Fatty acid synthase (FASN) is a multi-functional enzyme with a central role in the synthesis of long-chain fatty acid and has been considered as a positional candidate gene for BTA 19 quantitative trait loci (QTL) affecting milk-fat content and fatty acid composition. In this study, we sequenced the FASN gene in several cattle breeds including Hanwoo and imported beef cattle, and identified novel DNA polymorphisms and their linkage relationship in Hanwoo. We found a significant frequency difference of the FASN (AF285607) g.17924 A$\rightarrow$G polymorphism between Hanwoo (70%) and other breeds and this polymorphism has been known for an association with fatty acid composition in Angus. Furthermore, by direct DNA sequencing in 18 unrelated Hanwoo, we identified 27 SNPs including nine novel variations in the FASN gene. Among 27 SNPs identified in the FASN gene, four SNPs were further genotyped in 100 Hanwoo and 96 imported beef cattle, and analyzed for haplotype construction and association with beef quality traits. We performed haplotype block and linkage disequilibrium studies using four selected SNPs. Two different haplotype blocks (block A: g.10568 C$\rightarrow$T and g.11280 G$\rightarrow$ A; block B: g.13125 C$\rightarrow$T and g.17924 G$\rightarrow$A) were constructed and the block A in particular had a very high r2 (0.936), which indicated a nearly complete linkage disequilibrium existed between the g.10568 C$\rightarrow$T and g.11280 G$\rightarrow$A polymorphisms. A total of four major haplotypes (frequency > 0.05) were identified with the four polymorphisms including TATG (0.36), CGCG (0.31), CGTA (0.19) and TACG (0.06). Statistical association analysis revealed that the g.10568 C$\rightarrow$T and g.11280 G$\rightarrow$A polymorphisms in the FASN were significantly associated with meat color (P=0.004) and texture (P=0.0114). The g.13125 C$\rightarrow$T and g.17924 G$\rightarrow$A polymorphisms in the FASN were also significantly associated with back-fat thickness and quantity index (P=0.0179 and 0.0495, respectively). Our findings suggested that the FASN gene polymorphisms may be used for determining the (unsaturated) fatty acid contents and carcass trait in the Hanwoo beef.

In order to save costs and enhance quality in construction without damaging the environment, the VE/LCC analysis method is increasingly used. This study was carried out to conduct a value analysis for the ecological restoration of a slope considering life cycle cost and performance. The construction conditions were classified into three types(A, B, C) according to the condition of each base. Three construction methods for slope ecological restoration were selected by each condition. Eventually, a value analysis was conducted for total nine conditions by analyzing the life cycle cost and performance. The gradient of slope and base of Condition 1 were below 1:1.2 and general soil, while condition 2 and 3 were below 1:1.0(reaping rock) and below 1:0.7(soft rock, blasted rock), respectively. A value analysis was conducted based on the value estimated via life cycle cost and performance analysis. The result showed that the B construction method had the highest value in Condition 1 as it showed 108.4, while A and C showed 90.3 and 45.8, respectively. When it comes to Condition 2, Construction Method A indicated the highest value as it showed 89.1(B: 47.5, C: 47.0). In Condition 3, Construction Method A(89.1) was the highest, while B and C showed 55.4 and 40.2, respectively. Based on the result of this study, in order to make a reasonable decision that can enhance quality and reduce costs in slope ecological restoration, the slope ecological restoration method must be reviewed in consideration of life cycle cost and performance.
Purpose: ((R)-1-(2-chlorophenyl)-N-1-[$^{11}$C]methyl-N(1-propyl)-3-isoquinoline carboxamide ((R)-PK11195) is a specific ligand for the peripheral type benzodiazepine receptor and a marker of activated microglia, used to measure inflammation in neurologic disorders. This experiment was carried out to determine the effect of organic fertilizer and seed mixture on productivity, botanical composition and forage quality in middle areas(Geumsan, 250m sea level) pasture. Total six experimental pastures (orchardgrass, timothy and tall fescue based seed mixture and with or without organic fertilizer) were established in autumn, 2014 and evaluated productivity and agronomic characteristics from 2015 to 2016. Plant heigh was higher in orchardgrass based mixture and orchardgrass and tall fescue based mixture were higher in $2^{nd}$ harvest. There was not found the significant difference among treatment(P<0.05) in dry matter (DM) content. Botanical composition in $1^{st}$ harvest was higher weed portion in orchardgrass based mixture and legumes was disappeared at $3^{rd}$ harvest in all treatments. Fresh and dry matter yield was higher in $2^{nd}$ year than $1^{st}$ year. In $1^{st}$ year, orchardgrass based mixture with organic fertilizer plot was the highest(P<0.05) DM yield, but tall fescue based mixture with organic fertilizer was the highest in $2^{nd}$ year. Content of crude protein (CP) was the highest in tall fescue based mixture and Timothy based mixture showed low in ADF (acid detergent fiber) and NDF (neutral detergent fiber) content. But the content if in virto dry matter digestibility was higher than others. In $2^{nd}$ year, the nutritive value of $2^{nd}$ and $4^{th}$ harvested grasses were higher and average RFV (relative feed value) value was the highest at $4^{th}$ harvest. According to this result, orchardgrass and tall fescue based mixture would be recommendable for farm who considering productivity and timothy based mixture would be recommendable for considering forage quality in middle areas of Korea.

