The Microstructural Properties Change Owing to the Sintering Condition of T42 High Speed Steel Produced by Powder Injection Molding Process (분말 사출 성형법으로 제조된 T42 고속도 공구강의 소결 조건에 따른 조직 특성 변화)

  • High speed steels (HSS) were used as cutting tools and wear parts, because of high strength, wear resistance, and hardness together with an appreciable toughness and fatigue resistance. Conventional manufacturing process for production of components with HSS was used by casting. The powder metallurgy techniques were currently developed due to second phase segregation of conventional process. The powder injection molding method (PIM) was received attention owing to shape without additional processes. The experimental specimens were manufactured with T42 HSS powders (59 vol%) and polymer (41 vol%). The metal powders were prealloyed water-atomised T42 HSS. The green parts were solvent debinded in normal n-Hexane at $60^{\circ}C$ for 24 hours and thermal debinded at $N_2-H_2$ mixed gas atmosphere for 14 hours. Specimens were sintered in $N_2$, $H_2$ gas atmosphere and vacuum condition between 1200 and $1320^{\circ}C$. In result, polymer degradation temperatures about optimum conditions were found at $250^{\circ}C$ and $480^{\circ}C$. After sintering at $N_2$ gas atmosphere, maximum hardness of 310Hv was observed at $1280^{\circ}C$. Fine and well dispersed carbide were observed at this condition. But relative density was under 90%. When sintering at $H_2$ gas atmosphere, relative density was observed to 94.5% at $1200^{\circ}C$. However, the low hardness was obtained due to decarbonization by hydrogen. In case of sintering at the vacuum of $10^{-5}$ torr at temperature of $1240^{\circ}C$, full density and 550Hv hardness were obtained without precipitation of MC and $M_6C$ in grain boundary.

Influences of Protease on the Removal of Protein Soils from Cotton Fabrics -Emphasis on the Characteristics of Enzyme and Soil Substrates- (단백질 분해 효소가 세척에 미치는 영향 -효소와 오염 기질의 특성을 중심으로-)

  • The influences of protease on the removal of various protein soils from cotton fabrics were studied. The human epidermal stratum corneum, hemoglobin and casein were used as protein soils. The soiled fabrics were denatured by steaming for 30 min. before washing and laundered using Terg-O-Tometer under washing conditions. The removal efficiency was evaluated by analysis of protein on the fabrics before and after washing by means of copper-Folin method. The relations between the removal and the characteristics of protease were discussed. Also the degradation of protein were examined by microscopy. The seperation of human epidermal stratum corneum after hydrolysis was examined by SDS-PAGE. The results obtained were as follow : 1. The protein from the soiled cotton fabric was removed effectively by adding protease. The removal of protein was increased in proportion to increasing of the enzyme concentration up to a certain point, but it began to decrease above the point. The removal effect was high in the order of casein>human epidermal stratum corneum>hemoglobin. Especially the protein was more effectively removed in ADS solution(pH 9.5) containing enzyme. 2. When protease was used with ADS. the removal of protein was efficiently showed in relatively short time(5~15min.) compared to using ADS only. It is due to the properties of this enzyme that reacts with very short time. 3. Even at low temperature the removal efficiency of enzyme was relatively higher compared with the activity of enzyme. The removal of protein soil was increased up to a maximum near $50^{\circ}C$, and then decreased. 4. The removal of protein by protease was improved with the increase of alkalinity in the pH range from 9.5 to 11.0 but it began to decrease above pH 11.0. 5. According to the increase of mechanical agitation, the removal effect was increased. But the removal efficiency of protease was more effective compared with the agitation in detergency. 6. According to the SDS-PAGE separation and micrograph it was confirmed that the human epidermal corneum was effectively hydrolysed by the enzyme added. So the fragments of protein were removed more efficiently by means of the interfacial reaction of AOS.

