Gravity Field Interpretation and Underground Structure Modelling as a Method of Setting Horizontal and Vertical Zoning of a Active Fault Core (활성단층의 3차원적인 규모를 결정하기 위한 중력장 데이터의 해석 및 지각구조 모델링: 양산단층에서의 예)

  • In order to estimate the vertical and horizontal structural in the Yangsan fault core line (Naengsuri area, Pohang), we carried out gravity field measurements and interpretation procedures such as Euler deconvolution method and curvature analysis in addition to the forward modelling technique (i.e. IGMAS+). We found a prominent gravity difference of more than 1.5 mGal across the fault core. This indicates a distinct density difference between the western and eastern crustal area across the Yangsan fault line. Comparing this gravity field interpretation with other existent geologic and geophysical survey data (e.g. LiDAR, trenching, electric resistivity measurements), It is concluded that (1) the prominent gravity difference is caused by the density difference of about 0.1 g/㎤ between the Bulguksa Granite in the west and the Cretaceous Sandstone in the east side, (2) the fault core is elongated vertically into a depth of about 2,000 meters and extended horizontally 3,000 meters to the NNE direction from Naengsuri area. Our results present that the gravity field method is a very effective tool to estimate a three -dimensional image of the active fault core.

Vinyl Addition Polymerization Behavior of Poly(5-hexyl-2-norbornene-co-5-methylester-2-norbornene)s Using Cationic ${\eta}^3$-Allyl Palladium Catalyst (양이온성 ${\eta}^3$-알릴 팔라듐 촉매를 사용한 폴리(5-헥실-2-노보넨-co-5-메틸에스터-2-노보넨)의 비닐 부가 중합 거동)

  • We synthesized the vinyl addition-type polynorbomene copolymers using two monomers [5-hexyl-2-norbornene (HNB) and 5-methyleste-2-norbornene(MES-NB)] by means of a cationic ${\eta}^3$-allyl palladium catalyst system{[(${\eta}^3$-allyl)palladium(tricyclohexylphosphine) trifluoroacetate] and [lithium tetrakis(pentafluorophenyl) borate ${\cdot}$2.5 etherate]}. The molecular weights and yields of copolynorbomenes polymerized in various conditions were measured to investigate an optimum polymerization conditions to obtain highly ester-functionalized polynorbomenes. As a Pd catalyst content increased, the molecular weights (Mw) of polymers decreased while polymer yields increased. Also, as a Li cocatalyst content increased, the Mw’s and yields of polymers increased at the same time. The Mw’s of copolymers were also controlled by chain transfer agents such as 1-hexone, 1-octene and 1-decene, and we found that longer 1-decene and 1-octene were more efficient to reduce the Mw’s of polynorbornenes than 1-hexene. On the other hand, the content of chain transfer agents did not give influence significantly on polymer yields. From the $^1$H-NMR and GPC analysis of HNB/MES-NB(feed ratio of 40/60 mol%) copolymer, we found that this copolymer had an about 25 mol% of ester portion and a high molecular weight of 270,000.

Elevated Platelets Enhance Cancer Cell Migration, Promote Hematogenous Metastasis and Associate with a Poor Prognosis in Advanced Non-small Cell Lung Cancer Cases

  • Although correlations between platelets and lung cancer has been recognized, effects on non-small cell lung cancer (NSCLC) metastasis remain to be determined in detail. In the present study, wound healing assays revealed a role of platelets in NSCLC cell migration. Thus the mean migration rate of lung adenocarcinoma A549 cells was significantly elevated after co-culture with platelets ($81.7{\pm}0.45%$ vs $41.0{\pm}3.50%$, P<0.01). Expression of GAPDH was examined by reverse transcription-polymerase chain reaction to study the effect of platelets on NSCLC cell proliferation. The result showed that the proliferation of A549 and SPC-A1 cells was not affected. Mouse models were established by transfusing A549 cells and SPC-A1 cells into mice lateral tail veins. We found tumor metastasis nodules in lungs to be increased significantly after co-transfusion with platelets (in A549, $4.33{\pm}0.33$ vs $0.33{\pm}0.33$, P=0.01; in SPC-A1, $2.67{\pm}0.33$ vs $0.00{\pm}0.00$, P=0.01). In addition, consecutive inoperable patients with newly diagnosed NSCLC (TNM stage III or IV) between January 2009 and December 2011 were retrospectively reviewed. Using the Kaplan-Meier method, NSCLC patients with a high platelet counts demonstrated a significantly shorter progression free survival compared with those with a low platelet count (> $200{\times}10^9/L$, 3 months versus ${\leq}200{\times}10^9/L$, 5 months, P=0.001). An elevated platelet count was also identified as an independent prognostic factor by Cox regression analysis for prgression free survival (adjusted hazard ratio: 1.69; 95% CI: 1.16, 2.46; P=0.006). This study suggested that platelets might contribute to the hematogenous metastatic process by promoting cancer cell migration, which eventually affects the prognosis of NSCLC.

