Food Consumption and Utilization Efficiency in Samia ricini Donovan Reared on Ricinus communis, lin. Leaves Supplemented with Cyanobacteria

  • Food consumption and conversion efficiency of eri silkworm Samia ricini Donovan were studied during $4^{th}$ and $5^{th}$ larval instars by feeding castor leaves fortified with 100, 200, 300, 400 and 500 ppm concentrations of aqueous extracts of cyanobacteria Anabaena variabilis. The nutritional indices viz., ingesta, digesta, approximate digestibility (%), reference ratio and efficiency parameters like ECI and ECD were recorded which were significantly high at 400 ppm concentration treated batches of $4^{th}$ instar larvae over control batches. The decline in nutritional efficiency parameters of $5^{th}$ instar treated larvae might be due to higher utilization of the digested food for metabolic activities. Significant difference of ECI to cocoon % and non-significant difference of ECD to cocoon% and shell were observed between the treatments and control. Cyanabacteria feed supplement contains antibiotic and nutritions factors which has reflective effect on the biological parameters in eri silkworm and therefore has greater application in commercial eri silkworm rearing.

  • High blood cholesterol levels and oxidized cholesterol are risk factors for cardiovascular disease, which displays high annual incidence. Although studies on sericulture products, including pupae, silk protein, and blood lymph, as hypocholesterolemic substances have been reported, insufficient research in this field has been focused on silkworm larvae. Six larval extracts (Low temperature distilled water, LW; hot temperature distilled water, HW; and 30-100% ethanol, E30-E100) were prepared, and their effects on cholesterol metabolism were examined. LW most potently reduced the risk of cholesterol-related disorders. Polyphenols were highly represented in LW, corresponding with its increased antioxidant potency. The cholesterol biosynthesis enzyme, 3-hydroxy-3-methyl-glutaryl-coenzyme A reductase (HMGCR) was strongly inhibited by LW. Hepatocytes over-expressed LDL receptor (LDLR) after LW stimulation, promoting cholesterol elimination from plasma. LW also increased ATP binding cassette transporter 1 (ABCA1) gene expression, upregulating HDL biogenesis. In conclusion, LW exhibited strong antioxidant activity, suppressed cholesterol biosynthesis, improved LDL uptake from plasma, and upregulated HDL biosynthesis. In aggregate, these activities could reduce blood cholesterol levels and prevent cardiovascular disease.

Effects of Serotonin Injection on Spawning and Modulation of Sperm Densities for Preventing Polyspermy to Achieve Larval Development in Eastern Gooeyduck Clam Panopea sp. Distributed on the East Coast of Korea (동해안 서식 동해코끼리조개(Panopea sp.)의 세로토닌 주사에 따른 산란 유발과 다정수정 방지를 위한 정자밀도에 따른 유생발달)

  • We examined the effects of serotonin on the spawning response, sperm motility, and D-shaped larva production in Eastern Gooeyduck clam Panopea sp. based on the sperm densities at fertilization and washing after mixing the eggs and sperm. The highest spawning induction was found showed in females and males injected with 1 mL of 2 mM serotonin. The spawning responses in females and males were higher at concentrations greater than 1 mM and 0.75 mM, respectively. Regarding the activities of sperm in sea water after serotonin injection, the sperm showed activity at >90.0% until 120 mins. We also examined the effects of sperm concentration at the fertilization and washing times after mixing the eggs and sperm. We confirmed that washing within 1 minute at a concentration of 1,500 sperms/mL or less can prevent egg destruction by polyspermy and secure a large number of D-phase larvae. These results should be useful for developing the aquaculture process for Eastern Gooeyduck clam, Panopea sp.

Biotoxic Cyanobacterial Metabolites Exhibiting Pesticidal and Mosquito Larvicidal Activities

