  • This study was designed to investigate the antioxidative activity on oxidation of human low density lipoprotein(LDL) of band 2 fractionated from culture broth of Streptomyces sp. BH-405. Antioxidative activity of band 2 obtained from fractionation of BH-405 culture purification was measured against $Cu^{2+}$ mediated human LDL oxidation by thiobarbituric acid reactive substance. $CuSO_4$ mediated oxidation of LDL was degraded at a much higher rate than native LDL. Band 2 at a concentration of 100 or 200 !lg/mL inhibited the oxidation of LDL induced by $CuSO_4$, The formation of conjugated dienes induced in the presence of 5 !1M CuS04 of the mouse macrophage and J744. The electrophoretic mobility of the LDL in addition of $200\mu\textrm{g}$ band 2 in the presence of $5\mu\textrm{m}$ $CuSO_4$ was lower than that of native LDL. LDL modified by copper mediated or cell mediated uptake was degraded by macrophage at much greater than native LDL, and band 2 was found as potential inhibitor of modification of 125I-labelled LDL by macrophage. phage.

A Study on Construction of Roundabouts considering the Effects for Adjacent Intersections in Urban Network (인접교차로 영향을 고려한 회전교차로 도입방안 연구)

  • Though many studies regarding roundabout have been recently conducted, most of them have focused on operational aspect. Moreover, majority of the previous researches analyzes operational effects of single roundabout, but seldom investigate the effects of multi-roundabouts constructed on road networks. In this study, we seek ways to construct multiple roundabouts on road network maximizing their operational effects. The analysis investigate influence of both adjacent signalized and unsignalized intersections as well as influence of the distance from those intersections to roundabouts. The results show that the optimal distance between two adjacent intersections were calculated to be 150m, and any two intersections located within 150m apart influence each other thus imposing operational restrictions on each other. In addition, those results are confirmed using simulation analysis conducted on the real urban network in Nonheon regional area, Incheon City.

  • Nosema spp. (NS) causes severe problems to honey bee colonies including the death of the highly infected ones. Searching for effective materials to control this parasite is very important. The objectives of this study were to identify the calendar for the prevalence of NS and other bee diseases, and to test the efficacy of three materials: diluted honey mixed with lemon juice (M1), chamomile extract mixed with sugar syrup (M2) and sutrivet mixed with sugar syrup (M3) against Nosema. To realize these objectives, diseases of brood and adult honey bees were surveyed over one year. Also, the efficacy of M1, M2 and M3 against Nosema was evaluated under field and laboratory conditions. The results showed that few diseases for immature and mature stages of honey bees were recorded. NS was detected during winter and spring in link with low temperature and high relative humidity. Under field conditions, M2 reduced the infection by 36.66% while M3 by 23.33% and finally M1 by 13.33%. In the laboratory, the highest efficacy was to M2 followed by M1 and finally M3. The three materials impacted the percentage of survived bees significantly higher than infected bees without any treatments over the experimental period. The study suggests the potential role of chamomile as a natural material to control NS.

Antimutagenic and Cytotoxicity Effects of Agaricus blazei Murill Extracts (아가리쿠스버섯(Agaricus blazei Murill) 추출물의 항돌연변이원성 및 세포독성 효과)

  • This study was performed to determine the antimutagenic and cytotoxic effect of Agaricus blazei Murill methanol extract on Salmonella typhimurium TA98, TA100 and human cancer cell lines using Ames test and cytotoxicity assay, respectively. In Ames test, methanol extract from A. blazei Murill did not exhibit any mutagenicity and most of the samples showed high antimutagenic effects against mutation induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG), 4-nitroquinoline-1-oxide(4NQO), 3-amino-1,4-dimethyl-5H-pyrido [4,3-b] indol(Trp-P-1) and $benzo({\alpha})pyrene(B({\alpha})P)$. The methanol extracts of A. blazei Murill$(200\;{\mu}g/plate)$ showed approximately 92.4%, 81.9% and 83.4% inhibitory effect on the mutagenesis induced by 4NQO, Trp-P-1 and $B({\alpha})P$ against TA98 strain, whereas 87.3%, 94.7%. 92.3% and 89.9% inhibitions were observed on the mutagenesis induced by MNNG, 4NQO, Trp-P-1 and $B({\alpha})P$ against TA100 strain. The solvent fractions of methanol extracts from A. blazei Murill except water fraction showed high antimutagenic effects of $70{\sim}90%$ against mutation induced by MNNG, 4NQO. Trp-P-1 and $B({\alpha})P$. In anticancer effects of A. blazei Murill extract and fraction against cancer cell lines including human breast adenocarcinoma(MCF7), human lung carcinoma(A549), human fibrosarcoma(HT1080), human hepatocellular carcinoma(Hep3B), human epitheloid carcinoma(HeLa), human gastric carcinoma(KATO III) and human chronic myelogenous leukemia(K562) were investigated. The treatment of 1 mg/mL A. blazei Murill extracts had the highest cytotoxicity with 91.9% against HeLa, followed by KATO III(88.7%), A549(86.5%) and Hpe3B(84.3%). Whereas 1 mg/mL treatment of A. blazei Murill extracts had only $10{\sim}40%$ cytotoxicity on human normal liver cell (WRL68).