Studies on betterment of flue-curing of tobacco by using Ethephon (Ethephon (2-Chloroethylphosphonic acid)을 이용한 담배 건조법개선에 관한 연구)

  • Five different levels of Ethephon were treated on tobacco cultivar yellow Special A and Coker and harvested for curing. Changes in dehydration properties, yellowing, chlorophyll, nicotine, sugar and starch contents during the processes of curing were studied in order to justify feasibility of adoption an idea of 3-stage-curing method instead of the 5-stage. Ethephon-treated-tobacco leaves showed marked degradation of chlorophyll, greater dehydration and greater reduction in nicotine contents while little change in either sugar, starch and other chemical components. They showed also shorter hours of curing period allowing possible curing response at a relatively low temperature than those without treatment. Thus they undergo only 3 stages of flue-curing processes, yellowing, calor-fixing and vein-drying, and need 72 hours to finish curing, but the ordinary leaves need 100 hours for it.

Changes in Mineral and Pectic Substances of Korean Mature-Green Mume (Prunus mume Sieb. et Zucc) Fruits Packaged in Plastic Films with Gas Absorbents during Storage (가스 제거제 첨가에 따른 필름 포장 청매실의 저장 중 무기질 및 펙틴 성분 변화)

  • Changes in mineral (Ca, Mg) contents and pectic substances of mature-green 'Nanko' Mume fruits hermetically packaged in 0.03 mm low density polyethylene (LDPE) films with and without gas absorbents were examined during storage at $25^{\circ}C$ for 10 days. Each packaging contained 10 g $Ca(OH)_2$ as a carbon dioxide scavenger, 30 g $KMnO_4$ as an ethylene scrubber or their mixture. In the presence of the ethylene scrubber, losses in mineral contents of alcohol insoluble solids and water soluble pectin were remarkably suppressed, whereas no significant difference was observed in the Ca content between the fresh fruit and those stored for 10 days. Fruits packaged with the ethylene absorbent retained higher amount of pectic substances than those with other packaging treatments. Degradation of the pectic substances into small molecules was also noticeably reduced when the ethylene scrubber was used. Overall results showed that the combination of the gas permeable film and the ethylene absorbent could be applied to mature-green Mume fruits as an effective packaging method to retard the texture softening during storage at the ambient temperature.

  • A new second generation advanced thermal imager, which can be used for battle tank sight has been developed by ADD. This system uses a $480\times6$ TDI HgCdTe detector, operating in the $7.7-10.3{\mu}m$ wavelength made by Sofradir. The IR optics has dual field of views such as $2.67\times2^{\circ}$ in NFOV and $10\times7.5^{\circ}$ in WFOV. And also, this optics is used for athermalization of the system. It is certain that our sensor can be used in wide temperature range without any degradation of the system performance. The scanning system to be able to display 470,000 pixels is developed so that the pixel number is greatly increased comparing with the first generation thermal imaging system. In order to correct non-uniformity of detector arrays, the two point correction method has been developed by using the thermo electric cooler. Additionally, to enhance the image of low contrast and improve the detection capability, we have proposed the new technique of histogram processing being suitable for the characteristics of contrast distribution of thermal imagery. Through these image processing techniques, we obtained the highest quality thermal image. The MRTD of the LWIR thermal sight shows good results below 0.05K at spatial frequency 2 cycles/mrad at the narrow field of view.

Molecular Characterization of a Novel 1,3-α-3,6-Anhydro-L-Galactosidase, Ahg943, with Cold- and High-Salt-Tolerance from Gayadomonas joobiniege G7

