The effects of MCC and palm oil at different levels on lipid metabolism were assessed in adolescent rat. Dietary fat levels were 20% and 40% (kcal/kcal) and MCC level were 0%, 2%, and 4% (wt/wt). The experimental period took 8 weeks. During the experiment, weight gain and food efficiency ration were not affected by dietary factors. The weight of thymus however, was lower in high fat groups than in middle fat groups. The contents of total lipid and total cholesterol in plasma and HDL-cholesterol in plasma and HDL-cholesterol were significantly affected by dietary factors. The concentrations of cholesterol in LDL and VLDL , and the triglyceride content of VLDL was higher in high fat groups than in middle fat groups. Adding MCC lowered the LDL triglyceride in the high fat groups. The analysis of plasma fatty acids generally reflected the composition of the dietary palm oil. MCC had significant effects on total lipid and triglyceride of feces, but not on total cholesterol . In conclusion, the level of MCC had little hypolipodemic effects on lipid metabolism in rats fed a high fat diet. the middle fat group that contained 4% MCC showed increased contents of fecal triglyceride than the others, indicating that MCC had an effect on lipid absorption. Therefore, the other physiological functions of MCC need to be tested for their useful applications.
We investigated the pathogenesis of liver tissue damage during the lipid peroxidation and fibrogenesis with the observation of correlations between the parameters of collagen synthesis (and deposition) and lipid peroxidation in liver fibrosis (cirrhosis) rats. Rats were randomly divided into two groups, normal and $CCl_4$-thiopental sod. intoxicated group. And the one group was treated intragastrically with the mixture of $CCl_4$-thiopental sod. 3 times per week for 3 weeks. The liver tissue and sera were used for the measurement of hydroxyproline (HYP), malonedialdehyde (MDA) and superoxide dismutase (SOD). Biochemical parameters such as aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), total-bilirubin and blood urea nitrogen (BUN) were measured. Additionally, the expression of collagen ${\alpha}1$(III) and $\beta$-actin mRNA was observed by RTPCR. The histological change in liver tissue was also observed by Masson's trichrome and H&E staining. Correlation analysis was carried by Spearman's rho method. All biochemical parameters except total-bilirubin were significantly higher in the $CCl_4$-thiopental sod. treated group than that of the normal group (p < 0.01). In the $CCl_4$-thiopental sod. treated group, Hyp as a parameter of collagen synthesis (deposition) and MDA as a metabolite of lipid peroxidation, were significantly elevated by 1.98 and 2.11 times higher than that of the normal group (p < 0.001) respectively. The activity of SOD in the $CCl_4$-thiopental sod. treated group is decreased significantly by 44.8% (p < 0.001). And collagen ${\alpha}1$(III) mRNA was more expressed in the $CCl_4$-thiopental sod. treated group than that of the normal group. However, the expression of $\beta$-actin mRNA is showed similar in both of groups. A good correlation was observed between the content of hyp and MDA concentration (r = 0.70, n = 40) in the two groups. And the correlation between the levels of hyp and SOD (r = -0.71, n = 25) is also reliable. However, no correlation were observed between MDA concentration and SOD (r = -0.40, n = 25) in the two groups. Elevated levels of MDA in $CCl_4$-thiopental sod. treated rats indicated enhancement of lipid peroxidation, which is accompanied by a decrease in SOD activity. Moreover, we could confirm that the parameters of collagen synthesis (and deposition) is in good correlation with the metabolite of lipid peroxidation (MDA) and the lipid peroxidation antagonizing enzyme (SOD). Hence, we propose that (1) lipid peroxidation and collagen synthesis (and deposition) could be enhanced by intragastrically application of $CCl_4$-thiopental sod. during a short terms. And (2) the intoxication of $CCl_4$-thiopental sod. could be used for monitoring of lipid peroxidation and collagen synthesis (and deposition) for test of antioxidant and antifibrotic agent.
