The Roles of Arachidonic Acid and Calcium in the Angiotensin II-induced Inhibition of $Na^+$ Uptake in Renal Proximal Tubule Cells

  • Angiotensin II (ANG II) has a biphasic effect on $Na^+$ transport in proximal tubule: low doses of ANG II increase the $Na^+$ transport, whereas high doses of ANG II inhibit it. However, the mechanisms of high dose ANG II-induced inhibition on $Na^+$ uptake are poorly understood. Thus the aim of the present study was to investigate signal transduction pathways involved in the ANG II-induced inhibition of $Na^+$ uptake in the primary cultured rabbit renal proximal tubule cells (PTCs) in hormonally defined serum-free medium. ANG II $(10^{-9}\;M)-induced$ inhibition of $Na^+$ uptake was blocked by losartan $(10^{-8}\;M,\;AT_1\;antagonist),$ but not by PD123319 $(10^{-8}\;M,\;AT_2\;antagonist)$ (P<0.05). ANG II-induced inhibition of $Na^+$ uptake was also completely abolished by neomycin $(10^{-4}\;M,$ PLC inhibitor), W-7 $(10^{-4}\;M,$ calmodulin antagonist), and $AACOCF_3\;(10^{-6}\;M,\;PLA_2\;inhibitor)$ (P<0.05). ANG II significantly increased $[^3H]arachidonic$ acid (AA) release compared to control. The ANG II-induced $[^3H]AA$ release was blocked by losartan, $AACOCF_3,$ neomycin, and W-7, but not by PD123319. ANG II-induced $[^3H]AA$ release in the presence of extracellular $Ca^{2+}$ was greater than in $Ca^{2+}-free$ medium, and it was partially blocked by TMB-8 $(10^{-4}\;M,$ intracelluar $Ca^{2+}$ mobilization blocker). However, in the absence of extracellular $Ca^{2+},$ it was completely blocked by TMB-8. In addition, econazole $(10^{-6}\;M,$ cytochrome P-450 monooxygenase inhibitor) and indomethacin $(10^{-6}\;M,$ cyclooxygenase inhibitor) blocked ANG II-induced inhibition of $Na^+$ uptake, but NGDA $(10^{-6}\;M,$ lipoxygenase inhibitor) did not affect it. In conclusion, $PLA_2-mediated$ AA release is involved in ANG II-induced inhibition of $Na^+$ uptake and is modulated by $[Ca^{2+}]_i$ in the PTCs.

