가용화제를 활용한 세라마이드 고함량의 수화 액정형 베시클 개발 (Preparation of Hydrated Liquid Crystalline Vesicles Containing High Content of Ceramide using a Solubilizer)

  • 세라마이드와 인지질, 콜레스테롤 등과의 상호적 자가회합이 가능한 최적의 조성을 구성하여 세라마이드 고함량의 수화 액정형 베시클을 제조하였다. 혼합조성을 달리하여 다양한 베시클을 제조한 결과, 에지 액티베이터인 sodium deoxycholate (SDOC)와 가용화제인 PEG-60 hydrogenated castor oil (HCO 60)을 혼합하여 베시클을 만들었을 때, 가장 작은 나노 사이즈의 입자가 만들어지고, 베시클 분산액은 약산성이면서 가장 안정한 상태를 유지하였다. 또한 편광현미경과 열분석을 통해 SDOC와 HCO 60의 첨가가 세라마이드 등 지질 성분의 결정성 억제에 영향을 미치는 것으로 확인되었다. 베시클 분산액은 8주 동안의 고온의 장기 보관에서도 외관 및 점도의 변화없이 안정성이 그대로 유지되었다.

8종 어란의 일반성분, 아미노산, n-3 지방산 및 무기질 비교 (Comparison of Proximate, Amino Acid, n-3 Fatty Acid and Mineral Compositions of Fish Roes of Eight Species)

  • This study was conducted to identify the food components and nutrition value of major fish roes on the market in Korea. The proximate compositions of the roes were 60.02-82.85% moisture, 14.61-29.21% protein, 1.24-14.59% lipid and 0.88-1.78% ash. The major total amino acids in the roes were glutamic acid, leucine, aspartic acid, lysine, and alanine. The major fatty acids were 22:6n-3 (docosahexenoic acid, 9.37-32.68%), 16:0 (5.96-21.39%), 18:1n-9 (12.64-25.30%), and 20:5n-3 (eicosapentaenoic acid, 3.79-16.99%). The mean major-mineral levels were phosphorus (291.63 mg/100 g edible portion), potassium (271.00 mg), sodium (175.86 mg), calcium (24.02 mg), and magnesium (22.15 mg). The mean trace-mineral levels were zinc (7.75 mg), iron (3.68 mg), and copper (0.81 mg). The results suggest that these fish roes are good sources of proteins, amino acids, n-3 fatty acids and minerals.

Determination of tyrosinase inhibitory activity and betanin content changes in beetroot (Beta vulgaris) extracts fermented by EM

  • Beet (Beta vulgaris) is a crop similar to sugar beet, chard and leaf beets, and its origin is the Mediterranean coast of southern Europe and Central Asia. Among the components contained in beet, betalain, the main component of the root, has been reported to prevent lipid peroxidation induced by active oxygen and free radicals due to its high radical scavenging ability. Among these, the betalain, betanin (Betanidin 5-O-${\beta}$-glucoside) contains both phenolic and cyclic amine groups, all of which are highly electron-donating and act as antioxidants and has tyrosinase inhibitory activity. Betanin accounts for about 75-95% of the total pigment found in the beet. EM stands for effective microorganisms and is a collection of beneficial microorganisms. EM includes yeast, lactic acid bacteria, mycelia, photosynthetic bacteria, actinomycetes, etc. Human patch test according to CTFA guidelines was observed to be a safe source of no stimulation when 5% (v/v) of the EM fermentation liquid was applied to the human body. In addition, beneficial microorganisms are synergistic in the process of co-existence and cultivation and it has the effect of increasing antioxidant capacity and inhibiting corruption. This study confirms the difference in tyrosinase inhibitory activity and betanin content of beetroot extracts and EM fermented beetroot extracts. Hence, these results confirm that EM fermented beetroot extracts are highly beneficial for the human body.

