Expression and Characterization of Protein Latcripin-3, an Antioxidant and Antitumor Molecule from Lentinula edodes C91-3

  • In this study, an anti-oxidant and anti-tumor protein Latcripin-3 of Lentinula edodes C91-3 was expressed in Escherichia coli. for the first time. According to the cDNA library, the full-length gene of Latcripin-3 was cloned by the methods of 3'-full rapid amplification of cDNA Ends (RACE) and 5'-full RACE. The structural domain gene of Latcripin-3 was inserted into the pET32 a(+). The functional protein of Latcripin-3 was expressed in Rosetta-gami (DE3) E. coli, evaluated by Western blotting and mass spectrometry. DPPH testing showed that the protein Latcripin-3 can scavenge free radicals remarkably well. The activity of functional protein Latcripin-3 on A549 cells was studied with flow cytometry and the MTT method. The MTT assay results showed that there was a decreases in cell viability in a dose-dependent and time-dependent manner in protein Latcripin-3 treated groups. Flow cytometry demonstrated that Latcripin-3 can induce apoptosis and block S phase dramatically in human A549 lung cancer cells as compared to the control group. At the same time, the cell ultrastructure observed by transmission electron microscopy supported the results of flow cytometry. This research offers new insights and advantages for identifying anti-oxidant and anti-tumor proteins.

홍삼대보탕(紅蔘大補湯)의 항암활성(抗癌活性) 및 항전이(抗轉移) 효과(效果)에 관(關)한 연구(硏究) (Study on Antitumor Activity of Hongsamdaibotang(HDT-C))

  • To evaluate the antitumor activity and antimetastatic effects of Hongsam -daibotang(HDT-C), studies were done experimentally. The results were obtained as follows: 1. In cytotoxicity against A549, SK-OV-3 and B16-BL6 concentration inhibi ting cell growth up to below 30% of control was recognized at $10^{-3}g/ml$ of HDT-C. 2. The T/C% was 145.4% in HDT-C treated group in S-180 bearing ICR mice. 3. In Inhibitory effect on activity of DNA topoisomerase I, the $IC_{50}$ was shown $100-200{\mu}g/ml$ of HDT-C. 4. The expressing $TNF-{\alpha}$ was increased in HDT-C treated group as compared with control. 5. The expressing MMP-9 was decreased in HDT-C treated group as compared with control. 6. HDT-C extracts exhibited efficient adhesive effect of A549, B16-BL6 cell to complex extracellular matrix. 7. In CAM assays, angiogenesis was significantly inhibited in HDT-C treated group than control group. 8. In pumonary colonization assay, a number of colonies in the lungs were decreased significantly in HDT-C treated group as compared with control group. 9. In hematological changes in B16-BL6 injected C57BL/6, numbers of WBC and were decreased insignificantly and also those of platelet were increased insignificantly in HDT-C treated group as compared with control. 10. In the histological changes of lung in B16-BL6 injected mice, infiltration of cancer cells were inhibited effectively in HDT-C treated groups whereas many cancer cells were infiltrated into erivascular and peribronchiol of control group. These results suggested that HDT-C extracts might be usefully applied for prevention and treatment of cancer.

갯기름나물의 근(根)과 경엽(莖葉)의 면역 및 항암 활성 비교연구 (A Comparative Study on the Immunizing and Anti-cancer Effect of the Root and the Aerial Part of Peucedanum Japonicum Thunb)

