Detection of Quantitative Trait Loci Affecting Fat Deposition Traits in Pigs

  • Quantitative trait loci (QTL) associated with fat deposition traits in pigs are important gene positions in a chromosome that influence meat quality of pork. For QTL study, a three generation resource population was constructed from a cross between Korean native boars and Landrace sows. A total of 240 F2 animals from intercross of F1 were produced. 80 microsatellite markers covering chromosomes 1 to 10 were selected to genotype the resource population. Intervals between adjacent markers were approximately 19 cM. Linkage analysis was performed using CRIMAP software version 2.4 with a FIXED option to obtain the map distances. For QTL analysis, the public web-based software, QTL express (http://www.qtl.cap.ed.ac.uk) was used. Two significant and two suggestive QTL were identified on SSC 6, 7, and 8 as affecting body fat and IMF traits. For QTL affecting IMF, the most significant association was detected between marker sw71 and sw1881 on SSC 6, and a suggestive QTL was identified between sw268 and sw205 on SSC8. These QTL accounted for 26.58% and 12.31% of the phenotypic variance, respectively. A significant QTL affecting IMF was detected at position 105 cM between markers sw71 and sw1881 on SSC 6.

돼지 PGK 2 유전자의 단일염기다형성 및 성장 형질과의 연관성 구명 (Association of Single Nucleotide Polymorphism (SNP) in the PGK 2 Gene with Growth Traits in Pigs)

  • 자돈의 생시 체중은 자돈의 이유 체중 및 생존율에도 크게 영향을 미친다. 또한 출하 시까지 돼지의 성장률 뿐만아니라 출하 일령 단축 및 출하 체중과도 밀접한 연관성이 있는 것으로 알려져 있다. 본 연구는 돼지 7번 염색체의 PGK 2 (phosphoglycerate kinase 2) 유전자의 promoter 영역 및 transcription 영역에 해당하는 DNA 염기서열을 유전체 구조분석을 통해 20개의 SNP (Single Nucleotide Polymorpism)를 발굴하였고, 발굴된 SNP 중 PGK 2 유전자의 전사 조절 영역 내 g.122 T>G 다형을 재래돼지와 Landrace를 이용한 $F_2$ 집단 268두 자돈의 성장 형질과의 연관성 분석을 실시한 결과, 생시 체중(p<0.01) 및 3주령 체중(p<0.001)에서 통계적으로 고도의 유의적 연관성이 있음을 확인 할 수 있었다. 따라서 본 연구를 통하여 확인된 PGK 2 유전자의 promoter 영역 내 성장 형질 관련 SNP는 건강한 자돈 및 종돈을 조기 선발하기 위한 유전자 marker로 활용 가능성을 제시하였다.

돼지 7번 염색체에서 육색 연관 QTL 확인 (Identification of Quantitative Trait Loci(QTL) for Meat Color Trait on Chromosome 7 in Pig)

  • 본 연구는 돼지의 염색체 7번에 존재하는 주요 경제형질에 관여하는 양적형질 유전자좌위(Quantitative trait loci; QTL)를 밝히기 위해 초성위체 표지인자를 이용하여 유전자지도(연관지도) 작성을 수행하였다. 기준집단의 조성은 이형접합성이 높은 기준집단을 조성하기 위하여 유전적 특성이 현격히 다른 우리나라의 재래돼지와 Landrace를 전형매 교배하여 생산된 $F_2$ 183두를 사용하였으며 기준집단에 대해 도축 24시간 후의 pH, 육색, 육즙손실량, 전단력, 가열감량, 조지방, 조회분, 수분, 조단백질 등 육질형질을 조사 및 분석하였다. 염색체 7번의 연관지도는 총 23개의 표지인자로 작성되었으며 암수평균 연관지도 길이는 154.6 cM 이었으며 수컷과 암컷의 연관지도 길이는 각각 169.2 cM과 141.4 cM로 수컷의 연관지도 길이가 암컷의 것보다 27.8 cM 더 길었다. 표지인자간의 최소간격은 SW175와 S0066 표지인자사이로 1.6 cM이었고, 최대간격은 SW2002와 SWR773표지인자사이로서 15.9 cM이었으며 평균간격은 7.02 cM이었다. 본 연구에서는 9개의 육질관련 형질 중 육색과 연관된 2개의 QTL이 확인되었는데 적색도(CIE-a)와 연관된 QTL은 45 cM 영역에서 1% 수준의 통계적 유의성을 나타내었고(Fig. 2), 이 영역에서 불과 3${\sim}$4 cM 떨어진 위치에서 황색도(CIE-b)와 연관된 QTL이 확인되었다. 향후에 본 연구에서 탐색된 QTL 영역에 대하여 고밀도의 유전자 지도 작성을 통하여 특정 형질과 연관된 부분을 계속해서 추적해 나간다면 육색형질을 조절하는 주유전자의 클로닝 및 특성 구명이 가능할 것으로 사료되며 궁극적으로는 돼지의 개량에 효율적으로 활용할 수 있는 표지인자(MAS)의 개발 등도 가능 할 것으로 사료된다.