  • 1,3-α-3,6-anhydro-L-galactosidase (α-neoagarooligosaccharide hydrolase) catalyzes the last step of agar degradation by hydrolyzing neoagarobiose into monomers, D-galactose, and 3,6-anhydro-L-galactose, which is important for the bioindustrial application of algal biomass. Ahg943, from the agarolytic marine bacterium Gayadomonas joobiniege G7, is composed of 423 amino acids (47.96 kDa), including a 22-amino acid signal peptide. It was found to have 67% identity with the α-neoagarooligosaccharide hydrolase ZgAhgA, from Zobellia galactanivorans, but low identity (< 40%) with the other α-neoagarooligosaccharide hydrolases reported. The recombinant Ahg943 (rAhg943, 47.89 kDa), purified from Escherichia coli, was estimated to be a monomer upon gel filtration chromatography, making it quite distinct from other α-neoagarooligosaccharide hydrolases. The rAhg943 hydrolyzed neoagarobiose, neoagarotetraose, and neoagarohexaose into D-galactose, neoagarotriose, and neoagaropentaose, respectively, with a common product, 3,6-anhydro-L-galactose, indicating that it is an exo-acting α-neoagarooligosaccharide hydrolase that releases 3,6-anhydro-L-galactose by hydrolyzing α-1,3 glycosidic bonds from the nonreducing ends of neoagarooligosaccharides. The optimum pH and temperature of Ahg943 activity were 6.0 and 20℃, respectively. In particular, rAhg943 could maintain enzyme activity at 10℃ (71% of the maximum). Complete inhibition of rAhg943 activity by 0.5 mM EDTA was restored and even, remarkably, enhanced by Ca2+ ions. rAhg943 activity was at maximum at 0.5 M NaCl and maintained above 73% of the maximum at 3M NaCl. Km and Vmax of rAhg943 toward neoagarobiose were 9.7 mg/ml and 250 μM/min (3 U/mg), respectively. Therefore, Ahg943 is a unique α-neoagarooligosaccharide hydrolase that has cold- and high-salt-adapted features, and possibly exists as a monomer.

Postmortem Changes in Muscle of Sea Water Acclimated Tilapia, Oreochromis niloticus (해수순치한 틸라피아 근육의 사후변화)

  • Cultivated tilapia (Oreochromis niloticus) in the fresh water were acclimated to the sea water to improve palatability of the fish meat. Physicochemical properties in the rigor mortis of those fish meats were investigated during storage at $0^{\circ}C,\;10^{\circ}C\;and\;20^{\circ}C$. The faster onset of rigor mortis was occurred in acclimated meat than fresh water cultivated meat. Both meats stored at $0^{\circ}C$ showed faster figro mortis than at $10^{\circ}C\;and\;20^{\circ}C$. Significant difference was not observed between the breaking strength and the rigor index. The breaking strength reached maximum over 12hrs after death and then gradually declined, and the rigor index was slowly increased and reached maximum over 18hrs after postmortem. Low temperature and acclimation to the sea water affected the degradation of adenosine triphosphate (ATP), accumulation of inosine monophosphate (IMP) or lactate. These results suggest that the palatability of tilapia muscle cultivated in the fresh water could be improved by acclimation to the sea water which induces the prerigor at the early state of postmortem and the physical changes of fish muscle.

Degradation from Polishing Damage in Ferroelectric Characteristics of BLT Capacitor Fabricated by Chemical Mechanical Polishing Process (화학적기계적연마 공정으로 제조한 BLT Capacitor의 Polishing Damage에 의한 강유전 특성 열화)

  • (Bi,La)$Ti_3O_{12}$(BLT) thin film is one of the most attractive materials for ferroelectric random access memory (FRAM) applications due to its some excellent properties such as high fatigue endurance, low processing temperature, and large remanent polarization [1-2]. The authors firstly investigated and reported the damascene process of chemical mechanical polishing (CMP) for BLT thin film capacitor on behalf of plasma etching process for fabrication of FRAM [3]. CMP process could prepare the BLT capacitors with the superior process efficiency to the plasma etching process without the well-known problems such as plasma damages and sloped sidewall, which was enough to apply to the fabrication of FRAM [2]. BLT-CMP characteristics showed the typical oxide-CMP characteristics which were related in both pressure and velocity according to Preston's equation and Hernandez's power law [2-4]. Good surface roughness was also obtained for the densification of multilevel memory structure by CMP process [3]. The well prepared BLT capacitors fabricated by CMP process should have the sufficient ferroelectric properties for FRAM; therefore, in this study the electrical properties of the BLT capacitor fabricated by CMP process were analyzed with the process parameters. Especially, the effects of CMP pressure, which had mainly affected the removal rate of BLT thin films [2], on the electrical properties were investigated. In order to check the influences of the pressure in eMP process on the ferroelectric properties of BLT thin films, the electrical test of the BLT capacitors was performed. The polarization-voltage (P-V) characteristics show a decreased the remanent polarization (Pr) value when CMP process was performed with the high pressure. The shape of the hysteresis loop is close to typical loop of BLT thin films in case of the specimen after CMP process with the pressures of 4.9 kPa; however, the shape of the hysteresis loop is not saturated due to high leakage current caused by structural and/or chemical damages in case of the specimen after CMP process with the pressures of 29.4 kPa. The leakage current density obtained with positive bias is one order lower than that with negative bias in case of 29.4 kPa, which was one or two order higher than in case of 4.9 kPa. The high pressure condition was not suitable for the damascene process of BLT thin films due to the defects in electrical properties although the better efficiency of process. by higher removal rate of BLT thin films was obtained with the high pressure of 29.4 kPa in the previous study [2].

