Effect of the Biological Activities of Ethanol Extract from Korean Traditional Kochujang Added with Sea Tangle (Laminaria longissima) (다시마 분말을 첨가한 전통고추장 에탄올 추출물의 생리활성 효과)

  • This study was carried out to investigate antimutagenic and anticancer effects of Korean traditional Kochujang added with sea tangle (Laminuia longissima). Most of the mineral content in Kochujang added with sea tangle was increased when compared to traditional Kochujang. In the Ames test, the inhibition rate of ethanol extract (200 $\mu\textrm{g}$/plate) of Kochujang added with 5% sea tangle in the S. typimurium TA100 strain showed 87.2% against the mutagenesis induced by MNNG. And also, the inhibition rate of ethanol extract (200 $\mu\textrm{g}$/plate) of Kochujang added with 5% sea tangle in the S. typhimurium TA98 and TA100 strains was 62.9% and 71.6%, respectively, against the mutagenesis induced by 4NQO. The suppression under the same condition against B($\alpha$)P and Trp-P-1 in the TA98 and TA100 strains showed 70.2% and 80.8%, and 62.9% and 73.1%, respectively. In the anticancer effects, the treatment of 1.0 mg/mL Kochujang added with 5% sea tangle showed strong cytotoxicity of 78.6% and 79.4% against KATOⅢ and HepG2, respectively.

Effects of diet with brown algae on the quality characteristics of eggs (갈조류 첨가 사료로 식이한 산란계 계란의 품질 특성)

  • This study was performed to determine the effect of brown algae on the egg quality of laying hens fed a non-supplemented diet (Control) and a diet supplemented with 10% Undaria pinnatifida (A), Laminaria religiosa (B), and Hijikia fusiforme (C), respectively. Isa Brown (28 weeks old) were raised for 8 weeks. The darkness, redness, and blueness of the egg yolks increased with the addition of brown algae, expecially Undaria pinnatifida (A). The fucoxanthin contents were observed to be only 21.3-34.8 mg% level in the brown algae but they were nearly traced in the egg yolks. Among the analyzed minerals, the Ca, Fe, K contents increased to 391.2, 6.3, and 450.5 mg%, respectively, but the amount of P was reduced to 110.5 mg%. The total mineral contents were higher in the egg yolk with Hijikia fusiforme (C) (1,155.1 mg%) than with any other supplement. The brown algae diet reduced the saturated fatty acid level from 38.3 to 32.1% but increased the unsaturated fatty acid level from 61.5 to 69.6% owing to the rise of the level of polyunsaturated fatty acids.

Assessment of the possibility as Cosmetics Materials by Brown algae from Jeju Island Using Supercritical Fluid System (초임계 유체 기술을 이용한 제주산 갈조류의 화장품 소재로서의 가능성 평가)

  • Although various biological extracts derived from the resources from Jeju are currently being developed for cosmetic materials, few studies using Brown algae from Jeju have been conducted. This study identified materials with high antioxidant levels and examined their whitening effects to assess their feasibility as potential cosmetic resources. Brown algae from Jeju (Undaria pinnatifida (Harvey) Suringar, Laminaria japonica, Ecklonia cava, Hizikia fusiforme (Harvey) Okamura (HF), and Sargassum fulvellum (Turner) C. Agardh (SF) were processed using a supercritical fluid system at 50℃ and 400 bar, and their total phenol content, antioxidant activity, tyrosinase inhibition, and primary skin irritation were measured. Of all the samples, only SF using the supercritical fluid system (SFS) displayed concentration-dependent behavior for the total phenol content (p<0.05). Among the samples, the antioxidant activity of SFS was significantly higher (p<0.05) and the one increased at higher concentrations (p<0.05). On the other hand, tyrosinase was inhibited to a greater extent with HF than the others. The primary skin irritation tests showed that none of the samples were skin irritants. This study confirmed the feasibility of using Phaeophyceae from Jeju as potential cosmetic materials, where the samples can be selected and utilized based on the required physicochemical properties.

