Our aim was to investigate the chemopreventive potential of saffron in DMBA-induced hamster buccal pouch carcinogenesis. Assessment was by monitoring the percentage of tumor bearing hamsters, tumor size as well as the status of detoxification agents, lipid peroxidation and antioxidants. Oral squamous cell carcinomas were induced in the buccal pouch of Syrian golden hamsters by painting them with 0.5% DMBA in liquid paraffin three times a week for 14 weeks. We observed 100% oral tumor formation with severe histopathological abnormalities in all the hamsters treated with DMBA alone, activities of phase I and phase II detoxification enzymes, lipid peroxidation and antioxidants being significantly altered. Though oral administration of saffron completely prevented the formation of tumors, we noticed severe hyperplasia and dysplasia in hamsters treated with DMBA, suggesting that tumors might eventually develop. Oral administration of saffron return detoxification enzymes, lipid peroxidation and antioxidants to normal ranges. The chemopreventive potential of saffron thus is likely due to antioxidant properties and modulating effects on detoxification in favour of the excretion of carcinogenic metabolites during DMBA-induced hamster buccal pouch carcinogenesis.
To determine the effects of endogenous and exogenous strogen on serum lipid levels, twenty nonsmoking healthy Korean women were participated in this experiment for 12 weeks. They were assigned to three groups : (1) eight women aged 22 to 30(yr) for the premenopausal(Pre) group, (2) eight, aged 49 to 60(yr) for the postmenoparusal(Pst) group, (3) four, aged 23 to 30(yr) for the oral contraceptive(OC) group which used triphasic OC formulation. Fasting blood samples representing every phase of the hormonal levels were obtained from the subjects of the Pre and the OC group. From the subjects of the Pst group, fasting blood samples were obtained once per three weeks for 12 weeks. All the serum data were adjusted for dietary effects, exercise, personality type and body mass index(BMI) by using analysis of covariation(ANCOVA). Serum lipid levels of the three groups were significantly different. While serum levels of triglycerides(TG)(p<0.0001), low density lipoprotein-chloesterol(LDL-C)/high density liporotein-cholesterol(HDL-C) ratio (LDC-C/HDL-C)(P<0.01) and total cholesterol (TC)/HDL-C ratio (TC/HDL-C)(P<0.001) were significatnly high in the Pst group, serum HDL-C(P0.001) level was significantly high in the Pre group. The OC group showed significantly low serum TC(P<0.0001) and LDL-C(P<0.0001) levels. There was no signidicant difference in the fluictuation of serum lipid levels during the menstrual cycle of the Pre group. However, in the OC group, serum TG level was significantly increased at phase 2(P<0.05) where exogenous estrogen administration was highest. Even though other serum lipid levels of the OC group were not significantly fluctuated according to the exogenous estrogen administration, there was a trend of increased levels of serum TC, LDL-C, LDL-C/HDL-C and TC/HDL-C and decreased level of HDL-C during the menstruation period. Also, serum TC level was high(P<0.005) and serum TG level was low (P<0.005) at the baseline of the OC group compared with the periods of OC administration. When screening and counseling the female population at risk for coronary heart disease(CHD), the result of this study suggest that in may be desirable to divide the population into several groups according to their personal physiological characteristics, such as age, OC administration, menstrual cycle and menopause, as well as general risk factors for CHD.

This study was conducted to investigate the effects of heating, fiber type used in solid-phase microextraction (SPME, two phase vs three phase) and storage time on the volatile compounds of porcine M. longissimus dorsi (LD). Volatile compounds were measured using a gas chromatography and mass spectrometry (GC/MS) with a quadrupole mass analyzer. Among the volatile compounds identified, aldehydes (49.33%), alcohols (24.63%) and ketones (9.85%) were higher in pre-heated loins ($100^{\circ}C$/30 min), whereas, alcohols (34.33%), hydrocarbons (22.84%) and ketones (16.88%) were higher in non-heated loins. Heating of loins induced the formation of various volatile compounds such as aldehydes (hexanal) and alcohols. The total contents of hydrocarbons, alcohols, and carboxylic acids were higher in two phase fibers, whereas those of esters tended to be higher in three-phase fibers (p<0.05). Most volatile compounds increased (p<0.05) with increased storage time. Thus, the analysis of volatile compounds were affected by the fiber type, while heating and refrigerated storage of pork M. longissimus dorsi increased the volatile compounds derived from lipid oxidation and amino acid catabolism, respectively.
Effect of C/N ratios on the cell growth, cell lipid, cell protein and fatty acids of Rhodotorula sp. was examined. At low concentration of nitrogen in cultural medium the growth of the yeast was retarded, whereas its growth was favorable at high concentration but lipid yield was decreased due to a high content of cell protein. In order to increase lipid yield, the optimum C/N ratio obtained was 45-50 (5% glucose, 0.0414% nitrogen). During the cultivation cell protein synthesis occurred mainly in initia stage and the lipid was accumulated rapidly at the late phase. The highest content of total fatty acid was observed at the optimum C/N ratio. The higher C/N ratio was the more unsaturation degree of fatty acid decreased. It was attributed principally to a decreased oleic acid with palmitic acid increased. No significant changes occurred in content of other fatty acids.
