98.15\%$ (0.25 mg/mL), respectively, as evaluated by electron donating ability. These results indicated that Echinacea angustifolia has strong anticancer and antioxidative effects in vitro.

Antimicrobial Activity of Houttuynia cordata Ethanol Extract against Major Clinical Resistant Microorganisms (주요 임상 내성균에 대한 어성초 에탄올 추출물의 항균효과)

  • The increase in resistance by pathogenic bacteria to multiple antimicrobial agents has become a significant treat, as the effective antimicrobial agents available for the patients infected by such resistant bacteria are reduced, or even eliminated. Several natural plant extracts have exhibited antibacterial and synergistic activity against various resistant microorganisms. Houttuynia cordata is frequently used by many traditional medicine practicioners for its antimicrobial, antiviral, and anti-inflammatory properties. This study investigated the antibacterial effects of H. cordata extract against clinical multi-resistant bacteria, and compared the two methods used for the antimicrobial susceptibility testing. Thirty isolates of Methicillin-resistant Staphylococcus aureus (MRSA, 10), Vancomycin-resistant Enterococcus faecium (VRE, 10), Carbapenem-resistant Acinetobacter baumannii (CRAB, 10) were included in this study. The antibacterial effect of H. cordata was tested by disk diffusion and microbroth dilution methods as per CLSI guidelines. In disk diffusion, all isolates (30) showed no inhibition to 30,000 ug/mL of H. cordata. But in the microbroth dilution method, $MIC_{90}$ of H. cordata was 4,096 ug/mL, 8,192 ug/mL and 4,096 ug/mL in MRSA, VRE and CRAB, respectively. These results demonstrate that H. cordata exhibits antibacterial activity against MRSA, VRE and CRAB. Moreover, the microbroth dilution method is a more effective method than disk diffusion to evaluate the antibacterial activity of natural products. The Disk diffusion method used to evaluate the antibacterial activity of natural products required new standard guidelines including inoculum concentration of bacteria.

An Empirical Study on the Improvement of In Situ Soil Remediation Using Plasma Blasting, Pneumatic Fracturing and Vacuum Suction (플라즈마 블라스팅, 공압파쇄, 진공추출이 활용된 지중 토양정화공법의 정화 개선 효과에 대한 실증연구)

  • The in-situ remediation of a solidified stratum containing a large amount of fine-texture material like clay or organic matter in contaminated soil faces limitations such as increased remediation cost resulting from decreased purification efficiency. Even if the soil conditions are good, remediation generally requires a long time to complete because of non-uniform soil properties and low permeability. This study assessed the remediation effect and evaluated the field applicability of a methodology that combines pneumatic fracturing, vacuum extraction, and plasma blasting (the PPV method) to improve the limitations facing existing underground remediation methods. For comparison, underground remediation was performed over 80 days using the experimental PPV method and chemical oxidation (the control method). The control group showed no decrease in the degree of contamination due to the poor delivery of the soil remediation agent, whereas the PPV method clearly reduced the degree of contamination during the remediation period. Remediation effect, as assessed by the reduction of the highest TPH (Total Petroleum Hydrocarbons) concentration by distance from the injection well, was uncleared in the control group, whereas the PPV method showed a remediation effect of 62.6% within a 1 m radius of the injection well radius, 90.1% within 1.1~2.0 m, and 92.1% within 2.1~3.0 m. When evaluating the remediation efficiency by considering the average rate of TPH concentration reduction by distance from the injection well, the control group was not clear; in contrast, the PPV method showed 53.6% remediation effect within 1 m of the injection well, 82.4% within 1.1~2.0 m, and 68.7% within 2.1~3.0 m. Both ways of considering purification efficiency (based on changes in TPH maximum and average contamination concentration) found the PPV method to increase the remediation effect by 149.0~184.8% compared with the control group; its average increase in remediation effect was ~167%. The time taken to reduce contamination by 80% of the initial concentration was evaluated by deriving a correlation equation through analysis of the TPH concentration: the PPV method could reduce the purification time by 184.4% compared with chemical oxidation. However, the present evaluation of a single site cannot be equally applied to all strata, so additional research is necessary to explore more clearly the proposed method's effect.

