Effects of Aucubin Isolated from Eucommia ulmoides on UVB-induced Oxidative Stress in Human Keratinocytes HaCaT

  • Ultraviolet B (UVB) radiation provokes the generation of reactive oxygen species (ROS) in the cells and skin, which induce oxidative stress in the exposed cells, leading to photoaging and cancer. Using the human keratinocytes HaCaT cell line, we investigated the photoprotective effects of aucubin isolated from Eucommia ulmoides. Pretreatment with aucubin markedly suppressed UVB-induced oxidative stress, which manifests as a decrease in intracellular lipid peroxidation, elevation of catalase activity, and reduced glutathione content. In addition, aucubin significantly reduced expression of matrix metalloproteinase-1 (MMP-1) protein (54%) and mRNA. Taken together, these results suggest that aucubin may offer protection against UVB-induced oxidative stress and may be used as a potential agent in prevention of UVB-induced photoaging.

유 가열 혼합육(계육, 돈육)의 휘발성 성분 및 각종 첨가물의 영향 (Effects of Various Additives on the Volatile Compounds of Cooked Oil with Mixture Meat(Chicken and Pork))

  • Effects of metal chelating agents and metal ions on the volatile substance of cooked oil with chicken and pork mixture meat were examined by chemical analysis and sensory test. The addition of Na-tripolyphosphate(Na-TPP) to chicken and pork mixture meat increased the amount of H2S among volatiles evolved during cooking but decreased that of volatile carbonyl compounds(VCC) This treatment enhanced meat flavor in cooked oil with chicken and pork mixture. It was recognized that the increase in Ha5 evolution was caused by the rise of pH value. On the contrary cupric ion produced a negative effect on the production of chicken and pork mixture meat flavor and this addition increased VCC and TBA value. Other metal chelating agents such as citric acid, phytic acid and EDTA, provided the same results as Na-TPP. It was supposed that these phenomena were attributable to the chelating action to metal prooxidant in mixture meat at could be concluded that a proper evolution of H2S and protection against lipid oxidation during cooking were important to produce an excellent chicken and pork mixture meat flavor.

Over-expression of Cu/ZnSOD Increases Cadmium Tolerance in Arabidopsis thaliana

  • Over-expression of a copper/zinc superoxide dismutase (Cu/ZnSOD) resulted in substantially increased tolerance to cadmium exposure in Arabidopsis thaliana. Lower lipid peroxidation and $H_2O_2$ accumulation and the higher activities of $H_2O_2$ scavenging enzymes, including catalase (CAT) and ascorbate peroxidase (APX) in transformants (CuZnSOD-tr) compared to untransformed controls (wt) indicated that oxidative stress was the key factor in cadmium tolerance. Although progressive reductions in the dark-adapted photochemical efficiency (Fv/Fm) and quantum efficiency yield were observed with increasing cadmium levels, the chlorophyll fluorescence parameters were less marked in CuZnSOD-tr than in wi. These observations indicate that oxidative stress in the photosynthetic apparatus is a principal cause of Cd-induced phytotoxicity, and that Cu/ZnSOD plays a critical role in protection against Cd-induced oxidative stress.

EFFECT OF DITHIOL MALONATE DERIVATIVES (DMDs) ON CARBON TETRACHLORIDE-INDUCED HEPATOTOXICITY IN PRIMARY CULTURES OF ADULT RAT HEPATOCYTES

  • Protective effects of dithiol malonate derivatives (DMDs), YH-100, YH-150 and YH-439 on carbon tetrachloride-induced hepatotoxicity were investigated in primary rat hepatocytes culture. Treatment of DMDs to hepatocytes culture did not affect total cytochrome P-450 content and ECOD and AHH activities. Protein and RNA synthesis was also similar to control. Meanwhile, DMDs significantly decreased LDH release and in vitro lipid peroxidation induced by $CCI_4$. Accumulation of cellular triglyceride and decreased secretion of VLDL from liver cells by $CCI_4$ treatment were also significantly protected.

