Living Cell Functions and Morphology Revealed by Two-Photon Microscopy in Intact Neural and Secretory Organs

  • Laser light microscopy enables observation of various simultaneously occurring events in living cells. This capability is important for monitoring the spatiotemporal patterns of the molecular interactions underlying such events. Two-photon excited fluorescence microscopy (two-photon microscopy), a technology based on multiphoton excitation, is one of the most promising candidates for such imaging. The advantages of two-photon microscopy have spurred wider adoption of the method, especially in neurological studies. Multicolor excitation capability, one advantage of two-photon microscopy, has enabled the quantification of spatiotemporal patterns of $[Ca^{2+}]_i$ and single episodes of fusion pore openings during exocytosis. In pancreatic acinar cells, we have successfully demonstrated the existence of "sequential compound exocytosis" for the first time, a process which has subsequently been identified in a wide variety of secretory cells including exocrine, endocrine and blood cells. Our newly developed method, the two-photon extracellular polar-tracer imaging-based quantification (TEPIQ) method, can be used for determining fusion pores and the diameters of vesicles smaller than the diffraction-limited resolution. Furthermore, two-photon microscopy has the demonstrated capability of obtaining cross-sectional images from deep layers within nearly intact tissue samples over long observation times with excellent spatial resolution. Recently, we have successfully observed a neuron located deeper than 0.9 mm from the brain cortex surface in an anesthetized mouse. This microscopy also enables the monitoring of long-term changes in neural or glial cells in a living mouse. This minireview describes both the current and anticipated capabilities of two-photon microscopy, based on a discussion of previous publications and recently obtained data.

Synthesis of CdSe-TiO2 Photocatalyst and Their Enhanced Photocatalytic Activities under UV and Visible Light

  • In this study, CdSe-$TiO_2$ photocatalyst were synthesized by a facile solvothermal method and characterized by X-ray diffraction (XRD), Scanning electron microscopy (SEM) with energy dispersive X-ray (EDX) analysis and UV-vis diffuse reflectance spectrophotometer. The photocatalytic activity was investigated by degrading methylene blue (MB) in aqueous solution under irradiation of UV light as well as visible light. The absorbance of degraded MB solution was determined by UV-vis spectrophotometer. The results revealed that the CdSe-$TiO_2$ photocatalyst exhibited much higher photocatalytic activity than $TiO_2$ both under irradiation of UV light as well as visible light.

Ellipso-Microscopic Observation of Titanium Surface under UV-Light Irradiation

  • The ellipso-microscopic observation of a titanium surface undergoing anodization in $0.05mol\;dm^{-3}$ of $H_2SO_4$ was conducted. During irradiation by ultra-violet (UV) light with a wavelength of 325 nm, the titanium surface allowed for the flow of a photo-induced current and showed up as a bright, patch-like image on an ellipso-microscopic view. The brightness and patch-pattern in the image changed with flowing photo-induced current. The changes in the brightness and the image corresponded to the formation and/or degradation of titanium oxide due to the photo-electrochemical reaction of the oxide. An in situ monitoring using the ellipso-microscope revealed that the film change was dependent on the irradiation light power, by UV-light increases the anodic current and results in the initiation of pitting at lower potentials as compared with the non-irradiated condition.

Characterization of Nanoscale Electroactive Polymers via Piezoelectric Force Microscopy

  • Piezoelectric force microscopy (PFM) is a powerful method to characterize inversed piezoelectric effects directly using conductive atomic force microscopy (AFM) tips. Piezoelectric domains respond to an applied AC voltage with a characteristic strain via a contact between the tip and the surface of piezoelectric material. Electroactive piezoelectric polymers are widely investigated due to their advantages such as flexibility, light weight, and microactuation enabling various device features. Although piezoelectric polymers are promising materials for wide applications, they have the primary issue that the piezoelectric coefficient is much lower than that of piezoelectric ceramics. Researchers are studying widely to enhance the piezoelectric coefficient of the materials including nanoscale fabrication and copolymerization with some materials. In this report, nanoscale electroactive polymers are prepared by the electrospinning method that provides advantages of direct poling, scalability, and easy control. The main parameters of the electrospinning process such as distance, bias voltage, viscosity of the solution, and elasticity affects the piezoelectric coefficient and the nanoscale structures which are related to the phase of piezoelectric polymers. The characterization of such electroactive polymers are conducted using piezoelectric force microscopy (PFM). Their morphologies are characterized by field emission-scanning electron microscope (FE-SEM) and the crystallinity of the polymer is determined by X-ray diffractometer.

