수(數) 종(種) 한약재(韓藥材)가 생쥐의 골수(骨髓) 및 비장세포(脾臟細胞)의 조혈촉진(造血促進)과 방사선(放射線) 방어(防禦)에 미치는 영향 (Hemopoietic and Radioprotective Effects of Several Herbs on Murine Splenocytes and Bone Marrow Cells)

  • Astragalus membranaceus Bunge (黃?), Atractylodes macrocephala Loidz (白朮), Angelica sinensis(Oliv) Kiels (當歸), Lycium chinense Mill (枸杞子), Ligustrum lucidum Ail (女貞子) are Herbs that are frequently used in a lot of prescriptions to reduce the side effects of anti-cancer therapies, especially like chemotherapy and radiotherapy. The radioprotective and hemopoietic effects of these Herbs on BALB/c splenocytes and bone marrow cells are measured. In order to evaluate the Hemopoietic effects, Thymidine uptakes and secretion of colony stimulating factors(CSFs) of splenocytes and myelocytes treated with herbs were measured. Radioprotective effects were accessed by the method of immunocompetence of murine lymphocytes cultured with herbs before irradiation and with LPS, ConA after irradiation. The results are as follows. 1. The orders of Proliferative effects of herbs on splenocytes were Atractylodes macrocephala Loidz (白朮), the higest of all, Astragalus membranaceus Bunge (黃?), Angelica sinensis(Oliv) Kiels (當歸). Lycium chinense Mill (枸杞子) and Ligustrum lucidum Ait (女貞子), the lowest of all. At Optimal concentration, the proliferation ratios of herb-treated splenocytes compared to non-treated ones were like these. Atractylodes macrocephala Loidz (白朮) 44.3, Astragalus membranaceus Bunge (黃?) 17.7, Angelica sinensis(Oliv) Kiels (當歸) 10, Lycium chinense Mill (枸杞子) 6.4, Ligustrum lucidum Ait (女貞子) 2.0.(p<0.05) 2. When splenocytes were cultured during different periods, Atractylodes. macrocephala Loidz (白朮) and Astragalus membranaceus Bunge (黃?) showed the higest proliferation on 3th day, Angelica sinensis(Oliv) Kiels (當歸), Lycium chinense Mill (枸杞子) on 4th day,(p<0.05) and Ligustrum lucidum Ait (女貞子) until 5th day but with no significant increase. 3. The orders of Proliferative effects of herbs on Bone Marrow(BM) cells were Atractylodes macrocephala Loidz (白朮), the higest of all, Astragalus memhranaceus Bunge (黃?), Angelica sinensis(Oliv) Kiels (當歸), Lycium chinense Mill (枸杞子) and Ligustrum lucidum Ait (女貞子), the lowest of all too. At Optimal concentration, the proliferation ratios of herb-treated BM cells compared to non-treated ones were like these. Atractylodes macrocephala Loidz (白朮) 21.7, Astragalus membranaceus Bunge (黃?) 9.9, Angelica sinensis(Oliv) Kiels (當歸) 4.9, Lycium chinense Mill (枸杞子) 2.3, Ligustrum lucidum Ait (女貞子) 1.4(p<0.05). 4. The secretion ratio of colony stimulating factors(CSFs) of each herb-treated group, compared to control, was Atractylodes macrocephala Loidz (白朮) 9.4, Astragalus membranaceus Bunge (黃?) 9.0, Angelica sinensis(Oliv) Kiels (當歸) 4.4, Lycium chinense Mill (枸杞子) 3.8 (p<0.05) but no significant increase in Ligustrum lucidum Ait (女貞子). 5. The mitogen(ConA, LPS) stimulated-lymphocytes cultured with each herb before irradiation of 1-3 Gy showed more enhanced proliferation than control(p<0.05). When compared to each non-irradiated group of all groups, the orders of percentage increase of irradiated group were Atractylodes macrocephala Loidz (白朮), the higest of all, Astragalus membranaceus Bunge (黃?), Angelica sinensis(Oliv) Kiels (當歸), Ligustrum lucidum Ait (女貞子), Lycium chinense Mill (枸杞子). Each percentage showed significant enhancement compared to control group(p<0.05). According to the results, Atractylcdes macrocephala Loidz (白朮), Astragalus membranaceus Bunge (黃?) are suggested to be the most effective hemopoietic and radioprotective herbs, and Angelica sinensis(Oliv) Kiels (當歸), Lycium chinense Mill (枸杞子) the next, but Ligustrum lucidum Ait (女貞子) showed lower effects than expected.

