Establishment of Microbial Criteria by Investigation of Microbial Contamination in Ready-to-Eat Foods (즉석섭취·편의식품류의 미생물 오염도 조사를 통한 기준·규격 재평가)

  • Internationally different food safety regulation and standards could cause troubles in trade unless those are built on scientific knowledge. In this research, we monitored the microbial population and analyzed the results to determine the level of microbial contamination in foodstuffs using relatively new statistical analysis (microbiological sampling plan, International Commission on Microbiological Specification for Foods). The goal of this research falls on establishing entirely new standards for various food categories addressed in the Korean "Food Code". Targets for monitoring were indicator organisms (i.e. total aerobic count, coliform and Escherichia coli) and foodborne pathogens (i.e. Bacillus cereus, Staphylococcus aureus and Clostridium perfringens) in ready-to-eat (RTE) products. As the result of the monitoring, total aerobic count, coliform, E. coli, and B. cereus in RTE products were found at the mean values of 2.10 log CFU/g, -0.60 log CFU/g, -1.33 log CFU/g and -1.23 log CFU/g, respectively. S. aureus was detected with the level of -1.35 log CFU/g only in fresh-cut food, while C. perfringens was -1.37 log CFU/g only in ready-to-cook food. Other samples did not have any food borne pathogens. Total aerobic count, B. cereus, S. aureus and C. perfringens satisfied the Food Code (the MFDS). On the basis of the analysis, we proposed a draft of microbial criteria for RTE products.

Rhizosphere Inhibition of Cucumber Fusarium Wilt by Different Surfactinexcreting Strains of Bacillus subtilis

  • Bacillus subtilis B006 strain effectively suppresses the cucumber fusarium wilt caused by Fusarium oxysporum f. sp. cucumerinum (Foc). The population dynamics of Foc, strain B006 and its surfactin over-producing mutant B841 and surfactin-deficient mutant B1020, in the rhizosphere were determined under greenhouse conditions to elucidate the importance of the lipopeptides excreted by these strains in suppressing Foc. Results showed that B. subtilis strain B006 effectively suppressed the disease in natural soil by 42.9%, five weeks after transplanting, whereas B841 and B1020 suppressed the disease by only 22.6% and 7.1%, respectively. Quantitative PCR assays showed that effective colonization of strain B006 in the rhizosphere suppressed Foc propagation by more than 10 times both in nursery substrate and in field-infected soil. Reduction of Foc population at the cucumber stems in a range of $0.96log_{10}ng/g$ to $2.39log_{10}ng/g$ was attained at the third and the fifth weeks of B006 treatment in nursery substrate. In field-infected soil, all three treatments with B. subtilis suppressed Foc infection, indicated by the reduction of Foc population at a range of $2.91log_{10}ng/g$ to $3.36log_{10}ng/g$ at the stem base, one week after transplanting. This study reveals that the suppression of fusarium wilt disease is affected by the effective colonization of the surfactin-producing B. subtilis strain in the rhizosphere. These results improved our understanding of the biocontrol mechanism of the B. subtilis strain B006 in the natural soil and facilitate its application as biocontrol agent in the field.

Studies on Dorsal Aspect Target Strengths of Rock Bream, Oplegnathus Fasciatus and Dusky Spinefoot, Siganus Fuscescens (돌돔과 독가시치의 등방향 반사강도에 관한 연구)

  • In order to obtain fundamental data for estimation of fisheries resource by echo sounder, we carried out the measuring of dorsal aspect Target strengths for rock bream and dusky spinefoot fishes that were caught much around the Jeju Island and in South Sea of Korea. The appropriate equations share the common form. TS=A+20 log L, where TS is the average dorsal aspect target strength in decibels, L is the fish total length in centimeters, and the coefficient A is determined by a least mean squares regression analysis. For rock bream, the result is TS=-72.97+20 log L and, for dusky spinefoot it is TS=-63.16+20 log L And, we have investigated the bearing range of maximum dorsal aspect target strength for all of rock bream and dusky spinefoot by the echo sounder with transducer of which frequency is 200kHz. They are $-12^\circ$-$-21^\circ$and $-1^\circ$--8 espectively, when the fishes is swimming down to the bottom. The maximum dorsal target strengths are -41.50dB at -18 or rock bream and -30.69dB at $-6^\circ$for dusky spinefoot.