Short Communication of Novel Application of Food Irradiation

  • Irradiation of food is not only used for sanitation purposes but can be used for processing techniques to reduce or eliminate toxic or undesirable compounds on food. Irradiation wag effective to reduce the allergenicity of food by modification of the structure of proteins causing allergy reactions. Volatile N-nitrosmaine was reduced or eliminated by irradiation in the model system study and the breakdown products by irradiation did not recombine under human stomach conditions (pH 2,3, and 4,37$^{\circ}C$). The possibility of residual chlorophyll b reduction by irradiation was also found, and the model study indicated that irradiation be used to destroy chlorophyll b, resulting in protection from photooxidation in oil without acceleration of lipid oxidation during irradiation. In this paper, several on-going research projects for the application of food irradiation as a new processing technique are introduced, including reduction of food allergens, breakdown of volatile N-nitrosamine and residual chlorophyll b.

갈조류 곰피(Ecklonia stolonifera)의 간 건강기능성 (Health beneficial effects of brown algae ecklonia stolonifera in liver)

  • People in Korea, a peninsular, have acquired a variety of marine food resources including seaweeds. Ecklonia stolonifera, a brown algae, is commonly dwelling in Korean coasts and their cultivation methods were developed for a mass-production. Recently, studies have revealed that Ecklonia stolonifera is a promising material for the development of health functional foods. In an effort to carefully review the current understating in the effects and mechanisms of Ecklonia stolonifera on liver functions by deduction from relevant literatures, the effective components were identified as phlorotannins, including dieckol, eckstolonol, eckol, phlorofucofuroeckol A, and phlorosterol. Their aiding action on the hepatic functions is categorized as follows. A) Regulation of oxidative stress by anti-oxidant capacity, B) Protection of hepatocytes from toxins, C) Prevention of alcoholic fatty liver and fibrogenesis, D) Regulation of chronic disease by improvement of inflammatory responses and lipid metabolisms, and E) indirect benefit conferred by a personal total wellness.

  • Oxidative stress and its consequent lipid peroxidation exert harmful effects, which have been currently involved in the generation of carbon tetrachloride(CC14)-induced fibrosis(cirrhosis). In this study, it was investigated whether dried extract of 田螺(Concha Cipangopaludinae: CC) is liver functional improvement, antioxidative and antifibrotic effect under the liver fibrotic(cirrhotic) condition by CC14 administration.   • This study has been undertaken to examine the effect of biphenyl dimethyl dicarboxylate (DDB) on rat liver drug metabolizing enzyme in order to understand the mechanism of DDB on improving hepatic toxicity in rat liver. After DDB was administered into male rats for different periods of time, mRNA level of CYP1A1 and CYP2B1 was measured by polymerase chain reaction (PCR). DDB treatment resulted in increase in CYP2B1 mRNA level whereas there was no change in CYP1A1 mRNA level. This effect of DDB was time dependent reaching maximal level by 2-day treatment. DDB dose response study showed that 50mg/kg DDB induced CYP2B1 mRNA to maximal level and DDB icreased CYP2B1 gene expression with dose-dependent manner. Based on studies of lipid peroxidation, serum ALT and AST levels and histopathologic examination showed DDB protection on CCl4 induced hepatotoxiccity.

어유섭취가 출생후 발달과정의 흰 쥐의 간과 뇌조직의 지질과산화와 그 관련기능에 미치는 영향 (Effect of Dietary Fish Oil on Lipid Peroxidation in Rats Liver and Brain During Postnatal Development)

