Effect of Veterinary Antibiotics on the Growth of Lettuce

  • Veterinary antibiotics (VAs) has been used to treat animal disease and to increase body weight. However, released VAs in the soil via spreading of compost can transport to plant and affect its growth. Main purpose of this research was i) to monitor VAs concentration in plant and ii) to evaluate inhibition effect of VAs residuals on the plant growth. Red lettuce (Lactuca sativa) was cultivated for 35 days in the pot soil spiked with 3 different concertation (0.05, 0.5, $5.0mg\;kg^{-1}$) of chlortetracycline (CTC) and sulfamethazine (SMZ). After 35 days of cultivation, concentration of CTC and SMZ in the plant was measured. Residual of CTC and SMZ was only quantified at the range of $0.007-0.008mg\;kg^{-1}$ and $0.006-0.017mg\;kg^{-1}$ in the leaf and root respectively when high concentration ($5.0mg\;kg^{-1}$) of antibiotic was spiked in the soil. Leaf length and root mass was statistically reduced when $0.05mg\;kg^{-1}$ of CTC was spiked in the soil while no statistical difference was observed for SMZ treatment. This result might indicated that high $K_{ow}$ and $K_d$ value are the main parameters for inhibiting plant growth. Antibiotics that has a high $K_{ow}$ causing hydrophobicity and easy to bioaccumulate in the lipid cell membrane. Also, antibiotics that has a high $K_d$ properties can be sorbed in the root causing growth inhibition of the plant. Overall, management of VAs should be conducted to minimize adverse effect of VAs in the ecosystem.

Inhibition Effects Against Plant Pathogenic Fungi and Plant Growth Promotion by Beneficial Microorganisms (유용 미생물을 활용한 식물 병원 곰팡이의 억제와 식물 생장촉진 효과)

  • The experiment was carried out to analyze the inhibition effect of plant pathogenic fungi and growth promotion activity induced by the bacterial strains isolated from peatmoss. Among the isolated bacterial strains, B10-2, B10-4, B10-5 and B10-6 which showed more than 30% inhibition rate against Botrytis cinerea and Rhizoctonia solani in vitro, were further analyzed in the greenhouse for the growth promotion activity on lettuce (Lactuca sativa), pak-choi (Brassica compestris L. ssp. chinensis) and Chinese cabbage (Brassica campestris L. ssp. pekinensis). The results showed the treatment of B10-4 on lettuce showed the highest growth promotion activity with the leaf area ($169.17cm^2$), fresh weight (leaf: 40.29 g, root: 8.80 g)and dry weight (leaf: 11.24 g, root: 4.17 g), which was about two folds as compared to control. On pak-choi, the growth promotion rate was the highest with the leaf area of $112.87cm^2$, leaf fresh weight of 60.70 g, root fresh weight of 3.37 g, leaf dry weight of 14.34 g, and root dry weight of 1.90 g. As a result of treatment of B10-13 on chinese cabbage, the growth promotion rate was the highest with the leaf area ($293.56cm^2$), fresh weight (leaf: 113.67 g, root: 2.40 g) and dry weight (leaf: 6.03 g, root: 0.53 g). The production of Indole Acetic Acid (IAA) and Indole-3-Butylic Acid (IBA) were also analyzed in these bacterial isolates. The IAA and IBA analyses were carried out in all bacterial isolates each day within the 5 days of incubation period. The highest production of IAA was observed with $112.57{\mu}g/mg$ protein in B10-4 after 3 days of incubation and IBA production was the highest in B10-2 with $58.71{\mu}g/mg$ protein after 2 days of incubation. Also, phosphate solubilizing activity was expressed significantly in B10-13 in comparison to that of other bacterial isolates. Bacterial identification showed that B10-2 was Bacillaceae bacterium and B10-5 was Bacillus cereus, B10-4 and B10-6 were Bacillus sp. and B-13 was Staphylococcus sp. by ITS sequence.

Occurrence of Fusarium Wilt Caused by Fusarium oxysporum f. sp. lactucae and Cultivar Susceptibility on Lettuce (Fusarium oxysporum f. sp. lactucae에 의한 상추 시들음병 발생 및 품종간 감수성 비교)

  • A wilt disease on lettuce was observed in 2006 and 2007 in commercial plastic house at main production areas of lettuce in Icheon, Yongin and Goyang of Gyeonggi Province. The disease was characterized by the wilting of lower leaves, accompanied by stunting symptoms of the whole plants. Old affected stems showed the black streak in the vascular system. The pathogen, Fusarium oxysporum f. sp. lactucae was isolated from stems and roots of diseased plants. Isolated pathogen also produced the microconidia and macroconidia with chlamydospores on carnation leaf agar medium. The pathogen easily invaded and made many chlamydospores on the roots of lettuce and also made dark streaking through the vascular in the lettuce stems. The density of Fusarium sp. in the severely diseased field soil was more higher populations than that in the healthy and less diseased field soil. The minimum population of pathogen would be above $10^3$cfu/g soil to induce the Fusarium wilt on lettuce in plastic house. The results of pathogenicity test showed 'Sunpung' and 'Mipungpochap' was high susceptible to Fusarium pathogen isolates while some cultivar 'Mihongjeokchukmyeon' and 'Jinjachukmyeon' showed moderate resistance. Disease development for some lettuce was related to treated temperature, so the symptom was more severe above $25^{\circ}C$. Selection of appropriate lettuce cultivar and planting time should be related for the successful control of Fusarium wilt.