LC/MS-based Analysis of Bioactive Compounds from the Bark of Betula platyphylla var. japonica and Their Effects on Regulation of Adipocyte and Osteoblast Differentiation

  • Betula platyphylla var. japonica (Betulaceae), also known as Asian white birch, is an endemic medicinal tree, the bark of which has been used in Chinese traditional medicine for the treatment of various inflammatory diseases. In our continuing search for bioactive compounds from Korean natural resources, a phytochemical investigation of the bark of B. platyphylla var. japonica led to the isolation of 7-oxo-${\beta}$-sitosterol (1) and soyacerebroside I (2) from its ethanol extract as main components by liquid chromatography (LC)/mass spectrometry (MS)-based analysis. The structures of isolates were identified by comparison of $^1H$ and $^{13}C$ nuclear magnetic resonance spectroscopic data and physical data with the previously reported values and LC/MS analyses. To the best of our knowledge, this is the first study to demonstrate that the isolated compounds, 7-oxo-${\beta}$-sitosterol and soyacerebroside I, were isolated in B. platyphylla var. japonica. We examined the effects of the isolates on the regulation of adipocytes and osteoblast differentiation. These isolates (1 and 2) produced fewer lipid droplets compared to the untreated negative control in Oil Red O staining of the mouse mesenchymal stem cell line without altering the amount of alkaline phosphatase staining. The results demonstrated that both compounds showed marginal inhibitory effects on adipocyte differentiation but did not affect osteoblast differentiation.

살 오징어(Todarodes pacificus) 간췌장 유래 Aminopeptidase 활성획분에 의한 Alcalase 처리 멸치(Engrauris japonica) 가수분해물의 쓴맛 개선 최적화 (Optimization of Reduced Bitterness of Alcalase-treated Anchovy Engrauris japonica Hydrolysate by Aminopeptidase Active Fraction from Common Squid Todarodes pacificus Hepatopancreas)

  • This study used response surface methodology to investigate the optimal conditions to reduce the bitterness of alcalase-treated anchovy hydrolysate (AAH) by the aminopeptidase active fraction (AAF) derived from the common squid Todarodes pacificus hepatopancreas. The central composite design selected AAF/AAH ratio (X1, %) and hydrolysis time (X2, h) as independent variables, and the degree of hydrolysis (Y1) and bitterness (Y2) as dependent variables. The uncoded values of the multiple response optimization for independent variables were 3.4% for the AAF/AAH ratio and 9.2 h for the hydrolysis time. The predicted values of the yield and bitterness score of alcalase-AAF continuously treated anchovy hydrolysate (AAAH) under the optimized conditions were 68.9% and 4.6 points, respectively. Their measured values of 69.5% for yield and 4.6±0.5 points for bitterness were similar to the predicted values. The food components of AAAH were 91.4% (moisture), 7.5% (protein), 0.1% (lipid) and 0.6% (ash). The findings indicate the potential value for use as an anchovy seasoning base. The results also confirm that the bitterness of AAH was remarkably improved by AAF and implicates AAF derived from squid hepatopancreas as a good enzyme to catalyze reduced bitterness.

현미유가 생쥐의 골수로부터 M2로 유도한 대식세포의 미토콘드리아 호흡에 미치는 영향 (Effects of Dietary Rice Bran Oil on Mitochondrial Respiration in M2-induced Bone Marrow-derived Macrophages)

  • Previous studies have suggested that rice bran oil (RBO), an edible oil from the byproducts of rice milling, has anti-inflammatory effects in inflammation inducing macrophages, known as M1 subsets. Yet the effects of RBO on the counterpart M2 subsets, the "healing" macrophages, were poorly investigated to date. In this regard, recent studies on the molecular/cellular anti-inflammatory mechanisms of dietary components have demonstrated that mitochondrial respiration contributes to macrophage functioning. Therefore, the current study examined whether RBO regulates cytokine secretion by modulating mitochondrial metabolism in wound healing M2 subsets. Palm oil (PO), enriched with medium-chain fatty acids, served as a positive control. C57BL/6 mice were fed a diet containing either corn oil (CO), PO or RBO for 4 weeks, followed by purification of bone marrow-derived macrophages (BMDM) from their tibias and femurs. Cells were further polarized to M2-BMDM, and the expression of M2 marker (CD206) on cellular surfaces were not affected by dietary intervention. In addition, the secretion of anti-inflammatory cytokine (IL-10) in the culture supernatant was not affected by dietary lipids. Oxygen consumption rate, the indicator of mitochondrial respiration in M2-BMDM was not regulated by RBO intervention and PO treatment. Taken together, this study imply that RBO did not intervene both the regulation of inflammatory responses and mitochondrial respiration in M2 macrophages.

