Effects of Soyoligosaccharides and Varing Dietary Fats on Intestinal microflora. Lipid Profiles and Immune Responses in Rats (대두올리고당이 흰쥐의 장내 균총, 지질 대사 및 면역 능력에 미치는 영향)

  • The aim of this study was to evaluate the effects of soyoligosaccharides on intestinal microflora transit time lipid profiles and immune responses. Forty-eight male rats of Sprague-Dawley strain were divided into six groups(SYS: soybean oil-sucrose SYO: soybean oil-oligo SES; esame oil-sucrose SEO:sesame oil-oligo BFS : beef tallow-sucrose BFO : beef tallow-oligo) soyoligo-groups(SYO, SEO, BEO) were fed the diet containing 5% soyoligosaccharides but control groups not fed them and all groups were fed 20% fats for 4 weeks. The number of fecal bifidobacteria tended to increase after soyoligosaccharides feeding. The significant increase was found in SEO group compared to SES in that of fecal bifidobacteria,. Stool wet weight dry fecal weight and stool water content were increased significantly in soyoligo-group(SYO, SEO, BFO) compared to control groups(SYS, SES, BFS) and gastrointestinal transit time(TT) tended to increase. Soyoligosaccharides lowered plasma cholesterol in SEO group significantly more than did other groups. soyoligo-groups(SYO, SEO, BFO) tended to decrease liver triglycerides compared to control groups. Plasma IgG concentration increased in beef tallow groups(BFS, BFO) than that in other dietary fat groups. Soyoligosaccharides and kinds of fats had effects on plasma C3 concentration and mitogenesis of the spleen cells. Soyoligosaccharides had no effects on plasma leptin and insulin concentration but kinds of fats did so that sesame groups were decreased significantly in plasma leptin concentration but increased significantly in plasma insulin concentration compared to other dietary fat groups. In conclusion dietary soyoligosaccharides changed the composition of the intestinal microflora beneficially with sesame oil and partially had effect on plasma and liver lipid profiles .(Korean J Nutrition 33(6) : 597∼612, 2000)

Effects of Belly Dancing and Nutritional Education on Body Composition and Serum Lipids Profiles of Obese Women in a Study, 'Obesity Clinic Projects at Community Healthcenter' ('보건소 비만클리닉 시범사업'으로 실시한 Belly Dance와 영양교육이 비만여성의 신체조성 및 혈청지질 농도에 미치는 영향)

  • The effects of a 12-week weight reduction program applied to obese women at a community health center were studied. Middle aged women participated in the beginning of the program, but 34 persons finished the study (85% completion). Subjects practiced belly dancing three times a week for 60 minutes (60~85% HR max) per session, and nutritional education was carried out four times during the 12 week program. The weight, %body fat, and BMI significantly decreased by 5.6%, 9.5%, and 5.6%, respectively. The concentrations of serum triglycerides, total cholesterol, and LDL-C were also significantly reduced by 13.5%, 9.5%, and 10.4%, respectively. Insulin level was reduced by 19.8%, thus lowering the HOMA-IR index. Leptin level significantly was reduced by 32.35%, adiponectin concentration. Total ROS concentration increased while total antioxidant capacity in serum remained unchanged. In conclusion, a 12-week 'Obesity Clinic Program' belly dancing and nutritional education held at a Community Center seemed to have health-promoting effects through reducing body fats, plasma lipids, and improving insulin resistance.

Effects of Sunsik Prepared through Steaming-Drying Cycles on Blood Glucose and Inflammatory Marker in Rats Fed High Fat (증포 횟수를 달리하여 만든 선식을 첨가한 고지방식이가 비만 쥐의 혈당과 염증지표에 미치는 영향)

