Study on the Enzyme of Basidiomycetes(I) -The Effects of Iron Ions on the Light-Induced Mitochondrial $F_0F_1-ATPase$ of Lentinus edodes- (담자균류의 효소에 관한 연구(I) -표고버섯 중의 광감응성 Mitochondrial $F_0F_1-ATPase$의 철이온 효과-)

  • The effects of the iron ions for the light-induced mitochondrial $F_0F_1-ATPase$ of Lentinus edodes was studied. The enzyme activity was stimulated up to 202% by 0.1 mM $Fe^{2-}$ ion, but was inhibited by $Fe^{3+}\;and\;Mg^{2+}$. In the presence of 0.5 mM $Mg^{2+}$, the activity also increased 32% by 0.1 mM $Fe^{2+}$ ion, and decreased to a similar extent by $Fe^{3+}$ ion than by only $Fe^{3+}$ ion. Also, the activity was inhibited 53% by 5.0 mM $Fe^{2-}$ ion in the presence of 0.5 mM $Mg^{2+}$ ion and various concentration of $Fe^{3+}$ ion(mM). These results showed that $Fe^{2+}$ strongly stimulated the enzyme activity and its role for the enzyme was independent of $Mg^{2+}$ ion, but was dependent of $Fe^{3+}$ ion. From inactivation of the enzyme by addition of metal chelating agent, EDTA, it is suggested that the enzyme is to be metalloenzyme. The optimal pH and temperature of the enzyme in the presence of 0.1 mM $Fe^{2+}$ was 7.6 and $63^{\circ}C$, respectively.

A Study on Difference in Coercivity between Substituted Barium Ferrite Powder and Its Tape (자기기록용 치환형 Ba-Ferrite 분말과 Tape의 보자력 차이에 관한 연구)

  • A coercivity of barium ferrite (BaM) tape was found to be higher than the coercivity of BaM powder, This difference is in the range of 50 Oe to 600 Oe, which depends on substituted cations. physical properties of powder, and preparation conditions of the tape. The coercivity difference is attributed to both particle stacking and adsorption of organic solvent on particle surface. Regardless of substituted cations, the coercivity of longitudinally oriented BaFe tape was higher than that of BaM powder by a range of 100 Oe to 120 Oe, which is caused by particle stacking' During the active adsorption process on preparation of magnetic paint, where MEK (methyl ethyl ketone), TOL (toluene) and CHO (cyclohexanone) were used, chemisorption of solvents on the surface of substituted BaFe particle occurred to form polymeric compounds surrounding the particle. An increase in coercivity, caused by the solvent adsorption, was significant for Co-substutited BaM tape. Among these solvents CHO was the most effective one in increasing the coercivity of the Co-Ti substituted BaM tape.

Antimicrobial Activities of Ethosome-Encapsulated Palmitoyl Tripeptide (Ethosome에 캡슐화된 Palmitoyl Tripeptide의 항균효과)

  • Palmitoyl tripeptide (M330) showed higher antimicrobial activities than methyl paraben or phenoxy ethanol through minimum inhibitory concentration (MIC) test. However, when the M330 was added into cosmetic formulation, white precipitates formed due to the electrostatic interaction between M330 and carbopol (carboxy vinyl polymer) as a thickener in cosmetics, and the viscosity of cosmetics decreased sharply. Also, the antimicrobial activities of M330 in cosmetics became lower than those of methyl paraben or phenoxy ethanol. Thus, the encapsulation of M330 in ethosome vesicle was attempted in order to recover the declined antimicrobial activities of M330 in cosmetics and prevent the precipitates from forming. When ethosome-encapsulated M330 was added into cosmetics, the precipitates did not form, and the decrease in the viscosity of cosmetics was not large compared to the addition of unencapsulated M330. Challenge tests showed that antimicrobial activities against gram negative bacteria were improved by the encapsulation of M330, but the encapsulation was not effective against gram positive bacteria and fungus. A combination of M330 with EDTA showed synergistic inhibitory potential against C. albicans. After coencapsulation of M330 and EDTA in ethosome, antimicrobial activities proved to be higher than those of unencapsulated M330 and EDTA.

Anticancer Effects of Cisplatin in Combination with Paeonia Japonica in YD-10B Cells (YD-10B에서 Cisplatin과 백작약의 병용처리에 의한 항암 효과)

  • The present study investigated the anti-proliferate and anti-invasive of Phorbol 12-myristate 13-acetate (PMA)-induced matrix metalloproteinase (MMP-2) and MMP-9 activities of combined treatment with cisplatin and ethyl acetate fractions of Paeonia japonica. Cell Proliferation was detected by the MTS assay and the activity and mRNA expression of MMP-2/-9 were examined by zymography and RT-PCR. As results, cisplatin or p. japonica treatment of YD-10B cells resulted in a dose-dependent inhibition of cell growth. Also, the viability of YD-10B cells treated with combination of 200 μM cisplatin and 50 ㎍/ml p. japonica was inhibited to 50% in compared with the cisplatin alone. In PMA-treated YD-10B cells, co-treatment of 200 μM cisplatin with 50 ㎍/ml p. japonica significantly inhibited mRNA expression and protein activation of MMP-2/-9. Therefore, This study suggest that the combination treatment of cisplatin and p. japonica potentiates a promising anti-invasive agent and has more potential anti-cancer drug for oral cancer therapy than cisplatin alone.

