Anti-survival and pro-apoptotic effects of meridianin C derivatives on MV4-11 human acute myeloid leukemia cells

  • Meridianin C is a marine natural product with anticancer activity. Several meridianin C derivatives (compounds 7a-j) were recently synthesized, and their inhibitory effects on pro-viral integration site for Moloney murine leukemia virus (PIM) kinases, as well as their antiproliferative effects on human leukemia cells, were reported. However, the anti-leukemic effects and mechanisms of action of meridianin C and its derivatives remain largely unknown. The aim of the present study was to investigate the effects of meridianin C and its derivatives on MV4-11 human acute myeloid leukemia cell growth. The parent compound meridianin C did not markedly affect the viability and survival of MV4-11 cells. By contrast, MV4-11 cell viability and survival were reduced by meridianin C derivatives, with compound 7a achieving the most prominent reduction. Compound 7a notably inhibited the expression and activity of PIM kinases, as evidenced by reduced B-cell lymphoma-2 (Bcl-2)-associated death promoter phosphorylation at Ser112. However, meridianin C also suppressed PIM kinase expression and activity, and the pan-PIM kinase inhibitor AZD1208 only slightly suppressed the survival of MV4-11 cells. Thus, the anti-survival effect of compound 7a on MV4-11 cells was unrelated to PIM kinase inhibition. Moreover, compound 7a induced apoptosis, caspase-9 and -3 activation and poly(ADP-ribose) polymerase (PARP) cleavage, but did not affect death receptor (DR)-4 or DR-5 expression in MV4-11 cells. Compound 7a also induced the generation of cleaved Bcl-2, and the downregulation of myeloid cell leukemia (Mcl)-1 and X-linked inhibitor of apoptosis (XIAP) in MV4-11 cells. Furthermore, compound 7a increased eukaryotic initiation factor (eIF)-2α phosphorylation and decreased S6 phosphorylation, whereas GRP-78 expression was unaffected. Importantly, treatment with a pan-caspase inhibitor (z-VAD-fmk) significantly attenuated compound 7a-induced apoptosis, caspase-9 and -3 activation, PARP cleavage, generation of cleaved Bcl-2 and downregulation of Mcl-1 and XIAP in MV4-11 cells. Collectively, these findings demonstrated the strong anti-survival and pro-apoptotic effects of compound 7a on MV4-11 cells through regulation of caspase-9 and -3, Bcl-2, Mcl-1, XIAP, eIF-2α and S6 molecules.

Comparison between Affective Responses to Tactile Stimuli Based on the Presence of Visual Information Presentation (시각 정보 제시 여부에 따른 촉각 자극에 대한 정서 반응 비교)

  • Previous studies on texture and emotion have focused on identifying precisely which tactile stimuli trigger specific emotions. Despite the significant role of vision in tactile perception, research has so far only focused on the singular aspect of texture. In this study, we used tactile stimuli to investigate the effects of three variables-roughness, hardness, and visual blocking-on the affective responses to tactile perception. The experimental stimuli that can be encountered in daily life were selected based on the four conditions of "rough/hard," "rough/soft," "smooth/hard" and "smooth/soft" by crossing two roughness conditions (rough, smooth) and two hardness conditions (hard, soft). The experiment was divided into two sessions depending on whether or not visual blocking existed. Participants completed a session in which they evaluated a tactile stimulus after touching it without seeing it and then proceeded with a session in which they evaluated a stimulus after touching it with sight of it. The results of the repeated-measures ANOVA showed that individuals reported a more positive perception when touching stimuli with visual cues and more negative when touching stimuli without visual cues. Furthermore, the inclination to perceive smooth and soft stimuli more positively and rough stimuli more negatively was stronger when touching without visual cues. The results of this study suggest implications for enhancing the understanding of the interaction between emotion and visual information processing by elucidating how emotions are experienced differently in situations where visual information is provided and where it is not.

Morphological of Development Eggs, Larvae and Juveniles Rhinogobius giurinus in Botongcheon Stream (보통천에 서식하는 갈문망둑 Rhinogobius giurinus의 난발생 및 자치어 형태발달)

  • This study investigated the early life history of Rhinogobius giurinus in the Botongcheon stream and compared the differences with related species. The fertilized egg size (long×short) of the 3.46×1.17 mm (n=30) on average. The time required for hatching at the water temperature of 22.5±1℃ was 213 hours and 30 mins. Immediately after hatching, the egg yolk had an average total length of 2.34±0.05 mm (n=30), and the mouth and anus were not opened, and the number of myotomes was 23~25. 3 days after hatching (DAH), the preflexion larvae had an average total length of 2.70±0.09 mm (n=30), and live food intake began as the mouth and anus were opened. 22 DAH, the flexion larvae had an average total length of 5.26±0.16 mm (n=30), and the tip of the vertebrae began to bend upward. 48 DAH, the postflexion larvae had an average total length of 11.1±0.53 mm (n=30), and the tip of the vertebrae was completely bent 45° upward. 60 DAH the juveniles had an average total length of 14.9±0.69 mm (n=30) was reached to the integer with 6 first dorsal fins, 8~10 second dorsal fins, 9 anal fins, 15 caudal fins, and 5 ventral fins for each fin. As a result of the study, the melanophore differed from other postflexion larvae of the Gobiidae fishes in that it was deposited above the gill lid, on the back and body, and the xanthophore was deposited on the head and body.