Anti-oxidative Effect of Chungsimyeonja-um (CSYJE) via Nrf2/HO-1 Pathway Activity in Lipopolysaccharide (LPS) Induced RAW 264.7 Macrophages (대식세포에서 Nrf2/HO-1경로를 통한 청심연자음의 항산화효과)

  • Reactive oxygen species (ROS) plays an important role in maintaining homeostasis. However, excessive ROS production damages cellular components such as proteins, lipids, and nucleic acids and promotes skin aging. In this study, we confirmed the antioxidant effect of CSYJE to prevent excessive oxidative stress. First, DPPH and ABTS assays were performed to confirm the antioxidant effect of CSYJE and the radical scavenging activity was confirmed depending on the concentration. As a result of performing the MTT assay to confirm the cell viability, it was confirmed that there was no cytotoxicity at a concentration of 1,000 ㎍/mL. As a result of western blotting to confirm the expression levels of the antioxidant-related proteins nuclear-E2-related factor 2 (Nrf2) and Heme oxygenase-1 (HO-1), it was confirmed that the expression was increased in a concentration-dependent manner. After inducing ROS with lipopolysaccharide (LPS), an intracellular ROS-causing substance, DCF-DA was performed to confirm the inhibitory effect of ROS production, and the inhibition of ROS production was confirmed to concentration-dependent. Real-time RT-PCR was performed to confirm the mRNA expression level of inflammatory cytokines and inflammatory mediator caused by ROS generation, mRNA expression was reduced in a dose dependent manner. Therefore, this study confirmed the antioxidant effect of CSYJE through the Nrf2/HO-1 signaling pathway, which suggests that CSYJE can be used as an antioxidant cosmetic material by inhibiting free radicals.

Total Phenolics of Dried Platycodon grandiflorum and Its Effect on Growth of Human Cancer Cell Lines (건조 도라지 추출물 및 분획물의 총 페놀계 화합물 함량 및 인체 암세포 증식 억제효과)

  • We investigated the effects of extracts from dried Platycodon grandiflorum on total phenolic content and growth of cancer cell lines (HT-29 and AGS). Total phenolic contents of acetone/methylene chloride (A+M) extract and methanol (MeOH) fraction were 4.53 and 27.22 TAE mg/100 g, respectively. Among the fractions, n-butanol (n-BuOH) fraction contained the highest phenolic content. Treatments of crude extracts and fractions significantly inhibited the growth of HT-29 and AGS cancer cell lines (p<0.05). Among the fractions, n-BuOH fraction exhibited the highest inhibitory effect and was then sub-fractionated by reverse phase flash column chromatography (rfc). The rfc 1-3 exhibited a higher inhibitory effect on proliferation of both cancer cells. The rfc 1 contained the highest phenol content. Our results showed that n-BuOH fraction possessed a potent inhibitory effect on proliferation of human cancer cells. We suggested that this anticancer activity was partially related to the content of phenolic compounds.

Effects of Scutellaria baicalensis Extracts on Tyrosinase Gene Expression in B16 Melanoma Cells (B16 Melanoma Cell에서 티로시나아제 유전자 발현에 황금(Scutellaria baicalensis) 추출물이 미치는 효과)

  • To estimate the regulatory effects of Scutellaria baicalensis extracts on melanin biosynthesis, we evaluated the regulatory effects of the tyrosinase gene on B16 melanoma cells. The results revealed that methanolic extracts of Scutellaria baicalensis resulted in a high increase in the expression of the tyrosinase gene. Specifically, treatment with extracts at concentrations of $10\;{\mu}g/m{\ell}$ and $100\;{\mu}g/m{\ell}$ resulted in increases in tyrosinase gene expression rates of approximately 231% and 256%, respectively, when compared to the control. Moreover, the solvent fraction layers(methylene chloride, ethyl acetate, butyl alcohol, water) improved the expression of the tyrosinase gene, but to a lesser degree than the methanolic extracts. An MTT assay revealed, that the methanolic extract exhibited very low cytotoxicities at $10\;{\mu}g/m{\ell}$ and $100\;{\mu}g/m{\ell}$. Taken together, the results of this study indicated that the methanolic extracts of Scutellaria baicalensis was a very effective positive regulator of tyrosinase gene expression.

