Effect of Artemisia capillaries on Gene Expression of Lipid Metabolism in Rat (인진호(茵蔯蒿)가 지질대사에 관여하는 유전자의 발현에 미치는 영향)

  • Objectives : The purpose of this study is to evaluate the effect of Artemisia capillaries on gene expression of lipid metabolism in rats. Methods : The author performed several experimental items to analyze the total cholesterol and triglyceride in liver tissue, the gene expressions of CYP7A1 and HMG-CoA reductase. Results : 1. In Artemisia capillaries group, the levels of total cholesterol in liver tissue were significantly decreased. 2. In Artemisia capillaries group, the ratios of CYP7A1, HMG-CoA reductase were as same as the normal group. Conclusions : From the above results, Artemisia capillaries can be used to treat hyperlipidemia.

Imprint Cytology of Hepatic Angiomyolipoma -A Case Report - (간에 발생한 혈관근육지방종의 압착도말 세포 소견 -1예 보고-)

  • Angiomyolipoma of the liver is a rare benign tumor that's composed of variable mixtures of adipose tissue, smooth muscle and thick-walled blood vessels. We report here on the imprint cytologic features of a hepatic angiomyolipoma in a 47-year-old man. The smears showed spindle and epithelioid tumor cells in clusters, trabeculae and single cells. The spindle cells had elongated, cigar-shaped nuclei with finely granular chromatin and fibrillary cytoplasm. The epithelioid cells had round nuclei with a moderate amount of cytoplasm. Any adipose tissue was not found. Immunohistochemically, both the spindle and epithelioid cells revealed cytoplasmic positivity for smooth muscle actin and HMB-45.

  • Permanent cell culture lines derived from human cancer tissue are important experimental models in the study of human cancer cell proliferation. The in vitro effects of C. militaris and its extracted fractions on the human breast cancer (SK-BR3), liver cancer (SK-Hep1, HepG2), kidney cancer (p15), lymphoma (Jurkat) were studied. F1 (CCCA, crude cordycepin containing adenosine), F2 (ethanol precipitation), F3 (ethanol soluble supernatant) and F4 (fraction of through SK-1B) significantly stimulated in vitro cytotoxic in human cancer cell lines. Effect of Acute Heat Stress on Heat Shock Protein 70 and Its Corresponding mRNA Expression in the Heart, Liver, and Kidney of Broilers

  • The objective of this study was to investigate the expression and localization of heat shock protein 70 (Hsp70) and its mRNA in the heart, liver, and kidney of acutely heat-stressed broilers at various stressing times. Male AA broilers (n = 100) were randomly divided into 5 groups of 20 birds per group. After 30 d of adaptive feeding at ambient temperature, 80 experimental broilers were suddenly heat stressed by increasing the environmental temperature from $22{\pm}1^{\circ}C$ to $37{\pm}1^{\circ}C$. The 4 groups were heat stressed for 2, 3, 5, and 10 h, respectively. The localizations of Hsp70 protein and mRNA, determined by immunohistochemical staining and in situ hybridization, respectively, were demonstrated to be tissue dependent, implying that different tissues have differential sensibilities to heat stress. Intense Hsp70 staining was identified in the vascular endothelial cell of heart, liver and kidney, suggesting an association between expression of Hsp70 in vascular endothelial cell and functional recovery of blood vessels after heat shock treatment. Ante-mortem heat stress had a significant effect on the expression of Hsp70 protein and mRNA. The quantitation of Hsp70 protein and mRNA were both time and tissue dependent. During the exposure to heat stress, the heart, liver and kidney of broiler chickens exhibited increased amounts of Hsp70 protein and mRNA. The expression of hsp70 mRNA in the heart, liver and kidney of heat-stressed broilers increased significantly and attained the highest level after a 2-h exposure to elevated temperatures. However, significant elevations in Hsp70 protein occurred after 2, 5, and 3 h of heat stressing, respectively, indicating that the stress-induced responses vary among different tissues.

The Anti-obesity Effect of Seungyangjeseup-tang for High Fat Diet Induced Obese Mice (고지방식이 유도 비만 생쥐에 대한 승양제습탕의 항비만 효과)