교잡종 돼지에서 도체등급에 따른 돈육의 이화학적 특성 변화 (Physico-Chemical Characteristics of Crossbred Pigs with Carcass Grade)

  • 재래돼지 수컷과 랜드레이스 암컷을 교배하여 생산된 돼지 250두의 등급에 따른 돼지의 도체 특성과 돈육의 이화학적 특성을 비교한 결과는 다음과 같다. 생체중, 도체중, 지육율 및 등지방 두께는 E등급이 다른 네 등급에 비하여 유의적으로(P<0.05) 낮게 나타났다. 등급이 높을수록 가열 감량은 유의적으로(P<0.05) 많았으며, 수분 및 조지방 함량, PH 및 전단가는 등급간에 차이를 보이지 않았다. 육색 a*값은 C와D등급이 유의적으로 가장 높았으며, A등급이 유의적으로 가장 낮게 나타났으나 L*값과 b*값은 등급간에 차이를 보이지 않았다. 지방색 b*값은 A와 E 등급이 유의적으로 가장 높게 나타났으며, C 등급이 유의적으로(P<0.05) 가장 낮게 나타났으나 L*값과 a* 값은 등급간에 차이를 보이지 않았다. 가열육의 부착성과 씹힘성은 등급이 높을수록 유의적으로 (P<0.05)높게 나타났으나 경도, 응집성, 탄력성 및 파쇄성은 등급간에 차이를 보이지 않았다. 지방산조성은 등급이 높을 수록 palmitic산 함량이 유의적으로(P<0.05) 높고 불포화지방산/포화지방산 비율은 유의적으로(P<0.05) 낮게 나타났으며, 기타 다른 지방산들은 등급간에 유의적인 차이를 보이지 않았다.

돼지 Melanocortin Receptor 1(MC1R) 대립유전자 3의 신규 유전변이 탐색 (Detection of Novel Genetic Variations of the MG1R * 3 Allele in Pig(Sus scrofa))

  • 본 연구는 MCIR$^*$3 allele의 돼지에 있어서 유전적 변이를 관찰하기 위하여 수행하였다. 일반적으로 흑모색 바탕에 백색반점이나 백색띠를 갖고 있는 돼지의 MCIR 유전자의 유전자형은 E$^{D2}$로 나타낸다. 우성 백색계통의 E$^P$ 유전자형은 우성 흑모색 계통의 E$^{D2}$ 유전자와 frameshift mutation 관계가 있다. 돼지 MCIR 전체 번역지역을 증폭하기 위하여 oligonucleotide primer률 제작하여 PCR을 수행 하였다. 그 결과 길이가 963${\sim}$966 base pairs인 돼지 MCIR 유전자의 전체번역지역을 포함하는 산물을 얻었다. 이들 번역부위의 염기서열 결정하고 이들을 Clusta1 W 프로그램을 이용하여 정렬한 결과 23번 코돈{nt68)에서 Hampshire와 제주 재래혹돈은 염기 시토신(cytosine)이 3 개 그리고 Birl‘shire의 경우 염기 시토신(cytosine)이 2개 결실되어 있었다. 그 외에 3개의 missense mutations과 하나의 frameshift mutation이 발견되었다.

Association of functional sequence variants of the myosin heavy chain 3 gene with muscle collagen content in pigs

  • This study examined the association between functional sequence variants (FSVs) of myosin heavy chain 3 (MYH3) genotypes and collagen content in a Landrace and Jeju native pig (JNP) crossbred population. Four muscles (Musculus longissimus dorsi, Musculus semimembranosus, Musculus triceps brachii, and Musculus biceps femoris) were used for the analysis of meat collagen content, and the same animals were genotyped for the FSVs of the MYH3 gene by using PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism). Three FSVs of MYH3 genotypes were identified and had genotype frequencies of 0.358, 0.551, and 0.091 for QQ, Qq, and qq, respectively. QQ animals for the FSVs of the MYH3 genotypes showed higher collagen content in their M. longissimus dorsi (p < 0.001), M. semimembranosus (p < 0.001), M. triceps brachii (p < 0.001), and M. biceps femoris (p < 0.001) than qq homozygous animals. After the validation of this result in other independent populations, the FSVs of MYH3 genotypes can be a valuable genetic marker for improving collagen content in porcine muscles and can also be applied to increase the amount of collagen for biomedical purposes.

  • PIGK(phosphatidylinositol glycan, class K) is a subunit of GPI transamidase that cleaves the signal peptide in proproteins and replaces it with GPI. In addition, the structure and synthesis of GPI are critically involved in some of the cellular actions of insulin. Therefore, PIGK would be essential for mammalian development and many specific cellular functions as well as for metabolic activity of insulin associated with GPI. Two types of" full-length cDNAs of porcine PIGK were cloned through RT-PCR and RACE experiments. One is thought to be a normal form(consist of 395 amino acids) and the other is considered as an alternative spliced form(consist of 371 amino acids) which contains additional 63 bps in intron 7. Since a stop codon was contained within the insertion, the spliced form has a shorter coding sequence than that of normal form. A missense mutation (T314I) in exon 6 was detected and used for genotyping to estimate association with the growth and fat deposition traits for 545 $F_2$ animals(Korean native boars ${\times}$ Landrace). From the PCR-RFLP analysis using HpyCH4III, CT genotype showed highly significant relationship(P< 0.01) with carcass fat contents.