Effect of Green Tea Extract on Cisplatin- or Doxorubicin-Induced Cytotoxicity in Human Lung Cancer Cell Lines (사람 폐암 세포주에서 시스플라틴이나 독소루비신의 세포독성에 미치는 녹차 추출물의 영향)

  • Tea extract (TE) has been shown to have anti-tumor properties in a wide variety of experimental systems. We evaluated green tea extract (GTE) as a biochemical modulator for the antitumor activity of cisplatin and doxorubicin in the treatment of human lung cancer A549 cells. Cells were grown in RPMI-1640 medium supplemented with 10% (v/v) heat-inactivated fetal bovine serum and two antibiotics (100 units/mL penicillin and $100\;{\mu}g$/mL streptomycin). Two types of TE, epigallocatechin galate (EGCG) and GTE, were used in this experiment. The cells were seeded at $1{\times}10^4$ cells/well in the RPMI-1640 media with or without TE ($100\;{\mu}g$/mL) and then treated with different concentrations of doxorubicin ($0{\sim}14\;{\mu}g$/mL) or cisplatin ($0{\sim}35\;{\mu}g$/mL). After incubation in 5% $CO_2$ at $37^{\circ}C$ for 24 hr, cell viability was determined with a MTT assay. We used a Western blot to detect the influence of EGCG and GTE on the expression of p53 and caspase-3 genes in the A549 cells. A549 cell viability decreased to 15% with a $10\;{\mu}g$/mL concentration of cisplatin, and to 21% with a $8\;{\mu}g$/mL concentration of doxorubicin, as measured with the MTT assay. However, pre-treatment of the cells with EGCG ($100\;{\mu}g$/mL) or GTE ($100\;{\mu}g$/mL) resulted in decreased cell viability with $6\;{\mu}g$/mL of cisplatin and $4\;{\mu}g$/mL of doxorubicin. There was no apparent change in cell viability between EGCG or GTE administration in cisplatin- or doxorubicin-induced cytotoxicity in A549 cells. The levels of p53 and caspase-3 in the A549 cells increased with both EGCG and GTE treatment. We found that GTE could potentially affect cisplatin- or doxorubicin-induced cytotoxicity of A549 cells, which may be useful in the chemotreatment of cancer.

Anti-Xanthine Oxidase, Anti-Cholinesterase, and Anti-Inflammatory Activities of Fruiting Bodies of Phellinus gilvus (마른진흙버섯 자실체의 Xanthine Oxidase, Cholinesterase 및 염증 저해 효과)

  • Phellinus gilvus is a medicinal mushroom used that has been used in folk medicine in Asian countries for centuries. The aim of this study was to investigate the anti-xanthine oxidase, anti-cholinesterase, and anti-inflammatory activities of methanol (ME) and hot water (HW) extracts prepared from fruiting bodies of Ph. gilvus. ME and HW had good anti-xanthine oxidase (XO) activities compared to allopurinol, an inhibitor of xanthine oxidase. ME showed comparable and slightly lower inhibitory activity against acetylcholinesterase (AChE) and butyrylcholinesterase (BChE), respectively, than galanthamine, a standard AChE and BChE inhibitor. ME also showed a protective effect against glutamate-induced cytotoxicity at 40 mg/mL and 100 mg/mL in PC-12 cells. ME (0.5~2.0 mg/mL) significantly inhibited nitric oxide (NO) production in RAW 264.7 murine macrophage cells stimulated with lipopolysaccharide (LPS). Carrageenan-induced hind-paw edema in rats was significantly reduced 2~6 hr after treatment with 50 mg/kg of ME, which was comparable to administration of 5 mg/kg of indomethacin, the positive control. These results demonstrate that ME and HW of Ph. gilvus fruiting bodies possess good anti-xanthine oxidase, anti-cholinesterase, and anti-inflammatory activities.