집토끼 결장 각 부위의 구조에 대한 광학 및 주사전자현미경적 관찰 (Morphological Diversity of Various Divisions of the Rabbit Colon)

  • Structural differences in various divisions of the rabbit colon were investigated using light and scanning electron microscopy. For light microscopic study, various Portions of the colon from seven rabbits (2.5 kg body weight) were fixed in 10% neutral buffered formalin, and paraffin sections were stained with hematoxylin-eosin. Tissues for scanning electron microscopy were fixed in 1% glutaraldehyde-1.5% paraformaldehyde and postfixed in 1% $OsO_4$, dehydrated to 100% alcohol, transfered to isoamilacetate and dried by the critical point method. Subsequently, specimens were coated with gold and viewed with a JSM-35C scanning electron microscope. The colon displays a morphological diversity along its proximo-distal axis. Five regions can be discerned based on the macroscopic and microscopic characteristics. 1) The first segment immediately distal to the cecocolical junction possessing three teniae is approximately 5 cm ($4{\sim}6cm$) in length, and displays irregular folds of the mucosa oriented transversely similar to those of the cecum. 2) The second segment possessing three teniae is about 7 cm ($5{\sim}8cm$) in length, and is characterized by the papilla-like protrusions on the mucosal surface. 3) The third segment, possessing a single tenia is about 16 cm ($12{\sim}20cm$) in length, and also displays the papilla-like protrusions similar to the aforegoing segment. 4) Fusus coli, approximately 4 cm ($3{\sim}5cm$) in length, is free of teniae and exhibits longitudinal folds on the mucosal surface. These four portions together constitute the proximal colon. 5) The distal colon reaches a length of about 58 cm ($53{\sim}55cm$) and shows a pattern of surface irregularities with minor ridges on the mucosal folds.

느릅나무 근피 드레싱이 흰쥐에 유발된 욕창의 조직재생에 미치는 효과 (The Effects of the Ulmus Root-bark Dressing in Tissue Regeneration of Induced Pressure Ulcers in Rats)

  • Purpose: The purpose of this study was to examine the effects of the ulmus root-bark dressing on tissue regeneration in experimentally-induced pressure ulcers in rats. Method: A randomized pretest/post-test control group time-series study design was used. Thirty-three male Sprague-Dawley rats were used in this study. The rats were anesthetized with 100mg/kg of ketamine. Pressure ulcers were induced at 140mmHg for three hours using a personally-designed pressing apparatus. For four weeks, the ulmus root-bark dressing was applied every other day in the experimental group (n=18) and a wet gauze dressing in the control group (n=15). For data analysis, the statistical program SPSS WIN 12 was used. The wounds were examined by light microscopy andelectron microscopy. Result: There were significant statistical differences in the size of the pressure ulcers as time went by(p=0.006). It should be noted that there were no significant statistical differences in the number of capillaries. Using light microscopy the inflammatory infiltration and neovascularization in the dermis in the experimental group emerged densely in the early stages, but recovered rapidly at the latter stages. In addition, the reepithelization of the epidermis occurred earlier than in the control group. By electron microscopy, the cell organelles of the capillary endothelial cells and the basal lamina of capillaries in the experimental group showed a more rapid maturation during the latter stages, compared with the control group. Conclusion: According to this study, it can be concluded that the ulmus root-bark dressing is effective regarding the healing of pressure ulcers.

한국산 팥꽃나무과 잎과 엽병의 해부 및 분류학적인 검토 (Leaf and petiole anatomy of Thymelaeaceae Adans. in Korea and its taxonomic consideration)

  • 한국산 팥꽃나무과에 속하는 5속 9종(팥꽃나무속: 4종, 아마풀속: 1종, 삼지닥나무속: 1종, 피뿌리풀속: 1종, 산닥나무속: 2종)의 잎의 해부학적인 두 가지 형질(중륵 횡단면, 엽병 횡단면)을 광학현미경과 전자주사현미경으로 연구하였다. 잎의 중륵의 외형은 3개의 유형 즉, 주맥부의 돌출 양상에 따라 상측이 '평평한 유형', '함몰된 유형', 혹은 '돌출된 유형'으로 구분되었다. 엽병횡당면의 윤곽은 대부분은 반달형이거나 원형으로 분류군마다 다소 차이가 있었고, 날개가 뚜렷하거나 미약하게 발달한 유형, 혹은 날개가 없는 유형으로 구별되었다. 유관속조직, 표피상납질, 결정체, 탄닌 등, 추가적인 해부학적 형질에 대해 상세히 기재하였다. 끝으로 조사된 잎의 해부학적 형질이 한국산 팥꽃나무과 분류군의 동정에 유용한가에 대하여 검토하였다.