  • 1950-1960 년대의 녹색혁명은 1970년 Nobel 평화상수 상자 Norman Borlaug가 주도했었다. 제1 녹색혁명은 지구상의 가장 큰 문제점중의 하나인 기아를 해결하는지 크게 일조하였다. 많은 사람들을 굶주림으로부터 해방시켰지만 굶주리는 사람보다 더 많은 사람들이 매일 태어나고 있고, 지구의 인구증가는 지금까지 계속되고 있다. 언제까지 인구증가가 계속될 것인지, 인구증가에 따른 식량자원의 증가도 비례할 것인지는 오래전부터 인류의 관심의 대상이 되어 왔고, 자연스럽게 지구의 기아를 해결하는 제2의 녹색혁명은 과학자들의 연구를 집중시키는 결과를 낳게 되었다. 1980 년대에 미국 Monsanto 회사에서 유전공학적으로 개발한 Roundup-Ready 제초제저항성 및 Bt-살충 농작물을 선보이면서 제2의 녹색혁명이 시작되었다고 볼 수 있겠다. 이렇게 제2의 녹색혁명은 유전공학에 의한 CMO 식물$\cdot$작물에 의해 시작되었다. Monsanto 회사는 살충제 RoundUp 제초제 저항성 옥수수, 콩, 목화, Canola (유채꽃, rape seed) 등을 개발하여 이미 상업화하였다. 1960년대 쌀의 녹색혁명도 유전적으로 벼집이 짧은 품종에 의해 이루어졌다. 최근에 개발된 GMO "Golden Rice"는 비타민 A와 철분의 함량을 대폭 증대 시켜 세계 영양결핍 아동들의 건강과 시각을 향상시킬 것으로 기대되는데, "Golden Rice"는 제1회 금호국제과학상 수상자인 Potrykus (스위스공대) 교수가 개발하였다. 그러나 현재까지는 그 보급에 많은 장애물이 있다. 특히 GMO의 환경과 건강에 대한 안전성을 의심하는 사람들이 많다. 제2의 녹색혁명의 또 다른 분야는 식물의약 분야이다. GMO 개발에 적용되는 기술을 이용하면 taxol 같은 항암제, carotene 같은 항산화 영양제 등의 대량 생산이 가능해진다. 식물은 화학적 합성이 아주 까다로운 약제물질 등을 천연상태에서 합성하고 있기 때문이다. 또 식물은 lipoxygenase 효소계가 있어서 마치 천연물 석유제조공장과 같은 제조공정 capacity를 가지고 있다. 그러면 식물/식품 GMO는 안전한 것인가? 아니, KBS의 한 사회자가 말했듯이, 그리고 많은 소비자들이 믿는 것처럼 GMO는 위험한가? GMO에 대한 일반 사람들의 공포감은 Green Peace 당원들뿐만 아니라 일부 과학자들에 의해서도 조장되고 있다. 이러한 분위기 속에서 GMO에 의한 제2 녹색혁명은 Africa 대륙에서의 제1 녹색혁명이 지금도 지연되는 것과 같다고도 볼 수 있다. GMO의 환경에 대한 악영향은 과대 선전되어있는 것이 아닌가? 마치 GMO가 화학비료, 농약제보다 더 위험하다고 믿는 사람들도 많다. 나는 이러한 GMO 공포증이 과학적으로 그리고 "Risk Assessment"의 견지에서 볼 때 그 근거가 희박하다고 보여주는 몇 몇 실험 및 경험 사실들을 인용하려 한다. 그리고 올바른 Risk Assessment야 말로 한국의 21세기 BT 산업을 경쟁력 있게 하고 국민 년 소득 2만불 달성에 중요한 기여를 하게 될 것이라고 생각한다. 한국은 농토가 적고 천연자원이 빈약하다. GMO는 21세기의 생존 경쟁 산업이다. 제2의 녹색혁명은 얼마든지 가능하며, 한국은 부족한 농토와 빈약한 자원에도 불구하고 능력 있는 인적자원이 풍부하여 GMO 개발 연구에 국제적 경쟁력을 키울 수 있다. 그러나 GMO에 대한 논쟁만 하고 있으면 이미 때가 늦는다. 미국은 이미 GMO-BT 시장을 거의 완전 독점했으며, 타국에서의 논쟁과 불합리적으로 엄격한 GMO 관련 규정을 조장하고 환영한다.

Construction of Ovine Customer cDNA Chip and Analysis of Gene Expression Patterns in the Muscle and Fat Tissues of Native Korean Cattle (cDNA microarray를 이용하여 한우의 근육과 지방조직의 유전자 발현 패턴 분석 및 bovine customer cDNA chip 구성 연구)

  • To investigate the molecular events of controlling intramuscular fat (or marbling), which is an important factor in the evaluation of beef quality, we performed cDNA microarray analyses using the longissimus dorsi muscle and back fat tissues. For this study, we constructed normalized cDNA libraries: fat tissues in native Korean cattle (displaying 1,211 specific genes), and muscle tissues in native Korean cattle (displaying 1,346 specific genes). A bovine cDNA chip was constructed with 1,680 specific genes, consisting of 760 genes from muscle tissues and 920 genes from fat tissues. The microarray analysis in this experiment showed a number of differentially expressed genes, which compared the longissimus dorsi muscle (Cy5) with back fat tissue (Cy3). Among many specific differentially expressed genes, 12-lipoxygenase (oxidizing esterified fatty acids) and prostaglandin D synthase (differentiation of fibroblasts to adipocytes) are the key candidate enzymes that should be involved in controlling the accumulation of intramuscular fat. In this study, differentially and commonly expressed genes in the muscle and fat tissues of native Korean cattle were found in large numbers, using the hybridization assay. The expression levels of the selected genes were confirmed by semi-quantitative RT-PCR, and the results were similar to those of the cDNA microarray.