돼지 정액을 저장하는 동안 정자에 미치는 산화스트레스 (Oxidative Stress in Spermatozoa during Boar Semen Storage)

  • 돼지 정액을 저장하는 동안 산화스트레스의 발생은 정자의 질과 생존에 영향을 미치는 중요한 인자이다. 정액의 저장은 온도 변화, 동결보호제 등의 다양한 스트레스 인자에 노출되어 있다. 이러한 정자 내에서의 산화스트레스는 활성산소종의 생성에 의해 발생되며, 이는 지질, 단백질, DNA와 같은 세포를 구성하는 물질에 산화적으로 손상을 일으킨다. 활성산소종과 항산화물질의 균형있는 체계는 정자의 생존과 그 기능을 유지하는 데 중요한 역할을 한다. 정액을 장기간 보존하게 되면 활성산소종의 수준이 증가하여 정자의 운동성, 막 온전성, DNA 온전성에 영향을 미치게 된다. 또한, 활성산소종에 의해 유도된 지질과산화 반응은 정자막의 유동성과 안정성에 영향을 미쳐 정자의 운동성을 감소시킨다. 그리고, DNA의 산화적 손상은 DNA 단편화를 일으켜 정자의 DNA 온전성을 손상시킬 수 있다. 결론적으로, 정액을 보관하는 동안 발생되는 산화스트레스는 정자의 질과 기능을 유지하는 데 중요하다. 따라서, 산화스트레스의 기본적인 메커니즘과 정자의 기능에 미치는 영향을 이해하는 것은 산화스트레스로부터의 손상을 최소화하고, 효율적이고 기능적인 정자의 저장 방법을 개선하기 위한 효과적인 전략과 연구 개발을 위해 중요할 것으로 판단된다.

Atypical formations of gintonin lysophosphatidic acids as new materials and their beneficial effects on degenerative diseases

  • Fresh ginseng is prone to spoilage due to its high moisture content. For long-term storage, most fresh ginsengs are dried to white ginseng (WG) or steamed for hours at high temperature/pressure and dried to form Korean Red ginseng (KRG). They are further processed for ginseng products when subjected to hot water extraction/concentration under pressure. These WG or KRG preparation processes affect ginsenoside compositions and also other ginseng components, probably during treatments like steaming and drying, to form diverse bioactive phospholipids. It is known that ginseng contains high amounts of gintonin lysophosphatidic acids (LPAs). LPAs are simple lipid-derived growth factors in animals and humans and act as exogenous ligands of six GTP-binding-protein coupled LPA receptor subtypes. LPAs play diverse roles ranging from brain development to hair growth in animals and humans. LPA-mediated signaling pathways involve various GTP-binding proteins to regulate downstream pathways like [Ca2+]i transient induction. Recent studies have shown that gintonin exhibits anti-Alzheimer's disease and antiarthritis effects in vitro and in vivo mediated by gintonin LPAs, the active ingredients of gintonin, a ginseng-derived neurotrophin. However, little is known about how gintonin LPAs are formed in high amounts in ginseng compared to other herbs. This review introduces atypical or non-enzymatic pathways under the conversion of ginseng phospholipids into gintonin LPAs during steaming and extraction/concentration processes, which exert beneficial effects against degenerative diseases, including Alzheimer's disease and arthritis in animals and humans via LPA receptors.