Objective: This study was conducted to investigate the lipid-lowering effect and safety of Daeshiho-tang in patients with uncontrolled lipid levels by statins. Methods: We investigated patients who had an abnormal lipid profile even when taking statins and who were administered Daeshiho-tang at Kyung-Hee University Korean Medical Hospital for at least one day between January 2008 and December 2018. Their basal characteristics and examinations were reviewed retrospectively with respect to lipid profile, AST, ALT, GGT, BUN, and creatinine. The lipid profile was composed of total cholesterol, triglyceride, LDL-cholesterol, and HDL-cholesterol. Subgroup analysis was performed on each component of dyslipidemia. Results: Among 20 participants, there were 10 males and 10 females. The mean BMI was 23.52. Eighty five percent of the participants were diagnosed as having cerebral infarction. After the administration of Daeshiho-tang, total cholesterol and LDL cholesterol were significantly reduced, to 41.3 mg/dl and 33.95 mg/dl, respectively. In subgroup analyses, total cholesterol and LDL cholesterol were significantly decreased, to 63 mg/dl and 54.6 mg/dl, respectively. Liver and kidney function showed no significant difference after taking Daeshiho-tang. Conclusions: Daeshiho-tang as a decoction or powder had significant lipid-lowering effects on total cholesterol and LDL cholesterol in patients with dyslipidemia. The lipid-lowering effect on total and LDL cholesterol increased in patients with hypercholesterolemia and hyper-LDL-cholesterolemia, respectively. Based on the minimal changes in the liver and kidney function test, Daeshiho-tang would be safe enough to be used in clinics.
Objective : There is accumulating data that cardiovascular diseases, including hypertension, cerebro-vascular accident, coronary heart disease, relate to atherosclerotic changes. Increased serum levels of lipids could play a role in these circumstances. the SDPTG(the second derivative of photoplethysmogram waveform) is noninvasive methods for evaluating the pulse wave and is correlated with age and other risk factors for atherosclerosis. Methods : To study the effects of serum lipid levels on the SDPTG in the middle-age adults, data on serum lipid levels and the SDPTG were collected in 80 healthy middle age with varying serum lipid concentrations. Biological vascular age was assessed using the SDPTG. the SDPTG consists of an a, b, c, and d wave in systole and an e wave in diastole; an SDPTG aging index(Al) was calculated as (b-c-d-e)/a. Results : Serum triglyceride, total cholesterol, total lipid, phospholipid, HDL cholesterol levels were respectively and significantly correlated with SDPTG Al. Serum lipid levels and blood pressure are age-dependent and the SDPTG is blood pressure-dependent. Restricting analysis to age and blood pressure yield strong positive correlations between serum triglyceride, total lipid levels and SDPTG Al, but no correlations between serum total cholesterol, phospholipid levels and SDPTG Al. Conclusion : The results of this study suggest that preventive acupuncture programs or herbal medications for reducing serum lipid, especially triglyceride, levels in middle-age should include a evaluation of biological vascular aging with the second derivative of photoplethysmogram waveform.
Drosophila melanogaster (fruits fly) is a representative model system widely used in biological studies because its brain function and basic cellular processes are similar to human beings. The whole head of the fly is often used to obtain the key function in brain-related diseases like degenerative brain diseases; however the biomolecular distribution of the head may be slightly different from that of a brain. Herein, lipid profiles of the head and dissected brain samples of Drosophila were studied using electrospray ionization-mass spectrometry (ESI-MS). According to the sample types, the detection of phospholipid ions was suppressed by triacylglycerol (TAG), or the specific phospholipid signals that are absent in the mass spectrum were measured. The lipid distribution was found to be different in the wild-type and the microRNA-14 deficiency model ($miR-14{\Delta}^1$) with abnormal lipid metabolism. A few phospholipids were also profiled by comparison of the head and the brain in two fly model systems. The mass spectra showed that the phospholipid distributions in the $miR-14{\Delta}^1$ model and the wild-type were different, and principal component analysis revealed a correlation between some phospholipids (phosphatidylethanolamine (PE), phosphatidylinositol (PI), and phosphatidylserine (PS)) in $miR-14{\Delta}^1$. The overall results suggested that brain-related lipids should be profiled using fly samples after dissection for more accurate analysis.
Lipids play important roles in biological systems; they store energy, play a structural role in the cell membrane, and are involved in cell growth, signal transduction, and apoptosis. Phospholipids (PLs) in particular have received attention in the medical and lipidomics research fields because of their involvement in human diseases such as diabetes, obesity, atherosclerosis, and many cancers associated with lipid metabolic disorders. Here I review experimental strategies for PL analysis based on nanoflow liquid chromatography-electrospray ionization-tandem mass spectrometry (nLC-ESI-MSn). In particular, discussed are lipid extraction methods, nanoflow LC separation of PLs, effect of ionization modifiers on the ESI of PLs, influence of chain lengths and unsaturation degree of acyl chains of PLs on MS intensity, structural determination of the molecular structure of PLs and their oxidized products, and quantitative profiling of PLs from biological samples such as tissue, urine, and plasma in relation to cancer and coronary artery disease.