Inhibitory Effects of Ginger and Processed (Beopje) Ginger Extracts on HCl-ethanol Induced Gastritis in Rats (염산-에탄올에 의해 유발된 흰쥐 위염에 대한 생강 및 법제생강 추출물의 억제작용)

  • This study examined the protective effects of ginger and processed (Beopje) ginger extracts on gastritis induced by HCl-ethanol in SD-rats. Beopje (or Poje) is a branch of herbal medicines processed using a Korean traditional method to achieve specific pharmacological effects. Gastric lesions were induced in the rats by a treatment of 1 mL of HCl-ethanol (60% ethanol+150 mM HCl). The rats were divided into seven groups: Normal (1 mL of saline without HCl-ethanol treated group), Control (HCl-ethanol treated group), GL (35 mg/kg of ginger treated group), BGL (35 mg/kg of Beopje ginger treated group), GH (350 mg/kg of ginger treated group), BGH (350 mg/kg of Beopje ginger treated group) and Cimetidine (80 mg/kg of cimetidine treated group). The gastric injury inhibition rate was 40.2% and 64.9% in GL and BGL and 68.4% and 99.6% in GH and BGH respectively, showing significantly lower rates than the control (p<0.05). The level of gastric juice secretion decreased significantly in all ginger administered groups. The pH of the gastric juices of BGH increased and the acidity of BGH and cimetidine decreased significantly (p<0.05) compared to the other groups. Beopje ginger had stronger inhibitory effects on gastritis than ginger without the Beopje process. The protective effect on gastritis by the ginger and Beopje ginger extracts increased in a dose-dependent manner (p<0.05). These results suggest that ginger has inhibitory effects on HCl-ethanol induced gastritis in rats that can be improved through the Beopje process.

Effect of phosphorous acid on control of phytophthora blight of red-pepper and tomato, and downy mildew of cucumber in the greenhouse (온실에서 고추.토마토 역병 및 오이 노균병에 대한 아인산(phosphorous acid)의 방제 효과)

  • Control effects of phosphorous acid were investigated on three diseases. For Phytophthora blight of red pepper, protective and curative effects of phosphorous acid at the concentration of $1,408{\mu}g$ a. i./mL were 91.0% and 80.0%, respectively. In case of late blight of tomato, caused by Phytophthora infestans, protective and curative effects were 63.4% and 13.0% at the same concentration, respectively. However, the protective and curative effects of phosphorous acid increased by decreasing inoculum density of Phytophthora infestans. The protective effects of phosphorous acid on control of Phytophthora blight of red pepper was persisted for 4 days with high control efficacy (94.0%). The protective and curative effects of phosphorous acid ($1,408{\mu}g$ a. i./mL) on cucumber downy mildew were 82.0% and 62.0% respectively. The foliar application of phosphorous acid also promoted shoot growth and fresh weight of red pepper.

Antimicrobial Effect of Photodynamic Therapy Using Plaque Disclosing Agent (치면착색제를 이용한 광역동 치료의 항균 효과)

  • The aim of this study was to evaluate the antimicrobial effect of photodynamic therapy (PDT) using plaque disclosing agent, 10 - 20 mM erythrosine, as a photosensitizer. Multispecies cariogenic biofilms containing Streptococcus mutans, Lactobacillus casei and Candida albicans were formed on hydroxyapatite disc. 20 μM, 10 mM and 20 mM erythrosine were applied as a photosensitizer for 3 minutes, and then light-emitting diode (LED) irradiated for 24 seconds. Colony-forming unit (CFU) were measured and biofilms were observed using confocal laser scanning microscopy (CLSM). CFU were significantly decreased in the PDT groups using 10 - 20 mM erythrosine (10 mM, 20mM) and the results were also confirmed by CLSM. This study confirms the high antimicrobial effect of photodynamic therapy using plaque disclosing agent as a photosensitizer.

  • In Korea. the cultivating standard of stevia is not established yet. Because stevia is across-fertilizing plant. planting a cutting is its available propagation method. This study was made to know the effective propagation growth and adaptability of stevia in Jeju-do, according to the wintering effect and the planting density.