This experiment was conducted to compare the seed productivity of Spring sown Italian ryegrass(Lolium multiflorum Lam.) depending on the seeding rates(20kg/ha, 30kg/ha, 40kg/ha) in Gangwon region. The experiment was a randomized block design with three replications. The test plots were located in alpine areas of about 600 m above sea level in Gangwon province. The tested Italian ryegrass variety was 'Greencall' developed by the National Institute of Animal Science, RDA. Italian Ryegrass was sown on March 26, 2020, and the seed harvesting was on the 60th day(2 July) from heading date. The heading date was May 8 with no difference, There were no significant differences in the agronomic characteristics including plant height. 30kg/ha seed rate was the highest at 146.8 seed/spike and 40kg/ha seed rate was the lowest at 114.7 seed/spike for the number of seeds per spike. The number of spikes per unit area was the highest in 40kg/ha at 886/㎡ and the lowest in 20kg/ha at 750/㎡. The yield of seed and straw was the highest in 40kg/ha at 1,288kg/ha and 2,970kg/ha respectively, but there was no difference. From the above results, the production of Italian ryegrass seeds through spring sowing in the Gangwon region is not much than autumn seeding, requiring the input of various technologies to increase productivity in the future, and it is desirable to determine the production cost through economic analysis was evaluated.

This experiment was conducted to study on the growth characteristics and seed productivity of Italian ryegrass (Lolium multiflorum Lam., IRG) planted in the Spring in Gangwon Highland according to the seeding distance (20, 30 and 40 cm). The field was located in highland around 600 m above sea level. The experimental design was randomized block design with three replication and the tested IRG variety was 'Greencall' developed by National Institute of Animal Science (NIAS). IRG was sown on March 26, 2020, and harvested on July 2. The plant height was the shortest at 80.5 cm in the 40 cm seeding distance plot (P<0.05), and there was no significant difference between the 20 and 30 cm seeding distance. The number of spike per square meter (㎡) was significantly higher in the 20 cm seeding distance plot than that of 40 cm (937 vs. 571). The dry matter (DM) content of seed and straw after harvesting was 49.70 and 33.36 % on average, and there was no significant difference between treatments (P>005). However, there was a significant difference in the fresh and DM yield of seeds and straw (P<0.05). DM yield of seeds was significantly higher in 20 cm distance than that of 40 cm, and the yield of straw was the same trend. On the other hand, there was no significant difference in DM yield between 20 cm and 30 cm and also in the feed value of straw after seed harvesting among seeding distance. The average CP, ADF, NDF, and TDN contents were 6.91, 36.76, 61.75 and 59.86%, respectively, and the RFV value was 91. Considering the above results, the production of Italian ryegrass seeds sown in the spring in the highlands of the Gangwon is lower than that of autumn sowing, but it is judged that it needs to be reviewed in case it is unavoidable. In the future, there should be an economic analysis and the development of technology that can increase production.

This experiment was conducted to compare the seed productivity of the Italian ryegrass(Lolium multiflorum Lam.) varieties sown in the spring in Gangwon region. The experiment was randomized block design with three replications. The Experimental field was located in alpine areas of about 600 m above sea level in Gangwon province. The tested Italian Ryegrass varieties were 'Greenfarm', 'Greencall' and 'Kowinearly' developed by National Institute of Animal Science, RDA. Italian Ryegrass varieties were sown on March 26, 2020, and the harvest was on the 60th day of mean heading date, July 2. The heading date of Kowinearly was May 8, but Greenfarm and Greencall was May 4. The plant length was the largest in the Kowinearly variety. However, the Kowinearly suffered severe lodging. There was no significant difference in the length of spike among varieties, and the number of seeds per spike was the lowest in Greenfarm at 118.5 seed/spike. As for the seed weight per spike, the Greenfarm variety was significantly lower at 0.56 g/spike, but the 1,000 seed weight was the heaviest in the Greenfarm at 2.5g.. The number of spike per unit area was the highest in Greenfarm at 906/㎡. The dry matter content of seeds was the highest in Greenfarm at 54.3%, and for straw, Kowinearly was the highest at 35.3%. Seed productivity was not significant among varieties, and the average was 1,493 kg/ha. The yield of straw after seed production was also not significant among varieties (P>0.05), and the average was 3,172 kg/ha. From the above results, the production of Italian ryegrass seeds through spring sowing in the Gangwon region is not much than autumn seeding, requiring the input of various technologies to increase productivity in the future, and it is desirable to determine the production cost through economic analysis was judged.