  • This study compared the activity of the aerial part of P. japonicum with its root in order to examine the possibility of the medicinal use of the aerial part, which has not been used as medicine, in substitute for the root that has traditionally been used as medicine. For this purpose, the author measured the proliferation of Human $CD4^-$ T cells, which are related to immunity, the differentiation of HL-60 cells, and the contents of IL-6, IgE and $TNF-{\alpha}$ and compared their anti-cancer effect on Hep3B and A549 cells. The results of this study are as follows: 1. As for Human $CD4^-$ T cells, $1.0\;g/{\ell}$ methanol extract from the aerial part promoted the proliferation of the cells 1.8 times higher while $1.0\;g/{\ell}$ methanol extract from the root did by 1.76 times higher compared to the control group. 2. As for HL-60 cells, methanol extract and water extract from the aerial part showed differentiation 1.14 times higher and 1.12 times higher respectively while methanol extract and water extract from the root did 1.14 times higher and 1.07 times higher compared to tile control group. 3. Cell density was highest on Day 4 of culture in all samples, Methanol extracts from the aerial part and the root showed activities of $7.9{\times}10^3\;cells/m{\ell}$ and $7.5{\times}10^3\;cells/m{\ell}$ respectively, and water extracts from the aerial part and the root did activities of $5.3{\times}10^3\;cells/m{\ell}$ and $6.1{\times}10^3\;cells/m{\ell}$. 4. The secretion of IL-6 was highest on Day 4 of culture. Methanol extracts from the aerial part and the root showed secretions of $6.7{\times}10^{-3}\;pg/cells/m{\ell}$ and $7.2{\times}10^{-3}\;pg/cells/m{\ell}$ respectively, and water extracts from the aerial part and the root did secretions of as high as $7.0{\times}10^{-3}\;pg/cells/m{\ell}$ and $6.0{\times}10^{-3}\;pg/cells/m{\ell}$. 5. As for the production of IgE, water extract from the root effectively inhibited the product at $1,000\;{\mu}g/m{\ell}$, methanol extract from the root at $10\;{\mu}g/m{\ell}$ and $100\;{\mu}g/m{\ell}$, water extract from the aerial at $1,000\;{\mu}g/m{\ell}$, and methanol extract from the aerial part at $1,000\;{\mu}g/m{\ell}$. 6. According to the result of measuring the content of $TNF-{\alpha}$, methanol extracts from the root and the aerial part showed inhibition effect at $10\;{\mu}g/m{\ell}$, $100\;{\mu}g/m{\ell}$ and $1,000\;{\mu}g/m{\ell}$. 7. As for liver cancer cell Hep3B, $1.0\;g/{\ell}$ methanol extracts from the root and the aerial part showed inhibition effects of 78% and 70% respectively, and $1.0\;g/{\ell}$ water extracts from the root and the aerial part did inhibition effects of 56% and 59%. 8. As for lung cancer cell A549, $1.0\;g/{\ell}$ methanol extracts from the root and the aerial part showed inhibition effects of 75% and 70% respectively, and $1.0\;g/{\ell}$ water extracts from the root and the aerial part did inhibition effects of 48% and 45%. The results of this study presented above show that the aerial part of P. japonicum has immunizing and anti-cancer effects as high as its root, which has commonly been used as medicine. There should be more in-depth research on the aerial part of P. japonicum in the future.

광물석분의 항암 및 항전이 효과에 관한 연구 (Antitumor and Anti-metastatic Effect of Mineral Powder)

  • Antitumor and anti-metastatic effects of mineral powder(MP) were studied. In the present study, MP did not exhibit the any cytotoxic activity against leukemic cells such as L1210 and U937 tumor cell lines in vitro. Also, MP did not exhibit the any cytotoxic activity against solid cells such as A549 and B16-BL6 tumor cell lines in vitro. However, in vivo, MP exhibit a significant antitumor activity in BDF1 mice bearing Lewis lung carcinoma cells(LLC) with inhibition rates of 46 and $23\%$ at 200 and 100 mg/kg/day, respectively. Furthermore, in pulmonary colonization assay, MP exhibit the inhibitory effect of tumor metastasis. From these results, it was concluded that MP had antitumor and anti-metastatic activity suggesting its application for the prevention and treatment of cancer.

산수유에 함유된 항암물질의 정제 및 특성 (Purification and Characterization of Anticarcinogenic Compound from Corni fructus)

  • 암세포 성장 억제효과가 인정된 산수유 methanol 추출물의 chloroform층으로부터 silica gel column과 thin layer chromato-grapies를 이용하여 항암활성물질을 분리하고, IR, mass spectrometer, $^1H-NMR$$^{13}C-NMRs$을 통해 순수물질을 동정하였다. 이 물질은 triterpenoid로서 $C_{30}H_{48}O_3$의 구조식을 가진 화합물로 분자량이 456인 ursolic acid($3{\beta}$-hydroxyrus-12-ene-28-oic acid)로 동정되었다. 정제된 화합물을 A549 및 MCF-7 암세포주에 48시간동안 처리한 결과 농도 의존적으로 암세포주의 성장을 억제하였으며, 화합물을 처리하지 않은 대조구에 비하여 $30\;{\mu}g/mL$농도로 처리시 40% 이상 그 성장을 억제하였으며, $100\;{\mu}g/mL$농도로 처리시 90% 이상 그 성장을 억제하였다. 정제 화합물을 처리한 암세포를 광학현미경으로 관찰한 결과 뚜렷한 세포수의 감소와 함께 심한 형태학적 변화가 관찰되었다. 암세포 주에 정제시료를 $10\;{\mu}g/mL$농도로 처리하고, 15시간 배양한 후 세포주기를 분석한 결과 sub-G1 phase의 비율이 A549의 경우는 대조구에서 4.0%이었던 것이 11.7%로, MCF-7의 경우는 대조구에서 2.1%이었던 것이 처리구에서는 11.2%로 각각 증가된 것으로 나타나 정제물질은 apoptosis에 의한 세포사멸 유도를 시사하였다.