Study on the antioxidant, anti-inflammatory and Preservative features Water Chestnut (능실의 항산화, 항염증 및 방부 효과 연구)

  • The purpose of this study was to assess the antioxidant, anti-inflammatory and preservative effects of Water Chestnut from 70% ethanol extracts. The toxicity of extracts from Water Chestnut investigated using the RAW 264.7 cell showed higher than 90% of cell survival rate. The total content of polyphenol ethanol extract was $353.1{\pm}5.6mg/g$, while the total content of flavonoid was $26.2{\pm}1.4mg/g$. With a concentration level of $1{\sim}1000 ({\mu}g/m{\ell})$ ethanol extract of Water Chestnut the range of removal of 1, 1-diphenyl-2-picryl-hydrazyl (DPPH) radicals was $17.0{\pm}2.8%{\sim}88.6{\pm}0.6%$ respectively and the range of removal of 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radicals was also $2.3{\pm}0.8%{\sim}93.9{\pm}0.2%$ respectively. There were decreases in reactive oxygen species(ROS) generations ethanol extracts of Water Chestnut 1, 10, $100({\mu}g/m{\ell})$ and significance decrease at $100{\mu}g/m{\ell}$ (p<.01). As a result of measuring the Nitric Oxide(NO) generation amount of Water Chestnut extract 1, 10, $100({\mu}g/m{\ell})$ concentration exhibited significant (p<.001, p<.01) decreases. For the anti-bacterial features using a paper disk and the preservative features using petri film, no significance was found and therefore water chestnut extracts had not anti-bacterial or preservative features.

The Anti-aging Effects of Various Berries in the Human Skin Keratinocyte (HaCaT) Cells (피부각질형성세포에서 다양한 베리류의 피부노화개선 효과 비교)

  • Ultraviolet B (UV-B) irradiation is a negative factor that induces skin damage, inflammation, and aging. UVB irradiation induces the inflammatory response through interleukin (IL)-6 and IL-8 expression in keratinocytes. In addition, it induces the production of reactive oxygen species (ROS) and the activation of matrix metalloproteinase-1 (MMP-1), which plays an important role in collagen 1 degradation in the extracellular matrix. We investigated the antiaging effects of five kinds of berry in human skin keratinocyte (HaCaT) cells using juice of black raspberry (Rubus occidentalis), blueberry wild (Vacciniun angustifolium) and cultivar (Vacciniun corymbosum), black chokeberry (Aronia melanocarpa (Michx.) Elliott), and mulberry (Morus abla). HaCaT cells irradiated with UV-B exhibited increased ROS generation, as well as IL-6, IL-8, and MMP-1 gene expression, when compared to the control cells that were not irradiated with UV-B. However, pre-treatment of berry juice before UV-B irradiation significantly down-regulated the UV-B-induced ROS generation and inflammatory cytokine and MMP-1 expression. The results suggest that all berries have anti-aging effects including lowering inflammatory cytokine levels, ROS generation, and MMP-1 expression in HaCaT cells during UV-B irradiation.

[ $PFC^{3D}$ ] Modeling of Stress Wave Propagation Using The Hopkinson's Effect ($PFC^{3D}$ 상에서의 홉킨슨 효과를 이용한 응력파의 전파모델링)

  • An explosion modeling technique was developed by using the spherical discrete element code, $PFC^{3D}$, which can be used to model the dynamic stress wave propagation phenomenon. The modeling technique is simply based on an idea that the explosion pressure should be applied to a $PFC^{3D}$ particle assembly not in the form of an external force (body force), but in the form of a contact force (surface force). The stress wave propagation modeling was conducted by simulating the experimental approach based on the Hopkinson's effect combined with the spatting phenomenon that had previously been developed to determine the dynamic tensile strength of Inada granite. As a result, the stress wave velocity obtained by the proposed modeling technique was 4167 m/s, which is merely $3\%$ lower than the actual wave velocity of 4300 m/s for an Inada granite.

The effects of antioxidant and anti-aging treatment of UVB-irradiated human HaCaT keratinocytes with ethanol extracts of colored rice varieties (유색미 에탄올 추출물의 산화방지 효과와 UVB에 유도된 인간유래 피부각질형성세포에서의 항노화 효과)

  • In this study, we investigated the antioxidant activities and anti-aging efficacy in terms of suppression of matrix metalloproteinases (MMPs) in UVB-irradiated HaCaT cells by adding the ethanol extracts of Josaengheogchal (JE) and Shintoheug rice (SRE). In the electron-donating ability and ABTS radical-scavenging assays, we observed that both JE and SRE had scavenging activities and in a collagenase inhibition assay, both extracts showed inhibition effects of over 73% at $1,000{\mu}g/mL$ concentration. The expression of MMP-1 and -3, when the extracts were treated with UVB $50mJ/cm^2$, irradiated human HaCaT keratinocytes, was analyzed by western blotting and reverse-transcription polymerase chain reaction (RT-PCR). The results showed that MMP-1 and -3 proteins and mRNAs were downregulated in a concentration-dependent manner in response to both extracts. Therefore, we expect that these compounds have a potential for the use as functional ingredients with anti-aging effects in the cosmetic and food industries.