자외선 조사에 의한 Hairless Mouse 피부구조 변화에 대한 형태학적 연구 (Morphologic Study on the Changes of Skin Structure of Hairless Mouse by Ultraviolet Irradiation)

  • This study was observed of the skin that changed after irradiation of the ultraviolet A. All the mouse were hairless which the weight are about 25g and the ages $6\sim8$ weeks old. The mouse were divided into six groups; control, irradiated for 6 hours, 3 days, 7 days, 14 days, 21 days and 28 days. Each group was irradiated with ultraviolet that is $320nm\sim366nm$ of wavelengths. After irradiated, the skin was observed with the electron microscope and the light microscope. The results are as follow: 1) Light microscopy With following irradiation, the epidermis was not changed to most groups but at the 28 days group was thickened and deposit the melanocyte. The elastic fibers within the epidermis were thickened and twisted with following irradiation. 2) Eelectron microscopy The elastic fibers were slightly clumped at 6 hours group, mildly increased and partly aggregated in the 3 days group, branched and tangled at 7 days group, irregulated and electron density at 14 days group, sightly thickened and twisted at 21 days group, and randomly arranged, shortened, twisted, and electron density at 28 days group.

난황형성부터 성숙까지 메기(Silurus asotus) 난모세포의 미세구조 (Ultrastructural Changes of Oocytes from Vitellogenesis into Maturation in Korean Catfish, Silurus asotus)

  • This study was carried out to investigate the histomorphological changes and the electrophoretic patterns of egg components, obtained from 100 of 2-year-old female catfishes (Silurus asotus). Female Korean catfishes collected in the vicinity of Chollabukdo have synchronous ovaries which discontinously ovulate eggs during the breeding season (from late May until early July). The fishes were killed by severing the spinal cord just posterior to the head after immobilization with tricaine methanesulfonate (MS 222). Especially, the light microscopic and ultrastructural changes of ooplasm and follicular membranes of oocytes, were examined by means of light and transmission electron microscopy. The size of the nucleoli and number of the yolk granules increased as the oocyte groved. Yolk granules were deposited in the oocyte as fluid Due to tile presence of large early and late maturing oocytes, their ovaries were enlarged, transparent, granular and greenish in color. As the percentages of fish in late maturing oocyte (LMO) and ripe oocyte (RO) stage increased from March to April, mean gonadosomatic index (GSI) values (19.95%) increased. Zona radiata changed from squamous into cuboid in shape in the early maturing oocyte (EMO) stage. Processes, microvilli, from the zona radiata and from the oocyte grow, and make contact with each other in the pore canals of the zona radiata during vitellogenesis, but are withdrawn as the zona radials becomes more compact and devoid of pore canals during oocyte maturation. Seasonal changes in the microscopic appearance of the ovaries were well correlated with those in both gonadosomatic index and macroscopic appearance. The main cytological changes such as increase in size of cell, nucleus, nucleolus, and increase in number of nucleoli and mitochondria demonstrated with electron microscopy in the previtellogenic oocytes of Korean catfish, provided evidences for important synthetic processes in an early preparatory phase of oocyte development. The electrophoretic pattern of major band in mature stage was much thicker (24 k, 66 k, 90-110k dalton) than that in previtellogenic phase.

Emendation of Rhodomonas marina (Cryptophyceae): insights from morphology, molecular phylogeny and water-soluble pigment in an Arctic isolate

  • Rhodomonas (Cryptophyceae) and species assigned to this genus have undergone numerous taxonomic revisions. This also applies to R. marina studied here as it was originally assigned as a species of Cryptomonas and later considered a variation of R. baltica, the type species. Despite being described more than 130 years ago, R. marina still lacks a comprehensive characterization. Light and electron microscopy were employed to delineate a strain from western Greenland. The living cells were 18 ㎛ long and 9 ㎛ wide, elliptical in shape with a pointed to rounded posterior and truncated anterior in lateral view. Two sub-equal flagella emerged from a vestibulum, where also a furrow extended. In transmission electron microscopy, the furrow was associated with a tubular gullet and the pyrenoid embedded in a deeply lobed chloroplast. The chloroplast contained DNA in perforations and was surrounded by starch grains. A tubular nucleomorph was enclosed within the pyrenoid matrix. In scanning electron microscopy, the inner periplast consisted of rectangular plates with rounded edges and posteriorly these were replaced by a sheet-like structure. The water-soluble pigment was Crypto-Phycoerythrin type I (Cr-PE 545). A phylogenetic inference based on SSU rDNA confirmed the identity of strain S18 as a species of Rhodomonas as it clustered with congeners but also Rhinomonas, Storeatula, and Pyrenomonas. These genera formed a monophyletic clade separated from a diverse assemblage of other cryptophyte genera. To further explore the phylogeny of R. marina a concatenated phylogenetic analysis based on the SSU rDNA-ITS1-5.8S rDNA-ITS2-LSU rDNA region was performed but included only closely related species. The secondary structure of nuclear internal transcribed spacer 2 was predicted and compared to similar structures in related species. Using morphological and molecular signatures as diagnostic features the description of R. marina was emended.