  • Objectives This study was designed to evaluate the efficacy of Seungyangjeseup-tang on obesity by using 3T3-L1 cells and high fat diet mice. Methods In vitro, Seungyangjeseup-tang extract (SYJST) (10, 50, 100, 200, 400, $800{\mu}g/mL$) ware added in 3T3-L1 cells. SYJST cytotoxicity was measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assasy. Adipocyte differentiation was measured by Oil Red-O staining. In vivo, the experimental animals were divided into five groups: normal diet-fed normal group (N), high-fat diet (HFD)-fed control group (Con), HFD+SYJST 100 mg/kg group (SY100), HFD+SYJST 200 mg/kg group (SY200), and HFD+olistat 60 mg/kg as a positive drug control group (Orli). Markers of obesity, such as body weight, liver weight, food intake, serum total cholesterol (TC), triglycerides (TG), high density lipoprotein cholesterol (HDL-C), liver tissue TC, TG and fecal TC, TG were measured. Results In vitro, cytotoxicity was not significant compared with the control group. 3T3-L1 cell's differentiation was significantly decreased in Oil Red-O staining. In vivo, compared with controls, mice treated with SYJST demonstrate lower body and liver weight, and reduced food intake. In addition, SYJST increased TC, TG in the serum but not significance. And SYJST showed decreasing tendency TC, TG in the liver tissue. Furthermore, SYJST increased TC, TG in the facal but not significance. Conclusions Based on the results above, Seungyangjeseup-tang may reduce adipocyte differentiation, body fat, food intake, liver weight in obesity. This suggests that Seungyangjeseup-tang may be clinically useful in obesity treatment.

Effects of BCL oral administation and herbal acupuncture at BL18, BL19 on Liver function changes induced by Alcohol in the mice (죽력(竹瀝)(죽초액(竹酢液)) 경구투여(經口投與)와 간유(肝兪) 담유(膽兪) 약침(藥鍼)이 Alcohol 대사(代謝) 및 간기능(肝機能)에 미치는 영향(影響))

  • This dissertation was designed to evaluate the effect of BCL(refinded Bambusae Caulis in Liqua-men) oral administration and herbal acupuncture on alcohol metabolism and liver function. For this study. mice were damaged by a large quantity of alcohol and received treatment of either BCL 1 mg/kg in oral or BCL 250㎍/kg in herbal acupuncture-BL18 . BL19 bilateral. and then such parameters as GOT. GPT. catalase and superoxide dismustase(CuZn-SOD, Mn-SOD) were measured. The results of the experiments were summarized as follows. 1. Compared with control group, the proper degree of alcohol in serum was not significantly differ from oral administration group and herbal acupuncture group. 2. Compared with control group. the activity of GOT in serum was significantly reduced both oral administration and herbal acupuncture group. 3. Compared with control group. the activity of GPT in serum was significantly reduced both oral administration and herbal acupuncture group. 4. The activity of catalase in liver cell tissue, compared with control group. was not sigificantly affected either by oral administration and herbal acupuncture group. 5. The activity of CuZn-SOD in liver cell tissue was not significantly change in herbal acupuncture and oral administration group. The activity of Mn-SOD was significantly increased in oral administration group. while it was not the case in acupuncture group. In conclusion. we consider that BCL oral administration and herbal acupuncture is highly effetive in recovering alcohol metabolism and liver disfunction induced by alcohol.

Effects of grape pomace on the antioxidant defense system in diet-induced hypercholesterolemic rabbits

  • The effects of grape seeds extract and grape peels extract prepared from grape pomace on the activity of antioxidant enzymes, degree of lipid peroxidation in serum and liver tissue were investigated in rabbits fed on high cholesterol diet. New Zealand white rabbits were divided as follows ; 1) NOR (normal group); 2) CHOL (cholesterol group); 3) GSH (cholesterol + grape seed extract group); 4) GPE (cholesterol + grape peel extract); 5) GSP (cholesterol + grape seed powder); 6) GPP (cholesterol + grape peel powder); 7) GE (cholesterol + grape seed and peel extract); 8) GP (cholesterol + grape seed and peel powder). Eight groups of rabbits were studied for 8 weeks. At the end of the experimental period, rabbits were sacrificed and the liver tissue were removed. Then, GSH, GPx, GST, CAT and MDA in the liver were measured. In liver tissues, total glutathione contents (GSH), glutathione peroxidase (GPx) and catalase (CAT) activity, which was significantly higher by grape seed extract supplementation. The level of malondialdehyde (MDA) was lower in the serum of rabbits fed grape seed extract or grape peel powder plus cholesterol than in the serum of rabbits fed cholesterol alone. It is therefore likely that grape seed extract prepared from grape pomace functioned as antioxidants in vivo, negating the effects of the oxidative stress induced by 1% cholesterol diet. The grape seed extract was found effective in converting the oxidized glutathione into reduced glutathione, and in removing $H_2O_2$ that is created by oxidative stress. The grape peel powder was found to have small influence on reduced glutathione content, CAT and GPX activity, but it increased GST activity in liver tissues, resulting in promoting the combination of lipid peroxide and glutathione (GSH), and further, lowering the formation of lipid peroxide in the serum. Therefore, grape pomace (grape seed extract and grape peel powder) supplementation is considered to activate the antioxidant enzyme system and prevent damage with hypercholesterolemia.