Influence of the Mechanical Properties on Drapability in the Peach Skin-like Finished Fabrics (피치스킨 가공직물의 역학적 특성이 드레이프성에 미치는 영향)

  • The purpose of this study was to investigate the influence of the mechanical properties on drapability in the peach skin-like finished fabrics. For this study, the samples used were 50 kinds of peach skin-like finished fabrics. The mechanical properties such as tensile, shearing, bending, compressional, surface characteristic values, thickness and weight were measured with a KES-F system and drupe coefficient by drape tester. The relationship between the characteristic values and drape coefficicients of the peach skin-like finished fabrics results were obtained. 1. Peach skin-like finished fabric had $\pm$2o range of shearing, bending, compression, surface. properties, thickness and weight as compared with Japanese women's thin fabrics. The characteristic mixing values were better with the values of WC/T, W/T, etc. as compared with that of japanese women's thin fabrics. Accordingly, the peach skin-like finished fabrics had a little volume, excellent hanging and drapability as compared with japanese women's thin fabrics. 2. The drape coefficient of peach skin-like finished fabric had a high level of correlation between 2HB, G, WC, MIU, WT, MMD, 2HG, RT, W, B etc. of the mechanical properties. The blocked properties that contributed to the drape coefficient of peach skin-like finished fabrics were in the order of the bending> tensile> thickness> weight properties. This drape coefficients(DC) were found by measuring the mechanical properties according to the obtained regress on equate on. DC=99.0179+17.9023 log G -17.0543 log 2HG5+17.2104 log 2HG+35.7685 log 2HB+ 4.6082 log B-30.5906 log T+4.2308 log W 3. The contribution to the drape coefficient of the characteristic mixing values of peach skin-like finished fabric was in the order of > 2HB/W> 2HB/B> B/W The drape coefficients were found by measuring the characteristic mixing values according to the obtained regression equation. 4. The drape coefficients of peach skin-like finished fabrics were influenced by the differences between the bending of warp direction, bending of weft direction, shearing etc which in turn determine the level of hanging. The regression equation was as follows; 5. The drape coefficients of peach skin.like finished fabrics had a highly positive correlation with the node index. It has an negative correlation with number of nodes.

Studies on the Efficient Improvement of Measurement Methods of Stand Volume (임분재적(林分材積) 측정법(測定法)의 효율적(效率的) 개선방안(改善方案)에 관(關)한 연구(研究))

  • The purpose of this study is to develop the method of stand volume estimation by the plotless sampling method. The required data were obtained from 164 sampling plots in the red pine(Pinus densiflora) stands which were located in Kyeong-gi, Chung-nam, Chung-buk and Kang-won areas, and related factors were measured actually. The method of stand volume estimation and several tables were drivel from these data. 1. The relationship between the values of stand average height, basal area per ha, and basal area height obtained from the plotless sampling method and values measured actually could be described by the equation Y=bx, where b approached nearly 1.0 and there were no significant differences between them. Therefore stand volumes could be estimated by the plotless sampling method. 2. The estimated equations of the stand voulumes, which were estimated using factors to be measured by dendrometer, are as follows ; logV=-0.0208+0.8497 logGH, logV=-0.0028+0.7981 logG+0.9313 logH. Stand volume tables by these estimated equations were shown in table 4, 5 and estimation error percentages were 9.16% and 8.50% respectively. Effects of Preprocessing on Quality of Fermented Red Snow Crab Chionoecetes japonicus Sauce (전처리 방법에 따른 홍게(Chionoecetes japonicus) 어간장의 제조 및 품질변화)

