Pharmacological Effects of Lycium chinensis (구기자나무의 약물활성)

  • Pharmacological effects of water extracts of Lycii Fructus, Lycii Folium and Lycii Cortex Radicis from Lycium chinensis were investigated. Lycii Folium significantly protected the hepatic function from damages orally caused by $CCl_4 $administration in mice and had a strong hypoglycemic effect in hyperglycemic mice induced by streptozotocin. Lycii Fructus decreased the blood pressure rise associated with the growth of normal rats. Lycii Cortex Radicis had a strong hypoglycemic effect in hyrerglycemic mice induced by streptozotocin. Also, hypolipidemic effects in hyperlipidemic rats induced by 1% cholesterol fed-diet and 75% fructose were significantly observed by oral administration of water extracts of Lycii Fructus, Lycii Folium and Lycii Cortex Radicis.

Development of Extracts of Lycii folium Having High Antioxidant Activity (항산화 활성이 높은 구기엽 추출물 제조)

  • The aim of this study was to analyze the antioxidant properties of Lycii folium extracts prepared from different solvents. Lycii folium were extracted with water, 80% ethanol (80% EtOH), 80% methanol (80% MeOH) and 100% methanol (100% MeOH) in water bath at $40^{\circ}C$. The antioxidant activity of the extracts was evaluated using DPPH, hydroxyl and hydrogen radical scavenging activities, and SOD-liked activity. Total phenolic acid contents were 1.085 mg/mL in 100% MeOH, 1.382 mg/mL in 80% EtOH, 1.420 mg/mL in 80% MeOH and 1.084 mg/mL in water. DPPH radical scavenging activity of the extracts were 65.60% in 80% EtOH, 56.80% in 80% MeOH, 83.85% in 100% MeOH and 54.65% in water. Hydroxyl radical scavenging activities were 66.65% in 100% MeOH, 73.13% in 80% ethanol, 73.58% in 80% MeOH and 70.73% in water. Hydrogen radical scavenging activity of the extracts prepared from Lycii folium were 11.70% in 100% MeOH, 33.73% in 80% EtOH, 35.40% in 80% M eOH and 23.86% in water. SOD-liked activity of the extracts prepared from Lycii folium was 71.58% in 100% MeOH, 74.29% in 80% EtOH, 88.46% in 80% MeOH and 67.47% in water. Our result showed that Lycii folium extracts prepared from 80% methanol were found to be promising biomaterials with antioxidant effects.

The Effects of Lycii fructus and Lycii folium on the Liver in Ovariectomized Rat (구기자와 구기엽이 난소적출 흰쥐의 간에 미치는 영향)

  • We aimed to investigate whether the extracts of Lycii fructus, Lycii folium, and estradiol can attenuate on the level of cholesterol on serum and ultrastructure alteration of hepatocytes isolated from ovariectomized rats. Experimental groups are divided into five: Sham (sham-operated), OVX (ovariectomized animal), LCF (OVX, Lycii fructus), LCL (OVX, Lycii folium), and $E_2$ (OVX, estradiol). The body weights of OVX group showed increase compared with Sham, LCF, and LCL groups. The levels of total cholesterol and low-density lipoprotein (LDL) cholesterol were significantly decreased in the LCF and LCL groups compared with those of OVX group. Moreover, the High-density lipoprotein (HDL) cholesterol level was increased in the LCL group. As the results of electron microscopical observation, lipid droplets in the OVX group showed increase compared with Sham group. But in the LCF group not showed lipid droplets. In conclusion, administration of Lycii fructus or Lycii folium lowers the serum cholesterol content of the ovariectomized rats and also the damage of the hepatocytes and formation of lipid droplets.

Fermentation Properties of Yogurt Added by Lycii fructus, Lycii folium and Lycii cortex (구기자, 구기엽 및 지골피를 첨가한 요구르트의 발효 특성)

  • This experiment was carried out to examine the fermentation properties of yogurt with Lycii fructus, Lycii folium and Lycii cortex powder, and extract additives at concentrations of 0.5, 1.0, 2.0, 4.0, and 6.0%. Lactic acid bacteria was used in a mixed starter culture of Streptococcus salivarius ssp. thermophilus(ST36) and Lactobacillus delbrueckii ssp. bulgaricus(LB12). When the boxthorn was added with extract types, the changes of pH, acidity and lactic acid bacteria counts of yogurt during the fermenation of 3 hours were pH 5.64, titratable acidity 0.85%, 5.80xl0$\^$6/cfu/ml of viable cell counts for control yogurt, whereas those were pH 4.10∼5.06, titratable acidity 0.98∼1.27%, 1.80∼9.60x10$\^$7/ cfu/ml of viable cell counts for Lycii fructus extract yogurt. The lactose hydrolysis ratio was better for 1.0% Lycii fructus extract yogurt(42.00%) and 1.0% Lycii folium extract yogurt(41.46%) than for control yogurt(28.40%). Also, content of lactic acid of 1.0% Lycii fructus(11.9 times) and 1.0% Lycii folium extract yogurt(10.6 times) produced more than control yogurt(7.3 times). The viscosity of yogurt was better for boxthorn extract yogurt(1,027∼1,382 cps) than for control yogurt(975cps). The sensory scores of color, taste and overall acceptability of yogurt with 0.5, and 1.0% Lycii fructus extract additive were better than other groups. The yogurts made with increased Lycii fructus extract concentration(0.5∼6.0%), showed the increase of lactic acid, titratable acidity, number of lactic acid bacteria, viscosity and lactose hydrolysis rate compared to the treatments of 0.5, 1.0, 2.0, and 4.0% Lycii folium and Lycii cortex extract and powder yogurt. We gained excellent results from the yogurt to which Lycii fructus extract was added with 0.51.0% concentration.