Antioxidant Effects of Sagunja-Tang (Sijunzi-Tang) (사군자탕(四君子湯)의 항산화(抗酸化) 효과(效果))

  • Objectives : This study was carried out to research antioxidant effects of Sagunja-Tang(SA) through in vitro and vivo experiments, and tried to investigate the relation between oxidation of tissues and deficiency of Qi. Methods and results : HPLC analysis of glycyrrhizine - known to be the main compound of Radix Glycyrrhizae - was done to certify the quality of SA. Chemiluminescence was initiated by adding tort-butyl hydroperoxide (t-BHP) to rabbit polymorphonuclear leukocytes (neutrophils), and generated reactive oxygen species (superoxide anion) decreased significantly by SA as dose dependent manner. Cell injury during 60 minutes tissue incubation was initiated by adding t-BHP, a hydrophobic hydroperoxide and $H_2O_2$, an water soluble oxidant to rat renal cortical and liver slices. Percentage cell death and lipid peroxidation were estimated by measuring lactate dehydrogenase (LDH) and malondialdehyde (MDA), a product of lipid peroxidation. t-BHP induced % cell death of renal cortical slices and lipid peroxidation of renal cortical and liver slices were decreased significantly by SA. SA decreased significantly % cell death and lipid peroxidation of renal cortical and liver slices induced by $H_2O_2$, too. Acute renal and liver injury induced by $HgCl_2\;and\;CCl_4$, which initiated from free radical, were applied to mice and metabolic data were obtained. Data showed protective effects of SA on acute renal injury caused by decrease of glomerular filtration. SA protected acute liver injury too. Conclusions Through this study, we found that SA have antioxidant effects and tissue oxidation was similar to deficiency of Qi. And further studies have to be followed to certify the mechanisms.

Effects of Dietary Copper Exposure on Accumulation and Histopathological Change in Liver of Juvenile Rockfish, Sebastes schlegeli

  • Experiments were carried out to investigate the accumulation and the histopathological changes in liver of juvenile rockfish, S. schlegeli, after sub-chronic dietary Cu (0, 50, 125, 250 and 500mg/kg) exposure for 60 days. Cu accumulation in liver was significantly increased with dietary exposure period and concentration for 60 days, and has a linear relation with dietary exposure days. After 60 days of Cu dietary exposure, the Cu concentration in the liver was $75.9\pm12.05,\;126.29\pm22.11\;and\;360.44\pm45.26\;{\mu}g/g$ dry weight and was approximately 11-fold, 18-fold and 51-fold higher than in the control diet group at 125, 250 and 500 mg/kg Cu diet group. The accumulation factors were increased with the dietary exposure period in liver of rockfish. In the primary exposed stage, the effect of hepatic tissue in the rockfish exposed to dietary Cu observed enlargement of hepatocytes nuclei, activity of hepatic cells and the swelling of hepatic cells. While exposed time and concentration were increased, the distinct granulation, irregular shape and necrosis of hepatic cells were observed. It was observed that granule degeneration and necrosis showed a part of cells in hepatic tissue after 60 days at 500 mg/kg.

Effects of developmental iron deficiency and post-weaning iron repletion on the levels of iron transporter proteins in rats

  • BACKGROUND/OBJECTIVES: Iron deficiency in early life is associated with developmental problems, which may persist until later in life. The question of whether iron repletion after developmental iron deficiency could restore iron homeostasis is not well characterized. In the present study, we investigated the changes of iron transporters after iron depletion during the gestational-neonatal period and iron repletion during the post-weaning period. MATERIALS/METHODS: Pregnant rats were provided iron-deficient (< 6 ppm Fe) or control (36 ppm Fe) diets from gestational day 2. At weaning, pups from iron-deficient dams were fed either iron-deficient (ID group) or control (IDR group) diets for 4 week. Pups from control dams were continued to be fed with the control diet throughout the study period (CON). RESULTS: Compared to the CON, ID rats had significantly lower hemoglobin and hematocrits in the blood and significantly lower tissue iron in the liver and spleen. Hepatic hepcidin and BMP6 mRNA levels were also strongly down-regulated in the ID group. Developmental iron deficiency significantly increased iron transporters divalent metal transporter 1 (DMT1) and ferroportin (FPN) in the duodenum, but decreased DMT1 in the liver. Dietary iron repletion restored the levels of hemoglobin and hematocrit to a normal range, but the tissue iron levels and hepatic hepcidin mRNA levels were significantly lower than those in the CON group. Both FPN and DMT1 protein levels in the liver and in the duodenum were not different between the IDR and the CON. By contrast, DMT1 in the spleen was significantly lower in the IDR, compared to the CON. The splenic FPN was also decreased in the IDR more than in the CON, although the difference did not reach statistical significance. CONCLUSIONS: Our findings demonstrate that iron transporter proteins in the duodenum, liver and spleen are differentially regulated during developmental iron deficiency. Also, post-weaning iron repletion efficiently restores iron transporters in the duodenum and the liver but not in the spleen, which suggests that early-life iron deficiency may cause long term abnormalities in iron recycling from the spleen.