  • This study was conducted to evaluate the effects of Sunsik prepared by steaming -drying cycles- on blood glucose and inflammatory markers in high fat diet-induced obese rats. The experimental animals were divided into five groups and subjected to diets for 8 weeks; normal diet (control), high fat diet (HF), and high fat diet+10% supplementation of Sunsik (1HF, 3HF, 7HF, 9HF). The results showed body weight, food efficiency ratio, blood glucose and insulin levels of high fat diet groups were significantly higher than those of the control group. Increased glucose levels by high fat diet were reduced to normal levels by the four Sunsik supplementations. Insulin resistance (HOMA-IR) of the HF group was significantly higher than those of the control group, whereas supplementation with the four types of Sunsik reduced insulin resistance to similar levels as the control group. Increased insulin secretion and leptin levels of high fat diet-induced obese rats were significantly reduced by supplementation with the four types of Sunsik. C-reactive protein (CRP) of inflammation related components was significantly higher than the control group. However, increased CRP concentration by high fat diet was significantly reduced by supplementation with Sunsik.

  • This study was to investigate whether cultivated ginseng (CG), cultivated wild simulated ginseng (CWG) and cultivated red ginseng (CRG) extracts influences on the obesity. The saponin contents of 3 kinds of ginsengs were analysed by HPLC-ESI-TOF-MS. Total saponin contents were determined in CG on the most contents and since red ginseng has the highest PD (protopanaxadiol type) / PT (protopanaxatriol type) ratio, there may be differences between ginseng, wild ginseng, and red ginseng with respect to their pharmacological effects. Male C57BL/6J mice were fed a normal diet(N), HFD (60% Kcal fat, C), HFD with CG, CWG and CRG extracts (800 mg/kg) for 5 weeks. We observed change of total body weight, degree of hepatic lipid accumulation and immunohistochemical change of GLP-1 and insulin-secreting cells. Also this study attempts to use the physiological analysis method to analyze the changes of blood lipids, insulin and leptin concentration. The change of body weight and size of accumulated lipid droplets in liver lobules decreased in all of the experimental groups than the control(C) group. In the pancreas, the immunohistochemical density of insulin-secreting cells were significantly stronger in the CWG and CRG than C group. The levels of serum insulin and leptin significantly decreased 55.6%, 54.3% respectively in CWG and CRG. The changes of triglyceride, total cholesterol in serum decreased in CRG than the C group. Obesity related CG, CWG and CRG extracts might have contribute to improvement of obesity by regulating the levels of blood lipids and biochemical indicator of fat accumulation.

Antioxidant Activity of Rosa rugosa Thunberg and Effect on Serum Lipid Level in High Fat Diet-induced Mice (해당화 열매 추출 분말의 항산화 활성과 고지방 식이로 유도된 Mice의 혈청지질 수준에 미치는 영향)

  • Since Korean ancient times, powder of Rosa rugosa Thunberg has been used as a folk remedy. This study was conducted to verify the effects of Rosa rugosa Thunberg powder on in vitro antioxidant properties and serum lipid levels of high-fat diet-induced mice from a nutritional viewpoint. In the case of Rosa rugosa Thunberg powders, measurement of TPC, ABTS radical scavenging activity of trolox, DPPH radical scavenging activity, and measured value of FRAP were higher in ethanol extract than water extracts. For LDL-cholesterol concentration, mice fed 10% powder of Rosa rugosa Thunberg with high-fat diets showed a high numerical value compared with other groups (p<0.05). When testing for triglyceride concentrations in blood, mice fed 10% and 20% powder of Rosa rugosa Thunberg with high-fat diets showed the lowest numerical values (p<0.05). When testing for blood insulin concentrations, the high-fat diet group showed higher levels than compared to the control group (p<0.05). When testing for blood leptin concentrations, the high-fat diet group was $5.88{\pm}3.53ng/dL$, whereas mice fed 10% powder of Rosa rugosa Thunberg with high-fat diets showed a blood leptin level of $10.36{\pm}5.96ng/dL$ (p<0.05). Therefore, results prove that powder of Rosa rugosa Thunberg reduces triglyceride concentrations in the blood, and could be used as an excellent natural antioxidant in the future.

