귀전우(鬼箭羽)약침액이 대식세포의 Nitric Oxide와 Interleukin-6의 생성에 미치는 영향 (The Effects of Euonymi Ligunum Suberalatum Phamacopuncture on Nitric Oxide and Interleukin-6 Production in Macrophage)

  • Objectives : The objective of this study is to study the effects of Euonymi Lignum Suberatatum pharmacopuncture solution on NO and IL-6 production in macrophage. Methods : At first, the RAW 264.7 macrophage was subclutured. In order to evaluate cytotoxicity, MTT assay performed. Then, the cell was induced by LPS, INF-$\gamma$ and Experimental groups were divided into five(Normal, Control, Euonymi Lignum Suberatatum 100, 200, $300{\mu}g/m{\ell}$). Then Euonymi Lignum Suberatatum pharmacopuncture solution was put into cell. We measured IL-6, iNOS, NO. Results : The cytotoxic effect of Euonymi Lignum Suberatatum pharmacopuncture solution in RAW 264.7 macrophage was not appeared. $300{\mu}g/m\ell$ Euonymi Lignum Suberatatum pharmacopuncture solution inhibited IL-6 production in LPS, INF-$\gamma$-stimulated RAW 264.7 macrophages significantly. Euonymi Lignum Suberatatum pharmacopuncture solution inhibited iNOS revelation in LPS, INF-$\gamma$-stimulated RAW 264.7 macrophages. All group of Euonymi Lignum Suberatatum pharmacopuncture solution inhibited NO production in LPS, INF-$\gamma$-stimulated RAW 264.7 macrophages significantly. Conclusions : Our study demonstrated that Euonymi Lignum Suberatatum pharmacopuncture solution had an inhibition effect on NO production, iNOS revelation, IL-6 production. So Euonymi Lignum Suberatatum pharmaco puncture solution may have an Anti-inflammation effect.

침향(沈香)의 산지와 무역에 근거한 기원 연구 (A Research on the origin of Aquilariae Lignum based on its production area and trading status in history)

  • Objective : The purpose of this study is to determine the origin of Aquilariae Lignum. Method : Firstly identify the production areas of Aquilariae Lignum and its trading status with China in Chinese history through Chinese historical books such as Twenty-Five Histories(二十五史) and the records of the Chosun Dynasty and then, compare the distribution of the genus Aquilaria in the concerned areas. Result : Since the records in the NanfangCaomuZhuang(南方草木狀) written in 304 saying that Aquilariae Lignum was produced in Vietnam and had white flowers, Vietnam had led production and trading of Aquilariae Lignum until Qing Dynasty(淸代). Even though Thailand traded Aquilariae Lignum during Qing Dynasty, however, the volume was at a low level. Aquilariae Lignum from southern Thailand, Malaysia and Indonesia was rated as low quality and low-priced because of its fishy smell and strong flavor. Conclusion : These results show that the origin of Aquilariae Lignum comes from Vietnam and this species is distinguished from the ones of Indodesia or Malaysia.

침향(沈香)의 항알레르기 효과(效果)에 대한 연구(硏究) (Studies on the Antiallergic Effect of Aquillariae Lignum)