  • A freshwater bloom-forming cyanobacterium, Microcystis aeruginosa, and local soil isolate Scytonema sp. strain BT 23 were demonstrated to contain biotoxic secondary metabolites with pesticidal and mosquito larvicidal activities. A purified toxic constituent from M aeruginosa showed an absorption maximum at 230 nm and its toxicity symptoms, Rf value on TLC, and retention time observed ill an HPLC analysis were similar to those of the hepatotoxic heptapeptide microcystin-LR. The bioactive constituent of the Scytonema sp. was less polar in nature and exhibited two peaks at 240 and 285 m. When applied to two cruciffrous pests, Pieris brassicae and Plutella flostella, the crude extracts and toxic principles from the two cyanobacteria showed significant antifeedant activity in a no-choice bioassay, and at higher concenuations exhibited contact toxicity to the insect larvae. The purified toxin from M. aeruginosa was found to be more effective and produced 97.5 and $92.8\%$ larval mortality in the two pests, fo11owing 2 h of toxin treatment at a concentration of $25{\mu}g$ Per leaf disc (2.5 cm dia.). Meanwhile, similar treatment with the purified toxin from Sytonema sp. stain BT 23 only produced 73 and $78\%$ mortality in the two pests. The cyanobacterial constituents also showed significant activity against Culex and Anopheles larvae. The M. aeruginosa toxin ($20{\mu}g\;ml^-1$) caused 98.2 and $88.1\%$ mortality in the Culex and Anopheles larvae, respectively, while the purified toxin from the Sytonema sp. was less toxic and only produced a 96.3 and $91.2\%$ mortality, respectively, at a much higher concentration ($40{\mu}g\;ml^-1$). Accordingly, the current results point to certain hitherto unknown biological properties of cyanobacterial biotoxins.

Variation in Insecticide Susceptibilities of the Beet Armyworm, Spodoptera exigua (Hubner): Esterase and Acetylcholinesterase Activities (파밤나방(Spodoptera exigua (Hubner))의 살충제 감수성 변이-에스테라제와 아세틸콜린에스테라제 활력)

  • There was a great variation in insecticide susceptibilities among field and laboratory populations of the beet armyworm, Spodoptera exigua (Hiibner). Unselected laboratory population, which had been reared for 6-7 generations in our laboratory without exposure to insecticides, was more susceptible than its parental field population in all tested insecticides. Two selected laboratory populations with parathion or deltamethrin showed much higher insecticide tolerance than did the unselected laboratory population in their own selection insecticide. The variation of the insecticide susceptibilities was highly correlated with esterase and acetylcholinesterase activities. Field and the selected laboratory populations had lower acetylcholinesterase activities and higher esterase activities than did the unselected laboratory population. Acetylcholinesterase of the field and the selected laboratory populations had higher Km values than did that of the unselected. In a population, Km values were varied among different developmental stages; acetylcholinesterase of the fifth instar larvae had the highest Km value among those of the other larval stages. Twenty one esterase bands were separated on 6.5% nondenaturing polyacrylamide gel from the whole body extracts of the fifth instar larvae. E2, E7, E8, Ell, El6, and El7 esterase bands were developed more frequently in the insecticides-selected populations than in the unselected population. These results suggest that the variation of insecticide susceptibilities of the beet armyworm includes both biochemical mechanisms: target site insensitivity and enhanced activity of detoxification enzyme.

Development of "Bt-Plus" Biopesticide Using Entomopathogenic Bacterial (Xenorhabdus nematophila, Photorhabdus temperata ssp. temperata) Metabolites (곤충병원세균(Xenorhabdus nematophila, Photorhabdus temperata ssp. temperata)의 대사물질을 이용한 "비티플러스" 생물농약 개발)

  • Bacillus thuringiensis (Bt) is a bacterial biopesticide against insect pests, mainly lepidopterans. Spodoptera exigua and Plutella xylostella exhibit significant decreases in Bt susceptibility in late larval instars. To enhance Bt pathogenicity, we used a mixture treatment of Bt and other bacterial metabolites which possessed significant immunosuppressive activities. Mixtures of Bt with culture broths of Xenorhabdus nematophila (Xn) or Photorhabdus temperata ssp. temperata (Ptt) significantly enhanced the Bt pathogenicity against late larval instars. Different ratios of Bt to bacterial culture broth had significant pathogenicities against last instar P. xylostella and S. exigua. Five compounds identified from the bacterial culture broth also enhanced Bt pathogenicity. After determining the optimal ratios, the mixture was applied to cabbage infested by late instar P. xylostella or S. exigua in greenhouse conditions. A mixture of Bt and Xn culture broth killed 100% of both insect pests when it was sprayed twice, while Bt alone killed less than 80% or 60% of P. xylostella and S. exigua, respectively. Other Bt mixtures, including Ptt culture broth or bacterial metabolites, also significantly increased pathogenicity in the semi-field assays. These results demonstrated that the Bt mixtures collectively names "Bt-Plus" can be developed into potent biopesticides to increase the efficacy of Bt.