Freeze Treatment of Sludge for the Biogas Production from Brown Macroalgae (슬러지 냉동처리에 의한 갈조류로부터 바이오가스 생산)

  • In the present study, biogas was produced from the anaerobic digestion of marine macroalgae (Laminaria japonica) biomass. The optimal anaerobic condition for producing the sludge was the freeze treatment at $-70^{\circ}C$ for 20 min. Total amounts of hydrogen and methane gas produced were 667.28 mL/L and 3420.24 mL/L, respectively, which were 2.7 and 3.4 times greater than that in the control group. Freeze treatment of sludge produced the maximum biogas under an initial optimum pH of 7.0 and the maximum biomass at an initial optimum pH of 8.0. We confirmed that biogas production was greatly reduced under acidic conditions compared to that under alkaline conditions. Sludge was freeze treated, and the biomass and sludge production was optimal the total amounts of hydrogen and methane gas produced were 643.73 mL/L and 4291.6 mL/L, respectively, which were 2.6 and 4.3 times greater than in the control group. Also the results showed that under optimal conditions in a 5-L bioreactor, a maximum of 1605.03 mL/L of hydrogen and 4593.71 mL/L of methane gas could be produced by the substrate contained in the marine macroalgae biomass.

The Effect on the Quality of Yogurt Added Water Extracted from Sea Tangle (다시마 추출물이 요구르트 품질에 미치는 영향)

  • For the purpose of making a functional and stable yogurt, new type yogurt were prepared from 12% whole milk and 2% skim milk added hot-water extracted from sea tangle. The yogurt product were evaluated for acid production(pH, titratable acidity), number of viable cell, viscosity, degree of curd sedimentation, quality-keeping property and sensory property. By addition of hot-water extracted from 0.5% sea tangle, the titratable acidity of yogurt(1.89%) was higher than that of yogurt not added hot-water extract of sea tangle(1.53%). The propagation of lactic acid bacteria was stimulated by adding hot-water extracted from 0.5% sea tangle, and the number of viable cells were about 1.4${\times}$10$\^$9/CFU/$m\ell$. On the other hand, the number of viable cells in control were 4.4${\times}$10$\^$8/CFU/$m\ell$. Viscosity of yogurt made from adding hot-water extracted from 0.5% sea tangle(25 CPS) was higher than that of yogurt with only milk(22 CPS). When yogurt made from adding hot-water extracted from sea tangle was kept at 7$^{\circ}C$ for 15 days, its quality-keeping was relatively good. The sedimentation of curd was repressed a little by adding hot-water extracted from 0.5% and 0.1% sea tangle. The overall sensory score of yogurt made from adding hot-water extracted from 1% sea tangle was the best of tested yogurt.

Elucidation of Anti-tumor Initiator and Promoter Derived from Seaweed-3 : Anti-tumor Promoters of Ecklonia stolonifera Extracts (해조류 중의 anti-tumor initiator 및 promoter의 해석-3 : 곰피 추출물중의 발암 promotion억제 인자)

  • To elucidate anti-tumor promoter from seaweed, the anti-tumor promoting activity of Ecklonia stolonifera, Undaria pinnatifida and Laminaria japonica extracts were determined by Epstein-Barr virus (EBV)-early antigen (EA) induction caused by a tumor promoter, teleocidin B-4. The methanol extracts of seaweed were subsequently fractionated with diethyl ether, distilled water, chloroform and ethyl acetate. Among the solvent fractions tested, chloroform and ethyl acetate fraction of E. stolonifera showed a high anti-tumor promoting activity at the levels of 88.0 and $85.9\%$ by the addition of 20 ${\mu}g/m{\ell}$, respectively. To characterize anti-tumor promoters from solvent fractions of E. stolonifera, the effects of phenols, chlorophyll derivatives and carotenoids on the anti-tumor promoting activity were investigated. Phenols, such as bromophenol and phloroglucinol showed anti-tumor promoting activity of $57\~66\%$ at 20 ${\mu}g/m{\ell}$. Pigments, such as chlorophylls and carotenoids exerted high anti-tumor promoting activities. Chlorophyll a and pheophorbide a exhibited the activity of $77.4\%$ and $66.6\%$ at 5${\mu}M/m{\ell}$, respectively. The active compounds of carotenoids were tentatively identified as lutein and $\alpha-cryptoxanthin$ from the profiles of visible spectra and R_f value of their authentic compounds, and showed anti-tumor promoting activities of $76.9\%$ and $84.4\%$ at dose of 20 ${\mu}g/m{\ell}$, respectively.