Protective Efficacy and Immunogenicity of Rv0351/Rv3628 Subunit Vaccine Formulated in Different Adjuvants Against Mycobacterium tuberculosis Infection

  • Bacillus Calmette-Guerin (BCG) vaccine is the only licensed vaccine for tuberculosis (TB) prevention. Previously, our group demonstrated the vaccine potential of Rv0351 and Rv3628 against Mycobacterium tuberculosis (Mtb) infection by directing Th1-biased CD4+ T cells co-expressing IFN-γ, TNF-α, and IL-2 in the lungs. Here, we assessed immunogenicity and vaccine potential of the combined Ags (Rv0351/Rv3628) formulated in different adjuvants as subunit booster in BCG-primed mice against hypervirulent clinical Mtb strain K (Mtb K). Compared to BCG-only or subunit-only vaccine, BCG prime and subunit boost regimen exhibited significantly enhanced Th1 response. Next, we evaluated the immunogenicity to the combined Ags when formulated with four different types of monophosphoryl lipid A (MPL)-based adjuvants: 1) dimethyldioctadecylammonium bromide (DDA), MPL, and trehalose dicorynomycolate (TDM) in liposome form (DMT), 2) MPL and Poly I:C in liposome form (MP), 3) MPL, Poly I:C, and QS21 in liposome form (MPQ), and 4) MPL and Poly I:C in squalene emulsion form (MPS). MPQ and MPS displayed greater adjuvancity in Th1 induction than DMT or MP did. Especially, BCG prime and subunit-MPS boost regimen significantly reduced the bacterial loads and pulmonary inflammation against Mtb K infection when compared to BCG-only vaccine at a chronic stage of TB disease. Collectively, our findings highlighted the importance of adjuvant components and formulation to induce the enhanced protection with an optimal Th1 response.

급이 및 비급이 참돔의 색, 맛 및 영양성분 비교 (Comparison of Muscle Color, Taste and Nutrition Components Between Red Seabreams Cultured by Feeding and Starving)

  • 참돔의 효율적 이용을 위한 기초 연구로 급이 및 비급이 참돔의 색조, 맛 및 영양 특성에 대하여 살펴보았다. 비급이 참돔의 일반성분은 수분의 경우 72.7%, 조단백질의 경우 21.1%, 조지방의 경우 3.7% 및 조회분의 경우 1.4%로, 급이 참돔의 일반성분에 비하여 수분의 경우 약 3%가 높았고, 조지방의 경우 약 3%가 낮았으며, 조단백질 하량과 조회분 함량에서는 거의 차이가 없었다. 비급이 양식산 참돔 근육의 명도, 적색도, 황색도 및 색차는 각각 37.52, -1.47, 0.71 및 59.33으로, 급이 양식산 참돔 근육의 헌터 색조에 비하여 명도의 경우 미미한 정도에 높았던 반면 기타 적색도, 황색도 및 색차의 경우 차이가 인정되지 않았다. TCA 가용성 질소 함량은 비급이 참돔이 403.8 mg/100 g으로, 급이 참돔 보다 높았다. Taste value로 살펴 본 급이 및 비급이 참돔의 주요 유리아미노산은 두 종 모두가 glutamic acid, alanine, lysine 및 histidine이었다. 총 아미노산 함량은 비급이 참돔이 21.2 g/100 g으로, 급이 참돔보다 약간 높았다. 주요 구성 아미노산으로는 급이 유무에 관계없이 두 종의 참돔이 모두 glutamic acid(약 14.0%), aspartic acid(약 10%) 및 lysine(약 10%) 등으로 차이가 없었다. 비급이 참돔이 급이 참돔에 비하여 칼륨의 경우 높았고, 마그네슘의 경우 낮았으나, 기타 철 및 칼슘 함량의 경우 차이가 없었다. 관능검사 결과 비급이 참돔이 급이 참돔에 비하여 색조 및 냄새의 경우 차이가 없었으나, 맛 및 조직감의 경우 우수하다고 평가되었다.