  • 20세기 말까지만 해도 경제발전은 자원고갈과 자연환경파괴를 통한 대량생산 및 대량소비를 전제로 하여 이루어 졌다고 평가할 수 있다. 그 결과 세계의 자연환경은 급속히 훼손되었으며, 환경오염은 이제 인류의 생존을 위협하는 단계에 이르렀다. 이 같은 위기상황에 대처하기 위하여 지난 1980년대부터 선진국을 중심으로 환경경영에 대한 인식이 확산되기 시작하였다. 무엇보다도 자연환경 의 보존을 요구하는 사회적 압력은 기업으로 하여금 환경경영 의 필요성을 깨닫게 하였다. 환경 경영이란 기업들이 새로운 비교 우위를 창출하기 위하여 경 영 방식의 혁신에 있어서 자연환경을 초점으로 삼는 것이다. 환경경영이란 환경보전을 요구하는 이해 관계자들로부터의 요구에 기업이 이에 적극적으로 대응함으로써 한경보호와 경영성과를 동시에 달성하는 종합적인 경영을 의미한다. 환경경영은 이업의 전 활동에 걸친 전사적인 전략적 차원의 활동으로 기업경영이 어느 한 기능분야에 국한된 개념이 아니다. 궁극적으로 환경보호와 성장의 조화라는 기업의 목표달성을 위해 기업의 생산, 재무, 인사조직, 마케팅, 회계정보시스템등의 활동이 모두 통합되어야 할 것이다. Concentration-dependent in vitro Anti-osteoarthritis Effects of Mixed Formula - Pomegranate Concentrate Powder: Eucommiae Cortex: Achyranthis Radix 5:4:1 (g/g) on the Primary Cultured Rat Articular Chondrocytes

  • The objective of present study is to evaluate concentration-dependent in vitro anti-osteoarthritic (OA) effects of synergic mixed formula consisted of dried pomegranate juice concentrate powder, Eucommiae Cortex aqueous extract and Achyranthis Radix aqueous extract 5:4:1 (g/g) mixture on the primary cultured rat articular chondrocytes. First, any cytotoxic effect of mixture was observed using MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium Bromide) assay. Next, cyto-protective effect of test substances was evaluated by using the recombinant human interleukin $(rhIL)-1{\alpha}$ induced chondrocytes. In addition, anti-inflammatory effects were also observed on the lipopolysaccaride (LPS) treated chondrocytes through prostaglandin $E_2(PGE_2)$ productions and 5-lipoxygenase (LPO) activities, and inhibitory effects on matrix metalloproteinase (MMP)-2 and MMP-9 activities were observed on $rhIL-1{\alpha}$ treated chondrocytes with their extracellular matrix (ECM) related mRNA expressions. No obvious cytotoxic effects of mixture were demonstrated. Inflammatory damages of chondrocytes and related ECM degradations induced by treatment of LPS or $rhIL-1{\alpha}$ were significantly and concentration-dependently inhibited by pretreatment of mixture from a concentration level of 0.001 mg/ml to 1 mg/ml. In addition, mixture showed $IC_{50}$ for $rhIL-1{\alpha}-induced$ MMP-2 and MMP-9 activities as 44.01 and $162.47{\mu}g/ml$, and also showed $EC_{50}$ for $rhIL-1{\alpha}-induced$ inhibition of collagen type II, SOX9 and aggrecan mRNA expression as 8.61, 10.79 and $4.47{\mu}g/ml$, respectively. It is observed that mixture showed concentration-dependent anti-inflammatory and cytoprotective ECM preserved effects on the primary cultured rat articular chondrocytes without cytotoxicity.