Screening of Antagonistic Bacillus against Brown Rot in Dendrocalamus latiflorus and Preparation of Applying Bacterial Suspension

  • The aim of this study was to isolate biocontrol bacteria that could antagonize brown rot of Dendrocalamus latiflorus, optimize the culture conditions, and develop an effective biocontrol preparation for brown rot of D. latiflorus. This study isolated a bacterium with an antagonistic effect on bamboo brown rot from healthy D. latiflorus rhizosphere soil. Morphology, molecular biology, and physiological biochemistry methods identified it as Bacillus siamensis. The following culturing media and conditions improved the inhibition effect of B. siamensis: the best culturing media were 2% sucrose, 1.5% yeast extract, and 0.7% potassium chloride; the optimal culturing time, temperature, pH, and inoculation amount were 48 h, 30℃, 6, and 20%. The optimum formula of the applying bacterial suspension was 14% sodium dodecyl benzene sulfonate emulsifier, 4% Na2HPO4·2H2O, 0.3% hydroxypropyl methylcellulose thickener, and 20% B. siamensis. The pot experiment results showed the control effect of applying bacterial suspension, diluted 1,000 times is still better than that of 24% fenbuconazole suspension. The applying bacterial suspension enables reliable control of brown rot in D. latiflorus.

THE FRACTAL DIMENSION OF THE 𝜌 OPHIUCUS MOLECULAR CLOUD COMPLEX

  • We estimate the fractal dimension of the ${\rho}$ Ophiuchus Molecular Cloud Complex, associated with star forming regions. We selected a cube (${\upsilon}$, l, b) database, obtained with J = 1-0 transition lines of $^{12}CO$ and $^{13}CO$ at a resolution of 22" using a multibeam receiver system on the 14-m telescope of the Five College Radio Astronomy Observatory. Using a code developed within IRAF, we identified slice-clouds with two threshold temperatures to estimate the fractal dimension. With threshold temperatures of 2.25 K ($3{\sigma}$) and 3.75 K ($5{\sigma}$), the fractal dimension of the target cloud is estimated to be D = 1.52-1.54, where $P{\propto}A^{D/2}$, which is larger than previous results. We suggest that the sampling rate (spatial resolution) of observed data must be an important parameter when estimating the fractal dimension, and that narrower or wider dispersion around an arbitrary fit line and the intercepts at NP = 100 should be checked whether they relate to firms noise level or characteristic structure of the target cloud. This issue could be investigated by analysing several high resolution databases with different quality (low or moderate sensitivity).

B3GNT2, a Polylactosamine Synthase, Regulates Glycosylation of EGFR in H7721 Human Hepatocellular Carcinoma Cells

  • The epidermal growth factor receptor (EGFR) is an important surface receptor with N-glycans in its extracellular domain, whose glycosylation is essential for its function, especially in tumor cells. Here, we demonstrated that polylactosamine is markedly increased in H7721 hepatocellular carcinoma cells after treatment with EGF, while it apparently declined after exposure to all-trans retinoic acid (ATRA). In the study of the enzymatic mechanism of this phenomenon, we explored changes in the expression of poly-N-acetyllactosamine (PLN) branching glycosyltransferases using RT-PCR. Among the four glycosyltransferases with altered expression, GnT-V was most elevated by EGF, while GnT-V and B3GNT2 were most declined by ATRA. Next, we conducted co-immunoprecipitation experiments to test whether B3GNT2 and EGFR associate with each other. We observed that EGFR is a B3GNT2-targeting protein in H7721 cells. Taken together, these findings indicated that the altered expression of B3GNT2 will remodel the PLN stucture of EGFR in H7721 cells, which may modify downstream signal transduction.