Evaluation of Growth Characteristics and Lead Uptake of Lettuce under different application levels of Bottom Ash (저회의 시용수준에 따른 상추의 생육 및 납 흡수 특성 평가)

  • BACKGROUND: Most of the bottom ash(BA) from wood pellet-based thermal power plants that is not recycled is placed into landfill. BA has a function and structure similar to biochar. Hence, BA is classified as waste, but, it is predicted that BA can be used agricultural utilization. METHODS AND RESULTS: To investigate the effect of BA application on lettuce, growth characteristics and Pb contents were examined with BA application levels(0, 1, 2, 3 and 4 g/L), respectively, in hydroponic cultivation with Pb solution. Irrespective with BA application levels, the length, leaf number and fresh weight of lettuce in BA treatments were increased by 84.3~120, 36.2~39.0, and 215~322%, respectively, compared to the BA-0 treatment. The groups with BA treatments, Pb in the nutrient solution was adsorbed to the BA due to the surface area and functional groups of the BA, and the lettuce growth was maintained more smoothly than in the BA-0 treatment. BA application is considered to have created a favorable environment for lettuce growth in hydroponic cultivation with Pb solution. CONCLUSION(S): Although direct comparing the removal effect of heavy metal between BA and biochar is not present, the BA application in contaminated area suggested a significant meaning on the recycling waste, and increasing potential crop productivity by immobilizing heavy metal.

Effect of the Hydrolysate of Pigs Hoof on Plant Growth and Physico-chemical Properties (Pigs hoof 가수분해물의 이화학성 및 작물 생육에 미치는 효과)

  • This study was conducted to find the physico-chemical properties and the amino acid content of the pigs hoof hydrolysate, keratin protein and to investigate its fertilizer effect on the growth of crops. The keratin proteins such as pigs hoof were alkali-hydrolyzed to produce the hydrolysates. The chemical properties of the hydrolysate of pigs hoof was 6~7 of pH and $10{\sim}15dS\;m^{-1}$ of EC. Total amino acid contents released from the pigs hoof were 10.18%, respectively. The pot experiment was carried out for the cultivation of lettuce. The treatment design of these pot cultivation was composed of Control (compost + NPK), PHH-0.5, PHH-1.0, PHH-2.0 (${\times}2,000$ ; 1,000 ; 500 diluted solution of pig hoof hydrolysate). After lettuce cultivation, the pH values in all treatment soils were decreased than those in initial soils, and the exchangeable cation value was higher than that of control. In all PHH treatments, lettuce growth was better in the leaf length by 6~16% and the leaf width by 4~15% than in control. Therefore, the PHH solutions manufactured by hydrolysis process had plenty of amino acids, and among them PHH had the most abundant nutrients and amino acids with highest growth and yield effect on lettuce.

The Effect of Vermicast on the Growth of Vegetable Plants - Estimation of Optimum Mixture Ratios of Vermicast - (지렁이 분립이 엽채류의 생육에 미치는 영향 - 분립의 최적혼합비율의 추정 -)

  • This study was carried out to investigate the effect of different mixture ratios of vermicasts on the growth of leaf beet(Beta vulgaris var. cicla), young radish (Raphanus sativus L), spinach(Spinacia oleracea) and lettuce(Lactuca sativa) The mixture of ratios of vermicast(fermented pig manure with sawdust) and peatmoss were 100%+0%, 60%+40%, 40%+60% and 0%+100%(control), respectively. The bulk density, particle density, pH, electrolytic conductivity, ash, total nitrogen, cation exchange capacity, available phosphorous and exchangeable cations increased significantly compared to those grown in the 100% of peatmoss(control). The significantly higher values of plant length, leaf area, shoot dry weight, root weight, root length and biological yield obtained were $40{\sim}100%$ and 40% of vermicast in leaf beet and lettuce, respectively. The young radish and spinach showed that significantly differences of shoot dry weight, root weight and biological yield in the mixture ratios of vermicast were $40{\sim}60%$ and 40% in young radish and spinach, respectively.