Purpose : We investigated the effects of horseback riding and horseback riding machine exercise on the serum lipid profile in elderly individuals. Methods : Our study included 30 healthy elderly individuals who were randomly assigned to 2 groups: the horseback riding and the horseback riding machine exercise group, with each group comprising 15 individuals. The riding exercise program was performed 25 times a week over 12 weeks. The repeated measures analysis of variance was used to determine changes in serum lipid profiles before and after exercise 6 and 12 weeks after exercise. Results : An intergroup comparison of blood tests showed that triglyceride, total cholesterol and low-density lipoprotein-C levels were decreased in both groups after 12 weeks of the riding exercise program. However, both groups showed an increase in high-density lipoprotein-C levels, although this difference was not statistically significant. Conclusion : Horseback riding and horseback riding machine exercise showed a positive effect on the serum lipid profile in elderly individuals.

As a member of the focal adhesion complex of the plasma membrane, Src is a nonreceptor tyrosine kinase that controls cell adhesion and motility. However, how Src activity is regulated in the plasma membrane microdomain in response to components of the extracellular matrix (ECM) remains unclear. This study compared and investigated the activity of Src in response to three representative ECM proteins: collagen type 1, fibronectin, and laminin. Genetically encoded FRET-based Src biosensors for plasma membrane subdomains were used. FRET-based biosensors allow the real-time analysis of protein activity in living cells based on their high spatiotemporal resolution. The results showed that Src activity was maintained at a high level under all ECM conditions of the lipid raft, and there was no significant difference between the ECM conditions. In contrast, Src activity was maintained at a low level in the non-lipid raft membrane. In addition, the Src activity of lipid rafts remained significantly higher than that of non-lipid raft regions under the same ECM conditions. In conclusion, this study demonstrates that Src activity can be controlled differently by lipid rafts and non-lipid raft microdomains.
Juvenile visceral steatosis (JVS) mouse is an animal model of the systemic camitine deficiency. JVS mice first develop fatty liver following cardiac hypertrophy. hyperammonemia, etc. To clarify the relationship between fatty liver and other symptoms. lipid hydroperoxides levels of peripheral oragans in JVS mice at 1 month were determined by the use of phosphine derivatives. We also report here a new method to quantitate the lipid components level in fatty liver of JVS mice.
In order to evaluate anti -oxidant activities and protective effect against oxidatve damage, DPPH radical scavenging activity and lipid peroxidation inhibitory activity were measured among methanol extracts prepared from natural medicinal plants. Fourteen natural medicinal plants which were reported to have anti -oxidative or anti-inflammatory effects were selected based on our previous report. In addition to the total methanol extracts, n-hexane, dichloromethane, ethylacetate, n-butanol and water fractions were prepared from each total extract. DPPH radical scavenging assay was performed against 14 total extracts and all samples showed dose-dependent activities in various extent. Among those, 6 samples, methanol extracts of Euryale ferox, paeonia suffruticosa, Areca catechu var. dulcissima, Cinnamomun cassia, Alpinia katsumadai and Betula platyphlla var. japonica showed IC$\sub$50/ value lower than 6.0 $\mu\textrm{g}$/ml. The highest DPPH radical scavenging activity was found in ethylacetate fraction of paeonia suffruticosa with IC$\sub$50/ value of 1.1 $\mu\textrm{g}$/ml. Analysis of lipid peroxidation inhibitory activity on hydrogen peroxide-induced oxidative damage in Chinese hamster lung fibroblast (V79-4) cells revealed that the highest inhibitory effect was observed in methanol extract of Betula platyhpylla var. japonica. Lipid peroxidation inhibitory activity was observed as a dose-dependent manner in all samples used in this study. Among fraction samples, ethylacetate fraction of Alpinia katsumadai had the strongest inhibitory activity with IC$\sub$50/ value of 0.9 $\mu\textrm{g}$/ml.
