Application of Principle in Metal-Ligand Complexation to Remove Heavy Metals : Kind and Concentration Effects of Organic Ligands (금속-Ligand 착염형성에 의한 중금속(重金屬) 제거(除去) 방법(方法)에 관한 연구(硏究);유기 Ligand의 종류와 농도(濃度) 영향(影響))

  • This research was conducted to investigate the influence of kind and concentration of organic ligands such as humic (HA) or fulvic acid (FA) on the removal of Cu or Pb from the aqueous solution employing the principles in metal-ligand complexation. Increasing HA concentration enhanced the efficiency of Cu or Pb removal, but there existed upper critical concentrations capable of forming maximum HA-metal complex. which ranged 53-289 and 42-315mg/L for Cu and Pb, respectively. At these concentrations. efficiency of removal was 70 to 95 % for Pb, but 13 to 65 % for Cu. Amounts of Cu and Pb which complexed with 100mg HA were estimated to be 7.5 and 34.1mg, respectively. FA-metal complex forming reactions were fitted significantly to the empirical models of Freundlich for Cu and Langmuir for Pb. Fulvic acid precipitated nearly 100% of Pb in solution, but formed precipitates with Cu in only 13 to 29%. Comparing organic ligands. HA had a higher removal efficiency for Cu but FA had such for Pb. Metalligand complex formation was differed from kinds and concentrations of corresponding ligands and metals. Results demonstrated that this principle has a strong potential to be employed for treating heavy metals in aqueous solution.

  • Stimulatory effects of epidermal growth factor (EGF) and transforming growth $factor-{\alpha}$($TGF-{\alpha}$) on the growth of squamous cancer cell lines established from human oral cancer tissue with moderate differentiation were studied in vitro. After culturing in serum-free media for 24 hours, growth factors-EGF only, $TGF-{\alpha}$ only and EGF, $TGF-{\alpha}$ together-were added to the media and numbers of cells were analyzed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay and compared with the control at 96, 144 hours. Each of EGF and $TGF-{\alpha}$ showed statistically significant stimulatory effects on the growth of cells respectively. Dose-dependent relationship of the stimulatory effects were not clearly demonstrated. The effects of EGF were higher than those of $TGF-{\alpha}$ and combinative administration showed higher effects than those of single uses. In conclusion, EGF may play an important and major role in differentiation and growth of human oral squamous cancer cells. $TGF-{\alpha}$, produced from cells activated by EGF, also can stimulate the cell growth and could be an alternative ligand for EGF receptor.

  • Anion effects on structures of $Cd^{II}$ complexes containing 2,2'-bipyridine (2,2'-bpy) ligands have been studied, and compared with $Zn^{II}$-(2,2'-bpy) complexes. For each anion, different structures have been obtained in both $Zn^{II}$-(2,2'-bpy) and $Cd^{II}$-(2,2'-bpy). Polymeric structures of $Cd^{II}$-2,2'-bpy complexes can be produced by hydrogen bonding interactions as shown in $Zn^{II}$-2,2'-bpy complexes. In addition, the bigger size of a $Cd^{II}$ ion gives higher coordination numbers forming variety of structures, and it makes that chlorides can act as bridging ligands to form a one-dimensional structure. The compound $\mathbf{5}$ catalyzed efficiently the transesterification of a variety of esters with methanol, while the rest of the compounds have displayed very slow conversions. In addition, the emission bands of complexes $\mathbf{1}$, $\mathbf{2}$, $\mathbf{4}$, and $\mathbf{6}$ are blue-shifted compared to the corresponding ligand 2,2'-bpy, whereas $\mathbf{3}$ and $\mathbf{5}$ showed the similar emission observed for the ligand.

Development of an Improved Menopausal Symptom-Alleviating Licorice (Glycyrrhiza uralensis) by Biotransformation Using Monascus albidulus

  • Licorice (Glycyrrhiza uralensis) contains several compounds that have been reported to alleviate menopausal symptoms via interacting with estrogen receptors (ERs). The compounds exist mainly in the form of glycosides, which exhibit low bioavailability and function. To bioconvert liquiritin and isoliquiritin, the major estrogenic compounds, to the corresponding deglycosylated liquiritigenin and isoliquiritigenin, respectively, licorice was fermented with Monascus, which has been demonstrated to deglycosylate other substances. The contents of liquiritigenin and isoliquiritigenin in Monascus-fermented licorice increased by 10.46-fold (from 38.03 μM to 379.75 μM) and 12.50-fold (from 5.53 μM to 69.14 μM), respectively, compared with their contents in non-fermented licorice. Monascus-fermented licorice exhibited 82.5% of the ERβ binding activity of that observed in the positive control (17 β-estradiol), whereas the non-fermented licorice exhibited 54.1% of the binding activity in an in vivo ER binding assay. The increase in the ERβ binding activity was associated with increases in liquiritigenin and isoliquiritigenin contents. Liquiritigenin acts as a selective ligand for ERβ, which alleviates menopausal symptoms with fewer side effects, such as heart disease and hypertension, compared with a ligand for ERα. In addition, Monascus-fermented licorice contained 731 mg/kg of monacolin K, one of the metabolites produced by Monascus that reduces serum cholesterol. Therefore, Monascus-fermented licorice is a promising material for the prevention and treatment of menopausal syndrome with fewer side effects.