  • The inhibitory activity of Aquillariae Lignum (Thymelaeaceae) on type Ⅰ immediate hypersensitivity of the anaphylactic type in the wistar rat model of passive cutaneous anaphylaxis, an IgE-mediated, mast cell-dependent reaction. Administered orally at 250, 500 mg/kg body weight 1 h before the challenge, Aquillariae Lignum potently inhibited PCA in rats which disodium cromoglycate showed poor inhibitory activity. Aquillariae Lignum inhibited compound 48/80-induced anaphylaxis 100% with a dose of 0.5 g/kg body weight at 1 h before or 5 and 10 min after injection of compound 48/80. Aquillariae Lignum (0.05-1.6 mg/ml) also exhibited the dose-related inhibitory effect on compound 48/80-induced histamine release from rat_peritoneal mast cells. Moreover, it was clearly demonstrated that Aquillariae Lignum and disodium cromoglycate disodium cromoglycate potently inhibited such type Ⅰ allergic reactions as anaphylactic shocks, suggesting that these drugs, at least in part, share the same mechanism of action It is suggested that Aquillariae Lignum may exert a stronger inhibition on the mast cell degranulation process. Since Aquillariae Lignum (1.0 mg/ml) inhibited about 90% of histidine decarboxylase activity, the inhibitory activity of Aquillariae Lignum for histamine release was considered to be derived from the inhibition of histidine decarboxylase activity. It results from increased expression of the mRNA coding for histidine decarboxylase, as assessed by Northern blot analysis after a 12 h incubation to P-815 cells with dexamethasone plus 12-O-tetradecanoylphorbol-13-acetate. The addition of Aquillariae Lignum to P-815 cells with dexamethasone plus 12-O-tetradecanoyl-phorbol-13-acetate, significantly inhibited the histidine decarboxylase gene expression. Tumor necrosis $factor-{\alpha}$ was not constitutively expressed in P-815 cells. Substance P selectively activates the tumor necrosis $factor-{\alpha}$ gene expression in P-815 cells. Aquillariae Lignurm inhibited substance P-induced tumor necrosis $factor-{\alpha}$ gene expression. Furthennore, The effect of Aquillariae Lignum on the mRNA expression of novel protein kinase C ${\delta}$ a major isoform of mast cells, was examined by Northern blot analysis. The expression of novel protein kinase C ${\delta}$ mRNA in the presence of Aquillariae Lignum was significantly lower than in the absence of Aquillariae Lignum. These results suggest the possibility that the inhibition of allergic reaction by Aquillanae Lignum should be regulated by tumor necrosis $factor-{\alpha}$ and novel protein kinase C ${\delta}$.

침향(沈香)의 침수성(沈水性)에 관한 문헌적 연구 (A Philological study on Submersion Property of Aquilariae Lignum)

  • Objectives : The purpose of this study is searching for grounds of new regulations about submerging properties of Aquilariae Lignum described in the Korean Herbal Pharmacopoeia. Methods : Through the investigation of the various Chinese histories and herbal classics, the definitions of Aquilariae Lignum and documents on actual trade and discrimination methods by sinking under water. Results : In Jiaozhouyiwuzhi(交州異物志) written in the early 2nd century, Aquilariae Lignum was defined as heart wood part or knots sinking under water due to hard and dark deposition of resins. Also, which are not sinking were defined as zhan xiang(棧香), and which are floating were named as qian xiang(槧香). The definitions of Aquilariae Lignum had not been changed till Ching Dynasty era, but the classification and the names about not-sinking and floating Lignum materials were a little changed. Conclusions : These results indicate that it is reasonable to make a regulation that "Aquilariae Lignum sinks under water" in the property description of The Korean Herbal Pharmacopoeia.

Aquilaria 속 식물 분포도에 근거한 심향(沈香)의 학명 (The Scientific Name of Aquilariae Lignum based on distribution of Aquilaria spp.)

  • Objectives : The purpose of this study is correct the scientific name of Aquilariae Lignum in Korean Herbal Pharmacoepia. Methods : The production areas of Aquilariae Lignum and its trading status with China in Chinese history, Sanscrit-Chinese Translation Sutra, Naming year and the discovered district in main Aquilaria spp., Several nation's Pharmacoepia, The Plant List(TPL), Convention on International Trade in Endangered Species of Wild fauna and flora(CITES) and The International Union for Conservation of Nature(IUCN) were cross-checked. Results : The records in the Jiaozhouyiwuzhi written in the early 2nd century said that Aquilariae Lignum was produced in Vietnam. NanfangCaomuZhuang written in 304 said that Agarwood in Vietnam had white flowers. Vietnam had led production and trading of Aquilariae Lignum until Qing Dynasty. Aquilariae Lignum from Malaysia and Indonesia was not traded with China. In Sanscrit-Chinese Translation Sutra, India Aquilariae Lignum was translated as Vietnam Aquilariae Lignum. Aquilaria malaccensis was discovered from Malay-Peninsular in 1783, and has green or dirty yellow flowers. A. agallocha from North-Eastern India in 1814, white flowers. A. crasssna from Vietnam in 1914, white flowers. A. crassna is different from A. malaccensis in several ways, such as flower, fruit, seed and disribution. In several Nation's Pharmacoepia, A. crassna was a synonym of A. agallocha. But in TPL, CITES and IUCN, A. malaccensis was an accepted name, and A. agallocha was a synonym of A. malaccensis. Conclusions : These results show that the original species of Aquilariae Lignum in Korea Herbal Pharmacoepia should be reversed from A. agallocha to A. crassna Pierre ex Lacomte.