Uronate Compositions of Alginates from the Edible Brown Algae (식용 갈조류의 알긴산 조성)

  • Uronate compositions and molecular weights of alginates from the four kinds of brown algae, sea mustard (Undaria pinnatifida), sea tangle (Laminaria japonica), gulf weed (Sargassum fulvellum), and seaweed fusiforme (Hizikia fusiforme), in regard with the harvesting season were investigated. Sea mustard contained the highest amount of alginates in the four kinds of brown algae. D-Mannuronic acid to L-guluronic acid (M/C) ratio of the alginates was high in order of seaweed fusiforme, gulf weed, sea mustard, and sea tangle, and especially in water-soluble alginate. Molecular weights of the alginates were greater with the growing period ranging in $4,500\~4,800\;kDa$ for sea tangle, $4,000\~4,200\;kDa$ for sea mustard, $3,300\~3,400\;kDa$ for seaweed fusiforme, and $3,000\~3,200\;kDa$ for gulfweed. In water-soluble alginate of sea mustard, M/G ratio was much higher in sporophyll than in midrib and blade.

Quality and Antioxidant Characteristics of Granule Tea Prepared with Sea Tangle (Laminaria japonica) and Sea Mustard (Undaria pinnatifida) Powder as Affected by Extraction Method (추출방법을 달리한 미역 및 다시마 과립차의 품질 및 항산화 특성)

  • This study was conducted to evaluate the quality and anti-oxidant characteristics of different granule tea variants (WSMGT: granule tea prepared from sea mustard hot water extract; ASMGT: granule tea prepared from sea mustard autoclave extract; WSTGT: granule tea prepared from sea tangle hot water extract; and ASTGT: granule tea prepared from sea tangle autoclave extract) from sea tangle and sea mustard powder. The Color of WSTGT showed the highest $L^*$, $b^*$ values, and the lowest $a^*$ value. The water absorption binding agents in ASMGT were higher than those in WSMGT, WSTGT. and ASTGT. Binding agents on water absorption in ASMGT were higher than those of WSMGT, WSTGT, and ASTGT. The alginic acid content of ASMGT was highest. The total polyphenol and total flavonoid contents of ASMGT and ASTGT were highest. The electron donating ability and ferrous ion chelating activity in WSTGT greatly increased compared to those in the other granules tea variants. These results suggest that pressure extraction can be used in functional foods.

Studies on the Floristic Composition and Periodicity at Joomoonjin Sea Bank (주문진(主文津) 인공(人工) 방파제(防波堤)에 서식(棲息)하는 해조상(海藻相) 및 주기성(週期性)에 관한 연구(硏究))

  • Benthic marine algal community of a sea bank at Joomoonjin, in the eastern coast of Korea, was investigated monthly in view of floristic composition and seasonal periodicity during from August, 1989 to July 1990. Total number of benthic alga identified were 70 taxa, which composed of 7 species of Chlorophyta, 21 species of Phaeophyta and 42 species of Rhodophyta. The dominant species were Sargassum horneri, Underia pinnatifida, Laminaria japonica and Pachymeniopsis sp.. Vegetation, from the point of phygiognomy, was a mosaic type by these dominant species. The flora was compared with similarity index using S${\phi}$rensen coefficient and grouped by a cluster analysis. As a result, the algal community of this area could be divided into two clusters at similarity level 45% : Flora of water temperature-ascending period(January-July) and flora of water temperature-descending period(August-December). And also, the former flora was subdivided into two clusters at point of the $14^{\circ}C$ sea water temperature : winter flora(January-April) and spring-summer flora(May-July). The flora of this area was characterized as a mixed flora of cold and temperature zone by Cheney's index.

Production of Methane from Anaerobic Fermentation of Marine Macro-algae (해조류의 혐기성 발효를 이용한 메탄 생산)

  • Methane was produced from the anaerobic digestion of marine macro-algae. Elemental analysis was first performed to estimate the theoretical methane production of three macro-algae (Undaria pinnatifida, Laminaria japonica, Hizikia fusiformis). Three algae were found to contain C 34 ~ 36%, H 5%, O 37 ~ 43%, N 2 ~ 4%, S 0.4 ~ 0.7%, and ash 14~21%, and the theoretical methane content was in the range of 56 ~ 60%, which can produce 442 ~ 568 mL $CH_4$ per g of volatile solid (VS). Using the biological methane potential (BMP) test, we found that L. japonica resulted in the highest yield of methane (52%). Moreover, various operational conditions, such as algae amount, pH, salinity, particle size, and pre-treatment, were investigated in order to find an optimal condition of anaerobic digestion. At pH 8.0, the autoclaved L. japonica (5g VS/200 mL), when used without washing salt, produced 268.5 mL/g VS which is 65% of the theoretical methane productions. Furthermore, using a CSTR (with the working volume of 7 L out of the total volume of 10 L), we have successfully operated the reactor for 65 days and obtained maximum methane production rate of 1.4 L/day with purity of 70%.