Effects of the Biological Function of Yogurt Added with Lycium chinence Miller Extract (구기자(Lycium chinence Miller)추출액을 첨가한 요구르트의 생리활성 효과)

  • This experiment was carried out to investigate the biological function of Lycium chinence Miller yogurt. Antioxidant activity was higher in methanol extract yogurt than in water extract yogurt. The antioxidant activity was shown at 83.9% in Lycii folium extract yogurt, 47.0% in Lycii fructus extract yogurt and 54.0% in Lycii cortex extract yogurt. Angiotensin converting enzyme inhibitor activity was shown highly both in water extract and methanol extracts from the Lycii folium. The $\alpha$-glucosidase inhibitor activity was shown at the 4.0% concentration of Lycii folium extract yogurt, Lycii fructus extract yogurt and Lycii cortex extract. In an orally administrated rat, normal yogurt, Lycii fructus extract yogurt and Lycii cortex extract yogurt have no effect on blood cholesterol content. IgG production in blood is more increased in Lycii cortex extract yogurt than in normal yogurt and Lycii fructus extract yogurt.

Effects of Lycii Fructus and Lycii Folium Extracts on Osteoporosis in Ovariectomized Rats (구기자와 구기엽 추출물이 난소적출 흰쥐의 골다공증에 미치는 영향)

  • The effects of Lycii fructus and Lycii folium on osteoporosis and serum cholesterol levels were tested in ovariectomized (OVX) rats. Twenty-four female Sprague-Dawley rats were divided into four groups: Sham group (sham-operated), Control group (OVX, ovariectomized model), LCF group (Ovx+Lycii fructus extract), and LCL group (OVX+Lycium folium extract). After 8 weeks, the OVX ($330{\pm}5.39$ g), LCF ($315{\pm}2.99$ g), and LCL ($318{\pm}2.06$ g) groups showed increased body weight compared with sham group ($281{\pm}1.71$ g). The levels of serum osteocalcin (OC) also increased in the LCF ($444.6{\pm}26.9$ ng/mL) and LCL ($407{\pm}18.9$ ng/mL) groups compared with the OVX group ($107{\pm}3.52$ ng/mL). The activities of serum alkaline phosphatase (ALP) increased in the LCF ($108{\pm}2.7$ U/L) and LCL ($407{\pm}18.9$ ng/mL) groups compared with the OVX group ($95{\pm}2.9$ U/L). Stereomicroscopy found that the low bone density that developed in the OVX group was significantly reversed in the LCF and LCL groups after 8 weeks. We also obtained molecular-based in vivo evidence that supports a mechanism of action involving novel estrogen receptor ($ER{\alpha}$) modulator in the uterus. We found that expression of ER${\alpha}$ mRNA in the OVX rat uterus was elevated by Lycium chinense. These results suggest that Lycii fructus and Lycii folium administered to rats during 8 weeks after oophorectomy may partially recover postmenopausal osteoporosis or delay the progression of osteoporotic changes.

Effects of Antioxidant Activity and Changes in Vitamin C during Storage of Lycii folium Extracts Prepared by Different Cooking Methods (조리방법을 달리한 구기엽의 저장기간별 항산화 효과 및 비타민 C 함량의 변화)

  • Contents in polyphenol and vitamin C and their antioxidant activities during storage of Lycii folium prepared by different treatment methods were investigated. The antioxidant activity of extracts prepared from 80% MeOH was evaluated using DPPH radical scavenging and SOD-liked activities. Total phenolic acid contents were 1.34 mg/mL when dry, but extracts during 15-day storage after NaCl 0% treatment decreased. DPPH radical scavenging activity of extracts was 65.48% when dry and decreased during 15-day storage after dry and NaCl 0% treatment. SOD-liked activity was 98.63% in NaCl 0% during 15-day storage after dry. Contents of vitamin C decreased during 15-day storage after dry and NaCl 0% treatment. Consequently, our results indicate that Lycii folium prepared from dry treatment could be a promising biomaterial for the production of functional food for total phenolic acid and vitamin C.