Effect of Thermal History on the Physical Properties of Nylon66 (열 이력이 나일론66의 물성에 미치는 영향)

  • Nylon66 extrudates as a function of the extrusion number were prepared by a twin screw extruder. Chemical structures, thermal properties, melt index, crystal structures, mechanical properties such as the tensile strength, elongation at break and impact strength, and rheological property were measured by FT-IR, $^1H$-NMR, melt indexer, DSC, TGA, XRD, universal tensile tester, Izod impact tester, and rheometer. FT-IR and $^1H$-NMR characterizations indicated that the number of extrusions did not affect the chemical structure. The decrease in the molecular weight was checked by the melt index of extrudates. There were no effects of the thermal history on the melting and degradation temperature. The tensile and impact strength and modulus were found to be similar, regardless of the number of extrusions, but the elongation decreased significantly. The complex viscosity of extrudates at low frequencies decreased with the extrusion number. No structural changes after extrusion were confirmed from the fact that there was no change in the slope and shape of G'-G" plot.

Incubation conditions affecting biogenic amines degradation of probiotic lactic acid bacteria (프로바이오틱 유산균의 바이오제닉 아민 분해능에 영향을 미치는 배양 조건)

  • The purpose of this study was to investigate the inhibitory effect of antibacterial substances produced by probiotic lactic acid bacteria (LAB) against biogenic amines-producing bacteria and the influence of culture conditions on the antibacterial activity of bacteriocin and organic acid. The bacteriocin solutions of Lactobacillus plantarum FIL20 (64 AU/ml) and Lactobacillus paracasei FIL31 (128 AU/ml) showed strong antibacterial activity against Serratia marcescens CIH09 and Aeromonas hydrophilia RIH28, respectively. And the lactic acid contents in the cell-free culture supernatants (CFCS) obtained from FIL20 and FIL31 strains were $107.3{\pm}2.7mM$ and $129.5{\pm}4.6mM$, respectively. Therefore, the bacteriocin solution (200 AU/ml) and the CFCS ($200{\mu}l/ml$) produced by L. plantarum FIL20 and L. paracasei FIL31 significantly (P < 0.05) decreased the bacterial numbers and histamine and tyramine production ability of S. marcescens CIH09 and A. hydrophilia RIH28. The amounts of histamine and tyramine produced by the CIH09 strain under conditions of low initial pH (5.0) and incubation temperature ($15^{\circ}C$) was significantly reduced by treatment with bacteriocin solution and CFCS obtained from L. plantarum FIL20. In addition, the bacterial counts and biogenic amines contents of CIH09 strain were significantly decreased (P < 0.05) when sodium chloride (5%) or potassium nitrite (200 mg/g) were mixed with the antibacterial substances of L. plantarum FIL20. Consequently, the bacteriocin and organic acid solution of L. plantarum FIL20 and L. paracasei FIL31 can be used as a biological preservation to effectively control the production of biogenic amines by the application of hurdle technology.