  • We explored preprocessing-mediated quality changes in red snow crab fish sauce. A control (C) group and groups treated with autolysis (A), boiling (B), enzymatic hydrolysis (E), and addition of Aspergillus oryzae (K) were formed. The titratable acidity of the K group increased with storage time, whereas that of groups C, A, B, and E decreased. The total and amino nitrogen contents initially increased on storage of all samples, but decreased in later periods. The total plate count (TPC) of the K group was initially 5.26 log CFU/mL and increased to 7.28 log CFU/mL at 3 months of storage. The TPCs of the C, A, B, and E groups were initially <5.00 log CFU/mL and decreased with storage. The lactic acid bacteria count of the K group was initially 4.80 log CFU/mL and increased until month 5 to approximately 6.06 log CFU/mL. The K group scored higher in terms of sensory attributes than the other groups and maintained marketable scores for all relevant properties (color, flavor, off-odor, and overall acceptance). Furthermore, the free amino acid content of the K group was the highest among all groups at approximately 3,000 mg per 100 g. These results suggest that K treatment may be beneficial in the preparation of fermented fish sauce.

Assessment of Contamination Levels of Foodborne Pathogens Isolated in Major RTE Foods Marketed in Convenience Stores (편의점에서 판매중인 주요 RTE food 중 위해미생물의 오염도 평가)

  • Contamination levels of total aerobic bacteria, coliforms, Escherichia coli, Staphylococcus aureus, and Bacillus cereus In commercial Samgakkimbabs and sandwiches from southern Gyeounggi-do were monitored. Total aerobic bacteria counts in Samgakkimbabs and sandwiches were 3.50-5.54 and $3.88-6.29log_{10}CFU/g$, for coliforms 1.25-317 and $1.53-5.08log_{10}CFU/g$, for S. aureus 0.30-5.20 and $0.10-4.18log_{10}CFU/g$, and for B. cereus 0.88-2.48 and $0.22-2.18log_{10}CFU/g$, respectively. E. coli was not isolated from all Samgakkimbabs and sandwiches except from one sample of sandwich salad ingredient. Results indicate hygiene of commercial Samgakkimbabs and sandwiches was deleterious.

The relationships between lead exposure indicies and urinary δ-ALA by HPLC and colorimetric method in lead exposure workers (연노출근로자에 있어서 흡광광도법과 HPLC법에 의한 요중 δ-ALA 배설량과 연노출지표들 간의 관련성)

  • In order to compare the difference of the measurement of delta aminolevulinic acid(${\delta}$-ALA) in urine between HPLC method(HALA) and colorimetric method(CALA), and also to provide useful information for the new diagnostic criteria of ${\delta}$-ALA in urine in lead poisoning, if at all possible in the future, authors studied 234 male lead workers who were selected from 7 storage battery factories, 3 secondary smelting industries, and 2 litharge making industries. Study subjects were selected on the basis of blood Zinc protoporphyrin(ZPP) level from low to high concentration to cover wide range of lead exposure. Study variables for this study were ${\delta}$-ALA measured by two different methods, blood lead(PbB), and blood ZPP. The results were as follows: 1. There was very high correlation between ${\delta}$-ALA measured by two method(r = 0.989 : HALA = -0.8194 + 0.8110 ${\times}$ CALA), but the value of CALA was measured about 2mg/L greater than HALA. 2. While the correlations of ${\delta}$-ALA by two method with blood lead and blood ZPP were 0.46 and 0.37 respectively, they were increased to 0.63 and 0.57 if ${\delta}$-ALA values were log-transformed. 3. Simple linear regression of ${\delta}$-ALA measured by two method on ZPP were as follows: CALA = 2.0421 + 0.0341 ${\times}$ ZPP ($R^2=0.1385$ p = 0.0001) HALA = 0.8006 + 0.0280 ${\times}$ ZPP ($R^2=0.1389$ p = 0.0001) 4. Simple linear regression of ${\delta}$-ALA measured by two method on PbB were as follows: CALA = - 0.4134 + 0.1545 ${\times}$ PbB ($R^2=0.2085$ p = 0.0001) HALA = -1.2893 + 0.1287 PbB ($R^2=0.2154$ p = 0.0001), 5. Simple linear regression of log-transformed ${\delta}$-ALA by two method on ZPP and PbB were as follows: logHALA = 0.3078 + 0.0060 ZPP ($R^2=0.3329$ p = 0.0001) logCALA = 1.0189 + 0.0044 ZPP ($R^2=0.3290$ p = 0.0001) logHALA = -0.0221 + 0.0246 PbB ($R^2=0.4046$ p = 0.0001) logCALA = 0.7662 + 0.0184 PbB ($R^2=0.4108$ p = 0.0001) 6. The cumulative percent of colorimetric method to detect lead workers whose value of PbS and ZPP were over screening level such as $40{\mu}/dl$ and $100{\mu}/dl$ respectively was higher than HPLC method if cut-off level of ${\delta}$-ALA for screening of lead poisoning was 5 mg/L. But if cut-off level of ${\delta}$-ALA measured by HPLC was reduced to 3 mg/L which is compatible to 5 mg/L of ${\delta}$-ALA measured by colorimetric method, there were good agreement between two methods and showed dose-response relationship with other lead exposure indices such as PbB and ZPP.