Fermentation Properties and Functionality of Yogurt added with Lycium chinence Miller (구기자(Lycium chinence Miller) 첨가에 따른 요구르트의 발효 특성과 기능성)

  • This experiment was carried out to examine the fennentation properties of yogurts with or without Lyeii fructus, Lyeii folium and Lyeii cortex extract as additives at concentrations of 1.0%. The effects on promoting the fermentation by Lycii fructus, Lycii folium and Lycii cortex additives were higher and pH was below 4.06 when Streptococcus salivarius ssp. thermophilus and Lactobacillus delbrueckii ssp. bulgaricus, Lactobacillus cosei, and Lactobacillus acidophilus, Bifidobacterium longum and Streptococcus salivarius ssp. thermophilus were used. The acid production was higher when S. salivarius ssp. thermophilus and L. delbrueckii ssp. bulgaricw were used. The average lactic acid bacteria counts was 2.62 ${\times}$ $10^9$ cfu/ml in the yogurt added with Lycii fructus extract and fermentation with S. salivarius ssp. thermophilus and L. delbrueckii ssp. bulgaricw. The lactose hydrolysis ratio was higher in the milk added with Lycii fructus extract(36.11%), Lycii folium extract(37.76%) and Lycii cortex extract(32.70%) when S. salivarius ssp. thermophilus and L. delbrueckii ssp. bulgaricus were used. The isobutylic acid concentration was(34.39 to 37.72 mM) with S. salivarius ssp. thermophilus and L. delbrueckii ssp. bulgaricus. The viscosity of yogurt was 1,615 to 2,030 cP in yogurts added with skim milk and L. acidophilus; B. longum and S. salivarius ssp. thermophilus were used. The sensory scores of colors, tastes and overall acceptability of yogurt with Lycii cortex extract were shown 3.34 to 3.77 when fermented by L. cosei, L. acidophilus, B. longum and S. salivarius ssp. thermophilus. The cholesterol reducing effects were 17.38${\sim}$32.08% in all the yogurts and especially, greater effect(25.75 to 32.08%) for yogurts fermented with L. acidophilus KCTC3150 and L. salivarius subsp. salivarius CNU27. The inhibitory effects on the pathogenic bacteria by lactic acid bacteria added with Lycii fructus, Lycii folium and Lyeii cortex lower on S. typhimurium M-15, but higher on E. coli KCTC1021 and L. monocytogenes.

Effects of the Ethanol Extract from Lycii folium Leaves on Obesity and Blood Biochemical Indices in High-fat Diet Induced Obese Rats (구기엽 에탄올 추출물이 고지방 식이 유도 비만쥐의 비만과 혈중 생화학적 지표 조절에 미치는 영향)

  • This study was conducted to investigate the effects of ethanol extract from Lycii folium (L. folium) leaves on obesity index, serum lipids, homocysteine, leptin, ghrelin, and glucose in obese rats. Sprague-Dawley rats were administrated high-fat diets to induce obesity. Then, obese rats were divided into three diet groups: a basal-diet obese group (BO group), high-fat diet obese group (FO group), and high-fat diet plus L. folium extract obese group (FLEO group). Three groups were each fed experimental diet for 8 weeks. There were no significant differences in body weight and FER among the groups. However, obesity index such as Ro?hrer index, Lee index, and T.M. index of FLEO group was significantly decreased as compared to FO group. While serum triglyceride of BO group was significantly decreased as compared to FO group, there were no significant differences in serum lipids, homocysteine, leptin, ghrelin and glucose between FLEO group and FO group. In conclusion, these results indicated that ethanol extract from L. folium leaves might be beneficial with anti-obese effect by reduction of obesity indices in obese rats.

Moderating Effects of Skin Hyperpigmentation from Lycii fructus and Lycii folium Extracts (구기자 및 구기엽 추출물의 피부과색소 조절효과)

  • The objective of the present study was to evaluate the skin whitening effect of the extracts, Lycii fructus (LF), Dry-L. foilum (DLF) and Fresh-L. folium (FLF). Tyrosinase inhibition activities was 44% in DLF ethanol extracts at a $500{\mu}g/mL$. When the tyrosinase activities in B16F10 murine melanoma cell were tested, the activities in DLF ethanol extracts was 14% at a $50{\mu}g/mL$ concentration. The protein expression of microphthalmia-associated transcription factor, tyrosinase related protein 1 (TRP-1), TRP-2, and tyrosinase, which are all melanin related factors, showed that LF, DLF and FLF extracts inhibited the protein bio-synthesis in B16F10 melanoma cell. Especially the DLF extract showed greater decrease of protein expressions. Results indicate that the DLF extract tested in the present study had skin whitening activity and can be used as a function a ingredients for food and cosmetic compositions.