Investigation of Single Nucleotide Polymorphisms in Porcine Chromosome 2 Quantitative Trait Loci for Meat Quality Traits

  • Several studies have reported quantitative trait loci (QTL) for meat quality on porcine chromosome 2 (http://www.animalgenome.org/QTLdb/pig.html). For application of the molecular genetic information to the pig industry through marker-assisted selection, single nucleotide polymorphism (SNP) markers were analyzed by comparative re-sequencing of polymerase chain reaction (PCR) products of 13 candidate genes with DNA from commercial pig breeds such as Berkshire, Yorkshire, Landrace, Duroc and Korean Native pig. A total of 34 SNPs were identified in 15 PCR products producing an average of one SNP in every 253 bp. PCR restriction fragment length polymorphism (RFLP) assays were developed for 11 SNPs and used to investigate allele frequencies in five commercial pig breeds in Korea. Eight of the SNPs appear to be fixed in at least one of the five pig breeds, which indicates that different selection among pig breeds might be applied to these SNPs. Polymorphisms detected in the PTH, CSF2 and FOLR genes were chosen to genotype a Berkshire-Yorkshire pig breed reference family for linkage and association analyses. Using linkage analysis, PTH and CSF2 loci were mapped to pig chromosome 2, while FOLR was mapped to pig chromosome 9. Association analyses between SNPs in the PTH, CSF2 and FOLR suggested that the CSF2 MboII polymorphism was significantly associated with several pork quality traits in the Berkshire and Yorkshire crossed F2 pigs. Our current findings provide useful SNP marker information to fine map QTL regions on pig chromosome 2 and to clarify the relevance of SNP and quantitative traits in commercial pig populations.

Individual Identification using The Multiplex PCR with Microsatellite Markers in Swine

  • The swine is one of the most widespread mammalian throughout the whole world. Presently, many studies concerning microsatellites in swine, especially domestic pigs, have been carried out in order to investigate general diversity patterns among either populations or breeds. Until now, a lot of time and effort spend into a single PCR method. But simple and more rapid multiplex PCR methods have been developed. The purpose of this study is to develop a robust set of microsatellites markers (MS marker) for traceability and individual identification. Using multiplex-PCR method with 23 MS marker divided 2 set, various alleles occurring to 5 swine breed (Berkshire, Landrace, Yorkshire, Duroc and Korea native pig) used markers to determine allele frequency and heterozygosity. MS marker found 4 alleles at SW403, S0227, SWR414, SW1041 and SW1377. The most were found 10 alleles at SW1920. Heterozygosity represented the lowest value of 0.102 at SWR414 and highest value of 0.861 at SW1920. So, it was recognized appropriate allele frequency for individual identification in swine. Using multiplex-PCR method, MS markers used to determine individual identification biomarker and breed-specific marker for faster, more accurate and lower analysis cost. Based on this result, a scientific basis was established to the existing pedigree data by applying genetics additionally. Swine traceability is expected to be very useful system and be conducted nationwide in future.

Purification of Pig Muscle Stem Cells Using Magnetic-Activated Cell Sorting (MACS) Based on the Expression of Cluster of Differentiation 29 (CD29)

  • The muscle stem cells of domestic animals are of interest to researchers in the food and biotechnology industries for the production of cultured meat. For producing cultured meat, it is crucial for muscle stem cells to be efficiently isolated and stably maintained in vitro on a large scale. In the present study, we aimed to optimize the method for the enrichment of pig muscle stem cells using a magnetic-activated cell sorting (MACS) system. Pig muscle stem cells were collected from the biceps femoris muscles of 14 d-old pigs of three breeds [Landrace×Yorkshire×Duroc (LYD), Berkshire, and Korean native pigs] and cultured in skeletal muscle growth medium-2 (SkGM-2) supplemented with epidermal growth factor (EGF), dexamethasone, and a p38 inhibitor (SB203580). Approximately 30% of total cultured cells were nonmyogenic cells in the absence of purification in our system, as determined by immunostaining for cluster of differentiation 56 (CD56) and CD29, which are known markers of muscle stem cells. Interestingly, following MACS isolation using the CD29 antibody, the proportion of CD56+/CD29+ muscle stem cells was significantly increased (91.5±2.40%), and the proportion of CD56 single-positive nonmyogenic cells was dramatically decreased. Furthermore, we verified that this method worked well for purifying muscle stem cells in the three pig breeds. Accordingly, we found that CD29 is a valuable candidate among the various marker genes for the isolation of pig muscle stem cells and developed a simple sorting method based on a single antibody to this protein.