악성 피부 종양에서의 Fibroblast Growth Factor 4 (FGF4) 발현 (Fibroblast Growth Factor 4 (FGF4) Expression in Malignant Skin Cancers)

  • Purpose: FGF4 (fibroblast growth factor 4) is a newly characterized gene which was found to be a transforming gene in several cancerous cells. FGF4 expression and amplification has been subsequently observed in several human cancers including stomach cancer, breast cancer, head and neck squamous cell carcinoma, lung cancer and bladder cancer. This study was designed to measure the protein expression of FGF4 in malignant skin cancers. Methods: We examined 8 normal skin tissues and 24 malignant skin tumor tissues which were 8 malignant melanomas, 8 squamous cell carcinomas and 8 basal cell carcinomas. The specimens were analyzed for the protein expression of FGF4 using immunohistochemical staining. To evaluate the amount of expression of FGF4, the histochemical score (HSCORE) was used. Results: FGF4 was expressed more intensely in malignant melanoma, followed by SCC and BCC in immunohistochemistry. The average HSCORE was 0.01 for normal skin, 2.02 for malignant melanoma, 1.28 for squamous cell carcinoma, and 0.27 for basal cell carcinoma, respectively. The expression of FGF4 in malignant melanoma and squamous cell carcinoma was increased in comparison with normal tissues and basal cell cancer, and the difference was statistically significant (p<0.05). The difference between malignant melanoma and squamous cell carcinoma was not statistically significant. Conclusion: These findings provide evidences that the expression of FGF4 plays an important role in malignant melanoma and squamous cell carcinoma progressions. This article demonstrates expression of FGF4 in human skin malignant tumors, and suggests that FGF4 is more expressed in highly aggressive skin tumors.

저온고압 추출공정을 이용한 매자나무 수피의 항산화 및 항암활성 증진 (Enhancement of the Antioxidant and Anticancer Activities of Berberis koreana Bark by Using a Low Temperature and High-Pressure Extraction Process)

  • 본 연구에서는 저온고압 추출공정을 이용하여 전통적인 기존 추출공정과 비교함으로써 저온고압 추출공정에 의한 매자나무 수피의 항산화활성과 항암활성의 증진을 확인하고자 연구를 수행 하였다. DPPH radical 소거 활성은 5분 저온고압 처리한 추출물이 93%로 가장 높은 활성을 나타내며 대조군인 ascorbic acid의 95%와 유사한 활성을 나타내었다. 저온고압 추출공정을 통한 매자나무 추출물이 일반 열수 추출물과 비교해 xanthine oxidase 저해활성이 높게 나타났으며 특히 5분 저온고압 처리한 추출물이 56.68%의 높은 저해 활성을 나타냈다. 따라서 저온고압 추출공정을 통해 매자나무 추출물의 항산화 활성의 증진이 이루어졌음을 알수 있다. 항암활성 측정 결과, 매자나무 수피 추출물의 높은 항암활성을 확인하였으며 5분 저온고압 처리한 추출물의 암세포 억제 활성이 가장 높게 나타남에 따라 저온고압 추출공정을 통해 항암활성의 증진이 가능함을 확인하였다. 세포독성 측정 결과 5분 저온고압 추출물이 정상 세포에 대한 독성이 가장 높은데 반해 15분 저온고압 처리한 추출물의 세포독성이 제일 낮은 것으로 미루어 저온고압 추출물의 세포독성이 저온고압 추출시간의 영향을 받는 것으로 사료된다. 따라서 매자나무 수피의 생체적용이 가능한 기능성 소재로서의 이용 가능성을 확인할 수 있으며 저온 고압 추출공정의 최적화를 통한 활성물질의 추출 극대화를 통해 높은 경제적 가치를 부여해 줄 수 있을 것으로 사료된다.

발계(Smilax china L.) 열수 및 에탄올 추출물의 항산화 및 항암활성 (Antioxidant and anticancer properties of hot water and ethanol extracts from the roots of Smilax china L.)