Effects of Beneficial Microorganisms and Mycorrhizal Fungus Colonized Rhizoplane on the Suppression of Root Rot Pathogen, Fusarium solani (근면 정착 유용 미생물과 균근균이 근부병원균, Fusarium solani의 발병억제에 미치는 영향)

  • The survival or colonization of beneficial organsisms and suppression of root rot of ginseng (Panax ginseng) by two distinct bacteria, Pseudomonas cepacia, Bacillus cereus and three mycorrhiza in pot soil were investigated and compared with uninoculated root. In separate inoculation, colonization of roots by P. cepacia was maintained at 6.25 (log cfu/g root) during growth for 10 days under pot culture conditions comparing to $5.62{\sim}6.19$ by mixed treatment with other organisms. Colonizations of P. cepacia were gradually decreased from 6.25 (log cfu/g root) in 10 days growth to 3.01 (log cfu/g root) in 270 days incubation period. This reduction was also investgated in combination treatments by B. cereus or F. solani. The numbers of Fusarium spp. were colonized high number in rhizosphere soil from 3.33 to 3.67 (log cfu/g root) in control within $10{\sim}60$days after treatment of pathogen F. solani, but it's numbers were markedly decreased in 270 days cultivation of plant from 3.33 to 1.02 (log cfu/g root) after treatment. In treatment of beneficial strains of P. cepacia and B. cereus, P. cepacia significantly suppressed the development of root rot from 4.3 in control to 1.2 in treatment, whereas B. cereus alone had no effect on the rate of disease suppression. The disease index $(1.8{\sim}2.3)$ in combination of two bacteria was reduced in plants inoculated with both P. cepacia and B. cereus comparing to the index (4.3) of control. As an effect of inoculation with mycorrhiza on disease suppression, suppression of root rot by F. solani was reduced to $1.2{\sim}1.6$ in disease index in treatment of Glomus albidum and Acaulospora longular comparing to 4.3 of control. In the treatment of bacterial strain P. cepacia and mycorrhizal fungus Glomus albidum, the disease suppression was apparent to 1.2 and 1.2 comparing to 4.3 of control in disease index respectively.

  • In this paper, a multi-velocity log system which uses ultrasonic pulsed doppler signal is developed. The output of the system is the absoluted velcocity of the ship. By using digitral signal processing, we get the vector velocity which displays the ship's speed about fore, aft, port and starboard. And this system give us the information about depth. This multi-velocity system has a large merit that is bottom and water track velocity. In addition, this has the high accuracy and can measure the water-depth according to the deep mode. And the fish finder, echo sounder or docking system will be made by applying the speed log system.