동의보감(東醫寶鑑) 중(中) 목통(木通)이 주약(主藥)으로 배오(配伍)된 처방(處方)의 활용(活用)에 대한 고찰(考察) (Study on Application of Lignum akebiae Main Blended Prescription from Dongeuybogam)

  • This report describes the remedial fields, symptoms, pathology, dosage, prescriptional constitution of 46 studies related to the use of Lignum akebiae main blended prescriptions from Dongeubogam. The following conclusions were reached through investigations on the prescriptions that use Lignum akebiae as a key ingredient. Lignum akebiae blended prescriptions are utilized 17.4% of prescriptions appear in the chapter of urine. Prescriptions that utilize Lignum akebiae as the main ingredient are used commonly in the treatment of the symptoms of a urine and a cystic disease related to the urinary organs, or the diseases of eye, ear, nose, throat and the symptoms of a tumor, abscess. Lignum akebiae is utilized for an etiological and pathologenic fators related to heat, dampness, phlegm and summer heat such as excessive fever of heart and bladder, fever of small intestine, lower energizer dampness and heat, fever due to blood deficiency, phlegm fire, affection by summer heat and dampness etc. The clinical dosage of Lignum akebiae is 1 don(about 3.75gram), according to symptoms, it has ranged at a wide variety of amounts from 3 pun(about 1.13gram) to 1.5 don(about 11.25gram). Lignum akebiae is widely applied with base prescriptions such as Dozuksan, Moktongchajuntang, Manjunmoktongtang. etc.

소목(蘇木)과 그 지표물질인 brazilin이 인간 유래 각질 형성 세포의 tight junction 유전자 발현에 미치는 영향 (Investigation of the Effect of Sappan Lignum and Brazilin on Expression of Tight Junction Related-genes in Human Keratinocyte)

  • The aim of this research was to determine the diverse effects of Sappan Lignum extract and brazilin on human keratinocyte HaCaT cells. We confirmed the antioxidant effect of Sappan Lignum extract and brazilin was analyzed by using an ABTS assay, confirming the efficacy of water extraction method. Also, we examined effect of Sappan Lignum extract and brazilin on the cell viability, using the MTS assay in HaCaT cells. mRNA expression levels of tight junction-related genes associated with skin barrier in HaCaT cells were analyzed using quantitative real-time PCR analysis. Sappan Lignum extract increased the cellular activity of HaCaT cells and the expression of the tight junction-related genes claudin 3, claudin 6, and ZO-2. Brazilin displayed the same effects as that of the extract on HaCaT cells activity and tight junction-related genes expression. Furthermore, dispase assay demonstrated altered cell-cell adhesion strength of Sappan Lignum extract or brazilin treated HaCaT cells. Sappan Lignum extract or brazilin might be an useful ingredient in skin-mosturizinng and anti-wrinkle cosmetics, given its effects of altering mRNA expression of tight junction-related genes and enhancing cell-cell adhesion strength of HaCaT cells.

拱辰丹의 "麝香 代入 沈香 或 木香"에 관한 古典文獻 硏究 (Study on the "Moschus substitute for Aquilariae Resinatum Lignum or Aucklandiae Radix" of Gongjin-Dan In The Classic Literature.)