Influence of Ligustrum lucidum and Schisandra chinensis Fruits on Antioxidative Metabolism and Immunological Parameters of Layer Chicks

  • The experiment was conducted to evaluate the effects of Ligustrum lucidum (LL) and Schisandra chinensis (SC) on the growth, antioxidative metabolism and immunity of laying strain male chicks. The results showed that diets supplemented with 1% of either LL or SC had no effects on the growth performance of chicks compared with the control. Furthermore, both LL and SC significantly reduced malondialdehyde (MDA) concentration of serum and heart of chicks (p<0.05). In addition, superoxide dismutase (SOD) activity of serum of the birds was significantly elevated by supplementation with SC (p<0.05). Glutathione reductase (GR) activity of heart and serum of the birds was significantly elevated by supplementation with LL or SC (p<0.05). LL supplementation significantly elevated antibody values against Newcastle Disease virus (NDV)(p<0.05) and lymphoblastogenesis (p<0.05) of the birds. The results suggest that diets supplemented with 1% of either LL or SC may improve immune function and antioxidant status of chicks.

  • The effect of Ligustrum Lucidum(LL) on the production of nitric oxide (NO) and superoxide by murine peritoneal macrophages were investigated. Stimulation of the cells with LL in the presence or absence of interferon-r(IFN-r) resulted in the increased accumulation of nitrite in the medium. To further examine the mechanism of LL induced. NO Synthesis, we evaluated the secretion of tumor necrosis $factor-{\alpha}(TNF-{\alpha})$ by LL in murine macrophages. Treatment of LL increased the secretion of bioactive $TNF-{\alpha}$ in cultured medium. In addition, LL induced NO production was decreased by the treatment of anti-murine $TNF-{\alpha}$. neutralizing antibodies, indicating that LL induced superoxide production was decreased by the treatment of anti-murine $TNF-{\alpha}$ neutralizing antibodies. These data suggested that LL induced superoxide production was related to $TNF-{\alpha}$ secretion. In conclusion, our results indicates that LL may enhance innate immune response and be applied as a immunoregulating drug improving phagocytosis.

생쥐에서 급성 폐렴에 대한 여정자 열매 추출물의 억제효과 (Ligustrum lucidum Fruits Extract Inhibits Acute Pulmonary Inflammation in Mice)

  • Traditionally, Ligustrum lucidum fruits (LL) is one of the well-known oriental herb used in the treatment of skin and lung inflammation. This study investigated anti-inflammatory effects of LL in the pathogenesis of acute pulmonary inflammation in mice. Acute pulmonary inflammation was induced by intratracheal instillation of cigarette smoke condensate (CSC) and lipopolysaccharide (LPS) 5 times within 12 days in mice. LL extract was administered orally at a dose of 50 or 200 mg/kg. Administration of LPS and CSC significantly elevated airway hyperresponsiveness (AHR) to mice, and increased in the levels of inflammatory cells and mediators in mice. However, the LL extract significantly reduced the elevated AHR, and the increase of neutrophils, $CD4^+/CD3^+$ cells and $CD8^+/CD3^+$ cells, along with reducing the expression of TNF-${\alpha}$, IL-6, and MIP-2. Moreover, the LL extract alleviated the infiltration of inflammatory cells in expanded airway walls histologically. These results indicate that the LL extract has an inhibitory effects on acute pulmonary inflammation and AHR in murine model, and plays a crucial role as a immunomodulator which possess anti-inflammatory property.

광나무의 주성분, Ursolic acid와 Oleanolic acid의 항노화 효능 (Excellent Anti-aging Effects of Ursolic acid and Oleanolic acid Present in Ligustrum lucidum)

  • 광나무(L. lucidum)는 ursolic acid와 oleanolic acid를 다량 포함하고 있다. 본 연구에서는 광나무 열매, 줄기, 잎 세 부위 추출물의 항주름 효능을 평가하였다. 광나무 추출물은 human skin fibroblasts에서 독성이 없을 뿐만 아니라 MMP-1과 MMP-2의 발현을 감소시키고 COL1A1의 발현을 증가시켰다. 이들 추출물은 모두 농도 의존적으로 COL1A1의 발현을 증가시켰으며 MMP-1과 MMP-2의 발현을 감소시켰다. 광나무 세 부위 추출물 가운데, 열매 부위에 가장 많은 양의 ursolic acid 와 oleanolic acid가 함유되어 있었으며 가장 강한 COL1A1 upregulating 효과와 MMP-1과 MMP-2 downregulating 효과를 나타냈다. 이처럼 항주름 효능을 보이는 광나무 열매 추출물은 기능성 화장품 소재로 개발될 수 있는 가능성이 있다.