Anti-adipogenesis Effects of 3 Herbal Formula on Blood Stasis (어혈처방 3종이 지방전구세포 분화에 미치는 영향)

  • Objectives : Blood stasis (BS) is related to be caused by blood circulation and stagnation which are cancer, atherosclerosis and hyperlipidemia in traditional medicine. We extracted 3 kinds of BS formula; Seogakjihwang-tang (SGT), Tonggyuhawlhyul-tang (THT), Hyulbuchukeo-tang (HCT). This study was conducted to investigate whether the 3 kinds of herbal formula extracts have inhibitory efficacy association with anti-adipogenesis. Methods : To investigate the anti-adipogenesis, we used the mouse fibroblast cell line, 3T3-L1 which differentiated into adipocytes in response to insulin, IBMX and dexamethasone (MDI). Cytotoxicity of herbal formula extracts were examined by CCK-8 kit. Intracellular lipid droplets were detected by Oil-Red-O staining. Triglyceride (TG) and leptin were measure using elisa kit. Results : The yield of water extracts was 14.62% (SGT), 21.27% (THT), 20.02% (HCT). Lipid accumulation was reduced significantly by 3 kinds of herbal formula compared to control. Especially, THT and HCT decreased lipid droplet, respectively at all concentration. The TG and leptin were also inhibited by 3 kinds of herbal formula. The IC50 of TG were $280.51{\mu}g/m{\ell}$ (SGT), $52.62{\mu}g/m{\ell}$ (THT), $313.99{\mu}g/m{\ell}$ (HCT). The IC50 of leptin were $348.76{\mu}g/m{\ell}$ (SGT), $164.02{\mu}g/m{\ell}$ (THT), $257.00{\mu}g/m{\ell}$ (HCT). THT was better than other herbal formula on anti-adipogenesis. Conclusion : kinds of herbal formula inhibited adipogenesis in MDI-induced 3T3-L1 adipocytes, as indicated by the significant reduction in TG and leptin concentration without cytotoxicity. Therefore, 3 kinds of herbal formula for BS might act as a therapeutic agent for preventing lipid diseases, such as obesity and atherosclerosis.

Effect of velvet antler on the function of adipocytes(3T3-L1) and its association with IGF-1 (녹용(鹿茸)이 지방세포(脂肪細胞)(3T3-L1)와 성장인자(成長因子)에 미치는 영향(影響))

  • Objectives: These experimental studies were designed to investigate the effects of antler's extracts on the expression of leptin, IGF-l and IGF-1 receptor on cultured 3T3-L1 cells. Methods: A mouse adipoblast cell line, 3T3-L1, was cultured with or without a differentiation medium containing Isobutylmethylxanthin, Dexametasone and Insulin before adding extracts of antler of various concentratio(10, 50, ]00, 200${\mu}g/ml$). The expression of leptin and IGF-1 receptor was measured by western blot assay, and expression of IGF-1 was determined by F ACS analysis. Results and Conclusion: The 3T3-L1 cells' differentiation did not show a significant induction by extracts of antler. The expression of leptin was significantly decreased depend on antler's concentration. The expression of IGF-1 showed a slightly increase by extracts of antler, whereas that of IGF-receptor showed a tendency to increase. The total amount of intracellular triglyceride showed a tendency to diminish as the concentration of antler's extract increase.

Effect of Silk Fibroin Hydrolysate on Adipocyte Metabolism in db/db Mice (실크 피브로인 산 가수분해물이 db/db mice의 지방세포 대사에 미치는 영향)

  • Effect of the acid hydrolysate of silk fibroin on obesity was investigated in obese(057BL/KsJ-db/db) mice. After 8 weeks feeding of 1%(w/w) or 3%(w/w) fibroin hydrolysate, the extents of reduction in body weight were significantly higher than that of obese control. The weight reduction in female mice was higher than that in male mice. Plasma leptin in male mice increased up to 1.8-fold higher level than obese control by feeding hydrolysate. In case of female mice, however, it rather decreased with increased feeding concentration of hydrolysate. From the results of high glycine and serine contents of peptide fractions contained in fibroin hydrolysate, it was inferred that fibroin peptides might affect xylosyltransferase(XT) activity on chondroitin sulfate synthesis causing to change susceptibility of adipocytes to hormones such as insulin followed by the reduced leptin synthesis in female mice. The result of the higher lipolysis in hydrolysate-fed group than obese control indicated that the reduction in body weight was due to the increased lipolytic activities in male and female mice in common.