  • 식품 연구에서 흔히 사용되는 물과 주정(70% 에탄올)을 사용하여 추출된 발계(SCR)의 열수 추출물(SCRW)과 에탄올 추출물(SCRW)의 총 플라보노이드 및 폴리페놀 함량, 항산화 활성, 항암활성을 측정하였다. SCRW와 SCRE는 DPPH(SCRW; 72.9%, SCRE; 42.4%)와 ABTS(SCRW; 81.6%, SCRE; 50.3%) 자유 라디칼을 효과적으로 소거하였으며, 양성 대조군인 BHA(53.8% DPPH, 49.5% ABTS)보다 더 높은 소거활성을 나타냈다. 또한 인체 유래 신장 정상세포(HEK293)에 대해서는 높은 생존율을 나타내 독성이 없음을 확인한 반면, 인체 유래의 암세포(유방암세포; MCF-7, 폐암세포; A549, 위암세포; AGS)에 대해서는 대조군으로 사용된 항암제 CPA와 유사하거나 더 높은 세포성장 억제효과가 확인되었다. 특히 SCRE는 AGS에 대하여 $1,000{\mu}g/mL$ 농도에서 53.6%로 탁월한 위암 세포 성장저해 효과를 나타냈다. 본 연구 결과를 통해, 발계 추출물의 높은 항산화 및 항암 활성이 확인되어 약리활성에 대한 기초연구 자료로 제시 할 수 있을 것으로 사료되며, 기능성 소재로써의 이용 가치를 높일 수 있을 것으로 판단된다.

생더덕과 발효더덕의 유용생리활성 비교 (Comparison of Biological Activities of Fermented Codonopsis lanceolata and Fresh Codonopsis lanceolata)

  • Both fresh Codonopsis lanceolata and lactic acid bacteria fermented Codonopsis lanceolata were extracted with water at $100^{\circ}C$, and tested for anticancer activity using several human cancer cell lines. The fermented extracts inhibited the growth of hepatocellular carcinoma cells up to 90%, compared to 75% for fresh Codonopsis lanceolata. The extracts of cytotoxicity on human normal lung cells (HEK293) were as low as 15%. Especially, human hepatocellular carcinoma cell were more efficiently inhibited than other cells. This extract also inhibited $\alpha$-glucosidase activity up to 60% at 1.0mg/$m{\ell}$. This fermented extracts showed the inhibition potency on tyrosinase by 25% at 1.0mg/$m{\ell}$. From the results, the fermented Codonopsis lanceolata enhanced several biological activities up to $20{\sim}30%$, compared to those from fresh Codonopsis lanceolata. It implies that fermentation process could be one of useful methods of utilizing low quality Codonopsis lanceolata. Because this process could yield high amounts of biologically active compounds by the help of microbial growth.

각종악성종양환자(各種惡性腫瘍患者)의 Ferrokinetics 및 동대사(銅代謝)에 관(關)한 연구(硏究) (Studies on Ferrokinetics and Copper Metabolism in Various Malignant Tumors)

  • Anemia is a usual finding in advanced malignant diseases. Various mechanisms were reported as to be involved in the development of anemia of this kind, and they may differ in individual cases. Tumor anemias may be due, for instance, to chronic blood loss, shortened life span of the red blood cells or a decreased hemopoiesis in the bone marrow. The serum iron and copper levels, total iron binding capacity, apparent half survival of $^{51}Cr$-labelled red blood cells were measured along with the ferrokinetic studies using $^{59}Fe$ in 64 patients with various malignant tumors. Following were the results: 1. The serum iron levels were decreased in all cases. There existed no correlation between the serum iron levels and the severity of the diseases. 2. The serum copper levels were increased, particularly in lung cancer, rectal cancer, hepatoma and various sarcomas. There was also no correlation between the serum copper levels and the severity of the diseases. 3. The serum iron levels appeared to be inversely proportional to the serum copper levels. 4. The total iron binding capacities were within normal limits in all cases. There were also no correlations between the total iron binding capacities, serum iron levels and the severity of the diseases. 5. The patients could be classified according to the ferrokinetic patterns, namely, that of iron deficiency anemia in 10 cases, that of refractory anemia in 6 cases, normal in 1 case and that of atypical abnormal in 9 cases. 6. Apparent half survival time of $^{51}Cr$-labelled red blood cells were definitely shortened in half of the cases.