Development of Suicide Prevention Programs for Mental Health Professionals Working with Children and Adolescens at High Risk of Suicide (아동·청소년 자살고위험군 자살예방평가 및 프로그램)

  • Korea has the highest suicide rate among Organization for Economic Cooperation and Development countries, with increasing rates observed among children and adolescents with low levels of life satisfaction. Patients in these age groups exhibit particularly turbulent and impulsive behaviors, which make suicide highly contagious and fatal. The loss of meaningful work or activities increases the risk of suicide, especially in young people. Following revisions to the Mental Health Welfare Act (2016), which has included occupational therapists among mental health professionals, a multidisciplinary approach including occupational therapy has been emphasized upon in mental health services. Screening tools for preventing suicide in children and adolescents include the Suicidal Ideation Scale, Beck's Suicidal Ideation Scale, Columbia-Suicide Severity Rating Scale, Reasons for Living Scale for Adolescents, and the Student Emotional and Behavioral Characteristics Test. The Canadian Occupational Performance Measure, which is an occupational therapy evaluation tool, is also used. Various suicide prevention programs have been proposed across academic disciplines; however, due to the urgent nature of high-risk groups, there is a need for timely services. Currently, most existing programs focus primarily on the cognitive-behavioral aspects. In this study, we aimed to introduce diverse suicide prevention programs for mental health professionals working with high-risk children and adolescents in order to equip them with the relevant information and help apply their learnings effectively in different situations.

Computer Vision Approach for Phenotypic Characterization of Horticultural Crops (컴퓨터 비전을 활용한 토마토, 파프리카, 멜론 및 오이 작물의 표현형 특성화)

  • This study explored computer vision methods using the OpenCV open-source library to characterize the phenotypes of various horticultural crops. In the case of tomatoes, image color was examined to assess ripeness, while support vector machine (SVM) and histogram of oriented gradients (HOG) methods effectively identified ripe tomatoes. For sweet pepper, we visualized the color distribution and used the Gaussian mixture model for clustering to analyze its post-harvest color characteristics. For the quality assessment of netted melons, the LAB (lightness, a, b) color space, binary images, and depth mapping were used to measure the net patterns of the melon. In addition, a combination of depth and color data proved successful in identifying flowers of different sizes and distances in cucumber greenhouses. This study highlights the effectiveness of these computer vision strategies in monitoring the growth and development, ripening, and quality assessment of fruits and vegetables. For broader applications in agriculture, future researchers and developers should enhance these techniques with plant physiological indicators to promote their adoption in both research and practical agricultural settings.

Survey of College Students' Perceptions of COVID-19, which has been Converted into a Class 4 Infectious Disease (제4급감염병으로 전환된 코로나19에 대한 대학생의 인식 조사)

  • This study conducted a survey using Google on 282 college students to investigate college students' awareness of COVID-19, which has been converted into a class 4 infectious disease. As a result, the average level of interest in COVID-19 information was 2.85, and the average level of interest in COVID-19 information was 2.85. 54% did not know that the level had changed, showing that interest in COVID-19 was not high. In terms of personal hygiene attitude, the average of those who are not currently wearing a mask was 3.95, but the average of those who wear a mask when necessary was 3.94, washing hands for more than 30 seconds, washing hands frequently, and coughing etiquette were observed well. Additionally, awareness of infection was observed by others. Rather, they thought of their symptoms as connected to COVID-19 and were still conscious of COVID-19 infection (average 3.85). Additionally, COVID-19 was recognized as a threatening infectious disease that could cause a resurgence and repeat a pandemic. Since the crisis stage regarding COVID-19 can change at any time in the WithCorona era, this study sought to provide basic data for successful response to future crises by investigating trends in changes in people's perceptions.