Effect of the MeOH Extract of Adventious Root Culture of Panax ginseng on Hyperlipidemic Rat Induced by High Fat-rich Diet (조직배양산삼 부정근 메탄올 추출물이 식이성 고지혈증에 미치는 영향)

  • We have investigated the effect of MeOH extract of the adventious root culture of Panax ginseng on hyperlipidemic rats induced by fat-rich diet. We also analyzed and compared the content ginsenoisdes of the cultured ginsengs by HPLC. After oral administration of the MeOH extract to hyperlipidemic rats for four weeks, the variables including body weight, cholesterol, HDL, LDL, and triglyceride levels in serum were measured. One of the cultured ginseng roots (CBN3) decreased cholesterol and LDL-cholesterol and increased HDL-cholesterol levels in serum. HPLC analysis demonstrated that CBN3 contains more $Rb_1$ and Rd ginsenosides than any other cultured ginsengs and cultivated ginseng. These results suggest that CBN3 with high contents of $Rb_1$ and Rd may be useful in lowering blood cholesterol.

Photosynthetic carbon fixation by tropical coral reef phytoplankton assemblages: a UVR perspective

  • Photosynthetic carbon fixation regulates air-sea $CO_2$ fluxes in the waters of coral reefs. However, little has been documented on the effects of solar UV radiation (UVR, 280-400 nm) upon photosynthetic behaviors of phytoplankton dwelling in these ecosystems. In order to evaluate the aforesaid, surface dwelling tropical coral reef phytoplankton assemblages collected from the South China Sea were exposed to solar radiation (i.e., photosynthetically active radiation [PAR] + UV radiation A [UVA] + UV radiation B [UVB], 280-700 nm; PAR + UVA, 320-700 nm; and PAR, 400-700 nm) under static or simulated-mixing conditions. Under the static condition, UVA and UVB significantly reduced the carbon fixation with the maximum of 22.4 and 15.3%, respectively; while lower UVR-related photosynthetic inhibition was observed in case of phytoplankton samples being subjected to mixing. At a moderate level of mixing (i.e., circulation time 80 min), the UVA and UVB caused inhibition were lowered by 52.1 and 79.6%, respectively. Based on this it could be stated that vertical mixing induced by winds and/or tides in the natural environments could reduce the inhibitory effect of solar UVR on phytoplankton productivity in the coral reefs water.

Piezoelectric and Dielectric Properties of Low Temperature Sintering PMN-PZN-PZT Ceramics according to the Milling Time (밀링 시간에 따른 저온소결 PMN-PZN-PZT 세라믹스의 압전 및 유전특성)

  • In this paper, in order to develop low temperature sintering ceramics for multilayer piezoelectric actuator application, PMN-PZN-PZT ceramics were fabricated using $LiCO_3,\;Bi_2O_3$ and CuO as sintering aids. And also, their piezoelectric and dielectric properties were investigated according to the milling time. All the specimens sintered at $930\;^{\circ}C$ showed tetragonal phases without secondary phases. With increasing milling time, piezoelectric and dielectric characteristic of specimens increased up to 60 hours milling time and then decreased due to the agglomeration of fine particle. Accordingly, it seems that 60 hour is optimum milling condition. At the sintering temperature of $930\;^{\circ}C$ and milling time of 60 hour, density, dielectric constant(${\varepsilon}_r$), electromechanical coupling factor (kp), mechanical quality factor (Qm), piezoelectric d constant showed the optimum value of $7.95\;g/m^3$, 1382, 0.546, 1749, 330 pC/N, respectively for multilayer piezoelectric actuator application.

Characterization of ATPase Activity of Chaperonin from the Hyperthermophilic Archaeon Pyrococcus horikoshii (초고열성 고세균 Pyrococcus horikoshii 유래 샤페로닌의 ATPase 활성 특성)

  • ATP drives the conformational change of the group II chaperonin from the open lid substrate-binding conformation to the closed lid conformation to encapsulate an unfolded protein in the central cavity. It is thought that the folding activity of group II chaperonin is strongly correlated with the ATP-dependent conformational change ability. In order to confirm the dependence of the reaction temperature and ATP concentration of PhCpn, the ATPase activities were measured under different reaction temperatures and ATP concentrations. The maximal ATPase activity of PhCpn was observed at 80℃ and 3 mM ATP concentration. As a result of ATPase activity according to the type of salt ions, the highest activity was observed at 300 mM LiCl among the univalent cations and 5 mM MgCl2 among the divalent cations, respectively. The values of Km and Vmax for ATP substrate were estimated as 2.17 mM and 833.3 μM/min, respectively. This results provide the enzymatic information of PhCpn when the prolonged and high activities of pharmaceutical and industrial proteins (or enzymes), by using chaperonin molecules, are required.