Effects of Ligustrum lucidum Fruits on Growth Performance, Antioxidation and Meat Quality in Arbor Acres Broilers

  • This study was conducted to evaluate the effects of Ligustrum lucidum (LL) on growth performance, antioxidation, and meat quality in broilers. 270 birds (1 d old) were allotted to 3 treatments with 6 replicates per treatment. The feeding program included a starter diet from d 1 to 21 of age and a grower diet from d 22 to 42 of age. The birds were given a basal corn-soybean meal diet supplemented with 0 (the control), 5 or 10 g of LL/kg, respectively. The results showed that in the starter, grower and overall phase, broilers fed with 5 or 10 g of LL/kg had better (p<0.01) average daily gain, but there were no differences in feed: gain between treatments. In the starter and overall phase, average daily feed intake of LL groups was greater (p<0.05) than that of the control, but this difference was not observed in the grower phase. Supplementation of LL significantly increased (p<0.05) superoxide dismutase activity and total antioxidant capability in serum of chickens. In breast muscle, birds fed 5 g of LL/kg had an increase (p<0.05) in superoxide dismutase activity. The LL supplementation significantly decreased (p<0.05) malondialdehyde contents. Adding 5 or 10 g of LL/kg to the diet significantly increased pH value and reduced drip loss of meat (p<0.05). The results of this study indicated that dietary LL could improve growth performance, increase pH value and reduce drip loss of meat by decreasing lipid peroxidation and by improving antioxidative status in broilers.

반응표면분석법을 이용한 여정자의 페놀계 항산화 성분에 대한 마이크로웨이브 추출조건 최적화 (Optimization of Microwave Extraction Conditions for Antioxidant Phenolic Compounds from Ligustrum lucidum Aiton Using Response Surface Methodology)

  • 본 연구에서는 지역별 여정자 추출물의 항산화 활성 및 총 페놀과 총 플라보노이드 함량을 비교분석한 결과 해남산이 제주산과 중국산보다 우수함을 확인할 수 있었다. 총 페놀과 총 플라보노이드 함량은 국내 해남산이 중국산에 비해 각각 2배와 1.6배의 높은 값을 보였고, 해남산 여정자 추출물의 항산화는 DPPH법과 ABTS법에서 중국산에 비해 약 2배 이상의 높은 활성을 나타내었다. 활성이 높은 해남산 여정자의 추출공정을 최적화하기 위해서 반응표면분석을 이용하여 여정자의 추출온도와 추출시간에 따른 전자공여능, 총 페놀 함량, 총 플라보노이드 함량을 조사하였다. 중심합성계획에 따라 추출온도($X_1$), 추출시간($X_2$)을 독립변수($X_i$)로 하고 추출물의 특성 즉, 전자공여능($Y_1$), 총 페놀 함량($Y_2$), 총 플라보노이드 함량($Y_3$)을 종속변수로 하여 추출 최적화를 수행하였다. 그 결과 $R^2$은 0.8859~0.9517로 10% 이내에서 유의성이 인정되었으며, 전체 모델의 유의 확률은 0.0026~0.0206으로 모두 통계적으로 유의한 값을 나타내었다. 이들 추출물의 특성을 모두 만족시키는 최적 추출조건은 추출온도 $131.68^{\circ}C$, 추출시간 5.49분으로 나타났다. 예측된 최적 추출조건의 임의의 점에서 실험한 결과, 각 종속 변수들의 예측값이 전자공여능은 35.38%, 총 페놀 함량은 6,045.27 mg/100 g, 총 플라보노이드 함량은 103.24 mg/100 g이었고, 실제값은 전자공여능이 35.18%, 총 페놀 함량이 6,055.68 mg/100 g, 총 플라보노이드 함량이 103.72 mg/100 g으로 예측값과 실제값은 유사하게 나타났다. 따라서 마이크로웨이브 장치를 이용하여 여정자의 페놀계 화합물 추출조건을 최적화함으로써 단시간에 시료의 유용성분을 효과적으로 추출할 수 있었고, 여정자의 유용성분 열수추출 방법을 이용하여 다양한 식품소재로써 활용 가능성을 제시하게 되었다.