  • We compared the nutritional requirements of whole larvae of the black seabream Acanthopagrus schlegeli, sweet fish Plecoglossus altivelis, olive flounder Paralichthys olivaeus and rock fish Sebastes schlegeli. The larvae were 20, 30, 14 and 5 DAH (or spawning) of black seabream, sweet fish, olive flounder and rock fish, respectively. Specific ${\alpha}$-amylase activity (mU/mg protein) was highest (8,324.9 mU/mg protein) in rock fish larvae (P<0.05). Specific trypsin-like protease activity was highest (11,330.1 mU/mg protein) in black seabream larvae (P<0.05), which also exhibited the highest activity, 685.5 mU/mg dry weight (P<0.05). The specific activities per mg protein and mg dry weight of black seabream were the highest (187.4 mU/mg protein and 11.3 mU/mg dry weight, respectively) (P<0.05). A/P, P/L and A/L ratios of rock fish were 1.47, 90.3 and 133.1, respectively (P<0.05). We present here basic larval digestive enzymatic nutritional requirement data.

Comparison of silkworm powder from 3 Bombyx mori varieties on alcohol metabolism in rats

  • Increased alcohol consumption is a burden on the world because it is associated with various health problems. However, the effects of silkworms on alcohol metabolism have not been studied yet. The hard-to-eat mature silkworms have become easier to ingest recently due to the development of technology, steam-lyophilising mature silkworm larvae. In this study, we investigated and compared the effects of SMSPs from three silkworm varieties, Baekokjam, Golden-silk and Yeonnokjam weaving white, golden, and light green cocoons on alcohol metabolism in vivo. Sprague-Dawley rats pretreated with three SMSPs (0.1 g/kg or 1 g/kg body weight) or normal diet (AIN-76A) for 2 weeks were subjected to intragastric administration of absolute ethanol (3 g/kg body weight, 3 h). Three SMSPs did not affect the final body weight and liver weight. All 3 SMSPs were effective to reduce the enzymes in alcohol metabolism, alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH), and liver damage and enzymes involved in liver damage, aspartate aminotransferase (AST) and alanine aminotransferase (ALT). Among SMSP from 3 varieties of silkworm, preadministration of 1 g/kg Baekokjam SMSP showed the most effective suppressive effect on the activities of ADH, ALDH, AST and ALT. The Baekokjam SMSP contained higher amounts of beneficial amino acids than Golden-silk or Yeonnokjam SMSP. These results suggest that Baekokjam SMSP might be used as a new and promising candidate for improving alcohol metabolism and liver injury through promoting rapid alcohol metabolism.

  • Amphibian population declines globally. Aquatic contamination by organic pollutants including endocrine disrupters has been suspected to the one of the reason for distinction of amphibia which has obligate aquatic life style during larval period. Amphibians have been widely accepted as animal model for the study of endocrine disruption in aquatic ecosystem at molecular as well as individual levels. There are increasing need for toxicological data in amphibians at multiple endpoints for management of contamination and development of safety guideline for important EDs in aquatic media. Alkylphenols have been widely used in agricultural, industrial, and housekeeping activities, contaminating the aquatic media and evoking endocrine disruption in aquatic animals. In this review, we summarized data concerning the endocrine disruption by alkylphenol organic pollutants on amphibians according to route, concentration, terms, and developmental stage of exposure together with mechanism of endocrine disruption.

Histochemical Study on the Hemocytes during Metamorphosis (배추흰나비의 變態에 따른 血球細胞의 組織化學的 硏究)

  • Insect hemocytes during metamorphosis were studies by histchemical and autoradiographic methods is Pieris rapae (Lepidoptera). The hemocytes were classified into six types, prohemocytes, plasmatocytes, podocytes, granular hemocytes, spherule cells and oenocytoids on the basis of the transitions in shapes and inclnsions of cytoplasms. Proteins, nucleic acids, polysaccharides and lipids in the hemocytes were detected histochemically from larval to pupal stages to learn the rise and fall of them during growth and metamorphosis. Most of the granules consisted of glycogen, neutral mucopolysaccharides and mucoprotein in addition to some granules of neutral fats and phospholipids were found in the granular hemocytes and spherule cells. Mitotic figures and DNA synthetic activities were observed in every type of hemocytes from 2nd to 5th instars, suggesting the all types of hemocytes originated from the prohemocytes. The cytoplasmic filaments of plasmatocytes and pdocytes extended very long in prepupa and pupa and the vermiform cells were the transformed plasmatocytes due to their further differentiation.