Effect of the Erigeron annuus In Vitro Antioxidant Properties and Extract on Serum Lipid in Mice (개망초 추출 분말의 In Vitro에서의 항산화 특성과 고지방식이로 유도된 Mice의 혈청지질 수준에 미치는 영향)

  • This study was conducted to verify the effects of Erigeron annuus powder on serum lipid levels of high-fat diet-induced mice from a nutritional viewpoint. Erigeron annuus powder has been used as a folk remedy since ancient times in Korea. There was no significant difference in the weight of the kidneys and spleens of the mice. The high-fat diet group had a significantly higher kidney weight compared to other groups (p<0.05). In the group of mice fed 20% Erigeron annuus powder with a high-fat diet, the concentration of serum LDL-cholesterol was high (p<0.05), whereas the concentration of triglyceride was remarkably lower compared to other groups (p<0.05). The group fed 10% Erigeron annuus with a high-fat diet had the lowest concentration of blood phospholipids (p<0.05) as well as the highest alkaline phosphatase and alanine aminotransferase levels in blood (p<0.05). There was no difference in blood insulin concentration. However, blood leptin concentration was significantly higher ($5.88{\pm}3.53ng/dL$) in mice fed a high-fat diet compared to other groups (p<0.05). Measurements of Erigeron annuus revealed that TPC, $ABTS^+$ radical scavenging activity of trolox, DPPH radical scavenging activity, and the measured value of FRAP were higher in the ethanol extract than in the water extract. Especially, the antioxidant activity effects were excellent for the $ABTS^+$ radical scavenging activities of trolox and FRAP values of Erigeron annuus. Therefore, Erigeron annuus powder showed antioxidant activity. Hence, Erigeron annuus powder drastically lessened triglyceride concentration in blood in high-fat diet-induced mice. Thus, the powder is considered to have utility in the food processing industry. Additional related experiments are ongoing.

Effect of reduced energy density of close-up diets on metabolites, lipolysis and gluconeogenesis in Holstein cows

  • Objective: An experiment was conducted to determine the effect of reduced energy density of close-up diets on metabolites, lipolysis and gluconeogenesis in cows during the transition period. Methods: Thirty-nine Holstein dry cows were blocked and assigned randomly to three groups, fed a high energy density diet (HD, 1.62 Mcal of net energy for lactation $[NE_L]/kg$ dry matter [DM]), a medium energy density diet (MD, $1.47Mcal\;NE_L/kg\;DM$), or a low energy density diet (LD, $1.30Mcal\;NE_L/kg\;DM$) prepartum; they were fed the same lactation diet to 28 days in milk (DIM). All the cows were housed in a free-stall barn and fed ad libitum. Results: The reduced energy density diets decreased the blood insulin concentration and increased nonesterified fatty acids (NEFA) concentration in the prepartum period (p<0.05). They also increased the concentrations of glucose, insulin and glucagon, and decreased the concentrations of NEFA and ${\beta}-hydroxybutyrate$ during the first 2 weeks of lactation (p<0.05). The plasma urea nitrogen concentration of both prepartum and postpartum was not affected by dietary energy density (p>0.05). The dietary energy density had no effect on mRNA abundance of insulin receptors, leptin and peroxisome proliferator-activated $receptor-{\gamma}$ in adipose tissue, and phosphoenolpyruvate carboxykinase, carnitine palmitoyltransferase-1 and peroxisome proliferator-activated $receptor-{\alpha}$ in liver during the transition period (p>0.05). The HD cows had higher mRNA abundance of hormone-sensitive lipase at 3 DIM compared with the MD cows and LD cows (p = 0.001). The mRNA abundance of hepatic pyruvate carboxy-kinase at 3 DIM tended to be increased by the reduced energy density of the close-up diets (p = 0.08). Conclusion: The reduced energy density diet prepartum was effective in controlling adipose tissue mobilization and improving the capacity of hepatic gluconeogenesis postpartum.