A Study on the Effect of Educational Service Quality on Educational Satisfaction in Cooking Educational Institutions - Focusing on the moderating effect of mentoring - (조리교육기관의 교육서비스품질이 교육만족도에 미치는 영향관계 연구 - 멘토링 조절효과를 중심으로 -)

  • This study identified the relationship between empathy, tangibility, certainty, reliability, and educational satisfaction, which are sub-factors of educational service quality, and analyzed how educational service quality affects educational satisfaction through the moderating effect of mentoring. A total of 381 response samples were constructed for empirical analysis to analyze demographic characteristics, student use patterns, validity and reliability, correlation, and hypothesis verification, and a total of six hypotheses were established and verified using variables selected based on previous studies and preliminary surveys. First, as a result of the verification of Hypothesis 1, it was found that all constituent factors of education service quality had a positive (+) significant effect on education satisfaction. Second, as a result of the moderating effect of mentoring in Hypothesis 2, it was found that education satisfaction was increased in the confidence, reliability, and mentoring of education service quality. Therefore, empathy, tangibility, certainty, and reliability of education service quality are factors necessary to increase education satisfaction, and academy operators and managers are required to reflect this and operate strategies to increase the competitiveness of the academy.

Influence of COVID-19 Knowledge, Risk Perception, Anxiety on Preventive Behaviors of Nursing Students (간호대학생의 코로나19 관련 지식, 감염 위험지각, 불안이 감염예방 행위에 미치는 영향)

  • The purpose of this study was to investigate COVID-19 Knowledge, Risk Perception, Anxiety and Preventive Behaviors, and to identify the influencing factors on Preventive Behaviors of nursing students. Data were collected using questionnaires from 342 nursing students in Gwangju, November 2021. The collected data was analyzed by descriptive statistics, pearson's correlation coefficient and stepwise multiple regression using IBM SPSS 21.0 program. The total mean scores were as follows: COVID-19 Knowledge, 13.25±1.38; Risk Perception; 4.57±1.34, Anxiety; 44.55±9.66, Preventive Behaviors; 7.66±1.55. Preventive Behaviors was found to have significant positive correlations with COVID-19 Knowledge and Risk Perception). The most influential factors influencing the Preventive Behaviors of nursing students were Experience in education related COVID-19 prevention, Risk Perception, COVID-19 Knowledge. In conclusion, in order to develop the Preventive Behaviors of nursing students, it is necessary to apply a program that improve the COVID-19 prevention education and Knowledge, and to control Risk Perception according to the individual level.

A Study on the Effect of Start-up Support Program and Entrepreneurship on Start-up Execution Willingness - For Prospective Start-ups in Jeolla Province - (창업지원프로그램과 창업가정신이 창업실행의지에 미치는 영향에 관한 연구 - 전남지역 예비창업자 대상으로 -)

  • This study analyzed the impact on entrepreneurship, passion, and start-up execution willingness to start a business for 200 prospective start-ups in Jeollanam-do. As a result of this study, it was found that there was an effect at the statistically significant level (p<.05) in the relationship between the start-up support program and start-up willingness. In the mediating effect analysis of passion for entrepreneurship support programs, it was confirmed that passion played a complete mediating role in the relationship between mentoring and start-up willingness and a partial mediating role in the relationship between the start-up support fund and start-up willingness. In the analysis of the mediating effect of passion on entrepreneurship, it was confirmed that passion played a partial mediating role in the relationship between initiative and start-up execution willingness, and also in the relationship between risk sensitivity and start-up execution willingness. This shows that when proper initiative and risk tolerance are recognized, passion enhances the start-up willingness to run into the start-up market. Based on these results, we suggested theoretical implications and policy support directions to increase the willingness of prospective founders to implement.

A Comparative Study on the Subjective Happiness, Clinical Practice Stress and Satisfaction with Clinical Practice of the Nursing Student's in Korea and Mongolia (한국과 몽골 간호대학생의 주관적 행복감, 임상실습 스트레스, 임상실습 만족도에 대한 비교 연구)

  • This study is to prepare basic data to improve the clinical practice satisfaction of nursing students in Mongolia and Korea by comparatively studying subjective happiness, clinical practices stress and clinical practice satisfaction between Korean nursing students and Mongolian nursing students. A survey was conducted with 187 nursing students from one college in Gyeongnam and 100 nursing students from the National Medical University of Mongolia and data analysis were using the SPSS 21.0 program. The result was Korean nursing students had lower subjective happiness and clinical practice satisfaction than Mongolian nursing students, but they were not statistically significant and clinical practice stress was statistically significantly higher in Mongolian nursing students. In addition, it was found that the lower the clinical practice stress and the higher the subjective happiness, the higher the satisfaction with clinical practice among Korean nursing students and in Mongolian nursing students, the lower the clinical practice stress, the higher the clinical practice satisfaction. Therefore, it is necessary to conduct a comparative study on clinical practice education in nursing education institutions in Korea and Mongolia to develop a joint program to improve clinical practice satisfaction and to verify the effectiveness.