Compatibility effects of ginseng and Ligustrum lucidum Ait herb pair on hematopoietic recovery in mice with cyclophosphamide-induced myelosuppression and its material basis

  • Background: Ginseng (G) and Ligustrum lucidum Ait (LLA) are core traditional Chinese medicines in treating myelosuppression formula. The present study was designed to profile effect of G and LLA herb pair (G-LLA) on myelosuppressed mice. Methods: The mice myelosuppression model was established by intraperitoneal (i.p.) injection of cyclophosphamide (Cy). Hematopoietic function of bone marrow was measured by hemopoietic progenitor cell culture and peripheral blood count, and serum hemopoietic factors were tested by enzyme-linked immunosorbent assay. Bone marrow cell cycle was performed by flow cytometry. HPLC was used to measure 20 potential chemical components related to myelosuppression, including ginsenoside Rg1, Re, Rb1, Rc, Rb2, Rb3, Rd, Rk3, Rh4, 20 (S)-Rg3, 20 (R)-Rg3, Rk1, Rg5, salidroside, and so on. Results: G, LLA, and G-LLA improved the amount of peripheral blood cells and bone marrow cells of myelosuppressed mice (P < 0.01). They significantly increased the colony quantity of colony-forming unit-granulocyte macrophage, burst-forming unit-erythroid, colony-forming unit-erythroid, and colony-forming unit-megakaryocyte and amount of G2/M and S phase cells (P < 0.01). They also significantly decreased the amount of hematopoiesis-related cytokines (P < 0.01). The content of chemical components in G-LLA changed, and the change of rare saponin was the most obvious. Conclusion: These results show that G-LLA herb pair might produce synergistic or complementary compatibility effects on bone marrow suppression after chemotherapy. It suggests that the substance basis of G-LLA for treating bone marrow suppression may be effective chemical components.

인진, 울금, 구기자, 여정자 복합추출물(ACLL)의 항이상지질혈증 효능에 대한 실험적 연구 (Anti-dyslipidemic Effect of ACLL extract (Artemisia capillaris Thunb, Curcuma longa L, Lycium chinense, Ligustrum lucidum Aiton complex extract) on High Cholesterol Diet-induced Dyslipidemia in Mice)

  • Objectives : This study was aimed to investigate the effect of ACLL extract (Artemisia capillaris Thunb, Curcuma longa L, Lycium chinense, Ligustrum lucidum Aiton complex extract) in a dyslipidemia animal model induced by a high-fat diet. Methods : ACLL extracts was administered at 200 mg/kg, and 400 mg/kg to Apoe mice, where arthritis was induced by high cholesterol diet. After mice were treated with GO for 4 weeks, we measured AST, ALT, BUN and lipid levels (total cholesterol, HDL cholesterol, LDL cholesterol, triglyceride), atherogenic index(AI) and cardiac risk factor(CRF). Results : Serum AST, ALT, creatinine, BUN levels were not changed by ACLL extract do not show any toxic effects. Also, ACLL extract groups were found to have atherogenic Index and cardiac risk factor as well as lipid metabolism improvement (total cholesterol, LDL cholesterol and triglyceride decrease). Conclusions : We suggest that ACLL extract may have the control effects of Dyslipidemia by improving lipid metabolism.

제주도 자생식물들에 대한 미백 및 항산화 효능 탐색 (Preliminary Screening of Some Jeju Island Native Plants for Whitening and Antioxidant Activity)

  • 본 연구에서는 제주도에서 자생하는 37 종의 식물추출물에 대한 미백 및 항산화 효능을 조사하였다. 미백 효능은 in vitro tyrosinase 저해활성과 B16-F1 세포를 이용한 멜라닌 합성 저해효과를 조사하여 확인하였고, 항산화 효능은 DPPH (1,1-diphenyl-2-picrylhydrazyl)를 이용한 전자공여능 측정방법과 DCF-DA (dichlorofluorescin diacetate)를 이용한 V79-4 폐 섬유아세포에서 활성 산소종 소거작용 능력을 조사하여 측정하였다. 세포실험 조건에서 식물추출물들의 투여농도에 따른 세포독성 정도를 조사하기 위하여 MTT (3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide) assay를 수행하였다. 미백 효능조사 실험에서 우수한 결과를 나타내었던 8종의 식물 추출물들은 토끼를 이용한 국소 독성 시험 및 인체 피부 자극 임상 시험에 적용하여 안전성을 조사하였다. 연구결과에서 높은 미백과 항산화 효능을 동시에 나타내었던 비수리(Lespedeza cuneata), 제주광나무(Ligustrum lucidum (stem)), 산뽕나무(Morus bombycis (stem)), 꿀풀(Prunella vulgaris var. lilacina)의 식물추출물들은 화장품 원료화에 매우 유리한 조건을 갖고 있음을 확인하였다.