Whole-cell immobilization of the hydrocarbon rich microalgae, Botryococcus braunii and B. protuberans, in alginate beads under air-lift batch cultures resulted in a significant increase in chlorophyll, carotenoid, dry weight, and 1ipid contents at stationary and resting growth phases, as compared to free cells. Photosynthetic activity in both the species, of Botryococcus was enhanced, relative to free cells, at any growth phase of cultures. Immobilization exerted a protective influence on ageing of the cultures as reflected by higher chlorophyll and dry weight contents. Entrapment also stabilized the chlorophyll and carotenoid contents even at stationary and resting phases as compared to free cells in both the species.
The simple binary mixtures of pure phospholipids such as DPPC:DSPC and DPPC:DPPA were investigated with differential scanning calorimeter. The thermotropic properties of DPPC and DSPC mixtures did not deviate much from the ideal curves that was obtained on varying either the temperature or the relative proportions of phospholipids. This means that the two phospholipids are completely miscible in the liquid and solid phase. But the thermotropic properties of DPPC and DPPA mixture deviated much from the ideal phase diagram. It resulted from the repulsion between the head groups of acidic phospholipids. Heat capacity suggested by Harden McConell was calculated. The larger $C_p$ was, the sharper the endothermic peak became.

In this paper, we perform a direct comparison study of real experimental data for domain rearrangement and the Cahn-Hilliard (CH) equation on the dynamics of morphological evolution. To validate a mathematical model for physical phenomena, we take initial conditions from experimental images by using an image segmentation technique. The image segmentation algorithm is based on the Mumford-Shah functional and the Allen-Cahn (AC) equation. The segmented phase-field profile is similar to the solution of the CH equation, that is, it has hyperbolic tangent profile across interfacial transition region. We use unconditionally stable schemes to solve the governing equations. As a test problem, we take domain rearrangement of lipid bilayers. Numerical results demonstrate that comparison of the evolutions with experimental data is a good benchmark test for validating a mathematical model.
In order to study the lipid peroxidation caused by light and the protective action it in biological memberane, reverse-phase evaporation liposome (REV) was employed as a model memberance and the effect of several antioxidants and ginseng water extracts were tested. In the presence of photosensitizer, liposome was oxidized easily and the oxidation index dut to the peroxidation was increased. The oxidation index of liposome was increased according to the increase in temperature. When dl-${\alpha}$-tocopherol, ${\beta}$-carotene or L-ascorbic acid was added into the reaction mixture, the photooxidation of liposome was inhibited. Ginseng water extract and crude saponin inhibited the rate of oxidation index of liposome in low concentration but increase in high concentration. On the other hand, when lipid hydroperoxide of liposome was tested by ferrothiocyanate method, ginseng water extract and crude saponin acted as antioxidants.
lonophores, uniquely, create specific pathways of ion permeability in model and cell membranes. Calcium-transporting ionophores of microbiological origin, such as A23187 and ionomycin, have been used as experimental tools to elucidate the physiological role of calcium as a second messenger in many cell types. These ionophores are believed to bypass the initial ligand-receptor step in the activation of cells by increasing membrane permeability to calcium. In this report, we shall discuss several naturally occurring substances that share some properties of calcium-ionophores, primarily concentrating on oxidized fatty acids. We have previously demonstrated that oxidized linoteic and arachidonic acids, obtained either by lipoxygenase catalysis or nonenzymatic processes, significantly promote calcium translocation in a two-phase partition model and modulate calcium-transporting function in the isolated sarcoplasmic reticulum vesicles obtained from mammalian hearts. We have also confirmed that calcium-ionophoric properties are due not to their general amphiphilic nature of certain lipids, but to distinct structural characteristics. Although there are some skeptical views on the occurrence of ionophores in higher organisms, increasing evidence suggests that membrane lipids or their derivatives may serve as physiological calcium-ionophores. Abnormal accumulation of lipid peroxidation products(particularly end products), however, may be associated with the general oxidative damages as seen in many pathological conditions.
Mimicking the bacterial synthesis of magnetosomes, in which the functionalized surface of a cytoplasmic (lipid) membrane is considered to be stimulating the crystal growth of magnetite, we have successfully grown magnetite films at $30^{\circ}C$ using an arachic acid monomolecular layer as a functionalized surface. The lipid monomolecular layer was spread on an aqueous solution of FeCl$_2$ which was oxidized by flowing a mixed gas, with ratio $O_2$/$N_2$=1/2000, on the surface of the lipid layer. Mossbauer and X-ray diffraction analyses revealed that the Fe$_3$O$_4$ films contain small amounts of ferric hydroxyl impurity phases of ${\alpha}$-FeOOH and ${\tau}$-FeOOH. This is because the oxygen partial pressure at the ferrite/aqueous interface changed as the film (through which the gas penetrated) increased in thickness. Methods to obtain single phase magnetite films are proposed.