  • Objectives : The object of this paper is to seek and provide evidences for the possibilities of substitutional use of Aquilariae Resinatum Lignum and Aucklandiae Radix for Moschus as an ingredient of Gongjin-dan by clarifying the similarities and differences among these herbs in the classic literatures. Methods : We have found out the features of Moschus, Aquilariae Resinatum Lignum and Aucklandiae Radix by searching Qi, Flavor and Effect of each herb in 『Shennongbencaojing』, 『Zhengleibencao』, 『Bencaogangmu』, 『Dongeuibogam』, 『Bencaobeiyao』, 『Euijongsonik』 and 『Bangyakhappyeon』. And we have compared and analyzed the fomula of Gongjin-dan and other prescriptions in 『Beijiqianjinyaofang』, 『Shengjizonglu』, 『Shiyidexiaofang』, 『Dongeuibogam』, 『Euijongsonik』, 『Euibanghwaltu』 and 『Bangyakhappyeon』. Results : We could find out the similarities and differences in Qi, Flavor and Effect among Moschus, Aquilariae Resinatum Lignum and Aucklandiae Radix. And we could also find the examples of substitutional use of Aquilariae Resinatum Lignum as an ingredient of Gongjin-dan by comparing the formula of Gongjin-dan in the classic literatures as above. And in the other prescription, Aucklandiae Radix was also used instead of Moschus. Conclusions : All of three herbs have pungent in Flavor and warm in Qi. And these herbs also have similar effects in dispersing invading pathogenic factors and smoothening the flow of Qi. And according to 『Bencaobeiyao』, 『Euijongsonik』 and 『Bangyakhappyeon』, Aquilariae Resinatum Lignum and Aucklandiae Radix direct and regulate the Qi in common. The substitutional use of Aquilariae Resinatum Lignum and Aucklandiae Radix instead of Moschus in Gonjin-dan seems to be related to 'the ascent of Water and descent of Fire' effect of Gongjin-dan.

추출방법에 따른 소목 심재의 항산화 및 항당뇨 활성 평가 (Evaluation of Antioxidant and Anti-diabetic Effects of Sappan Lignum by Extraction Method)

  • Objectives : The heartwood of Sappan Lignum has been used since ancient times as an ingredient in folk medicines against anti-bacterial and anti-anemia purposes. Many bioactive constituents have been derived from this biomass such as chalcones and homoisoflavonoids. In the current investigation, the antioxidant and anti-diabetic properties using DPPH and $ABTS^+$ radicals scavenging, ${\alpha}-glucosidase$, and advanced glycation end products (AGEs) inhibition assays were evaluated by different extraction methods of Sappan Lignum. Methods : In our continuing investigation for bioactive natural ingredients, the antioxidant and ${\alpha}-glucosidase$ inhibitory properties of Sappan Lignum extracts were prepared from different extraction methods and the biological efficacies were investigated in vitro. The antioxidant properties were evaluated employing radical scavenging assays using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) ($ABTS^+$) radicals. In addition, the anti-diabetic effects of Sappan Lignum extracts were tested via ${\alpha}-glucosidase$ and AGEs formation inhibitory assay. The total phenolic contents were determined using a spectrophotometric method. Results : All the tested samples showed dose-dependent radical scavenging and ${\alpha}-glucosidase$ inhibitory activities. Among the tested extracts, the 80% methanolic extract of Sappan Lignum was showed the most potent activity with an $IC_{50}$ value of $82.3{\pm}1.7{\mu}g/m{\ell}$ against DPPH radical scavenging assay. While, $ABTS^+$ radical scavenging activity of 80% methanolic extract was higher than those of other extracts. Also, ${\alpha}-glucosidase$ inhibitory and AGEs formation effects of each extacts and total phenolic contents were evaluated. Conclusions : These results suggested that Sappan Lignum can be considered as a new effective source of natural antioxidant and anti-diabetic materials.

침향의 항알레르기 효과 (Antiallergic Effect of Aquilariae Lignum)

  • Effects of the aqueous extract of Aquilariae Lignum (Thymelaeaceae) on the allergic reactions were investigated. Oral administration of this extract (50, 250, and 500mg/kg) exhi bited a dose-dependent inhibition on passive cutaneous anaphylactic reactions in rats. Administrations of this extract (500mg/kg, i.p.) at 60 min before and 5, 10 min after the compound 48/80 treatment (8mg/kg, i.p.) decreased the mortality rates to 0, 0, and 14.2%, respectively. The aqueous extract of Aquilariae Lignum (0.05 ~ 1.6mg/ml) showed a dose-related inhibition on histamine release from rat peritoneal mast cells. The morphological examination also clearly showed that the aqueous extract of Aquilariae Lignum prevented the degranulation of mast cells in rats.