  • 어유를 섭취하였을 때, 체내 과산화물 생성과 항산화 기능을 조사하며, 그 결과를 다른 식이지방과 비교하기 위하여, 고등어유, 대두유, 쇠기름, 들기름, 채종유의 5 종의 지방을 먹이의 10% 되게 식이를 조제하여 70g 내외의 암, 수컷의 흰쥐에게 섭취시켰다. 출생후 초기 성장시에 적응도를 관찰하기 위하여, 위의 쥐들을 34일간 조제식이로 사육한후, 교배시켜 출생한 2 대째 쥐들을 수유시기(17일, 26일)과 이유시기(39일)에 희생시키고, 출산, 수유를 마친 어미쥐(나이 123일, 총 식이일수 81일)를 희생하여 간과 뇌조직의 지질과산화물, $\alpha$-tocopherol, glutathione 양을 정량하고, glutathione peroxidase, superoxide dismutase의 활성을 측정하였다. 간조직의 지질과산화물 값은 고등어유를 섭취한 어미쥐와 수유기(17, 26일)의 새끼쥐들에게서 타군에 비해 높았으나 39일의 새끼쥐에서는 채종유보다는 높았으나 대두유, 들기름군과 같은 수준이었다. 뇌조직에서는 간에 차이가 거의없었다. 간조직의 $\alpha$-tocopherol 농도와 환원형의 glutathione(GSH) 의 농도가 어미쥐에서 타군에 비해 현저히 낮았고, 새끼쥐에서도 $\alpha$-tocopherol 경우는 같은경향이나 그 정도가 덜 현저하며, GSH의 경우는 타군들과 차이가 없었다. 산화형 glutathioe (GSSG)의 농도는 어미쥐, 새끼쥐모두에서 식이지방에 따른 차이가 일관성 있게 나타나지 않았다. Glutathione peroxidase 활성은 간장과 뇌조직에서 성장과정중의 새끼쥐들에서 모두 약간씩 증가하였고, 채종유군에서 타군에 비해 증가정도가 낮아 활성이 39일에 현저히 낮았다. Superoxide dismutase 의 활성은 성장기간에 따른 변화는 적었고, 고등어유군이 타군에 비해 비교적 낮은값을 유지하였다. 어미쥐와 17일째 새끼쥐에서는 달리 26일과 39일째 새끼쥐의 간조직 glutathioe peroxidase 의활성이 지질과산화물 농도와 정의 상관관계가 보여져, 출생후 초기 성장과정에서 체내 적응성이 존재함을 나타내주었다.

어유섭취시 식이 비타민 E 수준에 따른 흰쥐 체내 비타민 E, A, 글루타치온 상태의 기간별 변화 (Periodic Changes in Vitamin E, A and Glutathione Status in Rats Fed Fish Oil Diet with Different Levels of Vitamin E)

  • 魚油섭취시 식이 vitamin E의 변화가 체내의 과산화물생성과 여러 종류의 생리적인 항산화물질의 상태에 주는 변동을 조사하기 위하여, 체중 $150pm15$g의 Sprague-Dawley종의 숫쥐를 실험식이에 따라 대조군, F0, FI, FII군으로 나누어 4,8,16주 동안 사육하였다. 식이지방수준은 10%(w/w)로 대조군 에게는 대두유를, 魚油군들(F)에게는 menhaden oil:대두우(9:1)의 혼합유를 주었고, vitamin E 수준은 대조군 30, F0군 2, FI군 30, FII군 140 T.E 이었다. F0군의 혈장 vitamin E는 극미량이었고, 간과지방조직에서도 제일 낮았으며, 기간이 길어짐에 따라 감소하였다. FI,FII군의 혈장 vitamin E 수준은 ml당으로는 대조군에 비하여 낮았으나, mg lipid 당으로는 같거나 높았거, 간에서는 8주에, 지방조직에서는 16주에 그 함량이 높은 현상을 보였고, 모든 조직에서 FII군이 FI군보다 높은 경향이었다. 혈장 TBARS값도 ml당으로 보면, FI,FII군이 대조군보다 낮은 추세였으나 mg lipid당으로는 높았고, FII군은 FI군에 비하여 낮은 경향이었다. 그러나 간조직 지질과산화물은 16주에 FII군만 제외하고, 어유군들이 대조군에 비하여 높아 FO>FI>FII>대조군 순이었다. 혈장과 간조직의 vitamin A, 혈액 GSH는 대체로 FII과 대조군이 비슷하게 제일 높고, FI,FO순으로 낮아지는 경향이있었다. 따라서 본 연구에서 식이지방 종류와 vitamin E의 수준이 체내 과산화물 생성뿐 아니라 체내의 여러 황산화기능 관련문질의 상태에 영향을 주고 있다고 결론을 지을 수 있다.