Isolation and Identification of Spoilage Bacteria on Organic and Conventional Fresh Produce in Korea (국내에 시판되고 있는 유기농산물과 일반농산물의 부패미생물 분리 및 동정)

  • This study was conducted to investigate spoilage bacteria on organic and conventional fresh produce in Korea. Three samples (perilla leaf, cabbage, and romaine lettuce) of organic and conventional fresh produce were stored at $4^{\circ}C$ for 14 days and examined for spoilage bacteria on TSA. Isolated bacteria from organic and conventional fresh produces were identified using 16S rRNA sequencing method. Population of total aerobic bacteria on conventional perilla leaf, cabbage, and romaine lettuce were 7.59, 7.01, and $5.84{\log}_{10}CFU/g$, and populations of total aerobic bacteria were 6.72, 6.15, and $5.85{\log}_{10}CFU/g$, for organic perilla leaf, cabbage, and romaine lettuce, respectively. Major spoilage bacteria of organic and conventional fresh produces were similar however their levels were little different. For example, a major spoilage bacterium resulting the highest level on conventional perilla leaf was Stenotrophomonas maltophilia whereas that was Microbacterium sp. for organic produce. From these results, microflora or spoilage microorganism could be different depending on their cultivation types as conventional or organic produces and this information might be used for developing effective preservation method for different types of fresh produce.

Absorption of sulfur dioxide gas with various crops and it's relation to leaf injury (아황산가스에 의(依)한 작물별파해엽율(作物別破害葉率) 및 가스흡수량조사(吸收量調査))

  • To study the effects of sulfur dioxide on the plant; Barly, wheat, soybean sweet potato, cucumber, egg plant, red pepper, tomato, lettuce, water melon, castor bean, grape and lily were exposed to the different levels of sulfur dioxide gas(0.1, 0.25, 0.50, $1.0mg/{\ell}/hr$) The symptoms of damage, ratios of destroyed leaf, sulfur content and absorption amounts of the gas by leaves were investigated 1. According to the increasing concentration of the gas ratios of destroyed leaf were increased in all plants. The ratios of destroyed leaf were shown by egg plant at $0.1mg/{\ell}/hr$ of $SO_2$ were 30 percent, and no visible injuries were shown by the wheat potato, castor bean, water melon, lily at $0.25mg/{\ell}/hr$. 2. Gray and red brown spots between the vein nerve shown by barly and wheat leaf; leaf burn by soybean, potato, sweat potato, castor bean, egg plant, red pepper, tomato and grape; leaf withering from the leaf tips by the lettuce, water melon, lily. 3. The volums of the gas absorption by cucumber, egg plant, red peper, castor he an were more than $10{\ell}/hr$, however less than $2{\ell}/hr$, be lettuce, water melon, grape, barly and wheat. 4. According to the increasing concentration of sulfur dioxide gas, sulfur contents in leaf were increased in all plants, however volums of absorption gas were decreaed. 5. According to the increase of sulfur content in leaf, ratios of destroyed leaf were increased. 6. Positive correlation was shown between total and water soluble sulfur content in leaf.

  • In the recirculating hydroponics, pH of nutrient solution was gradually decreased until the middle of growing-period, and then began to go up and reached the initial level at the latter of growing-period regardless of Cu·Zn content. Changes of pH was gently down for supply of humus in nutrient solution. The growth of lettuce was good with supply of humus, but it was decreased as Cu·Zn content was increased. Although leaf chlorosis was caused by excessive content of Cu·Zn as 5, 10 ppm, it was controlled by adding humus in nutrient solution, and fresh weight of lettuce was obviously improved by adding humus.

Efficiency of Removal of Indoor Pollutants by Pistia stratiotes, Eichhornia crassipes and Hydrocotyle umbellata

  • In this study, we compared efficiency of different aquatic plants in removing indoor pollutants and examined their potential to purify indoor air. Two liter of water in chamber was used as the control, while the other chambers containing water lettuce (Pistia stratiotes), water hyacinth (Eichhornia crassipes), and water coin (Hydrocotyle umbellata) were used as treatment groups. Temperatures inside all the chambers were maintained between 20 ℃ and 23 ℃. Humidity in the chambers with aquatic plants increased by 30% and 50% control respectively. The removal of formaldehyde per unit leaf area was examined in each aquatic plant. It turned out that water hyacinth removed the highest amount of formaldehyde, followed by water lettuce and water coin. Both water hyacinth and water lettuce increased the amount of removal of formaldehyde until the end of the experiment. In the case of airborne dust (PM 10) and fine dust (PM 2.5), water coin, which had the highest number of leaves, removed more PM 10 and PM 2.5 than the other aquatic plants, with statistically significant difference. In addition, both water coin and water hyacinth smoothly opened and closed stomata before and after the experiment. Consequently, as the aquatic plants were effective in controlling humidity and removing pollutants, they can be used as air purifying plants.