Physicochemical Properties and Biological Activities of Angelica gigas Fermented by Saccharomyces cerevisiae (유용 효모균주를 이용한 발효참당귀분말 추출물의 이화학적 특성 및 생리활성 효과)

  • The purpose of this study was to investigate the biological activities of an aqueous extract of Angelica gigas (Ag) fermented by Saccharomyces cerevisiae (Sc). First, the soluble solids of the F/3 group, in which the Ag was fermented by Sc for 3 days, decreased from $1^{\circ}Bx$ to $0.9^{\circ}Bx$. On the other hand, the pH increased with the number of days of fermentation. The result of a TLC experiment confirmed that it gradually decomposed into a low-molecular weight sugar form upon fermentation. The total phenolic compounds and flavonoid contents were higher in the fermented group than in the non-fermented group. K and Ca contents were increased by fermentation in the following order: F/3, NF, and F/0 groups. Decursin and decursinol angelate contents were highest in the F/3 group. The DPPH (${\alpha}$, ${\alpha}{\prime}$-diphenyl-${\beta}$-picrylhydrazyl) radical scavenging activity of the NF, F/0, and F/3 groups were 41.89%, 39.51%, and 60.26%, respectively. The inhibition activities of tyrosinase and lipoxygenase were stronger in the F/3 group than in the NF group. This experiment showed that the fermentation of Ag Nakai can lead to an increase in its antioxidant ability, physiological activity, whitening and anti-inflammatory effects. Thus, this oriental herbal medicine can be developed into a functional material that can be utilized in the development of cosmetic products in future.

3-Acetyl-11-Keto-Beta-Boswellic Acid from Boswellia serrata Attenuates Monosodium Iodoacetate-induced Osteoarthritis by Chondroprotective and Anti-inflammatory Effects (Monosodium iodoacetate로 유발된 골관절염 쥐에 유향(乳香) 성분 3-Acetyl-11-Keto-Beta-Boswellic Acid의 연골보호 및 항염증 효과)

  • Objectives : 3-Acetyl-11-keto-𝛽-boswellic acid (AKBA) is a major active compound in Boswellia serrata. We investigated the arthritic changes following AKBA administration in monosodium iodoacetate (MIA)-induced osteoarthritis rats. Methods : All rats were randomly divided into five groups: Normal, Control, INDO (indomethacin 2 mg/kg treated), AKBA30 (AKBA 30 mg/kg treated), and AKBA60 (AKBA 60 mg/kg treated); drugs were given 2 weeks before MIA injection. For all groups except the normal group, 50 µL of sterile saline with MIA (80 mg/mL) was injected into the right knee joint 2 weeks after drug administration. The drug administration was continued for 4 weeks from 1 week after osteoarthritis induction. The histomorphological changes of knee joint cartilage were observed by H&E staining. Also, the levels of glycosaminoglycan (GAG), cartilage oligomeric matrix protein (COMP), 5-lipoxygenase (5-LOX), 5-LOX-activating protein (FLAP), and leukotriene B4 (LTB4) in the knee joint were determined by the ELISA kits. The expressions of mitogen-activated protein kinases (MAPKs), inflammatory cytokines, and matrix metalloproteinases (MMPs) in knee joint were detected by Western blot. Results : Data show that levels of 5-LOX, FLAP, LTB4, and COMP were downregulated significantly in the AKBA treated groups when compared to those in the Control group. On the other hand, GAG levels were significantly elevated. As a result of Western blot, the AKBA-treated groups significantly inhibited phosphorylation of MAPKs. In addition, significant downregulation of the expression of inflammatory cytokines and MMPs was found in the AKBA-treated groups. Conclusion : Our findings suggest that administration of AKBA could exert better chondroprotective and anti-inflammatory effects for MIA-induced osteoarthritis rats.

Anti-osteoarthritis effect of Boswellia serrata gum resin extract in monosodium iodoacetate-induced osteoarthritic Sprague-Dawley rats (Monosodium iodoacetate 유도 골관절염 동물모델에서 보스웰리아 검레진 추출물의 항골관절염 효과 연구)