Pharmacokinetic Modelling and Simulation of the Counter-transport in the Hepatic Transport of Organic Anions (음이온계 약물의 간수송과정에 있어서 대향수송의 약물동력학적 모델링 및 시뮬레이션)

  • The purpose of the present study was to kinetically investigate the carrier-mediated uptake in the hepatic transport of organic anions, and to simulate the 'in vivo counter-transport' phenomena, using kinetic model which was developed in this study. The condition that the mobility of carrier-ligand complex is greater than that of free carrier is not essential for the occurrence of 'counter-transport' phenomenon. To examine the inhibitory effects on the initial uptake of organic anions by the liver, it is necessary to judge whether the true counter-transport mechanism (trans-stimulation) is working or not. Effects of bromophenol blue (BPB) or bromosulfophthalein (BSP) on the plasma disappearance curves of a 1-anilino-8-naphthalene sulfonate (ANS) were then kinetically analyzed based on a flow model, in which the ligand is eliminated only from the peripheral compartment (liver compartment). Moreover, 'in vivo counter-transport' phenomena were simulated based on the perfusion model which incorporated the carrier-mediated transport and the saturable intracellular binding. The 'in vivo counter-transport' phenomena in the hepatic transport of a organic anions were well demonstrated by incorporating the carrier-mediated process. However, the 'in vivo counter-transport' phenomena may be also explained by the enhancement of back diffusion due to the displacement of intracellular binding. In conclusion, one should be more cautious in interpreting data obtained from so-called 'in vivo counter-transport' experiments.

Oleoylethanolamide Exhibits GPR119-Dependent Inhibition of Osteoclast Function and GPR119-Independent Promotion of Osteoclast Apoptosis

  • Oleoylethanolamide (OEA), a bioactive lipid in bone, is known as an endogenous ligand for G protein-coupled receptor 119 (GPR119). Here, we explored the effects of OEA on osteoclast differentiation, function, and survival. While OEA inhibits osteoclast resorptive function by disrupting actin cytoskeleton, it does not affect receptor activator of nuclear factor-κB ligand (RANKL)-induced osteoclast differentiation. OEA attenuates osteoclast spreading, blocks actin ring formation, and eventually impairs bone resorption. Mechanistically, OEA inhibits Rac activation in response to macrophage colony-stimulating factor (M-CSF), but not RANKL. Furthermore, the OEA-mediated cytoskeletal disorganization is abrogated by GPR119 knockdown using small hairpin RNA (shRNA), indicating that GPR119 is pivotal for osteoclast cytoskeletal organization. In addition, OEA induces apoptosis in both control and GPR119 shRNA-transduced osteoclasts, suggesting that GPR119 is not required for osteoclast apoptosis. Collectively, our findings reveal that OEA has inhibitory effects on osteoclast function and survival of mature osteoclasts via GPR119-dependent and GPR119-independent pathways, respectively.

Production of polyclonal anti-$\beta$-adrenergic receptor antibody and it′s effects on receptor ligand binding

  • The analysis of membrane receptors for hormones and neurotransmitters has progressed considerably by pharmacological and biochemical means and more recently by the use of specific anti-receptor antibodies. A 14-mer peptide (from Phe102 to Leu115 of ${\beta}$2-adrenergic receptor) was synthesized and this peptide was coupled to carrier protein Keyhole Limpet Hemocyanin(KLH) by glutaraldehyde method. A 0.5mg of KLH-coupled peptide was emulsified with equal volume of complete Freund's adjuvant and injected via popliteal lymph node to each of the three Newzealnd White rabbits. Booster injections were repeated at 4 weeks interval for three times with incomplete Freund's adjuvants. One week after the final injection, serum was prepared from ear artery. Nonspecific immunoglobulins were removed by passing the serum through KLH-Sepharose 6B affinity matrix and further by incubation with bovine lung aceton powder. The titer of the antibody for synthetic peptide which was determined by enzyme linked immunosorbent assay(ELISA) was about l/l,000. The antibody produced in this study revealed 67kDa protein band in the western blot of partially purified guinea pig lung ${\beta}$-adrenergic receptor preparation. The antibody inhibited ${\beta}$-adrenergic antaginist [3H] Dihydroalprenolol binding to soluble ${\beta}$-adrenergic receptor by 25% while control sera did not show any inhibitory effects, The result of this study suggests that the peptide sequence selected in this study may play some important roles in adrenergic receptor-ligand interaction.