Physicochemical properties and anti-wrinkle effect of polysaccharides with different molecular weights from Gloiopeltis furcata (불등풀가사리 다당류의 분자량에 따른 이화학적 품질특성 및 피부 주름 개선 효과)

  • In this study, the physicochemical properties and anti-wrinkle effect of polysaccharides with different molecular weights from Gloiopeltis furcata were investigated. Crude polysaccharides were isolated by viscozyme treatment followed by ethanol precipitation and lyophilization. Crude polysaccharides were hydrolyzed by acid (0.1 N HCl) and the molecular weight fractions were generated by centrifugal filter (<10 kDa, 10 to 100 kDa, and 100 kDa>). The yield of polysaccharides with different molecular weight fractions was 8.4-39.6%. The major constituents in molecular weight fractions were total sugar (81.37-85.82%), uronic acid (27.89-32.85 g/100 g), sulfate (33.38-39.04%), and protein (0.35-3.16%) The L, a, and b value of the 100 kDa group were decreased, but viscosity increased. The oxygen radical absorbance capacity of the 100 kDa group at $180.07{\mu}M$ was the highest among groups. The protective effects of 100 kDa group at 0.5 and $5{\mu}g/mL$ against $H_2O_2$-induced cytotoxicity in L132 cell were 87.34% and 103.85%, respectively. The matrix metalloproteinase-1 activity of 100 kDa group decreased in a dose-dependent manner. The pro-collagen synthesis activity of 100 kDa group at $0.05-0.5{\mu}g/mL$ was 64.91-77.80%. The polysaccharides with different molecular weights from Gloiopeltis furcata investigated herein are useful as a potential candidate for cosmedical materials.

Protective Effect of Garlic (Allium sativum L.) Extracts Prepared by Different Processing Methods on DNA Damage in Human Leukocytes (마늘의 조리방법에 따른 DNA 손상 보호 효과의 비교)

  • DNA damage including base modifications, loss of base and breaks in DNA strands can occur by exposure to irradiation, smoking and several components of food. Unrepaired DNA damage is known to lead to cellular dysfunction, cell death, cancer, and other diseases such as arteriosclerosis and diabetes. The protective effect of garlic on oxidative stress induced DNA damage has been reported recently. In this study, we investigated the protective effect of garlic extracts prepared by different processing methods (raw garlic extracts, RGE; grilled garlic extracts, GGE; pickled garlic extracts, PGE) on leukocytic DNA damage using comet assay. Human leukocytes were incubated with ethanol and methanol extract of garlic at various concentrations (1, 5, 10, 50 ${\mu}g$/mL), followed by oxidative stimuli (200 ${\mu}M$ $H_2O_2$ or 200 ${\mu}M$ 4-hydroxynonenal (HNE)). The methanol and ethanol extracts of RGE, GGE, and PGE showed inhibitory activities of DNA damage induced by $H_2O_2$ or HNE. Especially methanol extract of RGE ($ED_{50}$; 13.3 ${\mu}g$/mL) had a higher antigenotoxic effect on $H_2O_2$ induced DNA damage than those of GGE (23.5 ${\mu}g$/mL) or PGE (24.5 ${\mu}g$/mL). HNE induced DNA damage tended to be effectively inhibited by the lower concentration of all garlic extracts. Therefore, garlic might have protective effects against oxidative DNA damage regardless of processing methods (raw, grilled, pickled) which are the general consumed forms of garlic in Korea.

Inhibitory Effects of Garlic Oil on Human Low Density Lipoprotein Oxidation

  • Growing evidence indicates that oxidized low density lipoprotein (LDL) may promote atherogenesis. Therefore, inhibition of LDL oxidation may impede this process. The inhibitory effected on the susceptibility of human LDL to $Cu^{2+}$ or macrophages induced oxidation was investigated by monitoring thiobarbituric acid reactive substances(TBARS). Organosulfur compounds of garlic oil contains diallyldisulfide, diallyltrisulfide, diallyltetrasulfide, and diallyl pentasulfide in order. Garlic oil inhibited LDL oxidation by $Cu^{2+}$, or macrophages in a dose dependently, with a 20~60 ${\mu}g$, as increased TBARS assay. Garlic oil, at 60 ${\mu}M$, almost completely inhibited macrophages induced increase in electrophoretic mobility of LDL. When compared with several other antioxidants, probucol showed highest ability, and then garlic oil showed a much higher ability than natural occurring antioxidants, ${\alpha}$-tocopherol and ascorbic acid. The results suggested that garlic oil might play the inhibitory effects in the process of LDL oxidation.