This experiment was conducted to the purpose of evaluating the effect of feeding on Korean Native Cattle to expand the production and utilization of whole crop rice (WCR). TMR (Total mixed ration) feed was prepared by using WCR produced in Pyeongtaek, Gyeonggi-do, and the growth capacity and meat quality characteristics of 16 Korean Native Cattle raised up to 29 months of age were investigated. The produced WCR silage for feed had a moisture content of 64.02% and a crude protein content of 7.54%, and was blended with about 45% during the growing season, and lowered to 35, 15 and 9%, respectively, in the fattening period (early, middle and late stages). The body weight of the WCR-TMR feeding group was significantly higher than that of the control in the middle and late fattening stage, and at the end (29 months of age), the control group was 631 kg/head, but the WCR-TMR feeding group was 647 kg/head, which was higher. The average daily gain was significantly higher in the WCR-TMR feeding group in the growing and early fattening period, and there was no difference in the mid- and late fattening period. In the whole period, 0.71 vs 0.75 kg/head/day, WCR-TMR feeding group was high. In terms of meat quantity, the back fat thickness of the control group (11.7 mm) was significantly thicker than that of the WCR-TMR fed group (9.3 mm) (P<0.05). There was no difference in Rib eye area, Carcass weight and Meat yield index (P>0.05). In terms of meat quality, the Marbling score was higher in the WCR-TMR feeding group (P<0.05), and there were no significant differences in Meat color, Fat color, Texture and Maturity. Considering the above results, TMR feeding mainly on whole crop rice silage for feed improved the productivity of livestock, but there was no significant difference in meat quality. Therefore, it is judged that it is necessary to produce and use the whole crop rice for feed in countries with poor forage conditions.
Purpose: $[^{11}C]6-OH-BTA-1$ ([N-methyl-$^{11}C$]2-(4'-methylaminophenyl)-6-hydroxybenzothiazole, 1), a -amyloid imaging agent for the diagnosis of Alzheimer's disease in PET, can be labeled with higher yield by a simple loop method. During the synthesis of $[^{11}C]1$, we found the formation of by-products in various solvents, e.g., methylethylketone (MEK), cyclohexanone (CHO), diethylketone (DEK), and dimethylformamide (DMF). Materials and Methods: In Automated radiosynthesis module, 1 mg of 4-aminophenyl-6-hydroxybenzothiazole (4) in 100 l of each solvent was reacted with $[^{11}C]methyl$ triflate in HPLC loop at room temperature (RT). The reaction mixture was separated by semi-preparative HPLC. Aliquots eluted at 14.4, 16.3 and 17.6 min were collected and analyzed by analytical HPLC and LC/MS spectrometer. Results: The labeling efficiencies of $[^{11}C]1$ were $86.0{\pm}5.5%$, $59.7{\pm}2.4%$, $29.9{\pm}1.8%$, and $7.6{\pm}0.5%$ in MEK, CHO, DEK and DMF, respectively. The LC/MS spectra of three products eluted at 14.4, 16.3 and 17.6 mins showed m/z peaks at 257.3 (M+1), 257.3 (M+1) and 271.3 (M+1), respectively, indicating their structures as 1, 2-(4'-aminophenyl)-6-methoxybenzothiazole (2) and by-product (3), respectively. Ratios of labeling efficiencies for the three products $([^{11}C]1:[^{11}C]2:[^{11}C]3)$ were $86.0{\pm}5.5%:5.0{\pm}3.4%:1.5{\pm}1.3%$ in MEK, $59.7{\pm}2.4%:4.7{\pm}3.2%:1.3{\pm}0.5%$ in CHO, $9.9{\pm}1.8%:2.0{\pm}0.7%:0.3{\pm}0.1%$ in DEK and $7.6{\pm}0.5%:0.0%:0.0%$ in DMF, respectively. Conclusion: The labeling efficiency of $[^{11}C]1$ was the highest when MEK was used as a reaction solvent. As results of mass spectrometry, 1 and 2 were conformed. 3 was presumed.