Synthesis of Highly Dispersible Metal Nanoparticles in P3HT:PCBM Layers and Their Effects on the Performance of Polymer Solar Cells (P3HT:PCBM 층 내 분산 가능한 금속 나노입자의 제조 및 이를 포함한 고분자 태양전지 소자의 특성에 관한 연구)

  • In this study, we prepare polymer solar cells incorporating organic ligand-modified Ag nanoparticles (O-AgNPs) highly dispersed in the P3HT:PCBM layer. Ag nanoparticles decorated with water-dispersible ligands (WAgNPs) were also utilized as a control sample. The existence of the ligands on the Ag surface was confirmed by FT-IR spectra. Metal nanoparticles with different surface chemistries exhibited different dispersion tendencies. O-AgNPs were highly dispersed even at high concentrations, whereas W-AgNPs exhibited significant aggregation in the polymer layer. Both dispersion and blending concentration of the Ag nanoparticles in P3HT:PCBM matrix had critical effects on the device performance as well as light absorption. The significant changes in short-circuit current density ($J_{SC}$) of the solar cells seemed to be related to the change in the polymer morphology according to the concentration of AgNPs introduced. These findings suggested the importance of uniform dispersion of plasmonic metal nanoparticles and their blending concentration conditions in order to boost the solar cell performance.

Synthesis and Properties of Tetraaza Macrocycles Containing Two 3-Pyridylmethyl, 4-Pyridylmethyl, or Phenylmethyl Pendant Arms and Their Nickel(Ⅱ) and Copper(Ⅱ) Complexes: Effects of the Pendant Arms on the Complex Formation Reaction

  • The synthesis and properties of 2,13-bis(3'-pyridylmethyl) $(L^3)$, 2,13-bis(4'-pyridylmethyl) $(L^4)$, and 2,13-bis(phenylmethyl) $(L^5)$ derivatives of 5,16-dimethyl-2,6,13,17-tetraazatrcyclo$[16.4.0.^{1.18}0^{7.12}]$docosane are reported. The 3- or 4-pyridylmethyl groups of $[ML^3](ClO_4)_2\;or\;[ML^4](ClO_4)_2$ (M = Ni(Ⅱ) or Cu(Ⅱ)) are not involved in coordination, and the coordination geometry (square-planar) and ligand field strength of the complexes are quite similar to those of $[ML^5](ClO_4)_2$, bearing two phenylmethyl pendant arms. However, the complex formation reactions of $L^3\;and\;L^4$ are strongly influenced by the pyridyl groups, which can interact with a proton or metal ion outside the macrocyclic ring. The macrocycle $L^5$ exhibits a high copper(Ⅱ) ion selectivity against nickel(Ⅱ) ion; the ligand readily reacts with copper(Ⅱ) ion to form $[CuL^5]^{2+}$ but does not react with hydrated nickel(Ⅱ) ion in methanol solutions. On the other hand, $L^3\;and\;L^4$ form their copper(Ⅱ) and nickel(Ⅱ) complexes under a similar condition, without showing any considerable metal ion selectivity. The ligands $L^3\;and\;L^4$ react with copper(Ⅱ) ion more rapidly than does $L^5$ at pH 6.4. At pH 5.0, however, the reaction rate of the former macrocycles is slower than that of the latter. The effects of the 3- or 4-pyridylmethyl pendant arms on the complex formation reaction of $L^3\;and\;L^4$ are discussed.

Temperature and Ligand Effects on the Eu(Ⅲ) Emission Spectra in the Aqueous Solution (수용액에서의 Eu(Ⅲ) 착물의 형광스펙트럼에 영향을 미치는 온도 및 리간드 효과에 관한 연구)

  • Temperature effects on the fluorescence emission spectra of 0.01 M Eu(III) ion with ClO$_4$, Cl$^-$, NO$_3$ were studied. Relative intensity change of hypersensitive band ($^5D0\; {\to}\;^7F_2$) and nonhypersensitive band ($^5D0 \;{\to}\;^7F_1$) was quite remarkable with temperature and ligand. The relative intensity change was interpreted as the change of formation constant and used to calculation the enthalpy change of $Eu(H_2O)_X^{3+}$+ to EuL(H$_2O)_{X-1}^{2+}$ complex. $\Delta{H}$ of $Eu(H_2O)_X^{3+}$ to EuCl(H$_2O)_{X-1}^{2+}$ was roughly 15 kJ/mol and temperature independent, but $\Delta{H}$ of EuNO$_3(H_2O)_{X-1}^{2+}$ was changed with temperature; -11 kJ/mol at 25$^{\circ}C$ and 47 kJ/mol at 250$^{\circ}C$.