  • Purpose: The aim of this study was to investigate the anti-osteoarthritic effect of the ethanol extract of Boswellia serrata gum resin (FJH-UBS) enriched with keto-β-boswellic acid and 3-O-acetyl-11-keto-β-boswellic acid compared to the conventional Boswellia serrata extract by adding the process of removing oil with hexane, in the monosodium iodoacetate (MIA)-induced osteoarthritis rat model. Methods: Sprague-Dawley (SD) rats were orally administered 0, 40, or 80 mg of FJH-UBS/kg body weight (BW)/day for 5 weeks and injected with MIA intra-articularly into right knee joints on day 15 to induce osteoarthritis. Changes in the knee joint microarchitecture, cartilage degradation, the expression of inflammatory mediators, cytokines, and matrix metalloproteinases (MMPs) in serum and synovia were observed. Results: Oral administration of FJH-UBS (80 mg/kg BW/day) reduced MIA-induced knee swelling and cartilage degradation and increased the expression of type II collagen and aggrecan in articular cartilage. Furthermore, FJH-UBS administration reduced MIA-induced increases in the serum levels of prostaglandin E2, leukotriene B4, interleukin (IL)-1β, IL-6, and MMP-13, and MIA-induced increases in the mRNA expressions of inducible nitric oxide synthase, cyclooxygenase-2, 5-lipoxygenase, IL-1β, IL-6, TNF-α, MMP-2, MMP-9, and MMP-13 in the synovia of knee joints. Conclusion: These results indicate that FJH-UBS exerts its anti-osteoarthritic effects by suppressing the expressions of inflammatory cytokines and MMPs and, thus, cartilage degradation. Furthermore, they suggest that FJH-UBS has potential use as a functional food that improves joint and cartilage health.

  • Background: Bronchial asthma is characterized by airway hyperresponsiveness(BHR) and inflammation. The cyclooxygenase(COX) is believed to be one of the important enzymes in these inflammatory reactions. Recently, the COX was divided into two isoforms, COX1 and COX2. COX2 is induced by lipopolysaccharide and some cytokines at the inflammation site. Prostaglandin E2(PGE2), produced from COX2, may affect airway inflammation. The purpose of this study is to evaluate the effect of COX2 inhibitor on COX2 expression, plasma PGE2, airway resistance and histologic finding in an animal asthma model. Methods : Sprague-Dawley rats were divided into 3 groups. The normal control group did not receive any treatment, but the asthma control group was sensitized by ovalbumin but not treated with the COX2 inhibitor(nimesulide, Mesulid$^{(R)}$). The treatment group was sensitized and treated with nimesulide. Specific airway resistance(sRaw) before and after nimesulide ingestion was investigated. The PGE2 level in the plasma was examined and COX2 immunogold-silver stain on lung tissue was performed. Results: sRaw and eosionophilic infiltration on airway, which increased in the asthma control group, was compared to normal control(p=0.014). However, there was no difference in eosinophilic infiltration between asthma control and treatment groups(p=0.408) and no difference in COX2 expression on bronchiolar epithelium among the three groups. Plasma PGE2 levels were not statically different among the three groups. Conclusion: The role of COX2 in the allergen-induced BHR was not significant The effect of nimesulide was not observed on BHR, COX2 expression, and plasma PGE2 level. Therefore, COX2 may not be a major substance of allergic asthma.

Effect of Mulberry (Morus alba L.) Extract on Blood Flow Improvement (오디 추출물(Morus alba L.)의 혈행개선 효과)

  • The objective of this study was to investigate the beneficial effects of mulberry extract (MBE) on blood flow improvement. The $SC_{50}$ value for the DPPH radical scavenging activity of MBE was $89.36{\pm}5.46{\mu}g/mL$. Analysis of the cellular toxicity of MBE on RAW 264.7 and HepG2 cells showed no toxicity under a concentration of 2,500 ${\mu}g/mL$. We found that MBE inhibited the enzyme activity of cyclooxygenase (COX)-2 as well as oxidation of human LDL. Western blotting analysis showed that MBE inhibited protein expression of COX-2 and 5-lipoxygenase in RAW 264.7 cells. In addition, MBE inhibited protein expression of intercellular adhesion molecule-1 and vascular cell adhesion molecule-1 in human umbilical vein endothelial cells. Furthermore, MBE reduced the serum levels of total cholesterol and C-reactive protein in a concentration-dependent manner. These results both in vitro and in vivo suggest that MBE can be employed for the improvement of blood flow.
